2Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 4Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. 5Optimizing the CRISPR/Cas9 system for genome editing in grape by using grape promoters显示文摘The efficacy of the CRISPR/Cas9 system in grapevine(Vitis vinifera L.)has been documented,but the optimization of this system,as well as CRISPR/Cas9-mediated multiplex genome editing,has not been explored in this species.Herein,we identified four VvU3 and VvU6 promoters and two ubiquitin(UBQ)promoters in grapevine and demonstrated that the use of the identified VvU3/U6 and UBQ2 promoters could significantly increase the editing efficiency in grape by improving the expression of sgRNA and Cas9,respectively.Furthermore,we conducted multiplex genome editing using the optimized CRISPR/Cas9 vector that contained the conventional multiple sgRNA expression cassettes or the polycistronic tRNA-sgRNA cassette(PTG)by targeting the sugar-related tonoplastic monosaccharide transporter(TMT)family members TMT1 and TMT2,and the overall editing efficiencies were higher than 10%.The simultaneous editing of TMT1 and TMT2 resulted in reduced sugar levels,which indicated the role of these two genes in sugar accumulation in grapes.Moreover,the activities of the VvU3,VvU6,and UBQ2 promoters in tobacco genome editing were demonstrated by editing the phytoene desaturase(PDS)gene in Nicotiana benthamiana leaves.Our study provides materials for the optimization of the CRISPR/Cas9 system.To our knowledge,our simultaneous editing of the grape TMT family genes TMT1 and TMT2 constitutes the first example of multiplex genome editing in grape.The multiplex editing systems described in this manuscript expand the toolbox of grape genome editing,which would facilitate basic research and molecular breeding in grapevine.Chong Ren Yanfei Liu Yuchen Guo Wei Duan Peige Fan Shaohua Li Zhenchang Liang 2021Horticulture Research2021,8,1:6
6Active optical clock based on four-level quantum system显示文摘Active optical clock,a new conception of atomic clock,has been proposed recently.In this work,we propose a scheme of active optical clock based on four-level quantum system.The final accuracy and stability of two-level quantum system are limited by second-order Doppler shift of thermal atomic beam.To three-level quantum system,they are mainly limited by light shift of pumping laser field.These limitations can be avoided effectively by applying the scheme proposed here.Rubidium atom four-level quantum system,as a typical example,is discussed.The population inversion between 6S 1/2 and 5P 3/2 states can be built up at a time scale of 10-6 s.With the mechanism of active optical clock,in which the cavity mode linewidth is much wider than that of the laser gain profile,it can output a laser with quantum-limited linewidth narrower than 1 Hz in theory.An experimental configuration is designed to realize this active optical clock.ZHANG TongGang WANG YanFei ZANG XiaoRun ZHUANG Wei CHEN JingBiao 2013Chinese Science Bulletin2013,58,18:6

8Homologous and heterologous interactions between catalytic and regulatory subunits of Escherichia coli acetohydroxyacid synthaseⅠand Ⅲ显示文摘Homologous and heterologous interactions between acetohydroxyacid synthase (AHAS) I and Ⅲ from E. coli have been studied by surface plasmon resonance (SPR). The catalytic and regulatory subunits association for AHAS I (KD = 1.13 × 10-7 M) was stronger than that for AHAS Ⅲ (KD = 5.29 × 10-7 M). A strong heterologous association between regulatory and catalytic subunits and heterologous activation of catalytic subunits were observed. SPR results combined with enzyme kinetics indicate that the reconstituted heterologous enzymes had similar kinetic properties as homologous enzymes, implying that the regulatory subunit of AHAS I could be replaced by the regulatory subunit of AHAS Ⅲ and vice versa. 