2On-treatment monitoring of liver fibrosis with serum hepatitis B core-related antigen in chronic hepatitis B显示文摘BACKGROUND Non-invasive evaluation for liver fibrosis is clinically important,especially in patients with undetectable hepatitis B virus(HBV)DNA treated with nucleoside analogs.AIM To clarify the monitoring power of hepatitis B core-related antigen(HBcrAg)for hepatic histologic changes in patients with chronic hepatitis B(CHB)treated with entecavir.METHODS This prospective multicenter study used multiple ordinal and multivariate logistics regression analysis to assess variables associated with Ishak fibrosis score and regression for fibrosis regression,respectively,in 403 CHB patients,including 374 with entecavir for 72 weeks(291 underwent paired liver biopsy)and 29 as controls.RESULTS Level of HBcrAg correlated negatively with liver fibrosis staging(γ=-0.357,P<0.001)in hepatitis B e antigen(HBeAg)-positive patients,and positively with liver fibrosis staging in HBeAg-negative patients.Higher HBcrAg concentration was associated with younger age,HBeAg positive status,high HBV DNA loads,high level of hepatitis B surface antigen(HBsAg)and higher necroinflammation,but not with HBV genotype.Serum concentration of HBcrAg,basal core promoter/precore(BCP/PC)mutant,quantitation of HBsAg(qHBsAg)and platelet counts were independently associated with Ishak fibrosis score on multiple ordinal regression.HBV DNA was undetectable in 88.37%of patients treated with entecavir at week 72,while their level of HBcrAg was still detectable.A greater reduction in post-treatment HBcrAg concentration was associated with the regression of hepatic fibrosis and histological improvement.HBcrAg concentration>6.33 log IU/mL at baseline and logarithmic reduction>1.03 log IU/mL at week 72 were associated with a higher chance of regression of liver fibrosis and histological improvement,respectively.CONCLUSION HBcrAg level is associated with liver fibrosis progression.HBcrAg is an excellent monitor of hepatic histological changes,especially in CHB patients treated with nucleoside analogs.Xiu-Juan Chang Chao Sun Yan Chen Xiao-Dong Li Zu-Jiang Yu Zheng Dong Wen-Lin Bai Xiao-Dong Wang Zhi-Qin Li Da Chen Wen-Juan Du Hao Liao Qi-Yu Jiang Li-Jun Sun Yin-Yin Li Cui-Hong Zhang Dong-Ping Xu Yong-Ping Chen Qin Li Yong-Ping Yang 2019World Journal of Gastroenterology2019,25,32:12
3Relationship of particle stimulated nucleation,recrystallization and mechanical properties responding to Fe and Si contents in hot-extruded 7055 aluminum alloys显示文摘The variations of coarse intermetallic particles in hot-extruded 7055 aluminum alloys with 0.041 wt%Fe and 0.024 wt%Si increasing to 0.272 wt%Fe and 0.134 wt%Si were investigated.The particle stimulated nucleation(PSN)behaviors for different kind of coarse particles were detailly analyzed by EBSD.Moreover,the effect of PSN responding to Fe and Si contents on recrystallization and tensile properties of 7055 alloys was evaluated.With increasing Fe and Si contents,the size and number density of coarseη/S particles are reduced,while the number densities of coarse Al7Cu2 Fe and Mg2Si particles are both increased and the coarse Al7Cu2 Fe particles transform from rod-like to irregular.More PSN recrystallized grains with predominant orientations deviated from the extruded fiber textures are stimulated by the irregular Al7Cu2 Fe and Mg2Si particles,because a higher degree of local non-uniform deformation is produced.The rod-like Al7Cu2 Fe particles cause the greatest degree of local non-uniform deformation owing to the largest aspect ratio,but the shape also restricts the area of particle deformation zone(PDZ)resulting in fewer PSN recrystallized grains.The irregularη/S particles give rise to the lowest degree of local non-uniform deformation and fewest PSN recrystallized grains with the major orientations close to the extruded fiber textures.Consequently,despite the number and size of coarseη/S particles are reduced,the proportion of high angle grain boundaries(HAGBs)is increased and the extruded fiber textures are weakened with Fe and Si contents increasing,because of the increased Al7Cu2 Fe and Mg2Si particles.The strength is slightly declined by the weakened<111>//ED(extrusion direction)fiber texture,while the elongation is reduced for a larger number of coarse particles and more HAGBs with higher Fe and Si contents.Huan She Da Shu Anping Dong Jun Wang Baode Sun Hongchang Lai 2019Journal of Materials Science & Technology2019,35,11:10
