2Umbilical cord-derived mesenchymal stem cells:strategies,challenges,and potential for cutaneous regeneration显示文摘Umbilical cord mesenchymal stem cells(MSCs)are a unique,accessible,and non-controversial source of early stem cells that can be readily manipulated.As the most common pluripotent cell,bone marrow-derived MSCs display limitations with the progress of stem cell therapy.By contrast,umbilical cord-derived cells,which have plentiful resources,are more accessible.However,several uncertain aspects,such as the effect of donor selection or culture conditions,long-term therapeutic effects,product consistency,and potential tumorigenicity,are the bottleneck in this clinical therapy.MSCs are predicted to undergo an unprecedented development in clinical treatment when a generally acknowledged criterion emerges.In the current paper,we highlight the application of umbilical cord-derived MSCs in skin therapies based on our previous studies,as well as the achievements of our peers in this field.This paper focuses on the strategies,challenges,and potential of this novel therapy.Siming Yang Sha Huang Changjiang Feng Xiaobing Fu 2012Frontiers of Medicine2012,6,1:2

11Engineering the viscoelasticity of gelatin methacryloyl(GelMA)hydrogels via small“dynamic bridges”to regulate BMSC behaviors for osteochondral regeneration显示文摘The dynamic extracellular matrix(ECM)constantly affects the behaviors of cells.To mimic the dynamics of ECM with controllable stiffness and energy dissipation,this study proposes a strategy in which a small molecule,3,4-dihydroxybenzaldehyde(DB),was used as fast'dynamic bridges'to construct viscoelastic gelatin methacryloyl(GelMA)-based hydrogels.The storage modulus and loss modulus of hydrogels were independently adjusted by the covalent crosslinking density and by the number of dynamic bonds.The hydrogels exhibited self-healing property,injectability,excellent adhesion and mechanical properties.Moreover,the in vitro results revealed that the viscous dissipation of hydrogels favored the spreading,proliferation,osteogenesis and chondrogenesis of bone marrow mesenchymal stem cells(BMSCs),but suppressed their adipogenesis.RNA-sequencing and immunofluorescence suggested that the viscous dissipation of hydrogels activated Yes-associated protein(YAP)by stabilizing integrinβ1,and further promoted nuclear translocation of smad2/3 andβ-catenin to enhance chondrogenesis and osteogenesis.As a result,the viscoelastic GelMA hydrogels with highest loss modulus showed best effect in cartilage and subchondral bone repair.Taken together,findings from this study reveal an effective strategy to fabricate viscoelastic hydrogels for modulating the interactions between cells and dynamic ECM to promote tissue regeneration.Changjiang Liu Qifan Yu Zhangqin Yuan Qianping Guo Xiting Liao Feng Han Tao Feng Guoping Liu Runze Zhao Zhuang Zhu Haijiao Mao Caihong Zhu Bin Li 2023Bioactive Materials2023,,7:1

16Synthesis and Immobilization of Polystyrene-b-Polyvinyltriethoxysilane Micelles显示文摘Diblock copolymers polystyrene-block-polyvinyltriethoxysilane(PS-b-PVTES) were synthesized via atom transfer radical polymerization(ATRP), which self-assembled into spherical micelles in solvent of THF-methanol mixtures. The self-assembled micelles were immobilized by cross-linking reaction of VTES in a shell layer of micelles. The chemical structures of block copolymers and morphology of micelles were characterized in detail. 17Cellular modulation by the mechanical cues from biomaterials for tissue engineering显示文摘Mechanical cues from the extracellular matrix(ECM)microenvironment are known to be significant in modulating the fate of stem cells to guide developmental processes and maintain bodily homeostasis.Tissue engineering has provided a promising approach to the repair or regeneration of damaged tissues.Scaffolds are fundamental in cell-based regenerative therapies.Developing artificial ECM that mimics the mechanical properties of native ECM would greatly help to guide cell functions and thus promote tissue regeneration.In this review,we introduce various mechanical cues provided by the ECM including elasticity,viscoelasticity,topography,and external stimuli,and their effects on cell behaviours.Meanwhile,we discuss the underlying principles and strategies to develop natural or synthetic biomaterials with different mechanical properties for cellular modulation,and explore the mechanism by which the mechanical cues from biomaterials regulate cell function toward tissue regeneration.We also discuss the challenges in multimodal mechanical modulation of cell behaviours and the interplay between mechanical cues and other microenvironmental factors.Qiang Wei Shenghao Wang Feng Han Huan Wang Weidong Zhang Qifan Yu Changjiang Liu Luguang Ding Jiayuan Wang Lili Yu Caihong Zhu Bin Li 2021Biomaterials Translational2021,2,4:0

19Capacity of human umbilical cord-derived mesenchymal stem cells to differentiate into sweat gland-like cells:a preclinical study显示文摘Human umbilical cord-derived mesenchymal stem cells(hUC-MSCs)possess various advantageous properties,including self-renewal,extended proliferation potential,multi-lineage differentiation potential and capacity for differentiating into sweat gland-like cells in certain conditions.However,little is known about the effect of clinical-grade culture conditions on these properties and on the differentiative potential of hUC-MSCs.In this study,we sought to investigate the properties of hUC-MSCs expanded with animal serum free culture media(ASFCM)in order to determine their potential for differentiation into sweat gland-like cells.We found that primary cultures of hUC-MSCs could be established with ASFCM.Moreover,cells cultured in ASFCM showed vigorous proliferation comparable to those of cells grown in classical culture conditions containing fetal bovine serum(FBS).Morphology of hUC-MSCs cultured in ASFCM was comparable to those of cells grown under classical culture conditions,and hUC-MSCs grown in both of the two culture conditions tested showed the typical antigen profile of MSCs—positive for CD29,CD44,CD90,and CD105,and negative for CD34 and CD45,as expected.Chromosomal aberration assay revealed that the cells were stable after long-term culture under both culture conditions.Like normal cultured MSCs,hUC-MSCs induced under ASFCM conditions exhibited expression of the same markers(CEA,CK14 and CK19)and developmental genes(EDA and EDAR)that are characteristic of normal sweat gland cells.Taken together,our findings indicate that the classical culture medium used to differentiate hUC-MSCs into sweat gland-like cells can be replaced safely by ASFCM for clinical purposes.Siming Yang Kui Ma Changjiang Feng Yan Wu Yao Wang Sha Huang Xiaobing Fu 2013Frontiers of Medicine2013,7,3:0
