3Gut microbiota metabolite regulation of host defenses at mucosal surfaces:implication in precision medicine显示文摘The gut microbiota has a well-established role in the regulation of host homeostasis.Multiple factors control the composition and function of the microbiota.The westernization of diet,a shift away from nutrient-dense foods toward diets high in saturated fats,has been implicated in the rise of chronic inflammatory diseases such as inflammatory bowel disease(IBD).Diet is critical in the development and maintenance of a healthy microbiome,where dietary fiber(found in the highest amounts in fruits,vegetables,and legumes)is metabolized by the microbiome.In turn,the bacterial metabolites of dietary fiber,short chain fatty acids(SCFAs),regulate gut homeostasis.SCFAs engage G-protein coupled receptors(GPRs)and act as histone deacetylase inhibitors(HDACi)to module epithelial and immune cell functions in the intestines,where they generally promote an anti-inflammatory state.This review highlights the functions of SCFAs and their roles in the pathogenesis of IBD to provide insights into their potential therapeutic application for the treatment of IBD for the purposes of precision medicine.Anthony J.Bilotta Yingzi Cong 2019Precision Clinical Medicine2019,2,2:13
4Gut microbiota-derived metabolites in the regulation of host immune responses and immune-related inflammatory diseases显示文摘The gut microbiota has a critical role in the maintenance of immune homeostasis.Alterations in the intestinal microbiota and gut microbiota-derived metabolites have been recognized in many immune-related inflammatory disorders.These metabolites can be produced by gut microbiota from dietaiy components or by the host and can be modified by gut bacteria or synthesized de novo by gut bacteria.Gut microbiota-derived metabolites influence a plethora of immune cell responses,including T cells,B cells,dendritic cells,and macrophages.Som e of these metabolites are involved in the pathogenesis of immune-related inflammatory diseases,such as inflammatory bowel diseases,diabetes,rheumatoid arthritis,and systemic lupus erythematosus.Here,we review the role of microbiota-derived metabolites in regulating the functions of different immune cells and the pathogenesis of chronic immune-related inflammatory diseases.Wenjing Yang Yingzi Cong 2021Cellular & Molecular Immunology2021,18,4:10
5Deformation Behavior and Microstructure Evolution of Wrought Magnesium Alloys显示文摘There are many researches on the deformation behavior of wrought magnesium alloys, such as AZ31, AZ80, AZ91, and ZK60 magnesium alloys at different temperatures and strain rates, but few of them focuses on the deformation behavior of AZ41M and ZK60M alloys, especially under the twin-roll casting (TRC) state. Meanwhile, the existing researches only focus on the grain refinement law of the magnesium alloys under deformation conditions, the deformation mechanism has not been revealed yet. The hot compression behavior of AZ41M and ZK60M magnesium alloys under the temperature and strain rate ranges of 250-400 ℃ and 0.001-1 s-1 are studied by thermal simulation methods using Gleeble 1500 machine and virtual simulation using finite element analysis software. Simulation results show that sine hyperbolic law is the most suitable flow stress model for wider deformation conditions. The most reasonable selected deformation conditions of ZK60M alloy is 350 oC/0.1 s-1 for TRC and 350 oC/1 s-1 for conventional casting (CC), while AZ41M alloy is 300 oC/0.01 s-1 for TRC and 350 oC/0.1 s-1 for CC. Deformation behavior and dynamic recrystallization (DRX) mechanism of them are analyzed at the same deformation conditions. The microstructures of AZ41M and ZK60M alloys are observed at different deformed conditions by optical microscopy (OM) and electron back scatter diffraction (EBSD) and it reveals the flow behavior and deformation mechanism of them. Working harden and work soften contribute to the activation of basal, non-basal slip systems which promote DRX. 6Gα_s signaling controls intramembranous ossification during cranial bone development by regulating both Hedgehog and Wnt/β-catenin signaling显示文摘How osteoblast cells are induced is a central question for understanding skeletal formation. Abnormal osteoblast differentiation leads to a broad range of devastating craniofacial diseases. Here we have investigated intramembranous ossification during cranial bone development in mouse models of skeletal genetic diseases that exhibit craniofacial bone defects. The GNAS gene encodes Gαs that transduces GPCR signaling. GNAS activation or loss-of-function mutations in humans cause fibrous dysplasia(FD) or progressive osseous