| 2 | Evaluation of irrigation water salinity and leaching fraction on the water productivity for crops显示文摘In arid and semi-arid irrigated croplands,the excessive accumulation of soluble salts in the root zone is an extensive problem that seriously limits crop yield and water productivity(WP).To avoid affects the yield potential of crops,the application of extra irrigation for leaching of excessive salts from the root zone was required.Quantitative knowledge of effects of the irrigation water salinity and leaching fraction(LF)on the relative yield(RY)and the unit water productivity of crop evapotranspiration(UWPET)and the unit water productivity of irrigation water(UWPI)were becoming gradually important.This article provided theoretical models for estimating the UWPs(UWPET and UWPI)and optimizing leaching fraction according to irrigation water salinity.In the present study,eight levels of irrigation water salinity(ECw=0.25,0.50,0.75,1,2,3,4,and 5 dS/m)and 39 levels of LF values ranging from 0.04 to 0.80 were set and tested to assessing their effects on the RY and UWPs for four typical crops(barley,bean,wheat,and maize)with different salt tolerance levels.Almost every curve determined between the UWPs and LFs for the four crops had an inflection point.It was indicated that the UWPET and UWPI could be maximized by optimizing the LF under different irrigation water salinities.Furthermore,the linear regression relationships were established to estimate the maximum values of UWPs and their corresponding optimal LFs for four crops by using the irrigation water salinity.Moreover,the theoretical models for estimating the UWPs were validated by data of wheat from previous literature,and the models could be suitable with acceptable relative errors when LFs ranging from 0.07 to 0.17. | Songrui Ning Beibei Zhou Quanjiu Wang Wanghai Tao | 2020 | International Journal of Agricultural and Biological Engineering2020,13,1: | 2 |
| 3 | Novel dual inhibitor for targeting PIM1 and FGFR1 kinases inhibits colorectal cancer growth in vitro and patient-derived xenografts in vivo显示文摘Colorectal cancer(CRC) is the second most common cause of cancer-related death in the world. The pro-viral integration site for Moloney murine leukemia virus 1(PIM1) is a proto-oncogene and belongs to the serine/threonine kinase family, which are involved in cell proliferation, migration,and apoptosis. Fibroblast growth factor receptor 1(FGFR1) is a tyrosine kinase that has been implicated in cell proliferation, differentiation and migration. Small molecule HCI-48 is a derivative of chalcone, a class of compounds known to possess anti-tumor, anti-inflammatory and antibacterial effects. However,the underlying mechanism of chalcones against colorectal cancer remains unclear. This study reports that HCI-48 mainly targets PIM1 and FGFR1 kinases, thereby eliciting antitumor effects on colorectal cancer growth in vitro and in vivo. HCI-48 inhibited the activity of both PIM1 and FGFR1 kinases in an ATPdependent manner, as revealed by computational docking models. Cell-based assays showed that HCI-48inhibited cell proliferation in CRC cells(HCT-15, DLD1, HCT-116 and SW620), and induced cell cycle arrest in the G2/M phase through modulation of cyclin A2. HCI-48 also induced cellular apoptosis, as evidenced by an increase in the expression of apoptosis biomarkers such as cleaved PARP, cleaved caspase 3 and cleaved caspase 7. Moreover, HCI-48 attenuated the activation of downstream components of the PIM1 and FGFR1 signaling pathways. Using patient-derived xenograft(PDX) murine tumor models,we found that treatment with HCI-48 diminished the PDX tumor growth of implanted CRC tissue expressing high protein levels of PIM1 and FGFR1. This study suggests that the inhibitory effect of HCI-48 on colorectal tumor growth is mainly mediated through the dual-targeting of PIM1 and FGFR1kinases. | 6 | HITAC-seq enables high-throughput cost-effective sequencing of plasmids and DNA fragments with identity显示文摘DNA sequencing is vital for many aspects of biological research and diagnostics. Despite the development of second and third generation sequencing technologies, Sanger sequencing has long been the only choice when required to precisely track each sequenced plasmids or DNA fragments. Here, we report a complete set of novel barcoding and assembling system, Highly-parallel Indexed Tagmentation-reads Assembled Consensus sequencing(HITAC-seq), that could massively sequence and track the identities of each individual sequencing sample. With the cost of much less than that of single read of Sanger sequencing,HITAC-seq can generate high-quality contiguous sequences of up to 10 kilobases or longer. The capability of HITAC-seq was confirmed through large-scale sequencing of thousands of plasmid clones and hundreds of amplicon fragments using approximately 100 pg of input DNAs. Due to its long synthetic length, HITACseq was effective in detecting relatively large structural variations, as demonstrated by the identification of a~1.3 kb Copia retrotransposon insertion in the upstream of a likely maize domestication gene. | 7 | Sanguinarine suppresses cell proliferation, migration and invasion in nasopharyngeal carcinoma via inhibiting mTOR signaling显示文摘OBJECTIVE: To confirm the anti-NPC effect of sanguinarine(SA) through a series of wet experiments.METHODS: NPC cell viability was determined by proliferation experiment. Cell clone formation experiment, cell scratch test, transwell migration and invasion experiment and flow cytometry-based cell apoptosis assay were further performed. In addition, Western blotting was performed to investigate the cell signaling pathway. All the relevant experimental data were statistically processed using SPSS 16.0 software.RESULTS: The results showed that sanguinarine represented a time and dose dependent inhibition effects on NPC cell proliferation including the low differentiated CNE2 cells and high metastatic 5-8F cells, along with the cell cloning ability reduction. In addition, sanguinarine has a certain inhibitory effect on the invasion and migration of NPC cells. Mechanistically, sanguinarine displayed the anti-NPC effects mainly involved into the suppression of m TOR signaling and cell apoptosis, which is closely associated with the tumor growth and metastatic malignancy. | 10 | Prediction of high-density polyethylene pyrolysis using kinetic parameters based on thermogravimetric and artificial neural networks显示文摘Pyrolysis is considered an attractive option and a promising way to dispose waste plastics.The thermogravimetric experiments of high-density polyethylene(HDPE)were conducted from 105℃ to 900℃ at different heating rates(10℃/min,20℃/min,and 30℃/min)to investigate their thermal pyrolysis behavior.We investigated four methods including three model-free methods and one modelfitting method to estimate dynamic parameters.Additionally,an artificial neural network model was developed by providing the heating rates and temperatures to predict the weight loss(wt.%)of HDPE,and optimized via assessing mean squared error and determination coefficient on the test set.The optimal MSE(2.6297×10^(−2))and R^(2) value(R^(2)>0.999)were obtained.Activation energy and preexponential factor obtained from four different models achieves the acceptable value between experimental and predicted results.The relative error of the model increased from 2.4%to 6.8%when the sampling frequency changed from 50 s to 60 s,but showed no significant difference when the sampling frequency was below 50 s.This result provides a promising approach to simplify the further modelling work and to reduce the required data storage space.This study revealed the possibility of simulating the HDPE pyrolysis process via machine learning with no significant accuracy loss of the kinetic parameters.It is hoped that this work could potentially benefit to the development of pyrolysis process modelling of HDPE and the other plastics. | Xiaoxiao Yin Junyu Tao Guanyi Chen Xilei Yao Pengpeng Luan Zhanjun Cheng Ning Li Zhongyue Zhou Beibei Yan | 2023 | Frontiers of Environmental Science & Engineering2023,17,1: | 0 |
