1Microwave Exposure Impairs Synaptic Plasticity in the Rat Hippocampus and PC12 Cells through Over-activation of the NMDA Receptor Signaling Pathway显示文摘Objective The aim of this study is to investigate whether microwave exposure would affect the N-methyl-D-aspartate receptor(NMDAR) signaling pathway to establish whether this plays a role in synaptic plasticity impairment. Methods 48 male Wistar rats were exposed to 30 m W/cm2 microwave for 10 min every other day for three times. Hippocampal structure was observed through H&E staining and transmission electron microscope. PC12 cells were exposed to 30 m W/cm2 microwave for 5 min and the synapse morphology was visualized with scanning electron microscope and atomic force microscope. The release of amino acid neurotransmitters and calcium influx were detected. The expressions of several key NMDAR signaling molecules were evaluated. Results Microwave exposure caused injury in rat hippocampal structure and PC12 cells, especially the structure and quantity of synapses. The ratio of glutamic acid and gamma-aminobutyric acid neurotransmitters was increased and the intracellular calcium level was elevated in PC12 cells. A significant change in NMDAR subunits(NR1, NR2 A, and NR2B) and related signaling molecules(Ca2+/calmodulin-dependent kinase II gamma and phosphorylated c AMP-response element binding protein) were examined. Conclusion 30 m W/cm2 microwave exposure resulted in alterations of synaptic structure, amino acid neurotransmitter release and calcium influx. 2A structural view of the antibiotic degradation enzyme NDM-1 from a superbug显示文摘Gram-negative Enterobacteriaceae with resistance to carbapenem conferred by New Delhi metallo-β-lactamase 1(NDM-1)are a type of newly discovered antibioticresistant bacteria.The rapid pandemic spread of NDM-1 bacteria worldwide(spreading to India,Pakistan,Europe,America,and Chinese Taiwan)in less than 2 months characterizes these microbes as a potentially major global health problem.The drug resistance of NDM-1 bacteria is largely due to plasmids containing the blaNDM-1 gene shuttling through bacterial populations.The NDM-1 enzyme encoded by the blaNDM-1 gene hydrolyzes β-lactam antibiotics,allowing the bacteria to escape the action of antibiotics.Although the biological functions and structural features of NDM-1 have been proposed according to results from functional and structural investigation of its homologues,the precise molecular characteristics and mechanism of action of NDM-1 have not been clarified.Here,we report the threedimensional structure of NDM-1 with two catalytic zinc ions in its active site.Biological and mass spectroscopy results revealed that D-captopril can effectively inhibit the enzymatic activity of NDM-1 by binding to its active site with high binding affinity.The unique features concerning the primary sequence and structural conformation of the active site distinguish NDM-1 from other reported metallo-β-lactamases(MBLs)and implicate its role in wide spectrum drug resistance.We also discuss the molecular mechanism of NDM-1 action and its essential role in the pandemic of drug-resistant NDM-1 bacteria.Our results will provide helpful information for future drug discovery targeting drug resistance caused by NDM-1 and related metallo-β-lactamases.Yu Guo Jing Wang Guojun Niu Wenqing Shui Yuna Sun Honggang Zhou Yaozhou Zhang Cheng Yang Zhiyong Lou Zihe Rao 2011Protein & Cell2011,2,5:16
3Predictive value of K-ras and PIK3CA in non-small cell lung cancer patients treated with EGFR-TKIs: a systemic review and meta-analysis显示文摘Objective: A meta-analysis was performed to augment the insufficient data on the impact of mutative EGFR downstream phosphatidylinositol-3-kinase(PI3K) and mitogen-activated protein kinase(MAPK) pathways on the clinical efficiency of epidermal growth factor receptor tyrosine kinase inhibitor(EGFR-TKI) treatment of non-small cell lung cancer(NSCLC) patients.Methods: Network databases were explored in April, 2015. Papers that investigated the clinical outcomes of NSCLC patients treated with EGFR-TKIs according to the status of K-ras and/or PIK3 CA gene mutation were included. A quantitative meta-analysis was conducted using standard