| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Effects of dietary protein/energy ratio on growth performance,carcass trait,meat quality,and plasma metabolites in pigs of different genotypes显示文摘Background:The protein/energy ratio is important for the production performance and utilization of available feed resources by animals.Increased protein consumption by mammals leads to elevated feed costs and increased nitrogen release into the environment.This study aimed to evaluate the effects of dietary protein/energy ratio on the growth performance,carcass traits,meat quality,and plasma metabolites of pigs of different genotypes.Methods:Bama mini-pigs and Landrace pigs were randomly assigned to two dietary treatment groups(Chinese conventional diet with low protein/energy ratio or National Research Council diet with high protein/energy ratio;n = 24 per treatment) in a 2x2 factorial arrangement.Blood and muscle samples were collected at the end of the nursery,growing,and finishing phases.Results:We observed significant interactions(P< 0.05) between breed and diet for total fat percentage,intramuscular fat(IMF) content,protein content in biceps femoris(BF) muscle,and plasma urea nitrogen(UN) concentration in the nursery phase;for average daily gain(ADG),average daily feed intake(ADFI),dry matter,IMF content in psoas major(PM) muscle,and plasma total protein and albumin concentrations in the growing phase;and for drip loss and plasma UN concentration in the finishing phase.Breed influenced(P < 0.05) growth performance,carcass traits,and meat quality,but not plasma metabolites.Throughout the trial,Landrace pigs showed significantly higher(P < 0.05) ADG,ADFI,dressing percentage,lean mass rate,and loin-eye area than did Bama mini-pigs,but significantly lower(P< 0.05)feed/gain ratio,fat percentage,backfat thickness,and IMF content.Dietary protein/energy ratio influenced the pH value,chemical composition of BF and PM muscles,and plasma activities of glutamic-pyruvic transaminase and gamma-glutamyl transpeptidase,and plasma concentration of UN.Conclusions:Compared with Landrace pigs,Bama mini-pigs showed slower growth and lower carcass performance,but had better meat quality.Moreover,unlike Landrace pigs,the dietary protein/energy ratio did not affect the growth performance of Bama mini-pigs.These results suggest that,in swine production,low dietary protein/energy ratio may be useful for reducing feed costs and minimizing the adverse effects of ammonia release into the environment. | Yingying Liu Xiangfeng Kong Guoli Jiang Bi'e Tan Jinping Deng Xiaojian Yang Fengna Li Xia Xiong Yulong Yin | 2015 | Journal of Animal Science and Biotechnology2015,6,4: | 37 |

| 4 | Phylogenetic reconstruction of Chirita and allies (Gesneriaceae) with taxonomic treatments显示文摘Chirita D. Don, a large genus in the subfamily Cyrtandroideae of Gesneriaceae, has been the subject of much debate whether it is a natural group or not. In addition, the highly heterogeneous Chirita has also been very problematic with regard to delimitation and subdivision. Here we used the nrDNA internal transcribed spacer and cpDNA trnL-F for molecular phylogenetic analaysis, combined with morphological data. Our results suggest that Chirita is an artificial, polyphyletic genus. The most important character that defines Chirita, the dorso-ventrally oblique and bilamellar stigma, has evolved convergently in different clades of diandrous Cyrtandroideae. Chirita sensu stricto only includes the species of Chirita sect. Chirita, whereas Chirita sect. Microchirita is an independent clade located at the basal node of the phylogenetic tree. Chirita sect. Liebigia is closely related to Didymocarpus with an entire stigma unlike other species of Chirita. The species of Chirita sect. Gibbosaccus, Chiritopsis, Primulina, and Wentsaiboea form a monophyletic group that is sister to a strongly supported clade comprising four monotypic genera Paralagarosolen, Calcareoboea, Petrocodon, and Tengia. | 5 | An autotrophic nitrogen removal process:Short-cut nitrification combined with ANAMMOX for treating diluted effluent from an UASB reactor fed by landfill leachate显示文摘A combined process consisting of a short-cut nitrification (SN) reactor and an anaerobic ammonium oxidation upflow anaerobic sludge bed (ANAMMOX) reactor was developed to treat the diluted effluent from an upflow anaerobic sludge bed (UASB) reactor treating high ammonium municipal landfill leachate.The SN process was