1Immunoregulatory Effects of Ethyl-acetate Fraction of Extracts from Tetrastigma Hemsleyanum Diels et. Gilg on Immune Functions of ICR Mice显示文摘Objective To evaluate the effects of ethyl-acetate fraction (EAF) of extracts from Tetrastigma hemsleyanum Diels et. Gilg (TDG) on immune functions of ICR mice. Methods ICR mice were exposed to different doses of EAF for 15 or 30 days and then their immune functions were analyzed, including ConA-induced splenic lymphocyte transformation, SRBC- induced delayed type hypersensitivity response, serum hemolysin analysis, antibody-producing cells, peritoneal macrophage phagocytized chicken red blood cells, natural killer cell activity, and serum level of cytokines. Results EAF of extracts from TDG at different doses had various effects on immune functions of ICR mice. As compared with the controls, it increased the mouse spleen lymphocyte transformation induced by ConA, the left-hind voix pedis thickness and the number of plague forming cells (PFCs) at the dose of 1.82 mg/mL, 5.48 mg/mL, and 9.12 mg/mL, respectively; increased the ink clearance ability at the dose of 0.91 mg/mL, 1.82 mg/mL, 5.48 mg/mL, and 9.12 mg/mL, respectively; increased the phagocytosis index of mononuclear-macrophages and production of serum interferon-gamma (IFN-γ) at the dose of 5.48 mg/mL; and could promote the production of serum tumor necrosis factor-alpha (TNF-α) at the dose of 9.12 mg/mL. 3Massive presacral bleeding during rectal surgery: From anatomy to clinical practice显示文摘AIM: To investigate control of two different types of massive presacral bleeding according to the anatomy of the presacral venous system. METHODS: A retrospective review was performed in 1628 patients with middle or low rectal carcinoma who were treated surgically in the Department of Colorectal Surgery, Changhai Hospital, Shanghai, China from January 2008 to December 2012. In four of these patients, the presacral venous plexus (n = 2) or basivertebral veins (n = 2) were injured with massive presacral bleeding during mobilization of the rectum. The first two patients with low rectal carcinoma were operated upon by a junior associate professor and the source of bleeding was the presacral venous plexus. The other two patients with recurrent rectal carcinoma were both women and the source of bleeding was the basivertebral veins.RESULTS: Two different techniques were used to con-trol the bleeding. In the first two patients with massive bleeding from the presacral venous plexus, we used suture ligation around the venous plexus in the area with intact presacral fascia that communicated with the site of bleeding (surrounding suture ligation). In the second two patients with massive bleeding from the basivertebral veins, the pelvis was packed with gauze, which resulted in recurrent bleeding as soon as it was removed. Following this, we used electrocautery applied through one epiploic appendix pressed with a long Kelly clamp over the bleeding sacral neural foramen where was felt like a pit Electrocautery adjusted to the highest setting was then applied to the clamp to 'weld' closed the bleeding point. Postoperatively, the blood loss was minimal and the drain tube was removed on days 4-7. 4Prevention of Cistanche salsa Extract on Hepatic Fibrosis Induced by Carbon Tetrachloride in Rats显示文摘Objective To explore the antifibrotic effect of echinacoside on carbon tetrachloride(CCl4)-induced hepatic fibrosis in rats.Methods Male Wistar rats were randomly divided into normal control(n=8),model(n=14),and echinacoside treatment(n=14)groups.The hepatic fibrosis model was induced by CCl4compositor.The rats were ig administered with echinacoside at a daily dose of 50 mg/kg.The anti-oxidant status,liver function parameters,and hepatic hydroxyproline content were detected by chromatometry.The serum levels of hyaluronic acid(HA),type IV collagen(CIV),type III precollagen(PIIIP),and laminin(LN)were assayed with radioimmunoassay.The hpatic injury was detected by haematoxylin-eosine staining.The deposition of collagen was observed with Masson staining.Results Echinacoside increased the superoxide dismutase activity and reduced the levels of malondialdehyde,aspartate aminotransferase,alanine aminotransferase,HA,CIV,PIIIP,and LN in serum.Echinacoside could also reduce the hydroxyproline content in liver,alleviate hepatic injury,and inhibit collagen deposition.Conclusion Echinacoside possesses antihepatic fibrosis effect.YANG Feng-rui WEN Du-su FANG Bu-wu LOU Jian-shi MENG Lin 2013Chinese Herbal Medicines2013,5,3:10