4Nicotinamide overload may play a role in the development of type 2 diabetes显示文摘AIM: To investigate whether nicotinamide overload plays a role in type 2 diabetes. METHODS: Nicotinamide metabolic patterns of 14diabetic and 14 non-diabetic subjects were compared using HPLC. Cumulative effects of nicotinamide and N1-methylnicotinamide on glucose metabolism, plasma H2O2 levels and tissue nicotinamide adenine dinucleotide (NAD) contents of adult Sprague-Dawley rats were observed. The role of human sweat glands and rat skin in nicotinamide metabolism was investigated using sauna and burn injury, respectively. RESULTS: Diabetic subjects had significantly higher plasma N 1-methylnicotinamide levels 5 h after a 100-mg nicotinamide load than the non-diabetic subjects (0.89 ± 0.13 μmol/L vs 0.6 ± 0.13 μmol/L, P < 0.001). Cumulative doses of nicotinamide (2 g/kg) significantly increased rat plasma N 1-methy lnico-tinamide concentrations associated with severe insulin resistance, which was mimicked by N1-methy-lnicotinamide. Moreover, cumulative exposure to N1-methylnicotinamide (2 g/kg) markedly reduced rat muscle and liver NAD contents and erythrocyte NAD/NADH ratio, and increased plasma H2O2 levels. Decrease in NAD/NADH ratio and increase in H2O2 generation were also observed in human erythrocytes after exposure to N1-methylnicotinamide in vitro. Sweating eliminated excessive nicotinamide (5.3-fold increase in sweat nicotinamide concentration 1 h after a 100-mg nicotinamide load). Skin damage or aldehyde oxidase inhibition with tamoxifen or olanzapine, both being notorious for impairing glucose tolerance, delayed N1-methylnicotinamide clearance. 8The combination of PD-1 blockade with interferon-αhas a synergistic effect on hepatocellular carcinoma显示文摘Background The efficacy of immune checkpoint inhibitors(ICIs),such as programmed cell death protein-1(PD-1)or its ligand 1(PD-L1)antibody,in hepatocellular carcinoma(HCC)is limited,and it is recommended that they be combined with other therapies.We evaluated the combination of pegylated interferon-α(Peg-IFNα)with PD-1 blockade in HCC mouse models.Methods We analyzed the effects of Peg-IFNαon tumor-infiltrating immune cells and PD-1 expression in the HCC immune microenvironment and examined the underlying mechanism of its unique effect on the PD-1 pathway.The in vivo efficacy of anti-PD-1 and Peg-IFNαwas evaluated in both subcutaneous and orthotopic mouse models of HCC.Results The combination of Peg-IFNαwith PD-1 blockade dramatically enhanced T-cell infiltration,improved the efficacy of PD-1 antibody and prolonged mouse survival compared with PD-1 antibody monotherapy.Mechanistically,Peg-IFNαcould recruit cytotoxic CD8+T cells to infiltrate the HCC microenvironment by inducing tumor cells to secrete the chemokine CCL4.Nevertheless,the HCC microenvironment quickly overcame the immune responses by upregulating PD-1 expression in CD8+T cells via the IFNα-IFNAR1-JAK1-STAT3 signaling pathway.The combination of PD-1 blockade with Peg-IFNαcould restore the cytotoxic capacity of CD8+T cells and exerted a significant synergistic effect on HCC.Conclusion These results indicate that in addition to initiating the antitumor immune response itself,Peg-IFNαcan also generate a microenvironment favoring PD-1 blockade.Thus,the combination of Peg-IFNαand PD-1 blockade can be a promising strategy for HCC.Ying Zhu Mo Chen Da Xu Tian-En Li Ze Zhang Jian-Hua Li Xiang-Yu Wang Xin Yang Lu Lu Hu-Liang Jia Qiong-Zhu Dong Lun-Xiu Qin 2022Cellular & Molecular Immunology2022,19,6:6

11Effects of Two Curcuminoids on Candida albicans显示文摘Objective To investigate and compare the action of curcuminoids on the causal pathogens of Candida albicans growth. Methods The effects of curcumin (CUR) and demethoxycurcumin (DMC) on C. albicans growth were first investigated and compared by microcalorimetry coupled with multiple analytical methods. The quantitative thermo-kinetic parameters obtained from these curves were analyzed to show difference of the actions. Results By analyzing the main parameters screened from principal component analysis together with 50% inhibiting concentration values, it was demonstrated that both CUR and DMC showed good antifungal activities and CUR was stronger. It was further concluded from structure-activity relationship that the existence of methoxy group might enhance