| 1 | A Resource for Inactivation of MicroRNAs Using Short Tandem Target Mimic Technology in Model and Crop Plants显示文摘microRNAs (miRNAs)are endogenous small non-coding RNAs that bind to mRNAs and target them for cleavage and/or translational repression,leading to gene silencing.We previously developed short tandem target mimic (STTM)technology to deactivate endogenous miRNAs in Arabidopsis.Here,we created hundreds of STTMs that target both conserved and species-specific miRNAs in Arabidopsis,tomato,rice,and maize,providing a resource for the functional interrogation of miRNAs.We not only revealed the functions of several miRNAs in plant development,but also demonstrated that tissue-specific inactivation of a few miRNAs in rice leads to an increase in grain size without adversely affecting overall plant growth and development.RNA-seq and small RNAseq analyses of STTM156/157 and STTM165/166 transgenic plants revealed the roles of these miRNAs in plant hormone biosynthesis and activation,secondary metabolism,and ion-channel activity-associated electrophysiology,demonstrating that STTM technology is an effective approach for studying miRNA functions.To facilitate the study and application of STTM transgenic plants and to provide a useful platform for storing and sharing of information about miRNA-regulated gene networks,we have established an online Genome Browser (http://gffzzc750bc0108fa4044skckquvvqc6bb6v9u.ffgz.tsg.suse.edu.cn/designindex2.php) to display the transcriptomic and miRNAomic changes in STTMinduced miRNA knockdown plants. | Ting Peng Mengmeng Qiao Haiping Liu Sachin Teotia Zhanhui Zhang Yafan Zhao Bobo Wang Dongjie Zhao Lina Shi Cui Zhang Brandon Le Kestrel Rogers Chathura Gunasekara Haitang Duan Yiyou Gu Lei Tian Jinfu Nie Jian Qi Fanrong Meng Lan Huang Qinghui Chen Zhenlin Wang Jinshan Tang Xiaoqing Tang Ting Lan Xuemei Chen Hairong Wei Quanzhi Zhao Guiliang Tang | 2018 | Molecular Plant2018,11,11: | 7 |
| 2 | Microstructure, precipitates and mechanical properties of powder bed fused inconel 718 before and after heat treatment显示文摘IN718 alloy was fabricated by laser powder bed fusion(PBF) for examination of microstructure, precipitates and mechanical properties in the as-built state and after different heat treatments. The as-built alloy had a characteristic fine cellular-dendritic microstructure with Nb, Mo and Ti segregated along the interdendritic region and cellular boundary. The as-built alloys were then subjected to solution heat treatment(SHT) at 980°C or 1065°C for 1 h. SHT at 980°C led to the formation of δ-phase in the interdendritic region or cellular boundary. The segregation was completely removed by the SHT at 1065°C, but recrystallization was observed, and the carbides decorated along the grain boundaries. The as-built alloy and alloys with SHT at 980°C and 1065°C were two-step aged, which consisted of annealing at 720°C for8 h followed by annealing at 620°C for 8 h. Transmission electron microscopy revealed the precipitation of λ’ and λ' in all alloys after two-step aging, but the amount and uniformity of distribution varied. The Vickers hardness of the PBF IN718 alloy increased from 296 HV to 467 HV after direct aging. The hardness decreased to 267 HV and 235 HV after SHT at 980°C and 1065°C, respectively, but increased to 458 HV and 477 HV followed by aging. The evolution of Young’s modulus after heat treatment exhibited similar trend to that of hardness. | 3 | Clinical classification of symptomatic heterotopic pancreas of the stomach and duodenum:A case series and systematic literature review显示文摘BACKGROUND Heterotopic pancreas(HP)is an aberrant anatomic malformation that occurs most commonly in the upper gastrointestinal tract.While the majority of heterotopic pancreatic lesions are asymptomatic,many manifest severe clinical symptoms which require surgical or endoscopic intervention.Understanding of the clinical manifestations and symptoms of HP is limited due to the lack of large volume studies in the literature.The purpose of this study is to review symptomatic cases at a single center and compare these to a systematic review of the