5Alleviation by abscisic acid of Al toxicity in rice bean is not associated with citrate efflux but depends on ABI5-mediated signal transduction pathways显示文摘Under conditions of aluminum(Al) toxicity,which severely inhibits root growth in acidic soils, plants rapidly alter their gene expression to optimize physiological fitness for survival. Abscisic acid(ABA) has been suggested as a mediator between Al stress and gene expression, but the underlying mechanisms remain largely unknown. Here,we investigated ABA-mediated Al-stress responses, using integrated physiological and molecular biology approaches.We demonstrate that Al stress caused ABA accumulation in the root apex of rice bean(Vigna umbellata [Thunb.] Ohwi &Ohashi), which positively regulated Al tolerance. However,this was not associated with known Al-tolerance mechanisms. Transcriptomic analysis revealed that nearly one-third of the responsive genes were shared between the Al-stress and ABA treatments. We further identified a transcription factor, ABI5, as being positively involved in Al tolerance. Arabidopsis abi5 mutants displayed increased sensitivity to Al, which was not related to the regulation of AtALMT1 and AtMATE expression. Functional categorization of ABI5-mediated genes revealed the importance of cell wall modification and osmoregulation in Al tolerance, a finding supported by osmotic stress treatment on Al tolerance. 6Reducing the oxidative stress mediates the cardioprotection of bicyclol against ischemia-reperfusion injury in rats显示文摘Objective:To investigate the beneficial effect of bicyclol on rat hearts subjected to ischemia-reperfusion(IR) injuries and its possible mechanism.Methods:Male Sprague-Dawley rats were intragastrically administered with bicyclol(25,50 or 100 mg/(kg·d)) for 3 d.Myocardial IR was produced by occlusion of the coronary artery for 1 h and reperfusion for 3 h.Left ventricular hemodynamics was continuously monitored.At the end of reperfusion,myocardial infarct was measured by 2,3,5-triphenyltetrazolium chloride(TTC) staining,and serum lactate dehydrogenase(LDH) level and myocardial superoxide dismutase(SOD) activity were determined by spectrophotometry.Isolated ventricular myocytes from adult rats were exposed to 60 min anoxia and 30 min reoxygenation to simulate IR injuries.After reperfusion,cell viability was determined with trypan blue;reactive oxygen species(ROS) and mitochondrial membrane potential of the cardiomyocytes were measured with the fluorescent probe.The mitochondrial permeability transition pore(mPTP) opening induced by Ca2+(200 μmol/L) was measured with the absorbance at 520 nm in the isolated myocardial mitochondria.Results:Low dose of bicyclol(25 mg/(kg·d)) had no significant improving effect on all cardiac parameters,whereas pretreatment with high bicyclol markedly reduced the myocardial infarct and improved the left ventricular contractility in the myocardium exposed to IR(P<0.05).Medium dose of bicyclol(50 mg/(kg·d)) markedly improved the myocardial contractility,left ventricular myocyte viability,and SOD activity,as well decreased infarct size,serum LDH level,ROS production,and mitochondrial membrane potential in rat myocardium exposed to IR.The reduction of ventricular myocyte viability in IR group was inhibited by pretreatment with 50 and 100 mg/(kg·d) bicyclol(P<0.05 vs.IR),but not by 25 mg/(kg·d) bicyclol.The opening of mPTP evoked by Ca2+ was significantly inhibited by medium bicyclol.Conclusions:Bicyclol exerts cardioprotection against IR injury,at least,via reducing oxidative stress and its subsequent mPTP opening.Jie CUI Zhi LI Ling-bo QIAN Qin GAO Jue WANG Meng XUE Xiao-e LOU Iain C.BRUCE Qiang XIA Hui-ping WANG 2013Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2013,14,6:8
