| 1 | Effects of dietary supplementation of probiotic,Clostridium butyricum,on growth performance,immune response,intestinal barrier function,and digestive enzyme activity in broiler chickens challenged with Escherichia coli K88显示文摘Background:Colibacillosis caused by enterotoxigenic Escherichia coli(E.coli) results in economic losses in the poultry industry.Antibiotics are usually used to control colibacillosis,however,E.coli has varying degrees of resistance to different antibiotics.Therefore the use of probiotics is becoming accepted as an alternative to antibiotics.In this study,we evaluated the effects of Clostridium butyricum(C.butyricum) on growth performance,immune response,intestinal barrier function,and digestive enzyme activity in broiler chickens challenged with Escherichia coli(E.coif) K88.Methods:The chickens were randomly divided into four treatment groups for 28 days.Negative control treatment(NC) consisted of birds fed a basal diet without E.coli K88 challenge and positive control treatment(PC) consisted of birds fed a basal diet and challenged with E.coli K88.C.butyricum probiotic treatment(CB) consisted of birds fed a diet containing 2×10~7 cfu C butyricum/kg of diet and challenged with E.coli K88.Colistin sulfate antibiotic treatment(CS)consisted of birds fed a diet containing 20 mg colistin sulfate/kg of diet and challenged with E.coli K88.Results:The body weight(BW) and average day gain(ADG) in the broilers of CB group were higher(P< 0.05) than the broilers in the PC group overall except the ADG in the 14-21 d post-challenge.The birds in CB treatment had higher(P < 0.05) concentration of tumor necrosis factor-α(TNF-α) at 3 and 7 d post-challenge,and higher(P < 0.05)concentration of interleukin-4(IL-4) at 14 d post-challenge than those in the PC treatment group.The concentration of serum endotoxin in CB birds was lower(P<0.05) at 21 d post-challenge,and the concentrations of serum diamine oxidase in CB birds were lower(P < 0.05) at 14 and 21 d post-challenge than in PC birds.Birds in CB treatment group had higher(P< 0.05) jejunum villi height than those in PC,NC,or CS treatment at 7,14,and 21 d post-challenge.In comparison to PC birds,the CB birds had lower(P < 0.05) jejunum crypt depth during the whole experiment.The birds in CB or CS treatment group had higher(P < 0.05) activities of amylase and protease at 3,7,and 14 d post-challenge,and higher(P< 0.05) activity of lipase at 3,7 d post-challenge than PC birds.Conclusions:In all,these results indicate that dietary supplementation with C.butyricum promotes immune response,improves intestinal barrier function,and digestive enzyme activities in broiler chickens challenged with E.coli K88.There is no significant difference between the C.butyricum probiotic treatment and the colistin sulfate antibiotic treatment.Therefore,the C.butyricum probiotic may be an alternative to antibiotic for broiler chickens. | Ling Zhang Lingling Zhang Xiu'an Zhan Xinfu Zeng Lin Zhou Guangtian Cao An'guo Chen Caimei Yang | 2016 | Journal of Animal Science and Biotechnology2016,7,1: | 43 |
| 2 | Formability,microstructure evolution and mechanical properties of wire arc additively manufactured AZ80M magnesium alloy using gas tungsten arc welding显示文摘Wire arc additive manufacturing(WAAM)technology has been used to fabricate the multi-layer single-pass deposited wall of AZ80M magnesium(Mg)alloy by gas tungsten arc welding.The formability,thermal cycles,microstructural evolution and mechanical properties of the WAAM AZ80M Mg alloy were investigated.The results show that there was significant difference in the temperature variation and the geometries between the original several layers and the subsequent deposited layers.Owing to the arc energy input,the interpass temperature rised rapidly and then stabilized at 150℃.As a result,the width of the deposited wall increased and then kept stable.There were obvious differences in the microstructure of the WAAM AZ80M Mg alloy among the top zone,intermediate zone and bottom zone of deposited wall.During the arc deposition process,theβphase of the WAAM AZ80M Mg alloy redissolved due to the cyclic heat accumulation,and then precipitated in the grain boundary.The cyclic heat accumulation also led to weakening of dendrite segregation.From the substrate to the top zone,the hardness of the deposited wall decreased gradually,and the intermediate zone which was the main body of deposited wall had relatively uniform hardness.The tensile properties of the WAAM AZ80M Mg alloy were different between the vertical direction and the horizontal direction.And the maximum ultimate tensile strength of the WAAM AZ80M Mg alloy was 308 MPa which was close to that of the as-extruded AZ80M Mg alloy. | Yangyang Guo Gaofeng Quan Yinglong Jiang Lingbao Ren Lingling Fan Houhong Pan | 2021 | Journal of Magnesium and Alloys2021,9,1: | 10 |
