1Mesenchymal stromal cells from human perinatal tissues: From biology to cell therapy显示文摘Cell-based regenerative medicine is of growing interest in biomedical research. The role of stem cells in this context is under intense scrutiny and may help to define principles of organ regeneration and develop innovative therapeutics for organ failure. Utilizing stem and progenitor cells for organ replacement has been conducted for many years when performing hematopoietic stem cell transplantation. Since the first successful transplantation of umbilical cord blood to treat hematological malignancies, non-hematopoietic stem and progenitor cell populations have recently been identified within umbilical cord blood and other perinatal and fetal tissues. A cell population entitled mesenchymal stromal cells (MSCs) emerged as one of the most intensely studied as it subsumes a variety of capacities: MSCs can differentiate into various subtypes of the mesodermal lineage, they secrete a large array of trophic factors suitable of recruiting endogenous repair processes and they are immunomodulatory.Focusing on perinatal tissues to isolate MSCs, we will discuss some of the challenges associated with these cell types concentrating on concepts of isolation and expansion, the comparison with cells derived from other tissue sources, regarding phenotype and differentiation capacity and finally their therapeutic potential.Karen Bieback Irena Brinkmann 2010World Journal of Stem Cells2010,2,4:15

3Animal phylogeny and large-scale sequencing: progress and pitfalls显示文摘Phylogenomics, the inference of phylogenetic trees using genome-scale data, is becoming the rule for resolving difficult parts of the tree of life. Its promise resides in the large amount of information available, which should eliminate stochastic error. However, systematic error, which is due to limitations of reconstruction meth-ods, is becoming more apparent. We will illustrate, using animal phylogeny as a case study, the three most effi-cient approaches to avoid the pitfalls of phylogenomics: (1) using a dense taxon sampling, (2) using probabilistic methods with complex models of sequence evolution that more accurately detect multiple substitutions, and (3) removing the fastest evolving part of the data (e.g., species and positions). The analysis of a dataset of 55 animal species and 102 proteins (25712 amino acid positions) shows that standard site-homogeneous model inference is sensitive to long-branch attraction artifact, whereas the site-heterogeneous CAT model is less so. The latter model correctly locates three very fast evolving species, the appendicularian tunicate Oikopleura, the acoel Convoluta and the myxozoan Buddenbrockia. Overall, the resulting tree is in excellent agreement with the new animal phylogeny, confirming that 'simple' organisms like platyhelminths and nematodes are not necessarily of basal emergence. 6Next generation per-and poly-fluoroalkyl substances:Status and trends,aquatic toxicity,and risk assessment显示文摘Widespread application of poly-and per-fluoroalkyl substances(PFAS)has resulted in some substances being ubiquitous in environmental matrices.That and their resistance to degradation have allowed them to accumulate in wildlife and humans with potential for toxic effects.While specific substances of concern have been phased-out or banned,other PFAS that are emerging as alternative substances are still produced and are being released into the environment.This review focuses on describing three emerging,replacement PFAS:perfluoroethylcyclohexane sulphonate(PFECHS),6:2 chlorinated polyfluoroalkyl ether sulfonate(6:2 Cl-PFAES),and hexafluoropropylene oxide dimer acid(HFPO-DA).By summarizing their physicochemical properties,environmental fate and transport,and toxic potencies in comparison to other PFAS compounds,this review offers insight into the viabilities of these chemicals as replacement substances.Using the chemical scoring and ranking assessment model,the relative hazards,uncertainties,and data gaps for each chemical were quantified and related to perfluorooctane sulfonic acid(PFOS)and perfluorooctanoic acid(PFOA)based on their chemical and uncertainty scores.The substances were ranked PFOS>6:2 Cl-PFAES>PFOA>HFPO-DA>PFECHS according to their potential toxicity and PFECHS>HFPO-DA>6:2 Cl-PFAES>PFOS>PFOA according to their need for future research.Since future uses of PFAS remain uncertain in the face of governmental regulations and production bans,replacement PFAS will continue to emerge on the world market and in the environment,raising concerns about their general lack of information on mechanisms and toxic potencies.Hannah Mahoney Yuwei Xie Markus Brinkmann John P.Giesy 2022Eco-Environment & Health2022,1,2:2

8Pseudomonas exotoxin antisense RNA selectively kills hepatitis B virus infected cells显示文摘AIM: To present an approach for selectively killing retrovirus-infected cells that combines the toxicity of Pseudomonas exotoxin (PE) and the presence of reverse transcriptase (RT) in infected cells. METHODS: PE antisense toxin RNA has palindromic stem loops at its 5' and 3' ends enabling self-primed generation of cDNA in the presence of RT. The RT activity expressed in retrovirus-infected cells converts 'antisense-toxin-RNA' into a lethal toxin gene exclusively in these cells. RESULTS: Using cotransfection studies with PE-expressing RNAs and β-gal expressing reporter plasmids, we show that, in HepG2 and HepG2.2.15 hepatoma cells as well as in duck hepatitis B virus (DHBV) infected cells, HBV or DHBV-polymerase reverse transcribe a lethal cDNA copy of an antisense toxin RNA, which is composed of sequences complementary to a PE gene and eukaryotic transcription and translation signals.
