| 1 | Productive HBV infection of well-differentiated, hNTCP-expressing human hepatoma-derived(Huh7) cells显示文摘Feasible and effective cell models for hepatitis B virus(HBV) infection are required for investigating the complete lifecycle of this virus, including the early steps of viral entry. Resistance to dimethyl sulfoxide/polyethylene glycol(DMSO/PEG), h NTCP expression, and a differentiated state are the limiting factors for successful HBV infection models. In the present study, we used a hepatoma cell line(Hu7^(hDNTCPh)) to overcome these limiting factors so that it exhibits excellent susceptibility to HBV infection. To achieve this goal, different hepatoma cell lines were tested with 2.5% DMSO/4%PEG8000, and one resistant cell line(Huh7 D) was used to construct a stable h NTCP-expressing cell line(Hu7^(hDNTCPh)) using a recombinant lentivirus system. Then, the morphological characteristics and differentiation molecular markers of Hu7^(hDNTCPh) cells with or without DMSO treatment were characterized. Finally, the susceptibility of Hu7^(hDNTCPh) cells to HBV infection was assessed. Our results showed that Huh7 D cells were resistant to 2.5% DMSO/4% PEG8000, whereas the others were not. Hu7^(hDNTCPh) cells were established to express a high level of h NTCP compared to liver extracts, and Hu7^(hDNTCPh) cells rapidly transformed into a non-dividing, well-differentiated polarized phenotype under DMSO treatment. Hu7^(hDNTCPh) cells fully supported the entire lifecycle of HBV infection. | 2 | Phosphatidylserine-Specific Phospholipase A1 is the Critical Bridge for Hepatitis C Virus Assembly显示文摘The phosphatidylserine-specific phospholipase A1(PLA1A)is an essential host factor in hepatitis C virus(HCV)assembly.In this study,we mapped the E2,NS2 and NS5A involved in PLA1A interaction to their lumenal domains and membranous parts,through which they form oligomeric protein complexes to participate in HCV assembly.Multiple regions of PLA1A were involved in their interaction and complex formation.Furthermore,the results represented structures with PLA1A and E2 in closer proximity than NS2 and NS5A,and strongly suggest PLA1 A-E2,s physical interaction in cells.Meanwhile,we mapped the NS5A sequence which participated in PLA1A interaction with the C-terminus of domain 1.Interestingly,these amino acids in the sequence are also essential for viral RNA replication.Further experiments revealed that these four proteins interact with each other.Moreover,PLA1A expression levels were elevated in livers from HCV-infected patients.In conclusion,we exposed the structural determinants of PLA1A,E2,NS2 and NS5A proteins which were important for HCV assembly and provided a detailed characterization of PLA1A in HCV assembly. | Qi Yang Min Guo Yuan Zhou Xue Hu Yun Wang Chunchen Wu Min Yang Rongjuan Pei Xinwen Chen Jizheng Chen | 2019 | Virologica Sinica2019,34,5: | 4 |
| 3 | Host HDAC4 regulates the antiviral response by inhibiting the phosphorylation of IRF3显示文摘Class II HDACs, such as HDAC4, are critical regulators of the immune response in various immune cells;however, its role in innate immunity remains largely unknown.Here, we report that the overexpression of HDAC4 suppresses the production of type I interferons triggered by pattern-recognition receptors (PRRs). HDAC4 repressed the translocation of transcription factor IRF3 to the nucleus, thereby decreasing IRF3-mediated IFN-β expression. In particular, we also determined that HDAC4 can be phosphorylated and simultaneously block the phosphorylation of IRF3 at Ser386 and Ser396 by TBK1 and IKKε, respectively, by interacting with the kinase domain of TBK1 and IKKε. Furthermore, IFN-β may stimulate the expression of HDAC4. | 4 | A practical method utilizing multi-spectral LiDAR to aid points cloud matching in SLAM显示文摘Light Detection and Ranging(LiDAR)sensors are popular in Simultaneous Localization and Mapping(SLAM)owing to their capability of obtaining ranging information actively.Researchers have attempted to use the intensity information that accompanies each range measurement to enhance LiDAR SLAM positioning accuracy.However,before employing LiDAR intensities in SLAM,a calibration operation is usually carried out so that the intensity is independent of the incident angle and range.The range is determined from the laser beam transmitting time.Therefore,the key to using LiDAR intensities in SLAM is to obtain the incident angle between the laser beam and target surface.In a complex environment,it is difficult to obtain the incident angle robustly.This procedure also complicates the data processing in SLAM and as a result,further application of the LiDAR intensity in SLAM is hampered.Motivated by this problem,in the present study,we propose a Hyperspectral LiDAR(HSL)-based-intensity calibration-free method to aid point cloud matching in SLAM.HSL employed in this study can obtain an eight-channel range accompanied by corresponding intensity measurements.Owing