7The role of non-coding RNAs in drug resistance of oral squamous cell carcinoma and therapeutic potential显示文摘Oral squamous cell carcinoma(OSCC),the eighth most prevalent cancer in the world,arises from the interaction of multiple factors including tobacco,alcohol consumption,and betel quid.Chemotherapeutic agents such as cisplatin,5-fluorouracil,and paclitaxel have now become the first-line options for OSCC patients.Nevertheless,most OSCC patients eventually acquire drug resistance,leading to poor prognosis.With the discovery and identification of non-coding RNAs(ncRNAs),the functions of dysregulated ncRNAs in OSCC development and drug resistance are gradually being widely recognized.The mechanisms of drug resistance of OSCC are intricate and involve drug efflux,epithelial-mesenchymal transition,DNA damage repair,and autophagy.At present,strategies to explore the reversal of drug resistance of OSCC need to be urgently developed.Nano-delivery and self-cellular drug delivery platforms are considered as effective strategies to overcome drug resistance due to their tumor targeting,controlled release,and consistent pharmacokinetic profiles.In particular,the combined application of new technologies(including CRISPR systems)opened up new horizons for the treatment of drug resistance of OSCC.Hence,this review explored emerging regulatory functions of ncRNAs in drug resistance of OSCC,elucidated multiple ncRNA-meditated mechanisms of drug resistance of OSCC,and discussed the potential value of drug delivery platforms using nanoparticles and self-cells as carriers in drug resistance of OSCC.Xiang Meng Qiu-Yue Lou Wen-Ying Yang Yue-Rong Wang Ran Chen Lu Wang Tao Xu Lei Zhang 2021Cancer Communications2021,41,10:8

9Activation of Toll-like receptors signaling in non-small cell lung cancer cell line induced by tumor-associated macrophages显示文摘Background:Inflammation is often linked with the progress and poor outcome of lung cancer.The understanding of the relationship between tumor-associated macrophages(TAMs) and lung cancer cells involves in the underlying mechanism of inflammatory cytokine production.Toll-like receptors(TLRs) are engaged in promoting the production of pro-inflammatory cytokines and play an important role in tumor immunology.Methods:To investigate the mechanisms by which TAMs influence the production of pro-inflammatory cytokines in lung cancer cells,we established an in vitro coculture system using TAMs and human nonsmall cell lung cancer(NSCLC) cell line SPC-A1.Levels of interleukin(IL)-1β,IL-6 and IL-8 in SPC-A1 were evaluated by RT-PCR and cytometric bead array assay after being cocultured with TAMs.Expression changes of TLRs and TLRs signaling pathway proteins in SPC-A1 were further confirmed by RT-PCR and western blot.The level changes of IL-1β,IL-6 and IL-8 in SPC-A1 were also detected after the stimulation of TLRs agonists.Results:We found that the phenotype markers of TAMs were highly expressed after stimulating human monocyte cell line THP-1 by phorbol-12-myristate-13-acetate(PMA).Higher mRNA and supernate secretion levels of IL-1β,IL-6 and IL-8 were detected in SPC-A1 after being cocultured with TAMs.We also found that TLR1,TLR6 and TLR7 were up-regulated in SPC-A1 in the coculture system with TAMs.Meanwhile,TLRs signaling pathway proteins were also significantly activated.Moreover,pre-treatment with agonist ligands for TLR1,TLR6 and TLR7 could dramatically promote inductions of IL-1β,IL-6 and IL-8.Conclusions:These findings demonstrated that TAMs may enhance IL-1β,IL-6 and IL-8 expressions via TLRs signaling pathway.We conclude that TAMs contribute to maintain the inflammation microenvironment and ultimately promote the development and progression of lung cancer.Xing Ke Meng Wu Jianfang Lou Shuping Zhang Peijun Huang Ruihong Sun Lei Huang Erfu Xie Fang Wang Bing Gu 2015Chinese Journal of Cancer Research2015,27,2:6
