| 1 | Tumor growth inhibition effect of hIL-6 on colon cancer cells transfected with the target gene by retroviral vector显示文摘AIM To observe the tumor inhibitory effects bytransfecting IL-6 cDNA into colon cancer cell lineHT-29 with retroviral vector pZIP cDNA.METHODS Human IL-6 gene was reconstructedin retrovirus vector and transfected into incasingcells PA317 by lipofectamine mediated method,the clones of the cells transferred with hlL-6were selected by G418,and targeted HT-29 cellswere infected with the virus granules secretedfrom PA317 and also selected by G418.Test genetranscription and expression level byhybridization,ELISA and MTT assay,etc.Analyze tumor inhibitory effects according to thecell growth curve,plating forming rate andtumorigenicity in nude mice.RESULT Successfully constructed andtransfected recombinant expressing vectorspZIPIL-6 cDNA and got positive transfected celllines.The colon cancer cell line(HT-29 IL-5)transfected with the hlL-5 gene by retroviralvector was established.The log proliferationperiod and the doubling time of this cell line wasbetween 4 to 7 days and 2.5 days according tothe direct cell count,the cell proliferation wasobviously inhibited with MTT assay,the platinginhibitory rate was 50% by plating efficiencytest.When HT-29 IL-6 cells were inoculated intothe nude mice subcutaneously,carcinogenicactivity of the solid tumor was found superior tothe control group and the size of tumor was notsignificantly enlarged.Injection of combinationvirus fluid containing 11.-6 gene intotransplantation tumors could inhibit the growthand development of the tumor.CONCLUSION IL-6 could inhibit the growth andproliferation of colon cancer cells by retroviralvector-mediated transduction. | Xiao B Jing B Zhang YL Zhou DY Zhang WD | 2000 | World Journal of Gastroenterology2000，6，1： | 11 |
| 2 | Effects of partial portal vein arterialization on the hilar bile duct in a rat model显示文摘BACKGROUND: Liver revascularization is frequently required during the enlarged radical operation for hilar cholangio carcinoma involving the hepatic artery. Researchers have carried out a number of experiments applying partial porta vein arterialization (PVA) in clinical practice. In this study we aimed to establish a theoretical basis for clinical application o partial PVA and to investigate the effects of partial PVA on ra hilar bile duct and hepatic functions. METHODS: Thirty rats were randomly and equally assigned into 3 groups: control (group A), hepatic artery ligation+bile duct recanalization (group B), and partial PVA+bile duc recanalization (group C). Proliferation and apoptosis o rat hilar bile duct epithelial cells, arteriolar counts of the peribiliary plexus (PBP) of the bile duct wall, changes in serum biochemistry, and pathologic changes in the bile duc were assessed 1 month after operation. RESULTS: The proliferation of hilar bile duct epithelial cells in group B was greater than in groups A and C (P<0.01). No apoptotic hilar bile duct epithelial cells were detected in any of the groups. The PBP arteriolar counts of the hilar bile duc wall were similar in groups A and C (P>0.05), but the coun was lower in group B than in group A (P<0.01). No statistically significant differences in alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase and albumin were found in the 3 groups. The gamma-glutamyltransferase value was higher in group B than in groups A and C (P<0.01) The hepatic tissues of groups A and C showed no significan abnormality. Chronic inflammatory changes in the hilar bile duct walls were observed only in group B. | 3 | ^(14)C measurement of forest soils in Dinghushan Biosphere Reserve显示文摘Organic carbon in forest soils of Qingyunsi and Wukesong profiles can be divided into fast and slow components. Δ14C values of these profiles decrease with increasing of depth. The Δ14C values in 30—40 cm depth interval of Wukesong profile are decreasing sharply until a very low value,showing that a strong geological environment change occurred about 1 560 years ago. The 14C apparent ages of Wukesong profile show that the coniferous and broad-leaf mixed forests around Wukesong profile have been developing since 425 a BP, which is consistent with historical documents. | 5 | Impact of Low Frequency Electro-acupuncture on Glucose and Lipid Metabolism in Unmarried PCOS Women:A Randomized Controlled Trial显示文摘Objective:To explore the effect of electro-acupuncture(EA)on glucose and lipid metabolism in unmarried patients with polycystic ovary syndrome(PCOS).Methods:Fifty-four PCOS patients were equally randomized into true acupuncture group and sham acupuncture group(control)for totally 16 weeks of treatment by random method with a computerized randomization program.Patients in true acupuncture group accepted traditional acupuncture methods with EA and two sets of acupoint groups were used alternatively.The first set consisted of Zhongji(CV 3),Qihai(CV 6),Guilai(ST 29),Sanyinjiao(SP 6),Yinlingquan(SP 9),Hegu(LI4)and Baihui(GV 20),and the second set consisted of Tianshu(ST 25),ST 29,CV 3,CV 6,SP 6,Taichong(LR 3),Neiguan(PC)6 and GV 20.Patients in the sham acupuncture group accepted shallow acupuncture methods through EA without electricity at 4 non-meridian points in each shoulder and upper arm.Outcome measures included body mass index(BMI),waist-hip-ratio(WHR),oral glucose tolerance test(OGTT),insulin release test,glucose and lipid metabolism indicators such as total cholesterol(TC),triglycerides,high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol,adiponectin,leptin,visfatin,resistin,and interleukin(IL-6).Results:Twenty-six subjects in the true acupuncture group and 20 subjects in the sham group completed the clinical trial.After 16 weeks of treatment,no significant difference in the outcome measures were observed between the two groups(P>0.05).However,as compared with baseline data,a reduction in weight,BMI,hipline,WHR,fasting glucose,homeostatic model assessment of insulin sensitivity,visfatin and HDL-C,and an increase in resistin and IL-6 were observed in the true acupuncture group(P<0.05).In addition,a reduction in visfatin and an increase in TC were also observed in the sham group(P<0.05).Conclusions:Acupuncture may have a beneficial effect in the treatment of PCOS by improving glucose and lipid metabolism.Moreover,the sham acupuncture may be not completely ineffective.Sham acupuncture may improve some of the aspects of the glucose and lipid metabolism of PCOS patients through a placebo effect.(Registration Nos.ChiCTR-TRC-12002529 and NCT01812161) | DONG Hao-xu WANG Qing WANG Zhi WU Xiao-ke CHENG Ling ZHOU Zhong-ming YANG Li Yl Ping HUANG Dong-mei | 2021 | Chinese Journal of Integrative Medicine2021，27，10： | 4 |
