3Overexpression of carbonic anhydrase Ⅱ and Ki-67 proteins in prognosis of gastrointestinal stromal tumors显示文摘AIM: To investigate the expression and prognostic value of carbonic anhydrase Ⅱ (CA Ⅱ) and Ki-67 in gastrointestinal stromal tumors (GISTs). METHODS: One hundred and thirteen GIST patients admitted to Chinese People's Liberation Army General Hospital from January 2004 to December 2010 were retrospectively followed up, and immunohistochemistry was used to detect CA Ⅱ, Ki-67 and CD117 expression in tumor samples. The survival rates of the patients were analyzed using the Kaplan-Meier method. Log-rank test, χ 2 test and Cox proportional hazards model were used to determine the relationships between CA Ⅱ, Ki-67 and CD117 expression and prognostic value in GISTs. RESULTS: The survival rates at 1, 3 and 5 years were 90.0%, 82.0% and 72.0% in all patients. However, in patients with positive CA Ⅱ or Ki-67, the survival rates were 92.0%, 83.0% and 77.0% or 83.0%, 66.6% and 53.0%, respectively. Compared with the negative groups, the survival rates in the positive groups were significantly lower (CA Ⅱ log-rankP = 0.000; Ki-67 logrank P = 0.004). Multivariate Cox analysis revealed that CA Ⅱ, CD117 and Ki-67 were considerable immune factors in prognosis of GIST patients (CA Ⅱ P = 0.043; CD117 P = 0.042; Ki-67 P = 0.007). Besides, tumor diameter, mitotic rate, tumor site, depth of invasion, complete resection, intraoperative rupture, and adjuvant therapy were important prognosis predictive factors. Our study indicated that CA Ⅱ had strong expression in GISTs and the prognosis of GISTs with high CA Ⅱ expression was better than that of GISTs with low or no expression, suggesting that CA Ⅱ is both a diagnostic and prognostic biomarker for GIST. CONCLUSION: CA Ⅱ and Ki-67 are significant prognostic factors for GISTs. 7Chromosome-scale genome assembly of sweet cherry(Prunus avium L.)cv.Tieton obtained using long-read and Hi-C sequencing显示文摘Sweet cherry(Prunus avium)is an economically significant fruit species in the genus Prunus.However,in contrast to other important fruit trees in this genus,only one draft genome assembly is available for sweet cherry,which was assembled using only Illumina short-read sequences.The incompleteness and low quality of the current sweet cherry draft genome limit its use in genetic and genomic studies.A high-quality chromosome-scale sweet cherry reference genome assembly is therefore needed.A total of 65.05 Gb of Oxford Nanopore long reads and 46.24 Gb of Illumina short reads were generated,representing~190x and 136x coverage,respectively,of the sweet cherry genome.The final de novo assembly resulted in a phased haplotype assembly of 344.29 Mb with a contig N50 of 3.25 Mb.Hi-C scaffolding of the genome resulted in eight pseudochromosomes containing 99.59%of the bases in the assembled genome.Genome annotation revealed that more than half of the genome(59.40%)was composed of repetitive sequences,and 40,338 protein-coding genes were predicted,75.40%of which were functionally annotated.With the chromosomescale assembly,we revealed that gene duplication events contributed to the expansion of gene families for salicylic acid/jasmonic acid carboxyl methyltransferase and ankyrin repeat-containing proteins in the genome of sweet cherry.Four auxin-responsive genes(two GH3s and two SAURs)were induced in the late stage of fruit development,indicating that auxin is crucial for the sweet cherry ripening process.In addition,772 resistance genes were identified and functionally predicted in the sweet cherry genome.The high-quality genome assembly of sweet cherry obtained in this study will provide valuable genomic resources for sweet cherry improvement and molecular breeding.Jiawei Wang Weizhen Liu Dongzi Zhu Po Hong Shizhong Zhang Shijun Xiao Yue Tan Xin Chen Li Xu Xiaojuan Zong Lisi Zhang Hairong Wei Xiaohui Yuan Qingzhong Liu 2020Horticulture Research2020,7,1:4
8Activation of anterior thalamic reticular nucleus GABAergic neurons promotes arousal from propofol anesthesia in mice显示文摘Propofol is widely used for the induction and maintenance of anesthesia,which causes a rapid loss of consciousness.However,the mechanisms underlying the hypnosis effect of propofol are still not fully understood.The thalamic reticular nucleus(TRN)is crucial for regulating wakefulness,sleep rhythm generation,and sleep stability,while the role of TRN in the process of propofol-induced anesthesia is still unknown.Here,we investigated the function of the anterior TRN in propofol general anesthesia.Our results demonstrated that the neural activity of anterior TRN is suppressed during propofol anesthesia,whereas it is robustly activated from anesthesia by recording the calcium signals using fiber photometry technology.The results showed that the activation of anterior TRN neurons by chemogenetic and optogenetic methods shortens the emergency time without changing the induction time.Conversely,chemogenetic or optogenetic inhibition of the TRN neurons leads to a delay in the recovery time.Our study showed that anterior TRN is crucial for behavioral arousal without affecting the induction time of propofol anesthesia.Yanjun Liu Bing Chen Yirong Cai Yuan Han Ying Xia Nanqi Li Bingqian Fan Tianjie Yuan Junli Jiang Po Gao Weifeng Yu Yingfu Jiao Wenxian Li 2021Acta Biochimica et Biophysica Sinica2021,53,7:3