9Construction of Rat Model of Hepatic Fibrosis with Blood Stasis Syndrome Integrated with Traditional Chinese Medicine(TCM)Syndrome and Western Medicine Disease显示文摘[Objective]According to the theory of blood stasis in traditional Chinese medicine( TCM),the animal modeling method of hepatic fibrosis integrated with pathology and symptoms was explored and improved,to construct a new type of rat model of hepatic fibrosis with blood stasis syndrome integrated with traditional Chinese and western medicine. [Method]The hepatic fibrosis model of blood stasis with blood stasis syndrome was constructed by jointing multi factors,including intragastric administration of ethanol,high fat and low protein feeding,joint injection of dimethylnitrosamine( DMN),bovine serum albumin( BSA) and norepinephrine( NE). The modeling method was further compared with traditional CC14 single-factor modeling method from the aspects of mortality,blood stasis syndrome of TCM,syndrome grading,general morphology of liver pathology,liver function changes,as well as expression levels of three kinds of collagen( type I and type III collagen,α-SMA) determined by immunohistochemical staining method,and four items of rat hepatic fibrosis( HA,P3 NP,LN,CⅣ content) determined by radio enzyme immunoassay. [Result]In blood stasis group,(1) the mortality of rats was 20%;(2) model rats appeared typical blood stasis syndromes of TCM such as ecchymosis,dark purple tongue,loose stool,and blood stasis syndrome grading was high;(3) fibrosis changes of liver such as dark white surface,dense gray nodules and brittle texture were observed in general morphological examination;(4) serological liver function tests found that ALT and AST of model rats,as well as TBIL,DBIL and IBIL contents increased significantly;(5) immunohistochemical staining demonstrated that the expression levels of three kinds of collagen(type I and type III collagen,α-SMA) increased significantly;(6) four items of rat serum hepatic fibrosis,HA,P3 NP,LN,CⅣ content,increased significantly;(7) the above results in blood stasis syndrome model group(except morality and liver function) were higher than those in CC14 modeling group,and the difference was statistically significant( P < 0. 05). [Conclusion]The new improved modeling method effectively reduces high mortality in traditional CC14 modeling method.In addition to low mortality,the model animal has dual characteristics of disease in western medicine and syndrome in TCM. 10Effects of Aqueous Extract of Plumbago zeylanica L. in the Reversal of DMN-induced Hepatic Fibrosis in Rat显示文摘[Objectives] This study was conducted to evaluate the action of the aqueous extract of a medicinal plant Plumbago zeylanica L. in the reversal of dimethylnitrosamine( DMN)-induced hepatic fibrosis in rats,and to provide a scientific basis for the utilization of P. zeylanica in treatment of hepatic fibrosis. [Methods] Hepatic fibrosis in rats was induced by intraperitoneal injection of DMN. The rats were then given the aqueous extract of P. zeylanica at high,medium or low concentration by gavage for five weeks. Serum level of alanine aminotransferase( ALT) was determined by lactate dehydrogenase( LDH)-release assay,and serum level of aspartate transaminase( AST) was measured by UV-malate dehydrogenase( MDH) assay. Serum levels of total bilirubin( TBIL),direct bilirubin( DBIL)and indirect bilirubin( IBIL) were measured by vanadate oxidation assay. Four indices of hepatic fibrosis( hyaluronic acid,laminin,procollagen type Ⅲ and collagen type IV) were determined by radioimmunoassay( RIA) assay. Morphological damage of liver tissue was observed by hematoxylin-eosin staining( H&E staining). Immunohistochemical staining was performed to determine the location and area of deposited collagen type I,collagen type Ⅲ and α-smooth muscle actin( α-SMA) in liver tissue. [Results]Compared with the negative control( rats with diseased liver and untreated with P. zeylanica aqueous extract),the serum levels of ALT,AST,TBIL,DBIL and IBIL were significantly decreased by P. zeylanica aqueous extract; the levels of the four serum indices of hepatic fibrosis were also obviously reduced. H&E staining showed that hepatic fibrosis in rats was obviously inhibited or even reversed by P. zeylanica aqueous extract. Immunohistochemical staining proved that the aqueous extract of P. zeylanica significantly reduced the area and coloration of collagen type I,collagen type Ⅲ and α-SMA in rat liver. 