literature in order to characterize common clinical manifestations and treatment of this disease.AIM To classify the common clinical manifestations of heterotopic pancreas.METHODS A retrospective review was conducted of pathologic samples containing heterotopic pancreas from 2000-2018.Review was limited to HP of the upper gastrointestinal tract due to the frequency of presentation in this location.Symptomatic patients were identified from review of the medical records and clinical symptoms were tabulated.These were compared to a systematic review of the literature utilizing Pub Med and Embase searches for papers pertaining to heterotopic pancreas.Publications describing symptomatic presentation of HP were selected for review.Information including demographics,symptoms,presentation and treatment were compiled and analyzed.RESULTS Twenty-nine patient were identified with HP at a single center,with six of these identified has having clinical symptoms.Clinical manifestations included,gastrointestinal bleeding,gastric ulceration with/without perforation,pancreatitis,and gastric outlet obstruction.Systemic review of the literature yielded 232 publications detailing symptomatic cases with only 20 studies describing ten or more patients.Single and multi-patient studies were combined to form a cohort of 934 symptomatic patients.The majority of patients presented with abdominal pain(67%)combined with one of the following clinical categories:(1)Dyspepsia,(n=445,48%);(2)Pancreatitis(n=260,28%);(3)Gastrointestinal bleeding(n=80,9%);and(4)Gastric outlet obstruction(n=80,9%).The majority of cases(n=832,90%)underwent surgical or endoscopic resection with 85%reporting resolution or improvement in their symptoms.CONCLUSION Heterotopic pancreas can cause significant clinical symptoms in the upper gastrointestinal tract.Better understanding and classification of this disease may result in more accurate identification and treatment of this malformation. | Michael T Le Compte Brandon Mason Keenan J Robbins Motoyo Yano Deyali Chatterjee Ryan C Fields Steven M Strasberg William G Hawkins | 2022 | World Journal of Gastroenterology2022,28,14: | 3 |

| 6 | Consumption of energy beverage is associated with attenuation of arterial endothelial flow-mediated dilatation显示文摘AIM To investigate whether consumption of an energy drink will acutely impair endothelial function in young healthy adults. METHODS Energy drinks are being consumed more and more worldwide, and have been associated with some deaths in adolescents and young adults, especially when consumed while exercising. After fasting and not smoking for at least 8 h prior, eleven medical students(9 males) received an electrocardiogram, blood pressure and pulse check, and underwent baseline testing(BL) of endothelial function using the technique of endothelium-dependent flow mediated dilatation(FMD) with high-resolution ultrasound(according to recommended guidelines of the University of Wisconsin Atherosclerosis Imaging Research Program Core Laboratory). The subjects then drank an energy beverage(EB), a 24-oz can of Monster Energy, and the above was repeated at 90 min after consumption. The relative FMD(%) was calculated as the ratio between the average post-cuff release and the baseline diameter. Each image was checked for quality control, and each artery diameter was measured from the media to media points by two experts, 3 measurements at the QRS complex, repeated on 3 separate beats, and then all were averaged.RESULTS Subjects characteristics averages(given with standard deviations) include: Age 24.5 ± 1.5 years, sex 9 male and 2 female, weight 71.0 ± 9.1 kg, height 176.4 ± 6.0 cm, BMI 22.8 ± 2.7 kg/m^2. The hemodynamics were as follows, BL vs EB group respectively(mean ± SD): Heart rate 65.2 ± 11.3 vs 68.2 ± 11.8 beats per minute, systolic blood pressure 114.0 ± 10.4 mmH g vs 114.1 ± 10.4 mmH g, diastolic blood pressure 68.8 ± 9.3 mmH g vs 70.6 ± 7.1 mmH g; all were not significantly different. However after drinking the EB, a significantly attenuated peak FMD response was measured(mean ± SD): BL group 5.9% ± 4.6% vs EB group 1.9% ± 2.1%; P = 0.03).