performed in an aerated upflow sludge bed (AUSB) reactor (working volume 3.05 L),treating about 50% of the diluted raw wastewater.The ammonium removal efficiency and the ratio of NO 2 N to NOx-N in the effluent were both higher than 80%,at a maximum nitrogen loading rate of 1.47 kg/(m 3 ·day).The ANAMMOX process was performed in an UASB reactor (working volume 8.5 L),using the mix of SN reactor effluent and diluted raw wastewater at a ratio of 1:1.The ammonium and nitrite removal efficiency reached over 93% and 95%,respectively,after 70-day continuous operation,at a maximum total nitrogen loading rate of 0.91 kg/(m 3 ·day),suggesting a successful operation of the combined process.The average nitrogen loading rate of the combined system was 0.56 kg/(m 3 ·day),with an average total inorganic nitrogen removal efficiency 87%.The nitrogen in the effluent was mostly nitrate.The results provided important evidence for the possibility of applying SN-ANAMMOX after UASB reactor to treat municipal landfill leachate. | Jie Liu Jian'e Zuo Tang Yang Shuquan Zhu Sulin Kuang Kaijun Wang | 2010 | Journal of Environmental Sciences2010,22,5: | 26 |
| 6 | Observation of ocean current response to 1998 Hurricane Georges in the Gulf of Mexico显示文摘The ocean current response to a hurricane on the shelf-break is examined. The study area is the DeSoto Canyon in the northeast Gulf of Mexico, and the event is the passage of 1998 Hurricane Georges with a maximum wind speed of 49 m/s. The data sets used for analy- sis consist of the mooring data taken by the Field Program of the DeSoto Canyon Eddy Intrusion Study, and simultaneous winds ob- served by NOAA (National Oceanic and Atmospheric Administration) Moored Buoy 42040. Time-depth ocean current energy density images derived from the observed data show that the ocean currents respond almost immediately to the hurricane with important differ- ences on and off the shelf. On the shelf, in the shallow water of 100 m, the disturbance penetrates rapidly downward to the bottom and forms two energy peaks, the major peak is located in the mixed layer and the secondary one in the lower layer. The response dissipates quickly after external forcing disappears. Off the shelf, in the deep water, the major disturbance energy seems to be trapped in the mixed layer with a trailing oscillation; although the disturbance signals may still be observed at the depths of 500 and 1 290 m. Verti- cal dispersion analysis reveals that the near-initial wave packet generated off the shelf consists of two modes. One is a barotropic wave mode characterized by a fast decay rate of velocity amplitude of 0.020 s-1, and the other is baroclinic wave mode characterized by a slow decay rate of 0.006 9 s-1. The band-pass-filtering and empirical function techniques are employed to the frequency analysis. The results indicate that all frequencies shift above the local inertial frequency. On the shelf, the average frequency is 1.04f in the mixed layer, close to the diagnosed frequency of the first baroclinic mode, and the average frequency increases to 1.07f in the thermocline. Off the shelf, all frequencies are a little smaller than the diagnosed frequency of the first mode. | 7 | Chimeric antigen receptor-engineered T-cell therapy for liver cancer显示文摘Background: Chimeric antigen receptor-engineered T-cell(CAR-T) therapy is a newly developed immunotherapy used in the treatment of cancers. Because CAR-T therapy has shown great success in treating CD19-positive hematological malignancies, its application has been explored in the treatment of solid tumors, such as liver cancer. In this review, we discuss the immune characteristics of liver cancer, the obstacles encountered during the application of CAR-T therapy, and preclinical and clinical progress in the use of CAR-T therapy in patients with liver cancer.Data sources: The data on CAR-T therapy related to liver cancers were collected by searching Pub Med and the Web of Science databases prior to December 2017 with the keywords 'chimeric antigen receptor','CAR-T', 'liver cancer', 'hepatocellular carcinoma', and 'solid tumor'. Additional articles were identified by manual search of references found in the primary articles. The data for clinical trials were collected by searching Clinical Trials.gov.Results: The liver has a tolerogenic nature in the intrahepatic milieu and its tumor microenvironment significantly affects tumor progression. The obstacles that reduce the efficacy