12Multi-scale three-dimensional variational data assimilation for high-resolution aerosol observations: Methodology and application显示文摘With an increasing number of air quality monitoring stations installed around the Chinese mainland,high-resolution aerosol observations become available,allowing improvements in air pollution monitoring and aerosol forecasting.However,the multi scales(especially small-scale)information included in high-resolution aerosol observations could not be effectively utilized by the traditional three-dimensional variational method(3DVAR).This study attempted to extend the traditional 3DVAR to a multi-scale 3DVAR with two iteration steps,two-scale-3DVAR(TS-3DVAR),to improve the effectiveness of assimilating high-resolution observations.In TS-3DVAR,the large-scale and small-scale components of observation information were decomposed from the original high-resolution observations using a Gaussian smoothing method and then assimilated using the corresponding large-scale or small-scale background error covariances which were derived from the partitioned background error samples.The data assimilation(DA)analysis field generated by TS-3DVAR is more accurate than 3DVAR in reproducing the field’s multi-scale characteristics,which could thus be used as the initial chemical field of the air quality model to improve aerosol forecasting.Particulate matter with an aerodynamic diameter of less than 2.5μm(PM_(2.5))and 10.0μm(PM_(10)) from the surface air quality monitoring stations from November 01 to November 30,2018 at 00:00 were assimilated daily to verify the effects of TS-3DVAR and 3DVAR on the aerosol analysis and forecast accuracy.The results showed that TS-3DVAR better constrained both large-scale and small-scale,especially the spatial wavelengths in a range of 54-216 km and those above 351 km.The average power spectra of the TS-3DVAR assimilation increment in the two wavelength ranges were 71.70%and 35.33%higher than those of 3DVAR.As a result,the TS-3DVAR was more effective than 3DVAR in improving the accuracy of the initial chemical field,and thereby the forecasting capability for PM_(2.5).In the initial chemical field,the 30-day average correlation coefficient(Corr)of PM_(2.5) of TS-3DVAR was 0.052(6.12%)higher than that of 3DVAR,and the root mean square error(RMSE)of TS-3DVAR was 3.446μg m^(−3)(16.4%)lower than that of 3DVAR.For the forecasting capability for PM_(2.5) mass concentration,the 30-day average Corr of TS-3DVAR during the 0-24 hour forecast period was 0.025(5.08%)higher than that of 3DVAR,and the average RMSE was 2.027μg m^(−3)(4.85%)lower.The positive effect of TS-3DVAR on the improvement of forecasting capability can last for more than 24 h.Zengliang ZANG Yanfei LIANG Wei YOU Yi LI Xiaobin PAN Zhijin LI 2022Science China Earth Sciences2022,65,10:2
13Recent progress in small molecule fluorescent probes for nitroreductase显示文摘Nitroreductase(NTR) is a member of flavin-containing enzymes that exists widely in bacteria. Hypoxia,which is a characteristic of locally advanced solid tumors, resulting from an imbalance between oxygen consumption and supply, can result in NTR overexpression. Using either nicotinamide adenine dinucleotide(NADH) or nicotinamide adenine dinucleotide phosphate(NADPH) as a source of reducing equivalents, NTR can catalyze the reduction of nitroaromatic compounds to the corresponding amines.Based on this reduction mechanism, NTR can be applied not only in the bioremediation and degradation of organic nitrogen compounds, but also in the development of NTR-targeted fluorescent probes to detect the hypoxic status of cancer cells. 16Highly stable lithium anode enabled by self-assembled monolayer of dihexadecanoalkyl phosphate显示文摘Li has been considered as the ultimate anode material for high energy density secondary Li batteries.However,its practical application has been limited due to its low Coulombic efficiency(CE)and the formation of lithium dendrites.Recently,we have developed a microspherical Li-carbon nanotube(Li-CNT)composite material passivated with octadecylphosphonic acid(OPA)self-assembled monolayer(SAM)exhibiting suppressed lithium dendrite formation and improved environmental/electrochemical stability.In this work,we demonstrated the significantly enhanced passivation effects of a SAM using dihexadecanoalkyl