9Hippocampus is more vulnerable to neural damages induced by repeated sevoflurane exposure in the second trimester than other brain areas显示文摘During the rapidly developing and sensitive period of the central nervous system (CNS), a harmful stimulus may have serious consequences. The effect of anesthetic exposure on the development of the offspring’s CNS during pregnancy is still unclear and has been widely concerned. In the present study, we compared the susceptibility of the hippocampus with those of other brain regions in offsprings when the mother mice were exposed to repeated sevoflurane. We found that other than affecting motor sensation, emotion, or social behavior of offspring mice, repeated sevoflurane exposure induced significant memory deficiency. Compared with other brain regions, the hippocampus, which is the key component of the brain serving for learning and memory, was more vulnerable to repeated sevoflurane exposure. We also found that repeated sevoflurane exposure to mother mice could inhibit the axon development of hippocampal neurons. We also predicted that N6-methyladenosine modification of mRNA might play an essential role in the vulnerability of the hippocampus to sevoflurane, while the underlying cellular mechanism needs to be explored in the future. 10A day-ahead scheduling framework for thermostatically controlled loads with thermal inertia and thermal comfort model显示文摘This paper proposes a day-ahead dispatch framework of thermostatically controlled loads(TCLs) for system peak load reduction. The proposed day-ahead scheduling framework estimates the user’s indoor thermal comfort degree through the building thermal inertia modelling. Based on the thermal comfort estimation, a dayahead TCL scheduling model is formulated, which consists of 3 stages: TCL aggregator estimates maximal controllable TCL capacities at each scheduling time interval by solving a optimization model;[ the system operator performs the day-ahead system dispatch to determine the load shedding instruction for each aggregator;and ′the TCL aggregator schedules the ON/OFFcontrol actions of the TCL groups based on the instruction from the system operator. 14Accelerating the discontinuous Galerkin method for seismic wave propagation simulations using multiple GPUs with CUDA and MPI显示文摘We have successfully ported an arbitrary highorder discontinuous Galerkin method for solving the threedimensional isotropic elastic wave equation on unstructured tetrahedral meshes to multiple Graphic Processing Units(GPUs)using the Compute Unified Device Architecture(CUDA)of NVIDIA and Message Passing Interface(MPI)and obtained a speedup factor of about 28.3 for the single-precision version of our codes and a speedup factor of about 14.9 for the double-precision version.The GPU used in the comparisons is NVIDIA Tesla C2070 Fermi,and the CPU used is Intel Xeon W5660.To effectively overlap inter-process communication with computation,we separate the elements on each subdomain into inner and outer elements and complete the computation on outer elements and fill the MPI buffer first.While the MPI messages travel across the network,the GPU performs computation on inner elements,and all other calculations that do not use information of outer elements from neighboring subdomains.A significant portion of the speedup also comes from a customized matrix–matrix multiplication kernel,which is used extensively throughout our program.Preliminary performance analysis on our parallel GPU codes shows favorable strong and weak scalabilities.Dawei Mu Po Chen Liqiang Wang 2013Earthquake Science2013,26,6:2
15Silk-based hydrogel incorporated with metal-organic framework nanozymes for enhanced osteochondral regeneration显示文摘Osteochondral defects(OCD)cannot be efficiently repaired due to the unique physical architecture and the pathological microenvironment including enhanced oxidative stress and inflammation.Conventional strategies,such as the control of implant microstructure or the introduction of growth factors,have limited functions failing to manage these complex environments.Here we developed a multifunctional silk-based hydrogel incorporated with metal-organic framework nanozymes(CuTA@SF)to provide a suitable microenvironment for enhanced OCD regeneration.The incorporation of CuTA nanozymes endowed the SF hydrogel with a uniform microstructure and elevated hydrophilicity.In vitro cultivation of mesenchymal stem cells(MSCs)and chondrocytes showed that CuTA@SF hydrogel accelerated cell proliferation and enhanced cell viability,as well as had antioxidant and antibacterial properties.Under the inflammatory environment with the stimulation of IL-1β,CuTA@SF hydrogel still possessed the potential to promote MSC osteogenesis and deposition of cartilage-specific extracellular matrix(ECM).The proteomics analysis further confirmed that CuTA@SF hydrogel promoted cell proliferation and ECM synthesis.In the full-thickness OCD model of rabbit,CuTA@SF hydrogel displayed successfully in situ OCD regeneration,as evidenced by micro-CT,histology(HE,S/O,and toluidine blue staining)and immunohistochemistry(Col I and aggrecan immunostaining).Therefore,CuTA@SF hydrogel is a promising biomaterial targeted at the regeneration of OCD.Zhicheng Cao Hongmei Wang Jialin Chen Yanan Zhang Qingyun Mo Po Zhang Mingyue Wang Haoyang Liu Xueyang Bao Yuzhi Sun Wei Zhang Qingqiang Yao 2023Bioactive Materials2023,,2:2