to the design of the laser,the eight-channel range and intensity were collected with the same incident angle and range.According to the laser beam radiation model,the ratio values between two randomly selected channels’intensities at an identical target are independent of the range information and incident angle.To test the proposed method,the HSL was employed to scan a wall with different coloured papers pasted on it(white,red,yellow,pink,and green)at four distinct positions along a corridor(with an interval of 60 cm in between two consecutive positions).Then,a ratio value vector was constructed for each scan.The ratio value vectors between consecutive laser scans were employed to match the point cloud.A classic Iterative Closest Point(ICP)algorithm was employed to estimate the HSL motion using the range information from the matched point clouds.According to the test results,we found that pink and green papers were distinctive at 650,690,and 720 nm.A ratio value vector was constructed using 650-nm spectral information against the reference channel.Furthermore,compared with the classic ICP using range information only,the proposed method that matched ratio value vectors presented an improved performance in heading angle estimation.For the best case in the field test,the proposed method enhanced the heading angle estimation by 72%,and showed an average 25.5%improvement in a featureless spatial testing environment.The results of the primary test indicated that the proposed method has the potential to aid point cloud matching in typical SLAM of real scenarios. | Changhui Jiang Yuwei Chen Wenxin Tian Ziyi Feng Wei Li Chunchen Zhou Hui Shao Eetu Puttonen Juha Hyyppä | 2020 | Satellite Navigation2020,1,1: | 2 |
| 5 | Repurposing of Antazoline Hydrochloride as an Inhibitor of Hepatitis B Virus DNA Secretion显示文摘Hepatitis B virus(HBV) belongs to Hepadnaviridae family and mainly infects hepatocytes, which can cause acute or chronic hepatitis. Currently, two types of antiviral drugs are approved for chronic infection clinically: interferons and nucleos(t)ide analogues. However, the clinical cure for chronic infection is still rare, and it is a huge challenge for all researchers to develop high-efficiency, safe, non-tolerant, and low-toxicity anti-HBV drugs. Antazoline hydrochloride is a first-generation antihistamine with anticholinergic properties, and it is commonly used to relieve nasal congestion and in eye drops. Recently, an in vitro high-throughput evaluation system was constructed to screen nearly 800 compounds from the Food and Drug Administration(FDA)-approved Drug Library. We found that arbidol hydrochloride and antazoline hydrochloride can effectively reduce HBV DNA in the extracellular supernatant in a dose-dependent manner, with EC_(50) of4.321 lmol/L and 2.910 lmol/L in HepAD38 cells, respectively. Moreover, the antiviral effects and potential mechanism of action of antazoline hydrochloride were studied in different HBV replication systems. The results indicate that antazoline hydrochloride also has a significant inhibitory effect on HBV DNA in the extracellular supernatant of Huh7 cells,with an EC_(50) of 2.349 lmol/L. | 6 | A Computational Study on Lateral Flight Stability of the Cranefly in Hover显示文摘The dynamic flight stability of hovering insects includes the longitudinal and lateral motion.Research results have shown that for the majority of hovering insects the same longitudinal natural modes are identified and the hovering flight in longitudinal is unstable.However,in lateral,the modal structure for hovering insects could be different and the stability property of lateral disturbance motion is not as robust as that of longitudinal motion.The cranefly possesses larger aspect ratio and lower Reynolds number,and such differences in morphology and kinematics may make the lateral dynamic stability different.In this paper,the lateral flight stability of the cranefly in hover is investigated by numerical simulation.Firstly,the stability derivatives are acquired by solving the incompressible Navier–Stokes equations.Subsequently,the dynamic stability characteristics are checked by analyzing the eigenvalues and eigenvectors of the linearized system.Computational results indicate that the lateral dynamic modal structure of cranefly is different from most other insects,consisting of three natural modes,and the weakly oscillatory mode illustrates the hovering lateral flight is nearly neutral.This neutral stability is mainly caused by the negative derivative of roll-moment vs.sideslip-velocity,which can be attributed to the weaker‘changingLEV-axial-velocity’effect.These results suggest that insects in nature may exhibit different dynamic stabilities with different morphological and kinematic parameters,which should be considered in the designs of flapping wing air vehicles. | Na Xu Shuaizhi Zhou Chunchen Zhang Xiaolei Mou | 2021 | Computer Modeling in Engineering & Sciences2021,,8: | 0 |