| 3 | Protective effects of Jiayan Kangtai granules on autoimmune thyroiditis in a rat model by modulating Th17/Treg cell balance显示文摘OBJECTIVE: To investigate the protective effects of Jiayan Kangtai(JYKT) granules, consisting of 9 Chinese herbs, in a rat model of autoimmune thyroiditis(AIT), and the possible underlying mechanism.METHODS: Female Lewis rats(6-8 weeks) were randomly apportioned to 5 groups of 10, including a normal control. AIT was induced in the untreated AIT-model group, and rats treated subsequently with daily low, medium, or high dose JYKT granules. After 12 weeks, plasma levels of thyroid autoantibodies and morphological changes in the thyroid were detected by enzyme-linked immunosorbent assay and histological examination, respectively. The presence of interleukin(IL)-6,IL23 p19, and IL-2 in thyroid tissue was assessed by immunohistochemical staining. The percentages of T helper(Th)17 cells and regulatory T cells(Tregs) in the peripheral blood were analyzed by flow cytometry. Relevant levels of cytokines and proteins were examined via bead-based multiplex flow cytometry and ELISA, respectively. Expressions of genes and proteins regulated by Th17 cells and Tregs were shown by real-time PCR and Western blot.RESULTS: Compared to the control, AIT-model rats had higher plasma concentrations of thyroid autoantibodies. The high-dose JYKT rats showed significantly lower levels of thyroid autoantibodies compared with the AIT model group. Rats in the AIT-JYKT groups also had fewer thyroid lesions and less lymphocytic infiltration, a lower percentage of Th17 cells, and a higher percentage of Tregs, compared with the AIT-model. Rats given high-dose JYKT had a significantly lower Th17/Treg ratio compared with the AIT model. | 5 | Effect of Jiawei Shenfu decoction on tumor necrosis factor-alpha and nuclear factor-kappa B in patients who have chronic heart failure with syndromes of deficiency of heart Yang显示文摘OBJECTIVE:To examine the clinical efficacy of Jiawei Shenfu decoction on tumor necrosis factor-alpha (TNF-cα) and nuclear factor-kappa B (NF-KB) levels in patients who have chronic heart failure with syndromes of deficiency of heart Yang.METHODS:A total of 63 patients with syndromes of deficiency of heart Yang (chronic heart failure)were enrolled.Patients were randomly divided into the control group and Jiawei Shenfu group.All patients received standard medications for treatment of chronic heart failure.Patients in the Jiawei Shenfu group were additionally provided Jiawei Shenfu decoction one dose daily.Treatments continued for 4 consecutive weeks.The primary endpoint was the change in plasma B-type natriuretic peptide (BNP),NF-KB,and TNF-cα levels during 4 weeks of treatment.RESULTS:At the 4-week follow-up,a significant reduction in BNP levels compared with baseline was observed in both groups,but the Jiawei Shenfu decoction group showed a significantly greater reduction than did the control group.The Jiawei Shenfu group also showed superior performance regarding the Minnesota Living with Heart Failure Questionnaire score,the Chinese medicine syndrome score,heart rate,left ventricular ejection fraction,and 6-min walking distance compared with the control group.The degree of changes in NF-KB and TNF-α levels in the Jiawei Shenfu group was more significant than that in the control group.CONCLUSION:Routine medicine combined with Jiawei Shenfu decoction for patients with heart Yang deficiency syndrome in chronic heart failure can improve the left ventricular ejection fraction and cardiac function,and reduce BNP levels.The mechanism may be related to inhibition of pro-inflammatory cytokines and the NF-KB-induced kinase pathway,leading to amelioration of the inflammatory response. | Yu Mei Guo Hangyuan Ye Lingling Bian Jiaping Ma Lijuan Zheng Chunli | 2019 | Journal of Traditional Chinese Medicine2019,39,3: | 6 |

