3Atorvastatin protects cardiac progenitor cells from hypoxia-induced cell growth inhibition via MEG3/miR-22/HMGB1 pathway显示文摘Heart failure (HF)induced by ischemia myocardial infarction (MI)is one of the major causes of morbidity and mortality all around the world.Atorvastatin,a hydroxyrnethylglutaryl coenzyme A reductase inhibitor,has been demonstrated to benefit patients with ischemic or non-ischemicinduced HF,but the mechanism is still poorly understood.Increasing evidence indicates that IncRNAs play important role in variety of human disease.However,the role and underlying molecular mechanisms remain largely unclear.In our work,we applied 0.5%02 to generate a hypoxia cardiac progenitor cell (CPC)model.Then,CCK8 and EdU assays were employed to investigate the role of atorvastatin in hypoxia CPC cell model.We found that hypoxia inhibits CPC viability and proliferation through modulating MEG3 expression,while atorvastatin application can protect CPCs from hypoxia-induced injury through inhibiting MEG3 expression.Then,we demonstrated that repression of MEG3 inhibited the hypoxia-induced injury of CPCs and overexpression of MEG3 inhibited the protective effect of atorvastatin in the hypoxia-induced injury of CPCs.Furthermore,our study illustrated that atorvastatin played its role in CPC viability and proliferation by modulating the expression of HMGB1 through the MEG3/miR-22 pathway.Our study, for the first time,uncovered the molecular mechanism of atorvastatin's protective role in cardiomyocytes under hypoxia condition,which may provide an exploitable target in developing effective therapy drugs for MI patients.Jinwen Su Ming Fang Bei Tian Jun Luo Can Jin Xuejun Wang Zhongping Ning Xinming Li 2018Acta Biochimica et Biophysica Sinica2018,50,12:6

6Omics-based integrated analysis identified ATRX as a biomarker associated with glioma diagnosis and prognosis显示文摘Objective:ATRX is a multifunctional protein that is tightly regulated by and implicated in transcriptional regulation and chromatin remodeling.Numerous studies have shown that genetic alterations in ATRX play a significant role in gliomas.This study aims to further determine the relationship between ATRX and glioma prognosis and identify possible mechanisms for exploring the biological significance of ATRX using large data sets.Methods:We used The Cancer Genome Atlas(TCGA)database and 130 immunohistochemical results to confirm the difference in ATRX mutations in high-and low-grade gliomas.An online analysis of the TCGA glioma datasets using the cBioPortal platform was performed to study the relationship between ATRX mutations and IDH1,TP53,CDKN2 A and CDKN2 B mutations in the corresponding TCGA glioma dataset.In combination with clinical pathology data,the biological significance of the relationships were analyzed.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analyses and annotations of all adjacent genes in the network were performedin the Database for Annotation,Visualization and Integrated Discovery(DAVID)and R language.A protein-protein interaction(PPI)network was constructed,and the interactions of all adjacent nodes were analyzed by the String database and using Cytoscape software.Results:In the selected TCGA glioma datasets,a total of 2,228 patients were queried,21%of whom had ATRX alterations,which co-occurred frequently with TP53 and IDH1 mutations.ATRX alterations are associated with multiple critical molecular events,which results in a significantly improved overall survival(OS)rate.In low-grade gliomas,ATRX mutations are significantly associated with multiple important molecular events,such as ZNF274 and FDXR at mRNA and protein levels.A functional cluster analysis revealed that these genes played a role in chromatin binding and P53,and a link was observed between ATRX and IDH1 and TP53 in the interaction network.ATRX and TP53 are important nodes in the network and have potential links with the blood oxygen imbalance.Conclusions:ATRX mutations have clinical implications for the molecular diagnosis of gliomas and can provide diagnostic and prognostic information for gliomas.ATRX is expected to serve as a new therapeutic target.Yingbin Xie Yanli Tan Chao Yang Xuehao Zhang Can Xu Xiaoxia Qiao Jianglong Xu Shaohui Tian Chuan Fang Chunsheng Kang 2019Cancer Biology & Medicine2019,16,4:5