10Preoperative carcinoembryonic antibody is predictive of distant metastasis in pathologically T1 colorectal cancer after radical surgery显示文摘AIM:To identify the predictors of distant metastasis in pathologically T1(pT1)colorectal cancer(CRC)after radical resection. METHODS:Variables including age,gender,preoperative carcinoembryonic antibody(CEA)level,tumor location,tumor size,lymph node status,and histological grade were recorded.Patients with and without metastasis were compared with regard to age,gender,CEA level and pathologic tumor characteristics using the independent t test orχ 2 test,as appropriate.Risk factors were determined by logistic regression analysis. RESULTS:Metastasis occurred in 6(3.8%)of the 159 patients during a median follow-up of 67.0(46.5%) mo.The rates of distant metastasis in patients with pT1 cancer of the colon and rectum were 6.7%and 2.9%, respectively(P<0.001).The rates of distant metastasis between male and female patients with T1 CRC were 6.25%and 1.27%,respectively(P<0.001).The most frequent site of distant metastasis was the liver. Age(P=0.522),gender(P=0.980),tumor location(P =0.330),tumor size(P=0.786),histological grade(P =0.509),and high serum CEA level(P=0.262)were not prognostic factors for lymph node metastasis.Univariate analysis revealed that age(P=0.231),gender(P =0.137),tumor location(P=0.386),and tumor size (P=0.514)were not risk factors for distant metastasis after radical resection for T1 colorectal cancer.Postoperative metastasis was only significantly correlated with high preoperative serum CEA level(P=0.001).Using multivariate logistic regression analysis,high preoperative serum CEA level(P=0.004;odds ratio 15.341; 95%CI 2.371-99.275)was an independent predictor for postoperative distant metastasis. 11Identifying changes in the wheat kernel proteome under heat stress using iTRAQ显示文摘Wheat(Triticum aestivum L.) is one of the three major global food crops. Hightemperature stress can affect its yield and quality. Studies of the effect of hightemperature stress on wheat kernel development are important because they can reveal the stability of wheat quality and lead to the genetic improvement of wheat quality traits. In this study, the isobaric tags for relative and absolute quantitation(iTRAQ)method was adopted to analyze changes in the protein expression profile of wheat cultivars under high temperature stress. The protein content of wheat grain increased under heat stress, while the SDS-sedimentation value and starch content decreased.Grain filling was deficient under high temperature stress, which reduced thousandkernel weight but did not affect wheat kernel length. The 207 differentially expressed proteins identified in Gaocheng 8901 under heat stress were associated with energy metabolism, growth and development, and stress response. Gene Ontology enrichment analysis showed that the annotated proteins that were differentially expressed in Gaocheng 8901 under heat stress were involved mainly in stimulus response, abiotic stress response, stress response, and plasma membrane. A set of 78 differentially expressed proteins were assigned to 83 KEGG signaling/metabolic pathways. KEGG pathway enrichment analysis showed that this set of proteins was significantly enriched in members of 51 pathways, and the proteins participated mainly in protein synthesis in the endoplasmic reticulum, starch and sucrose metabolism, and reaction on ribosomes. Five differentially expressed proteins were involved in protein–protein interaction networks that may greatly influence the yield and quality of wheat grain. 