| 9 | Methionine deficiency and its hydroxy analogue influence chicken intestinal 3-dimensional organoid development显示文摘Methionine and its hydroxy analogue(MHA)have been shown to benefit mouse intestinal regeneration.The intestinal organoid is a good model that directly reflects the impact of certain nutrients or chemicals on intestinal development.Here,we aimed to establish a chicken intestinal organoid culture method first and then use the model to explore the influence of methionine deficiency and MHA on intestinal organoid development.The results showed that 125-mm cell strainer exhibited the highest efficiency for chicken embryo crypt harvesting.We found that transforming growth factor-b inhibitor(A8301)supplementation promoted enterocyte differentiation at the expense of the proliferation of intestinal stem cells(ISC).The mitogen-activated protein kinase p38 inhibitor(SB202190)promoted intestinal organoid formation and enterocyte differentiation but suppressed the differentiation of enteroendocrine cells,goblet cells and Paneth cells.However,the suppression of enteroendocrine cell and Paneth cell differentiation by SB202190 was alleviated at the presence of A8301.The glycogen synthase kinase 3 inhibitor(CHIR99021),valproic acid(VPA)alone and their combination promoted chicken intestinal organoid formation and enterocyte differentiation at the expense of the expression of Paneth cells and goblet cells.Chicken serum significantly improved organoid formation,especially in the presence of A8301,SB202190,CHIR99021,and VPA,but inhibited the differentiation of Paneth cells and enteroendocrine cells.Chicken serum at a concentration of 0.25%meets the requirement of chicken intestinal organoid development,and the beneficial effect of chicken serum on chicken intestinal organoid culture could not be replaced by fetal bovine serum and insulin-like growth factor-1.Moreover,commercial mouse organoid culture medium supplemented with A8301,SB202190,CHIR99021,VPA,and chicken serum promotes chicken organoid budding.Based on the chicken intestinal organoid model,we found that methionine deficiency mimicked by cycloleucine suppressed organoid formation and organoid size,and this effect was reinforced with increased cycloleucine concentrations.Methionine hydroxy analogue promoted regeneration of ISC but decreased cell differentiation compared with the results obtained with L-methionine.In conclusion,our results provide a potentially excellent guideline for chicken intestinal organoid culture and insights into methionine function in crypt development. | Youli Wang Qihang Hou Yuqin Wu Yanwei Xu Yan Liu Jing Chen Lingling Xu Yuming Guo Shuai Gao Jianmin Yuan | 2022 | Animal Nutrition2022,,1: | 4 |
| 10 | Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. | 14 | Pore Structure and Permeability Characterization of Highrank Coal Reservoirs: A Case of the Bide-Santang Basin, Western Guizhou, South China显示文摘The methods of nuclear magnetic resonance(NMR)spectroscopy,mercury injection porosimetry(MIP),and gas-water relative permeability(GWRP)were used to reveal the pore structure and permeability characteristics of high-rank coal reservoirs in the Bide-Santang basin,western Guizhou,South China,to provide guidance for coalbed methane(CBM)exploration and exploitation and obtain direct insights for the development of CBM wells.The results indicate that the coal reservoirs in the study area are characterized by well-developed adsorption pores and poorly developed seepage pores.The bimodal NMR transverse relaxation time(T2)spectra and the mutation in the fractal characteristic of the MIP pore volume indicate poor connectivity between the adsorption pores and the seepage pores.As a result,the effective porosity is relatively low,with an average of 1.70%.The irreducible water saturation of the coal reservoir is relatively high,with an average of 66%,leading to a low gas relative permeability under irreducible water saturation.This is the main reason for the low recovery of high-rank CBM reservoirs,and effective enhanced CBM recovery technology urgently is needed.As a nondestructive and less time-consuming technique,the NMR is a promising method to quantitatively characterize the pores and fractures of coals. | GUO Chen QIN Yong MA Dongmin XIA Yucheng BAO Yuan CHEN Yue LU Lingling | 2020 | Acta Geologica Sinica(English Edition)2020,94,2: | 3 |