phosphate(DHP),a molecule that is comprised of double hydrophobic alkyl chains and forms a denser SAM on surfaces with large curvature.As a result,the DHP SAM delivers superior environmental and electrochemical stability to the OPA passivated Li-CNT material.In specific,the DHP passivated Li-CNT composite(DHP-Li-CNT)delivers a high CE of 99.25%under a 33.3%depth of discharge(DOD)at 1 C,when it is paired with a LiFePO4 cathode.The evolution of the SAM during cycling and the effects of DOD and current density on the CE of the DHP-Li-CNT anode have also been investigated.The improved SAM passivation constitutes an important step in achieving the goal of practically applicable Li anodes.Lei Zheng Feng Guo Tuo Kang Jin Yang Ya Liu Wei Gu Yanfei Zhao Hongzhen Lin Yanbin Shen Wei Lu Liwei Chen 2020Nano Research2020,13,5:1
17The research of propagation characteristic and formation of double half-Gaussian hollow beams显示文摘A new kind of hollow beams, double half-Gaussian hollow beams,was put forward. With the help of the Collins formula, the analytical equation of propagation and transformation of the hollow laser beams in free space was deduced. The simulation shows that the intensity exhibits the three-dimensional trap distribution in the near-field, while the double half-Gaussian hollow beams turn into solid laser beams when propagating a certain distance, which shows the characteristics of self-focus. The double half-Gaussian hollow beams were obtained by means of the dual-reflecting splitting optical system. 20A three-dimensional variational data assimilation system for a size-resolved aerosol model: Implementation and application for particulate matter and gaseous pollutant forecasts across China显示文摘A three-dimensional variational(3DVAR)data assimilation(DA)system is presented here based on a size-resolved sectional aerosol model,the Model for Simulating Aerosol Interactions and Chemistry(MOSAIC)within the Weather Research and Forecasting model coupled to Chemistry(WRF-Chem)model.The use of this approach means that both gaseous pollutants such as SO2,NO2,CO,and O3 as well as particulate matter(PM2.5,PM10)observational data can be assimilated simultaneously.Two one-month parallel simulation experiments were conducted,one with the assimilation of surface hourly concentration observations of the above six pollutants released by the China National Environmental Monitoring Centre(CNEMC)and one without assimilation in order to verify the impact of assimilation on initial chemical fields and subsequent forecasts.Results show that,in the first place,use of the DA system can provide a more accurate model initial field.The root-mean-square error of PM2.5,PM10,SO2,NO2,CO,and O3 mass concentrations in analysis field fell by 29.27μg m-3(53.5%),34.5μg m-3(50.9%),30.36μg m-3(64.2%),8.91μg m-3(39.5%),0.46 mg m-3(47.4%),and 15.11μg m-3(51.0%),respectively,compared to a background field without assimilation.At the same time,mean fraction error was reduced by 42.6%,53.1%,45.2%,43.1%,69.9%,and 48.8%,respectively,while the correlation coefficient increased by 0.51,0.55,0.48,0.38,0.47,0.65,respectively.Secondly,the results of this analysis reveal variable benefits from assimilation on different pollutants.DA significantly improves PM2.5,PM10,and CO forecasts leading to positive effects that last more than 48 h.The positive effects of DA on SO2 and O3 forecasts last up to 8 h but that remains relatively poor for NO2 forecasts.Thirdly,the influence of assimilation varies in different areas.It is possible that the positive effects of DA on PM2.5 and PM10 forecasts can last more than 48 h across most regions of China.Indeed,DA significantly improves SO2 forecasts within 48 h over north China,and much longer CO assimilation benefits(48 h)are found in most regions apart from north and east China and across the Sichuan Basin.DA is able to improve O3 forecasts within 48 h across China with the exception of southwest and northwest regions and the O3 DA benefits in southern China are more evident,while from a spatial distribution perspective,NO2 DA benefits remain relatively poor.Daichun WANG Wei YOU Zengliang ZANG Xiaobin PAN Hongrang HE Yanfei LIANG 2020Science China Earth Sciences2020,63,9:1