12FLT3 and NPM1 mutations in Chinese patients with acute myeloid leukemia and normal cytogenetics显示文摘Mutations of fms-like tyrosine kinase 3(FLT3) and nucleophosmin(NPM1) exon 12 genes are the most common abnormalities in adult acute myeloid leukemia(AML) with normal cytogenetics.To assess the prognostic impact of the two gene mutations in Chinese AML patients,we used multiplex polymerase chain reaction(PCR) and capillary electrophoresis to screen 76 AML patients with normal cytogenetics for mutations in FLT3 internal tandem duplication(FLT3/ITD) and exon 12 of the NPM1 gene.FLT3/ITD mutation was detected in 15(19.7%) of 76 subjects,and NPM1 mutation in 20(26.3%) subjects.Seven(9.2%) cases were positive for both FLT3/ITD and NPM1 mutations.Significantly more FLT3/ITD aberration was detected in subjects with French-American-British(FAB) M1(42.8%).NPM1 mutation was frequently detected in subjects with M5(47.1%) and infrequently in subjects with M2(11.1%).FLT3 and NPM1 mutations were significantly associated with a higher white blood cell count in peripheral blood and a lower CD34 antigen expression,but not age,sex,or platelet count.Statistical analysis revealed that the FLT3/ITD-positive group had a lower complete remission(CR) rate(53.3% vs.83.6%).Survival analysis showed that the FLT3/ITD-positive/NPM1 mutation-negative group had worse overall survival(OS) and relapse-free survival(RFS).The FLT3/ITD-positive/NPM1 mutation-positive group showed a trend towards favorable survival compared with the FLT3/ITD-positive/NPM1 mutation-negative group(P=0.069).Our results indicate that the FLT3/ITD mutation might be a prognostic factor for an unfavorable outcome in Chinese AML subjects with normal cytogenetics,while NPM1 mutation may be a favorable prognostic factor for OS and RFS in the presence of FLT3/ITD.Lei WANG Wei-lai XU Hai-tao MENG Wen-bin QIAN Wen-yuan MAI Hong-yan TONG Li-ping MAO Yin TONG Jie-jing QIAN Yin-jun LOU Zhi-mei CHEN Yun-gui WANG Jie JIN 2010Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,10:4
13Wideband mid-infrared thermal emitter based on stacked nanocavity metasurfaces显示文摘Efficient thermal radiation in the mid-infrared(M-IR)region is of supreme importance for many applications including thermal imaging and sensing,thermal infrared light sources,infrared spectroscopy,emissivity coatings,and camouflage.The ability to control light makes metasurfaces an attractive platform for infrared applications.Recently,different metamaterials have been proposed to achieve high thermal radiation.To date,broadening the radiation bandwidth of a metasurface emitter(meta-emitter)has become a key goal to enable extensive applications.We experimentally demonstrate a broadband M-IR thermal emitter using stacked nanocavity metasurface consisting of two pairs of circular-shaped dielectric(Si;N;)–metal(Au)stacks.A high thermal radiation can be obtained by engineering the geometry of nanocavity metasurfaces.Such a meta-emitter provides wideband and broad angular absorptance of both p-and s-polarized light,offering a wideband thermal radiation with an average emissivity of more than 80%in the M-IR atmospheric window of 8–14μm.The experimental illustration together with the theoretical framework establishes a basis for designing broadband thermal emitters,which,as anticipated,will initiate a promising avenue to M-IR sources.Tun Cao Meng Lian Kuan Liu Xianchao Lou Yaoming Guo Dongming Guo 2022International Journal of Extreme Manufacturing2022,4,1:3

16Room-temperature chlorine gas sensor based on CdSnO_(3) synthesized by hydrothermal process显示文摘The perovskite-structure CdSnO_(3) was obtained by calcinating CdSnO_(3)·3H_(2)O precursor at 550℃,which was synthesized by hydrothermal process at 170℃for 16 h.The phase and microstructure of the obtained CdSnO_(3) powders were characterized by X-ray diffraction(XRD),scanning electron microscopy(SEM)and transmission electron microscopy(TEM).The CdSnO_(3) powders exhibit uniformly cubic structure with side length of about 100 nm.The effects of working temperature and concentration of detected gas on the gas response were studied.The selectivity of chlorine gas against other gases and response-recovery time of the sensor were also investigated.The results reveal that the CdSnO_(3) gas sensor has enhanced sensing properties to 1-10 ppm chlorine gas at room temperature;the value of gas response can reach 1338.9 to 5 ppm chlorine gas.Moreover,the sensor shows good selectivity and quick response behavior(23 s)to chlorine gas,indicating its application in detecting chlorine gas at room temperature in the future.Xiaohua ZHAO Zhenzhen LI Xiangdong LOU Meng LI Ning ZHANG 2013Journal of Advanced Ceramics2013,2,1:3