statistical methods. Odds ratios(ORs) for objective response rate(ORR) and hazard ratios(HRs) for progression-free survival(PFS) and overall survival(OS) were calculated.Results: Mutation in K-ras significantly predicted poor ORR [OR =0.22; 95% confidence interval(CI), 0.13-0.35], shorter PFS(HR =1.56; 95% CI, 1.27-1.92), and shorter OS(HR =1.59; 95% CI, 1.33-1.91) in NSCLC patients treated with EGFR-TKIs. Mutant PIK3 CA significantly predicted shorter OS(HR =1.83; 95% CI, 1.05-3.20), showed poor ORR(OR =0.70; 95% CI, 0.22-2.18), and shorter PFS(HR =1.79; 95% CI, 0.91-3.53) in NSCLC patients treated with EGFR-TKIs.Conclusion: K-ras mutation adversely affected the clinical response and survival of NSCLC patients treated with EGFRTKIs. PIK3 CA mutation showed similar trends. 4Elevated Platelet Activating Factor Level in Ischemia-Related Arrhythmia and Its Electrophysiological Effect on Myocardium显示文摘Objective The mechanism through which platelet activating factor (PAF) induces cardiac electrical activity and arrhythmia is not well understood and previous studies have suggested a potential involvement of ion channels in its action. The present study was aimed to clarify the role of PAF in fatal arrhythmias following acute myocardia infarction (AMI) and the underlying mechanism. Methods (1) Blood PAF levels were measured among 72 AMI patients at the time of diagnosis with AMI and 48 h later, and their electrocardiogram (ECG) was recorded continuously. (2) Ischemia simulation and surface electrocardiogram were conducted in 20 pigs and their PAF levels were measured. (3) PAF perfusion and standard microelectrode recording were performed on guinea pig papillarymuscles. Results In both humans and pigs, elevated PAF levels were detected in AMI and simulated ischemia, respectively, and even higher PAF levels were found when fatal arrhythmias occurred. In guinea pig myocardium, PAF induced a shortening of action potential duration at 90% level of repolarization (APD 90 )under non-ischemic conditions and a more pronounced shortening under early simulated ischemic conditions. Conclusion AMI and ischemia are associated with increased PAF levels in humans and pigs, which are further raised when fatal arrhythmia follows. 5Effect of traditional Chinese medicine compounds on rumen fermentation, methanogenesis and microbial flora in vitro显示文摘This study was conducted to investigate the effects of traditional Chinese medicine compounds(TCMC)on rumen fermentation, methane emission and populations of ruminal microbes using an in vitro gas production technique. Cablin patchouli herb(CPH), Atractylodes rhizome(AR), Amur Cork-tree(AC) and Cypsum were mixed with the weight ratios of 1:1:1:0.5 and 1:1:1:1 to make up TCMC1 and TCMC2,respectively. Both TCMC were added at level of 25 g/kg of substrate dry matter. In vitro gas production was recorded and methane concentration was determined at 12 and 24 h of incubation. After 24 h, the incubation was terminated and the inoculants were measured for pH, ammonia nitrogen, volatile fatty acids(VFA). Total deoxyribonucleic acid of ruminal microbes was extracted from the inocula, and populations were determined by a real-time quantitative polymerase chain reaction. Populations of total rumen methanogens, protozoa, total fungi, Ruminococcus albus, Fibrobacter succinogenes and Ruminococcus flavefaciens were expressed as a proportion of total rumen bacterial 16 S ribosomal deoxyribonucleic acid. Compared with the control, the 2 TCMC decreased(P < 0.05) total VFA concentration,acetate molar proportion, acetate to propionate ratio, gas and methane productions at 12 and 24 h,hydrogen(H) produced and consumed, and methanogens and total fungi populations, while the 2 TCMC increased(P < 0.05) propionate molar proportion. Traditional Chinese medicine compound 1 also decreased(P ≤ 0.05) R. flavefaciens population. From the present study, it is inferred that there is an effect of the TCMC in suppressing methanogenesis, probably mediated via indirect mode by channeling H2 utilized for methanogenesis to synthesis of propionate and direct action against the rumen microbes involved in methane formation. 