| 2 | Nickel accumulation and its effects on the survival rate of Spodoptera litura Fabricius under continuous nickel stress显示文摘The artificial diets mixed with various concentrations of nickel were offered to the larvae of the phytophagous insect Spodoptera litura Fabricius for 3 generations. Nickel accumulations in the 6th instar larvae, pupae and newly emerged adults of the corresponding generations of S. litura were investigated by Inductively Coupled Plasma-Atomic Emission Spectrometer (ICP-AES), and the effects of nickel accumulations on the survival rate of S. litura were also evaluated by individual rearing. The results showed that nickel accumulated in the 6th instar larvae, pupae and adults of S. litura, and the accu- mulated nickel in all the tested developmental stages within a generation increased with the increase of the nickel doses in the treated diets and showed significant dose-dependent relationship with the nickel doses in the artificial diets. The results also indicated that the nickel accumulations in the 6th instar larvae, pupae, and newly emerged adults from the 3rd generation were higher than those from the 2nd generation, which were also higher than those from the 1st generation. Nickel concentrations in pupae and adults were significantly lower than those in larvae, which indicated that the excessive nickel might be excreted during metamorphosis. | 3 | Secretory/releasing proteome-based identification of plasma biomarkers in HBV-associated hepatocellular carcinoma显示文摘For successful therapy, hepatocellular carcinoma (HCC) must be detected at an early stage. Herein, we used a proteomic approach to analyze the secretory/releasing proteome of HCC tissues to identify plasma biomarkers. Serum-free conditioned media (CM) were collected from primary cultures of cancerous tissues and surrounding noncancerous tissues. Proteomic analysis of the CM proteins permitted the identification of 1365 proteins. The enriched molecular functions and biological processes of the CM proteins, such as hydrolase activity and catabolic processes, were consistent with the liver being the most important metabolic organ. Moreover, 19% of the proteins were characterized as extracellular or membrane-bound. For validation, secretory proteins involved in transforming growth factor-β signaling pathways were validated in plasma samples. Alphafetoprotein (AFP), metalloproteinase (MMP)1, osteopontin (OPN), and pregnancy-specific beta-1-glycoprotein (PSG)9 were significantly increased in HCC patients. The overall performance of MMP1 and OPN in the diagnosis of HCC remained greater than that of AFP. In addition, this study represents the first report of MMP1 as a biomarker with a higher sensitivity and specificity than AFP. Thus, this study provides a valuable resource of the HCC secretome with the potential to investigate serological biomarkers. | 5 | Effect of Jianpiyiqi granule on ocular myasthenia gravis in children prepared with a formula from Traditional Chinese Medicine显示文摘OBJECTIVE: To investigate the effect of Jianpiyiqi granule, prepared according to a Traditional Chinese Medicine(TCM) formula, on the ocular type of myasthenia gravis(MG) caused by a defect in synaptic transmission at the neuromuscular junction.METHODS: A total of 155 children with ocular MG were recruited from January 2008 to January 2015.All individuals received ineffective glucocorticoid treatment prior to admission, and were given Jianpiyiqi granules(two doses per day) for 12 months.Plasma levels of acetylcholine receptor autoantibodies(ACh R-Ab), cytokines, immune parameters and clinical score were analyzed.RESULTS: After intervention with Jianpiyiqi granule for 12 months, the levels of cytokines [including interleukin(IL)-6, IL-12, IL-17], immune parameters[including immunoglobulin G(Ig G), Ig M, free triiodothyronine, free thyroxine] and ACh R-Ab were sig-nificantly decreased(P = 0.042, P = 0.049, P = 0.011,P = 0.02, P = 0.04, P = 0.03, P = 0.01; F = 21.60, P =0.000). In contrast, IL-4 levels were significantly increased(P = 0.049). The absolute clinical score after treatment declined significantly compared with before treatment(F = 33.24, P = 0.000). | 9 | Ocean current observation and spectrum analysis in central Chukchi Sea during the summer of 2008显示文摘During the summer of 2008,the third CHINARE Arctic Expedition was carried out on board of Xuelong Icebreaker in the central Chukchi Sea.A submersible mooring system was deployed and recovered at Station CN-01(71 40.024'N,167 58.910'W) with 33 days of the current profile records,and continuous observation of temperature and salinity data were collected.This mooring station locates in the blank of similar observation area and it is the first time for our Chinese to finish this kind of long-term mooring work in this area.This mooring system finished integrated hydrological observations with long-term continuous record of the whole profile velocity for the first time.Based on time series analysis of temperature,salinity,velocity and flow direction,we get the following main results.