17Brain-wide Mapping of Mono-synaptic Afferents to Different Cell Types in the Laterodorsal Tegmentum显示文摘The laterodorsal tegmentum(LDT) is a brain structure involved in distinct behaviors including arousal,reward, and innate fear. How environmental stimuli and top-down control from high-order sensory and limbic cortical areas converge and coordinate in this region to modulate diverse behavioral outputs remains unclear.Using a modified rabies virus, we applied monosynaptic retrograde tracing to the whole brain to examine the LDT cell type specific upstream nuclei. The LDT received very strong midbrain and hindbrain afferents and moderate cortical and hypothalamic innervation but weak connections to the thalamus. The main projection neurons from cortical areas were restricted to the limbic lobe, including the ventral orbital cortex(VO), prelimbic, and cingulate cortices. Although different cell populations received qualitatively similar inputs, primarily via afferents from the periaqueductal gray area, superior colliculus, and the LDT itself, parvalbumin-positive(PV?) GABAergic cells received preferential projections from local LDT neurons.With regard to the different subtypes of GABAergic cells, aconsiderable number of nuclei, including those of the ventral tegmental area, central amygdaloid nucleus, and VO, made significantly greater inputs to somatostatinpositive cells than to PV?cells. 19The chromosome-scale genome of Phoebe bournei reveals contrasting fates of terpene synthase (TPS)-a and TPS-b subfamilies显示文摘Terpenoids,including aromatic volatile monoterpenoids and sesquiterpenoids,function in defense against pathogens and herbivores.Phoebe trees are remarkable for their scented wood and decay resistance.Unlike other Lauraceae species investigated to date,Phoebe species predominantly accumulate sesquiterpenoids instead of monoterpenoids.Limited genomic data restrict the elucidation of terpenoid variation and functions.Here,we present a chromosome-scale genome assembly of a Lauraceae tree,Phoebe bournei,and identify 72 full-length terpene synthase(TPS)genes.Genome-level comparison shows pervasive lineage-specific duplication and contraction of TPS subfamilies,which have contributed to the extreme terpenoid variation within Lauraceae species.Although the TPS-a and TPS-b subfamilies were both expanded via tandem duplication in P.bournei,more TPS-a copies were retained and constitutively expressed,whereas more TPS-b copies were lost.The TPS-a genes on chromosome 8 functionally diverged to synthesize eight highly accumulated sesquiterpenes in P.bournei.The essential oil of P.bournei and its main component,b-caryophyllene,exhibited antifungal activities against the three most widespread canker pathogens of trees.The TPS-a and TPS-b subfamilies have experienced contrasting fates over the evolution of P.bournei.The abundant sesquiterpenoids produced by TPS-a proteins contribute to the excellent pathogen resistance of P.bournei trees.Overall,this study sheds light on the evolution and adaptation of terpenoids in Lauraceae and provides valuable resources for boosting plant immunity against pathogens in various trees and crops.Xiao Han Junhong Zhang Shuang Han Sun Li Chong Guanliang Meng Minyan Song Yang Wang Shengcai Zhou Chengcheng Liu Luhuan Lou Xiongzhen Lou Longjun Cheng Erpei Lin Huahong Huang Qi Yang Zaikang Tong 2022Plant Communications2022,3,6:3