11Core muscle functional strength training for reducing the risk of low back pain in military recruits:An open-label randomized controlled trial显示文摘Background:Core muscle functional strength training(CMFST)has been reported to reduce injuries to the lower extremity.However,no study has confirmed whether CMFST can reduce the risk of low back pain(LBP).Objective:This study identified the effects of CMFST on the incidence of LBP in military recruits.Design,setting,participants and intervention:We performed a prospective,open-label,randomized,controlled study in a population of young healthy male naval recruits from a Chinese basic combat training program.Participants were randomly assigned to either the core group or the control group.In additional to normal basic combat training,recruits in the core group underwent a CMFST program for 12 weeks,while recruits in the control group received no extra training.Main outcome measures:At the beginning of the study and at the 12 th week,the number of participants with LBP was counted,and lumbar muscle endurance was measured.In addition,when participants complained of LBP,they were assessed using the visual analog scale(VAS)and Roland Morris Disability Questionnaire(RMDQ).Results:A total of 588 participants were included in the final analysis(295 in the core group and 293 in the control group).The incidence of LBP in the control group was about twice that of the core group over the 12-week study(20.8%vs 10.8%,odds ratio:2.161–2.159,P<0.001).The core group had better lumbar muscle endurance at 12 weeks than the control group([200.80±92.98]s vs[147.00±84.51]s,P<0.01).There was no significant difference in VAS score between groups,but the core group had a significantly lower RMDQ score at week 12 than the control group(3.33±0.58 vs 5.47±4.41,P<0.05).Conclusion:This study demonstrated that the CMFST effectively reduced the incidence of LBP,improved lumbar muscle endurance,and relieved the dysfunction of LBP during basic military training.Xin Wang Wen-juan Song Yi Ruan Bing-chu Li Can Lü Nian Huang Fan-fu Fang Wei Gu 2022Journal of Integrative Medicine2022,20,2:3
12Gene duplication drove the loss of awn in sorghum.显示文摘Loss of the awn in some cereals including sorghum is a key transition during cereal domestication or improvement that has facilitated grain harvest and storage.The genetic basis for the loss of awn in sorghum during domestication or improvement remains unknown.Here,we identified a transcription factor gene awn1 encoding an ALOG domain,which is responsible for awn loss during sorghum domestication or improvement.awn1 arose from a gene duplication from chromosome 10 that translocated to chromosome 3,recruiting a new promoter from the neighbouring intergenic region filled with'noncoding DNA',and recreating the first exon and intron.The awn1 acquires high expr`ession after duplication and represses the elongation of awns in domesticated sorghum.Comparative mapping revealed a high collinearity at awn1 paralog locus on chromosome 10 across cereals and awn growth and development was successfully reactivated on the rice spikelet by inactivating rice awn1 orthologue.Further RNA-seq and DAP-seq revealed that as a transcription repressor,AWN1 directly bound to the motif in the regulatory regions from three MADS genes related to flower development and two genes DL and LKS2 for the development of awn,downregulated the expressions of these genes,and then repressed the elongation of awn.The preexistence of regulatory elements in the neighbouring intergenic region of awn1 before domestication signified that noncoding DNA may serve as a treasure trove for evolution during adaptation to a changing world.Our results supported that gene duplication can promptly drive the evolution of gene regulatory network.Leina Zhou Can Zhu Xiaojian Fang Hangqin Liu Shuyang Zhong Yan Li Jiacheng Liu Yang Song Xing Jian Zhongwei Lin 2021Molecular Plant2021,14,11:2

14In-situ generation and global property profiling of metal nanoclusters by ultraviolet laser dissociation-mass spectrometry显示文摘Metal nanoclusters are promising nanomaterials with unique properties, but only a few ones with specific numbers of metal atoms can be obtained and studied up to now. In this study, we establish a new paradigm of in-situ generation and global study of metal nanoclusters with different sizes, constitutions, and charge states, including both accurate constitution characterization and global activity profiling. The complex mixtures of metal nanoclusters are produced by employing single-pulsed 193-nm laser dissociation of monolayer-protected cluster(MPC) precursors within a high-resolution mass spectrometry(HRMS). More than400 types of bare gold nanoclusters including novel multiply charged(2+ and 3+), S-/P-doped, and silver alloy ones can be efficiently generated and accurately characterized. A distinct size(1 to 142 atoms)-and charge(1+ to 3+)-hierarchy reactivity is clearly observed for the first time. 15Preparation,structure,luminescence properties of europium doped zinc spinel structure green-emitting phosphor ZnAl_2O_4:Eu^(2+)显示文摘The Zn_(1-x)Al_2 O_4:xEu^(2+) phosphor powders were synthesized by the solid-state reaction method.The synthesis temperature for ZnAl_2 O_4 was optimized,whereas the phase structure,TEM images,photoluminescence(PL) properties,the concentration quenching mechanism,the fluorescence decay curves,as well as the CIE chromaticity coordinates of the samples were investigated in details.Under the excitation at 379 nm,the phosphor exhibits an asymmetric broad-band green emission with a peak at 532 nm,which is ascribed to the 5 d-4 f transition of Eu2+.When the doping concentration of Eu2+ ions is 0.01,the luminescence intensity of the sample reaches the maximum value.It is further proved that the exchange interaction results in the concentration quenching of Eu2+ in the Zn_(1-x)Al_2 O_4:xEu^(2+) phosphor powders.The thermal quenching property of ZnAl_2 O_4:Eu^(2+)phosphor was investigated and the quantum efficiency(QE) values of the selected Zn_(0.99)Al_2 O_4:0.01 Eu^(2+) phosphor was measured and determined as 54.85%.The lifetime of the optimized sample Zn_(0.99)Al_2 O_4:0.01 Eu^(2+) is 3.0852 μs and the CIE coordinate of the sample was calculated as(0.3323,0.5538) with high-color-purity green emission.All properties indicate that the green-emitting ZnAl_2 O_4:Eu^(2+) phosphor powder has potential application in white LEDs.Can He Haipeng Ji Zhaohui Huang Xiaoguang Zhang Yan'gai Liu Minghao Fang Xiaowen Wu Xin Min 2018Journal of Rare Earths2018,36,9:2