heteroplasia(POH) that shows craniofacial hyperostosis or craniosynostosis, respectively. We find here that, while Hh ligand-dependent Hh signaling is essential for endochondral ossification, it is dispensable for intramembranous ossification, where Gαsregulates Hh signaling in a ligand-independent manner. We further show that Gαscontrols intramembranous ossification by regulating both Hh and Wnt/β-catenin signaling. In addition, Gαsactivation in the developing cranial bone leads to reduced ossification but increased cartilage presence due to reduced cartilage dissolution, not cell fate switch. Small molecule inhibitors of Hh and Wnt signaling can effectively ameliorate cranial bone phenotypes in mice caused by loss or gain of Gnas function mutations, respectively. 7Unsteady Flow Characteristic of Low-Specific-Speed Centrifugal Pump under Different Flow-Rate Conditions显示文摘To investigate the unsteady flow characteristics in centrifugal pump, the flow field in a low-specific-speed centrifugal pump with complex impeller is numerically simulated under different conditions. The RNG κ-ε turbulence model and sliding mesh are adopted during the process of computation. The results show that the interaction between impeller and volute results in the unstable flow of the fluid, which causes the uneven distribution of pressure fluctuations around the circumference of volute. Besides the main frequency and its multiple frequency of pressure fluctuations in the centrifugal pump, the frequency caused by the long blades of complex impeller also plays a dominant role in the low-frequency areas. Furthermore, there exists biggish fluctuation phenomenon near the tongue. The composition of static pressure fluctuations frequency on the volute wall and blade outlet is similar except that the fluctuation amplitude near the volute wall reduces. 8IL-33 induces immunosuppressive neutrophils via a type 2 innate lymphoid cell/IL-13/STAT6 axis and protects the liver against injury in LCMV infection-induced viral hepatitis显示文摘Viral hepatitis is still a public health problem affecting several million people around the world.Neutrophils are polymorphonuclear cells that have a critical role in antibacterial infection.However,the role of neutrophils in viral infection is not fully understood.By using a mouse model of lymphocytic choriomeningitis virus infection-induced viral hepatitis,we observed increased neutrophil recruitment in the liver accompanied by enhanced CD8+T-cell responses.Liver neutrophils expressed high levels of immunomodulatory cytokines,such as C-X-C chemokine ligand 2,arginase-1,inducible nitric oxide synthase and interleukin(IL)-10,demonstrating immunosuppressive properties.Depletion of neutrophils in vivo by a neutralizing antibody resulted in the exacerbation of liver injury and the promotion of T-cell responses at the immune contraction stage.IL-33 significantly induced neutrophil recruitment in the liver and attenuated liver injury by limiting effector T-cell accumulation.Mechanistically,we found that IL-33 promoted the expression of arginase-1 in neutrophils through the type 2 innate lymphoid cell(ILC2)-derived IL-13.Additionally,IL-13 increased the inhibitory effect of neutrophils on CD8+T-cell proliferation in vitro,partially through arginase-1.Finally,we found that IL-13 induced arginase-1 expression,depending on signal transducer and activator of transcription factor 6(STAT6)signaling.Therefore,IL-33 induced immunosuppressive neutrophils via an ILC2/IL-13/STAT6 axis.Collectively,our findings shed new light on the mechanisms associated with IL-33-triggered neutrophils in the liver and suggest potential targets for therapeutic investigation in viral hepatitis.Yuejin Liang Panpan Yi Denley Ming Kee Yuan Zuliang Jie Zakari Kwota Lynn Soong Yingzi Cong Jiaren Sun 2019Cellular & Molecular Immunology2019,16,2:7

10A study on solid density primary standard显示文摘The accuracy of solid density primary standard is decided mainly by the accuracy of the diameter measurement of silicon sphere. With traditional five-interferogram algorithm to unwrap phase for di- ameter measurement, the phase steps should be equal to π/2 exactly, but this is almost impossible to achieve in nanometer positioning technique. In order to overcome this defect, we have derived an im- proved five-interferogram algorithm, which not only keeps the high accuracy of traditional five-inter- ferogram algorithm, but also does not require absolute equal step to unwrap phase. Instead, the im- proved five-interferogram algorithm only needs measuring phase shifting. Based on the improved al- gorithm, we have developed a novel interferometer with special etalon, and the cavity length of this etalon can be changed by pressure from vertical direction to realize phase shifting. 