of CAR-T therapy in solid tumors include a lack of specific tumor antigens, limited trafficking and penetration of CAR-T cells to tumor sites, and an immunosuppressive tumor microenvironment. To overcome these obstacles, several strategies have emerged. In addition, several strategies have been developed to manage the side effects of CAR-T, including enhancing the selectivity of CARs and controlling CAR-T activity. To date, no clinical trials of CAR-T therapy against HCC have been completed. However, preclinical studies in vitro and in vivo have shown potent antitumor efficacy. Glypican-3, mucin-1, epithelial cell adhesion molecule, carcinoembryonic antigen, and other targets are currently being studied.Conclusions: The application of CAR-T therapy for liver cancer is just beginning to be explored and more research is needed. | 10 | Rotational Swashplate Pulse Continuously Variable Transmission Based on Helical Gear Axial Meshing Transmission显示文摘The current research on pulse continuously variable transmission(CVT) is mainly focused on reducing the pulse degree and making pulse degrees a constant value. Current research mainly confined to find out new design parameters by using the method of optimization, and reduce the pulse degree of pulse CVT and its range of variation. But the fact is that the reduction of the pulse degree is not significant. This article presents a new structure of mechanical pulse CVT-the rotational swashplate pulse CVT with driven by helical gear axial meshing. This transmission is simple and compact in structure and low in pulsatile rate (it adopts 6 guide rods), and the pulsatile degree is irrelevant to the transmission ratio. Theoretically, pulsatile rate could be reduced to zero if appropriate curved surface of the swashplate is used. Compared with the connecting rod pulse CVT, the present structure uses helical gear mechanism as transmission part and it avoids unbalanced inertial force in the former model. This paper analyzes the principle of driving of this transmission, presents its mechanical structure, and discusses its motion characteristics. Experimental prototype of this type of CVT has been manufactured. Tests for the transmission efficiency(when the rotational speed of the output shaft is the maximum) and the angular velocity of the output shaft have been carried out, and data have been analyzed. The experimental results show that the speed of the output shaft for the experimental prototype is slightly lower than the theoretical value, and the transmission efficiency of the experimental prototype is about 70%. The pulse degree of the CVT discussed in this paper is less than the existing pulse CVT of other types, and it is irrelevant to the transmission ratio of the CVT. | 14 | Spring-back deformation in tube bending显示文摘The spring-back of a bending metal tube was studied through extensive experiments and finite element method (FEM) analysis. An approximate equation for the spring-back angle of bending was deduced. It is noted that the mechanical properties of the material (in a tubular form) are quite different from those found in the standard tensile tests (when the materials are in bar forms). This is one of the major reasons that result in the discrepancies in the outcomes of experimental study,FEM calculations,and spring-back analysis. It is therefore of crucial importance to study the mechanical properties of the materials in their tubular forms. The experiments and FEM simulations prove that the spring-back angle is significantly affected by the mechanical properties of the materials. The angle decreases accordingly with plastic modulus,but changes inversely with the hardening index and elastic modulus. The spring-back angle is also affected by the conditions of tube deformation:it increases accordingly with the relative bending radius but changes inversely with the relative wall thickness. | 17 | Probabilistic ecological risk assessment for three chlorophenols in surface waters of China显示文摘Individual and combined assessment of risks of adverse effects to aquatic ecosystems of three chlorophenols(CPs),including 2,4dichlorophenol(2,4-DCP),2,4,6-trichlorophenol(2,4,6-TCP) and pentachlorophenol(PCP),were conducted.A probabilistic approach based on the concentrations of CPs in surface waters of China was used to determine the likelihood of adverse effects.The potential risk of CPs in surface waters of China was determined to be of concern,especially PCP and mixtures of