| 1 | Impact of Taurine on the proliferation and apoptosis of human cervical carcinoma cells and its mechanism显示文摘Background:Cervical cancer has the fourth highest incidence and mortality rate of all cancers in women worldwide;让seriously harms their physical and mental health.The aim of this study was to observe the roles and preliminary mechanism of Taurine(Tau)-induced apoptosis in cervical cancer cells.Methods:Cells from the human cervical cancer cell line SiHa were transfected with the recombinant plasmid pEGFP-N1-MST1(mammalian sterile 20-like kinase 1);then,the cell proliferation activity was analyzed by the MTT assay,cell apoptosis by flow cytometry,and the related protein levels by Western blotting.Results:Tau inhibited the proliferation of SiHa cells and induced apoptosis in these cells(the apoptotic rate was 21.95%in the Tau 160 mmol/L group and 30%in the Tau 320 mmol/L group),upregulated the expression of the MST1(control,0.53;Tau 40-320 mmol/L groups,0.84-1.45)and Bax(control,0.45;Tau 40-320 mmol/L groups,0.64-1.51)proteins(P<0.01),and downregulated the expression of Bcl-2(control,1.28,Tau 40-320 mmol/L groups,0.93-0.47)(P<0.01).The overexpression of MST1 promoted the apoptosis of SiHa cells,enhanced the apoptosis-inductive effects of Tau(P<0.01),upregulated the expression of the proapoptotic proteins p73,p53,PUMA(p53 upregulated modulator of apoptosis),and caspase-3,and promoted the phosphorylation of YAP(Yes-associated protein).Conclusions:Tau inhibited the proliferation and induced the apoptosis of cervical cancer SiHa cells.The MST1 protein plays an important role in the Tau-induced apoptosis of cervical cancer cells. | Hua Li Wen-Jing Ruan Li-Qiao Liu Hui-Fang Wan Xiao-Hong Yang Wei-Feng Zhu Le-Han Yu Xia-Li Zhang Fu-Sheng Wan | 2019 | Chinese Medical Journal2019,,8: | 18 |

| 4 | Emerging role of Hippo pathway in gastric and othergastrointestinal cancers显示文摘More evidence has underscored the importance of Hippo signaling pathway in gastrointestinal tissue homeostasis, whereas its deregulation induces tumorigenesis. Yes-associated protein 1(YAP1) and its close paralog TAZ, transcriptional co-activator with a PDZbinding motif, function as key effectors negatively controlled by the Hippo pathway. YAP1/TAZ exerts oncogenic activities by transcriptional regulation via physical interaction with TEAD transcription factors. In various cancers, Hippo pathway cross-talks with pro- or anti-tumorigenic pathways such as GPCR, Wnt/β-catenin, Notch and TGF-β signaling and is deregulated by multiple factors including cell density/junction and micro RNAs. As YAP1 expression is significantly associated with poor prognosis of gastric and other gastrointestinal cancers, detailed delineation of Hippo regulation in tumorigenesis provides novel insight for therapeutic intervention. | 7 | HBx通过HIPPO信号通路导致小鼠肝癌的发生机制显示文摘目的观察HIPPO信号通路在小鼠感染乙型肝炎病毒X(hepatitis B virus X,HBx)不同时间的变化,初步探讨HBx导致肝癌发生的机制。方法将转染HBx的肝前体细胞通过门静脉注射入小鼠体内,构建能稳定表达HBx的动物模型,分别于30、90、180、360 d取肝脏组织,HE染色观察肝脏组织病理特征,RT-PCR、Western blot以及免疫组化法检测肝脏组织中HBx的表达,RT-PCR检测HIPPO信号通路相关因子MST1、YAP1 mRNA的表达,Western blot检测MST1、YAP1、p-MST1、p-YAP1蛋白的表达。结果成功构建能在小鼠肝脏中稳定表达HBx的动物模型并发展为肝癌,Western blot结果显示,与对照组相比,HBx组小鼠肝脏组织在术后30 d即出现MST1表达下调,p-MST1表达上调(P<0.01);YAP1表达上调,p-YAP1表达下调(P<0.05),RT-PCR与Western blot结果一致。这种改变一直持续至肿瘤发生。结论HBx可以导致小鼠肝癌发生,其机制可能与长期影响HIPPO信号通路的表达有关。 | 张思遥 黄欣 王薛 杜彬 毋楠 周梦瑶 冯涛 | 2020 | 第三军医大学学报2020,42,7: | 2 |