11Study on Airflow Characteristics in the Semi-Closed Irregular Narrow Flow channel显示文摘The air-jet loom is widely used in the textile industry. The interaction mechanism of airflow and yarn is not clear in such a narrow flow channel, the gas consumption is relatively large, the yarn motion is unstable and the weft insertion is often interrupted during the operation. In order to study the characteristics of the semi-closed flow field in profiled dents, the momentum conservation equation is modified and the model parameters and boundary conditions are set. Compared with the different r, the ratio of profiled dent's thickness and gap, the results show that the smaller the r is, the smaller the velocity fluctuations of the airflow is. When the angle of profiled dents ? is close to zero, the diffusion of the airflow will be less. The experiment is also conducted to verify the result of the simulation with a high-speed camera and pressure sensor in profiled dents. 13Numerical Analysis of Chevron Nozzle Effects on Performance of the Supersonic Ejector-Diffuser System显示文摘The supersonic nozzle is the most important device of an ejector-diffuser system.The best operation condition and optimal structure of supersonic nozzle are hardly known due to the complicated turbulent mixing,compressibility effects and even flow unsteadiness which are generated around the nozzle extent.In the present study,the primary stream nozzle was redesigned using convergent nozzle to activate the shear actions between the primary and secondary streams,by means of longitudinal vortices generated between the Chevron lobes.Exactly same geometrical model of ejector-diffuser system was created to validate the results of experimental data.The operation characteristics of the ejector system were compared between Chevron nozzle and conventional convergent nozzle for the primary stream.A CFD method has been applied to simulate the supersonic flows and shock waves inside the ejector.It is observed that the flow structure and shock system were changed and primary numerical analysis results show that the Chevron nozzle achieve a positive effect on the supersonic ejector-diffuser system performance.The ejector with Chevron nozzle can entrain more secondary stream with less primary stream mass flow rate.Fanshi Kong Yingzi Jin Toshiaki Setoguchi Heuy Dong Kim 2013Journal of Thermal Science2013,22,5:5

15Experimental Study on a Centrifugal Pump with an Open Impeller during Startup Period显示文摘Transient performance of fluid machinery during transient operating periods,such as startup and stopping,has been drawn more attentions recently due to the growing engineering needs.In this paper,the transient behavior of a prototype centrifugal pump with an open impeller during rapid startup period is studied experimentally.The variations of the rotational speed,flow rate,head,and shaft power during rapid startup period are recorded in experiments at different discharge valve openings.In addition,the non-dimensional flow rate and head are also used to analyze the transient behavior.The research result shows that the rise characteristic of the rotational speed is not basically changed by working points,while mainly depends on the startup characteristics of the driving motor.Compared with the rapid rise of the rotational speed,the flow rate rises slowly in the initial stage of startup.Moreover,the flow rate lags behind the rotational speed to rise to final stable value,and the delay becomes more severe with the increase of the discharge valve opening.The shaft power impact phenomenon generally exists in the process of startup.The non-dimensional analysis shows that the non-dimensional head is very high at the very beginning of startup,and quickly falls to the minimum,then gradually rises to final stable value,while the non-dimensional flow rate always shows the rise tendency during whole startup period.In conclusion,it is found from the non-dimensional results that the quasi-steady analysis is unable to accurately assess the transient flow during startup period.Yuliang Zhang Zuchao Zhu Yingzi Jin Baoling Cui Yi Li Huashu Dou 2013Journal of Thermal Science2013,22,1:5