CPs.The risks of adverse effects were examined as the joint probabilities of exposure and response.The joint probability for PCP was 0.271 in the worst case and 0.111 in the median case,respectively.Based on the cumulative probability,5% of aquatic organisms included in the assessment would be affected 21.36% of the time in the worst case and 5.99% of the time in median case,respectively.For the mixtures of CPs,the joint probability were 0.171 in the worst case and 0.503 in median case,respectively and 5% of species would be affected 49.83% of the time for the worst case and 12.72% in the median case,respectively.Risks of effects of the individual CPs,2,4-DCP and 2,4,6-TCP were deemed to be acceptable with a overlapping probability of < 0.1 with 5% of species being affected less than 4% of the time. | Liqun Xing Hongling Liu John E Giesy Xiaowei Zhang Hongxia Yu | 2012 | Journal of Environmental Sciences2012,24,2: | 7 |

| 19 | Pore throat characteristics of tight sandstone of Yanchang Formation in eastern Gansu,Ordos Basin显示文摘An important factor to evaluate reservoir quality is the pore-throat size.However,the strong heterogeneity makes it difficult to characterize the pore-throat distribution in tight reservoirs.The field emission scanning electron microscope(FESEM),high pressure mercury injection and rate-controlled mercury injection are used to investigate the pore-throat size distribution in tight sandstone reservoirs of Member 7 of the Yanchang Formation in eastern Gansu,Ordos Basin,and studies of the pore throat size controlling on physical property of the tight sandstone reservoirs are also carried out.The result shows that the pore type is mainly dominated by the residual intergranular pore,dissolution pore,micropore and a few micro-fractures;the high-pressure mercury injection experiment indicates that the pore-throat size ranges from 0.0148 μm to 40mm,the pore throat more than 1 mm is less;the ratecontrolled mercury injection experiment reveals that for samples with different physical properties,the pore radius mainly varies from 80 μm to 350 μm;the throat radius exhibits the strong heterogeneity,and is from 0.12 μm to 30μm;the pore-throat size can be effectively characterized by combination of high-pressure and rate-controlled mercury injections,and it varies from 0.0148 μm to 350 μm.The permeability is mainly controlled by the large pore throat(>R_(50))which accounts for a small proportion;in the tight sandstone with the permeability greater than 0.1 mD,the permeability is mainly controlled by the micropore and mesopore;in the tight sandstone with the permeability smaller than 0.1 mD,the permeability is mainly controlled by the nanopore and micropore;the proportion of small pore throat increases with reduction of permeability,it is important that the small pore throat influences the reservoir storage property though its effect on permeability are small. | Hao Wu Chunlin Zhang Youliang Ji Rui'e Liu Shang Cao Sheng Chen Yunzhao Zhang Ye Wang Wei Du Gang Liu | 2018 | Petroleum Research2018,3,1: | 7 |
| 20 | Aquatic Species Dominate Organic Matter in Qinghai Lake during the Holocene:Evidence from Eolian Deposits around the Lake显示文摘Total organic carbon(TOC) in lake sediments and its stable carbon isotope(δ^(13)C_(org)) are widely applied to investigate paleoenvironmental changes even though their implications are complicated and multi-explanatory. Organic geochemistry studies of lake sediments from Qinghai Lake have been investigated, but some interpretations are controversial. In this study, TOC of one Holocene eolian section and δ^(13)C_(org) of three eolian sections were studied around Qinghai Lake. Results indicate that the TOC content in eolian deposits was low during the Early Holocene, and gradually increased to high values during the Middle and Late Holocene. The variation in TOC content of eolian deposits is different from that in the lacustrine sequence from Qinghai Lake during the Holocene. The δ^(13)C_(org) values in the eolian sections were relatively stable, with oscillation amplitudes of ~4‰(ranging from-25.8‰ to-22.1‰), in contrast to ~10‰ variation s in δ^(13)C_(org) values(varying from-30‰ to-20‰) in lacustrine sediments. Through comparison of TOC and δ^(13)C_(org) values between eolian deposits and lacustrine sediments, we can confirm indications that the organic matter in Qinghai Lake sediments during the Holocene was primarily a contribution of the aquatic species in the lake.