8Carbon and nitrogen stable isotope analyses of mammal bone fossils from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River,China显示文摘Based on AMS 14 C dating data,carbon and nitrogen isotope analyses were conducted on mammal bone collagen of deer,cattle and pigs from the Zhongba site in the Three Gorges Reservoir region of the Yangtze River.These analyses were conducted to reconstruct palaeodiets of mammals,palaeoecology,palaeoenviroment and previous human activities in the study area.Results show that the collagen loss of bone did not change the in vivo isotopic composition of carbon and nitrogen stable isotopes,and most of the bone fossils were well preserved.The bone collagen of samples from deer had a mean δ 13 C of-23.1‰ and a mean δ 15 N of 4.7‰,suggesting that deer subsisted in a closed habitat and fed on branches and leaves.The bone collagen of cattle had a mean δ 13 C of-19.6‰ and a mean δ 15 N of 5.2‰,which indicates that cattle subsisted in an open habitat and fed on grasses and stems.The δ 13 C values show that both deer and cattle fed on C 3 plants and lived in the same ecosystem,but the t-test results show that deer δ 13 C and δ 15 N values were both more negative than those of cattle,indicating that they inhabited different niches.The δ 13 C and δ 15 N values of cattle partially overlapped those of deer,suggesting some competition in diets between them.The t-tests show that the δ 13 C and δ 15 N values of pigs were more positive than those of cattle and deer,which signifies that pigs occupied a higher trophic level compared to cattle and deer.The wide range of pig δ 13 C values demonstrates that pig trading had been taking place from early Neolithic Age to late Bronze Age.There were no significant differences in deer δ 13 C and δ 15 N values among different archaeological periods,making it clear that climatic,ecological and environmental conditions were kept relatively stable from 2200 to 4200 a BP.This stability may have been responsible for the extensive and complete cultural layers at the Zhongba site.The minimum number of samples required to estimate the mean δ 13 C values of deer,pigs and cattle are 8,73 and 16,respectively,and for mean δ 15 N values of deer,pigs and cattle,the minimum numbers are 4,5 and 6,respectively.TIAN XiaoSi ZHU Cheng SUN ZhiBin SHUI Tao HUANG YunPing FLAD Rowan Kimon LI YuMei 2011Chinese Science Bulletin2011,56,2:2
9Enhancing carrier transport in flexible CZTSSe solar cells via doping Li strategy显示文摘The passivation of non-radiative states and inhibition of band tailings are desirable for improving the open-circuit voltage(V_(oc))of CZTSSe thin-film solar cells.Recently,alkali metal doping has been investigated to passivate defects in CZTSSe films.Herein,we investigate Li doping effects by applying Li OH into CZTSSe precursor solutions,and verify that carrier transport is enhanced in the CZTSSe solar cells.Systematic characterizations demonstrate that Li doping can effectively passivate non-radiative recombination centers and reduce band tailings of the CZTSSe films,leading to the decrease in total defect density and the increase in separation distance between donor and acceptor.Fewer free carriers are trapped in the band tail states,which speeds up carrier transport and reduces the probability of deep-level defects capturing carriers.The charge recombination lifetime is about twice as long as that of the undoped CZTSSe device,implying the heterojunction interface recombination is also inhibited.Besides,Li doping can increase carrier concentration and enhance build-in voltage,leading to a better carrier collection.By adjusting the Li/(Li+Cu)ratio to 18%,the solar cell efficiency is increased significantly to 9.68%with the fill factor(FF)of 65.94%,which is the highest FF reported so far for the flexible CZTSSe solar cells.The increased efficiency is mainly attributed to the reduction of V_(oc)deficit and the improved CZTSSe/Cd S junction quality.These results open up a simple route to passivate non-radiative states and reduce the band tailings of the CZTSSe films and improve the efficiency of the flexible CZTSSe solar cells.Qiong Yan Quanzhen Sun Hui Deng Weihao Xie Caixia Zhang Jionghua Wu Qiao Zheng Shuying Cheng 2022Journal of Energy Chemistry2022,,12:2