17Chromatographic fingerprinting and quantitative analysis for the quality evaluation of Xinkeshu tablet显示文摘A simple, sensitive and accurate method based on high performance liquid chromatography (HPLC) with diode array detector (DAD) was developed and validated for systematic quality evaluation of one type of traditional Chinese medicine preparations named Xinkeshu (XKS) tablet. In this study, the chromatographic fingerprints of XKS tablet were developed first, 23 peaks were selected as the common peaks to evaluate the similarities among different batches of XKS samples, which were manufactured in a long time span of three years. Additionally, simultaneous quantification of six markers in XKS tablet, including Danshensu, Protocatechuic aldehyde, Puerarin, Daidzin, Salvianolic acid B and Daidzein, was performed. The validation results showed that the developed method was specific, accurate, precise and robust. The preliminary explanation on why a close similarity between fingerprints did not exactly mean similar contents of chemical components in samples was given. The contribution of each chromatographic peak to similarity was also evaluated. 18Effects of rotor structure on performance of small size axial flow fans显示文摘Small size axial flow fans are used as a cooling component for computers,electronic equipment and other electronic components.With the increasing power of electrical equipment,the demand for lower noise and higher ventilation of cooling fan is also increasing.Traditional methods of improving ventilation by raising the fan's rotation speed causes a decrease in efficiency and an increase in noise.In this paper,different structures of fans were simulated,and as a result,the counter-rotating fan can achieve higher pressure,efficiency and facilitate ventilation in a smaller space.Furthermore,some other conclusions are as follows:(1) Higher pressure rise can be obtained by a counter-rotating fan than by the two-stage rotor fan in the same axial length.Meanwhile,the counter-rotating fan has a broader work scope.(2) The main noise type of the counter-rotating fan is rotating noise;the small peak pulse caused by vortex noise mainly due to the eddy current produced by small eddies.(3) When the distance of counter-rotating fans is smaller than 2 times the chord,the greater distance the greater total pressure of the circum-averages and along the axial direction,the total pressure begin to decline until the distance is three times the chord,so there is an optimal distance between rotors.The simulation results are of important significance to the quantitative analysis and optimization design of the counter-rotating fan.Pin Liu Yingzi Jin Yanping Wang 2011Journal of Thermal Science2011,20,3:5
19Baicalin inhibits colistin sulfate-induced apoptosis of PC12 cells显示文摘Baicalin,a type of flavonoid extracted from the dried root of Scutellaria baicalensis georgi,has been shown to effectively inhibit cell apoptosis.Therefore,we assumed that baicalin would suppress colistin sulfate-induced neuronal apoptosis.PC12 cells exposed to colistin sulfate(62.5–500μg/mL)for 24 hours resulted in PC12 cell apoptosis.In addition,caspase-3 activity,lactate dehydrogenase level and free radical content increased in a dose-dependent manner.Subsequently,PC12 cells were pretreated with baicalin(25,50 and 100μg/mL),and exposed to 125μg/mL colistin sulfate.Cell morphology markedly changed,and cell viability increased.Moreover,caspase-3 activity,lactate dehydrogenase level and free radical content decreased.Results indicated that baicalin inhibited colistin sulfate-induced PC12 cell apoptosis by suppressing free radical injury,and reducing caspase-3 activity and lactate dehydrogenase activity.Hong Jiang Pengfei Lv Jichang Li Hongjun Wang Tiezhong Zhou Yingzi Liu Wei Lin 2013Neural Regeneration Research2013,8,28:4
20Numerical and Experimental Study on Aerodynamic Performance of Small Axial Flow Fan with Splitter Blades显示文摘To improve the aerodynamic performance of small axial flow fan, in this paper the design of a small axial flow fan with splitter blades is studied. The RNG k-ε turbulence model and SIMPLE algorithm were applied to the steady simulation calculation of the flow field, and its result was used as the initial field of the large eddy simulation to calculate the unsteady pressure field. The FW-H noise model was adopted to predict aerodynamic noise in the six monitoring points. Fast Fourier transform algorithm was applied to process the pressure signal. Experiment of noise testing was done to further investigate the aerodynamic noise of fans. And then the results obtained from the numerical simulation and experiment were described and analyzed. The results show that the static characteristics of small axial fan with splitter blades are similar with the prototype fan, and the static characteristics are improved within a certain range of flux. The power spectral density at the six monitoring points of small axial flow fan with splitter blades have decreased to some extent. The experimental results show sound pressure level of new fan has reduced in most frequency bands by comparing with prototype fan.