(1) During the observation period,the mean surface current velocity is 70.2 cm/s eastward,and velocity reaches its maximum in average at 3 m level with magnitude 90.0 cm/s,direction 206.(2) In 9-30 m layers,the semidiurnal period variation is the most obvious,the flow direction is quite stable,and the flow is synchronous and consistent vertically.(3) Besides the semidiurnal period variation,the main variation in the upper layer is in 11-d period,with variations in period 5.5,5.5,and 3.7 d,which reflect the influences of sea surface wind change and maintenance.(4) Near the bottom,the temperature change is correlated and synchronized with the conductivity. | CHEN Hongxia WANG Huiwu SHU Qi WANG Daolong LIU Na | 2013 | Acta Oceanologica Sinica2013，32，3： | 2 |

| 11 | Long-Term Effects of Docosahexaenoic Acid-Bound Phospholipids and the Combination of Docosahexaenoic Acid-Bound Triglyceride and Egg Yolk Phospholipid on Lipid Metabolism in Mice显示文摘The bioavailability of docosahexaenoic acid(DHA) and eicosapentaenoic acid(EPA) depends on their chemical forms. This study investigated the long-term effects of DHA-bound triglyceride(TG-DHA), DHA-bound phospholipid(PL-DHA), and the combination of TG-DHA and egg yolk phospholipid(Egg-PL) on lipid metabolism in mice fed with a high-fat diet(fat levels of 22.5%). Male C57BL/6J mice were fed with different formulations containing 0.5% DHA, including TG-DHA, PL-DHA, and the combination of TG-DHA and Egg-PL, for 6 weeks. Serum, hepatic, and cerebral lipid concentrations and the fatty acid compositions of the liver and brain were determined. The concentrations of serum total triglyceride(TG), total cholesterol(TC), high-density lipoprotein cholesterol(HDL-c), and hepatic TG in the PL-DHA group and the combination group were significantly lower than those in the high-fat(HF) group(P < 0.05). Atherogenic index(AI) of the PL-DHA group was significantly lower than that of the combination group(P < 0.05). Hepatic TC level in the combination group was significantly lower than that in the HF group(P < 0.05), but no significant difference was observed between the combination group and the PL-DHA group. Both the PL-DHA and the combination groups showed significantly increased DHA levels in the liver compared with the HF group(P < 0.05). However, there were no obvious increases in the cerebral DHA levels in all DHA diet groups. These results suggest that PL-DHA was superior to the combination of TG-DHA and Egg-PL in decreasing the AI. | 17 | A genetic variant in pseudogene E2F3P1 contributes to prognosis of hepatocellular carcinoma显示文摘Certain pseudogenes may regulate their protein-coding cousins by competing for miRNAs and play an active biological role in cancer.However,few studies have focused on the association of genetic variations in pseudogenes with cancer prognosis.We selected six potentially functional single nucleotide polymorphisms(SNPs) in cancerrelated pseudogenes,and performed a case-only study to assess the association between those SNPs and the prognosis of hepatocellular carcinoma(HCC) in 331 HBV-positive HCC patients without surgical treatment.Log-rank test and Cox proportional hazard models were used for survival analysis.We found that the A allele of rs9909601 in E2F3P1was significantly associated with a better prognosis compared with the G allele[adjusted hazard ratio(HR) = 0.69,95%confidence interval(CI) = 0.56-0.86,P = 0.001].Additionally,this protective effect was more predominant for patients without chemotherapy and transcatheter hepatic arterial chemoembolization(TACE) treatment.Interestingly,we also detected a statistically significant multiplicative interaction between genotypes of rs9909601 and chemotherapy or TACE status on HCC survival(P for multiplicative interaction < 0.001).These findings indicate that rs9909601 in the pseudogene E2F3P1 may be a genetic marker for HCC prognosis in Chinese. | Yun Pan Chongqi Sun Mingde Huang Yao Liu Fuzhen Qi Li Liu Juan Wen Jibin Liu Kaipeng Xie Hongxia Ma Zhibin Hu Hongbing Shen | 2014 | The Journal of Biomedical Research2014，28，3： | 1 |