Their physicochemical properties were investigated using FT-IR, TEM,powder XRD, VSM, TGA, DLS and zeta potential. We found that CA-IONP dispersion was composed of monocrystalline particles while CS-IONP and FA-g-CS-IONP were composed of polycrystalline aggregates. All IONPs retained the crystalline structure of magnetite and exhibited the superparamagnetic behavior. Their saturation magnetization decreased with the increase in the amount of their organic coatings. Their drug loading capacities, drug release patterns and in vitro anticancer efficiencies were studied by using doxorubicin(DOX) as a model drug. DOX@CS-IONP and DOX@FA-g-CSIONP exhibited lower drug loading while showing higher water dispersity when compared with DOX@CA-IONP. All IONPs were surface charged and they tended to agglomerate in medium with high pH value and ionic strength. In the presence of chitosan or FA-g-CS coatings, their DOX release rate was slowed down compared with that of DOX@CA-IONP. Unloaded IONPs exhibited nearly no cytotoxicity on both cancer cells and normal cells in the presence of chitosan and FA-g-CS when compared with CA-IONP which presented high cytotoxicity. However, DOX@FA-g-CS-IONP showed significantly cytotoxicity on folate receptors(FRs) positive breast cancer cells while exhibiting nearly no cytotoxicity on FRs negative normal cells. | 17 | Validation of MODIS ocean-colour products in the coastal waters of the Yellow Sea and East China Sea显示文摘An extensive study collected in situ data along the Yellow Sea(YS) and East China Sea(ECS) to assess the radiometric properties and the concentration of the water constituents derived from Moderate Resolution Imaging Spectroradiometer(MODIS). Thirteen high quality match-ups were obtained for evaluating the MODIS estimates of Rrs(λ), chlorophyll a(Chl a) and concentrations of suspended particulate sediment matter(SPM). For MODIS Rrs(λ), the mean absolute percentage difference(APD) was in the range of 20%–36%, and the highest uncertainty appeared at 412 nm, whereas the band ratio of Rrs(λ) at 488 nm compared with that at 547 nm was highly consistent, with an APD of 7%. A combination of near-infrared bands and shortwave infrared wavelengths atmosphere correction algorithm(NIR-SWIR algorithm) was applied to the MODIS data, and the estimation accuracy of Rrs were improved at most of the visible spectral bands except 645 nm, 667 nm and 678 nm. Two ocean-colour empirical algorithms for Chl a estimation were applied to the processed data, the results indicated that the accuracy of the derived Chl a values was obviously improved, the four-band algorithms outperformed the other algorithm for measured and simulated datasets, and the minimum APD was 35%. The SPM was also quantified. Two regional and two coastal SPM algorithms were modified according to the in situ data. By comparison, the modified Tassan model had a higher accuracy for the application along the YS and ECS with an APD of 21%. | 19 | Amplicon-Based Illumina Sequencing and Quantitative PCR Reveals Nanoplankton Diversity and Biomass in Surface Water of Qinhuangdao Coastal Area, China显示文摘Aureococcus anophagefferens caused brown tides for three consecutive years from 2009 to 2011 in the coastal waters of Qinhuangdao, China, with numerous, widespread ecological and economic impact on ecosystems. To understand the population dy- namics of nanoplankton during the brown tides, sequences of the V9 region of the 18S rDNA gene, used as a marker, were analyzed by Illumina sequencing to assess nanoplankton biomass, and real-time fluorescence quantitative PCR was performed to analyze spa- tial variation in the 18S rDNA copy concentrations of nanoplankton off the Qinhuangdao coast in July, 2011. The results showed that A. anophagefferens and Minutocellus polymorphus were the dominant species in the local phytoplankton community during the brown tide in July 2011. The highest 18S rDNA copy concentrations of A. anophagefferens and M. polymorphus were detected at stations SHG and FN, respectively. The central area most strongly affected by the brown tide migrated southward from 2011 to 2013. Redundancy analysis (RDA) showed that the decreasing NOx concentration might provide suitable nutrient conditions for the A. anophagefferens outbreak. During the brown tide caused by A. anophagefferens, other phytoplankton, such as diatoms, cryptophytes, chlorophytes, dinoflagellates and other flagellates, could co-occur with it.