lipophilicity of the mother nucleus, which made it easier for the molecular to enter into the cell membrane of fungi to inhibit its growth. Conclusion This study provides a new method for screening new antifungal agents with high efficacy and low toxicity. Meanwhile, it contributes to the application of curcuminoids as food additive, colorant, and drug. 14Construction of LDPC Codes for the Layered Decoding Algorithm显示文摘The layered decoding algorithm has been widely used in the implementation of Low Density Parity Check (LDPC) decoders, due to its high convergence speed. However, the pipeline operation of the layered decoder may introduce memory access conflicts, which heavily deteriorates the decoder throughput. To essentially deal with the issue of memory access conflicts, we propose a construction algorithm of LDPC codes, to which a constraint condition is added in the Progressive Edge-Growth (PEG) algorithm. The constraint condition can guarantee that for our constructed LDPC codes, the sets of all the variable nodes connected to the consecutive layers do not share any common variable node, which can avoid the memory access conflicts. Simulation results show that the performance of our constructed LDPC codes is close to the several other LDPC codes adopted in wireless standards. Moreover, compared with the decoder for IEEE 802. 16e LDPC codes, the throughput of our LDPC decoder has large improvement, while the chip resource consumption is unchanged. 15High Potential Columnar Nanocrystalline AlN Films Deposited by RF Reactive Magnetron Sputtering显示文摘Columnar nanocrystalline aluminum nitride(cnc-AlN) thin films with(002) orientation and uniform texture have been deposited successfully on large silicon wafers by RF reactive magnetron sputtering.At the optimum sputtering parameters, the deposited cnc-AlN thin films show a c-axis preferred orientation with a crystallite size of about 28 nm and surface roughness(RMS) of about 1.29 nm. The cnc-AlN thin films were well transparent with an optical band gap about 4.8 e V, and the residual compressive stress and the defect density in the film have been revealed by Ramon spectroscopy. 16Effect of Sizing on the Interfacial Shear Strength of Carbon Fiber/Epoxy Resin Monofilament Composite显示文摘The single fi ber fragmentation test(SFFT) was used to measure the interfacial shear strength(IFSS) of sized and unsized CF800/epoxy resin monofilament composite in order to evaluate the effect of sizing respectively. Besides, the interfacial reinforcing mechanism was explored by analyzing the surface morphology of the carbon fi bers, the wettability between the carbon fi bers and the epoxy resin, and the chemical characteristics of the fi ber surface. Moreover, the effect of sizing on heat and humidity resistance of interface was investigated by aging test. 20Genetic Analysis of Chromosomal Loci Affecting the Content of Insoluble Glutenin in Common Wheat显示文摘In common wheat, insoluble glutenin(IG) is an important fraction of flour glutenin macropolymers, and insoluble glutenin content(IGC)is positively associated with key end-use quality parameters. Here, we present a genetic analysis of the chromosomal loci affecting IGC with the data collected from 90 common wheat varieties cultivated in four environments. Statistical analysis showed that IGC was controlled mainly genetically and influenced by the environment. Among the major genetic components known to affect end-use quality,1BL/1RS translocation had a significantly negative effect on IGC across all four environments. As to the different alleles of Glu-A1,-B1 and-D1 loci, Glu-A1 a, Glu-B1 b and Glu-D1 d exhibited relatively strong positive effects on IGC in all environments. To identify new loci affecting IGC, association mapping with 1355 DAr T markers was conducted. A total of 133 markers were found associated with IGC in two or more environments(P < 0.05), ten of which consistently affected IGC in all four environments. The phenotypic variance explained by the ten markers varied from 4.66% to 8.03%, and their elite alleles performed significantly better than the inferior counterparts in enhancing IGC. Among the ten markers, w Pt-3743 and w Pt-733835 reflected the action of Glu-D1, and w Pt-664972 probably indicated the effect of Glu-A1. The other seven markers, forming three clusters on 2AL, 3BL or 7BL chromosome arms, represented newly identified genetic determinants of IGC.
