1microRNA-218 suppresses the proliferation, invasion and promotes apoptosis of pancreatic cancer cells by targeting HMGB1显示文摘Objective: To detect the expression profiles of micro RNA-218(mi R-218) in human pancreatic cancer tissue(PCT) and cells and their effects on the biological features of human pancreatic cancer cell line PANC-1 and observe the effect of mi R-218 on the expression of the target gene high mobility group box 1(HMGB1), with an attempt to provide new treatment methods and strategies for pancreatic cancer.Methods: The expressions of mi R-218 in PCT and normal pancreas tissue as well as in various pancreatic cancer cell lines including As PC-1, Bx PC-3, and PANC-1 were determined with quantitative real-time reverse transcription polymerase chain reaction(q RT-PCR). The change of mi R-218 expression in PANC-1 cells was detected using qR T-PCT after the transfection of miR-218 mimic for 48 h. Cell Counting Kit-8(CCK-8) was applied for detecting the effect of mi R-218 on the activity of PANC-1 cells. The effects of mi R-218 on the proliferation and apoptosis of PANC-1 cells were analyzed using the flow cytometry. The effect of mi R-218 on the migration of PANC-1 cells was detected using the Trans-well migration assay. The HMGB1 was found to be a target gene of mi R-218 by luciferase reporter assay, and the effect of mi R-218 on the expression of HMGB1 protein in cells were determined using Western blotting.Results: As shown by q RT-PCR, the expressions of mi R-218 in PCT and in pancreatic cancer cell line significantly decreased when compared with the normal pancreatic tissue(NPT)(P<0.01). Compared with the control group, the miR-218 expression significantly increased in the PANC-1 group after the transfection of mi R-218 mimic for 48 h(P<0.01). Growth curve showed that the cell viability significantly dropped after the overexpression of mi R-218 in the PANC-1 cells for two days(P<0.05). Flow cytometry showed that the S-phase fraction significantly dropped after the overexpression of mi R-218(P<0.01) and the percentage of apoptotic cells significantly increased(P<0.01). As shown by the Trans-well migration assay, the enhanced mi R-218 expression was associated with a significantly lower number of cells that passed through a Transwell chamber(P<0.01). Luciferase reporter assay showed that, compared with the control group, the relative luciferase activity significantly decreased in the mi R-218 mimic group(P<0.01). As shown by the Western blotting, compared with the control group, the HMGB1 protein expression significantly decreased in the PANC-1 group after the transfection of mi R-218 mimic for 48 h(P<0.01).Conclusions: The mi R-218 expression decreases in human PCT and cell lines. mi R-218 can negatively regulate the HMGB1 protein expression and inhibit the proliferation and invasion of pancreatic cancer cells. 2Ethnobotany and diversity of medicinal plants used by the Buyi in eastern Yunnan, China显示文摘The Buyi are a socio-linguistic group in Yunnan Province of southwest China that have a long history of using medicinal plants as part of their indigenous medical system.Given the limited written documentation of the Buyi indigenous medical system,the objective of this paper is to document the medicinal plants of the Buyi and associated traditional knowledge and transmission.Field research was conducted in four villages in Lubuge Township of Luoping County in Yunnan Province using ethnobotanical methodologies including participatory observation,semi-structured interviews,key informant interviews,and focus group discussions to elicit information on medicinal plants.In total,120 informants(including 15 key informants who are healers)were interviewed.This study found that a total of 121 medicinal plant species belonging to 64 families are used by the Buyi including by local healers to treat different diseases.Among the medicinal plants recorded in this study,56 species(46%)have not previously been documented in the scientific literature as having medicinal value,highlighting the pressing need for ethnobotanical documentation in indigenous communities.The most frequently used medicinal part was the leaf(24.9%of documented plants),and the most common preparation method was decoction(62.8%of medicinal).Medicinal plants were mainly used to treat rheumatism(12.4%of plants),trauma and injuries(9.6%).The documented plants are also used for other non-medicinal purposes including food,fodder,fencing,and ornamental.In addition,35 of the medicinal plants are considered poisonous and are used by local Buyi healers for medicine.The traditional Buyi beliefs and practices associated with the documented medicinal plants likely contributes to their conservation in the environments and around Buyi communities.This study further highlights that ethnomedicinal knowledge of the Buyi is at risk of disappearing due to increased introduction and use of modern medicine in Buyi communities,livelihood changes,rapid modernization,and urbanization.Research,policy,and community programs are urgently needed to conserve the biocultural diversity associated with the Buyi medical system including ethnobotanical knowledge towards supporting both environmental and human wellbeing.Yong Xiong Xueyi Sui Selena Ahmed Zhi Wang Chunlin Long 2020Plant Diversity2020,42,6:3
3microRNA-218 promotes gemcitabine sensitivity in human pancreatic cancer cells by regulating HMGB1 expression显示文摘Objective: The purpose of this study was to examine the effect of gemcitabine(GEM) on micro RNA-218(mi R-218) expression in human pancreatic cancer cells.Methods: Quantitative reverse transcription polymerase chain reaction(q RT-PCR) was performed to examine the differences in mi R-218 expression between the GEM-sensitive Bx PC-3 pancreatic cancer cells and GEM-resistant PANC-1 cells. The effect of GEM on the expression of mi R-218 in PANC-1 cells was also investigated. PANC-1 cells were transfected either with HMGB1 si RNA to knock down the expression of HMGB1 or with the recombinant HMGB1 expression vector(pc DNA3.1-HMGB1) to overexpress HMGB1. The effect of ectopic expression of HMGB1 on the apoptosis of mi R-218-transfected and GEMtreated PANC-1 cells was examined by flow cytometric analysis.Results: The mi R-218 expression level was lower in GEM-resistant PANC-1 cells compared to GEMsensitive Bx PC-3 cells(P<0.05). The percentage of apoptotic PANC-1 cells was significantly increased in the mi R-218 mimic + GEM group compared to the mimic ctrl + GEM group and the normal control group(P<0.01). The HMGB1 expression level was markedly decreased in PANC-1 cells transfected with HMGB1 si RNA but was significantly increased in PANC-1 cells transfected with the recombinant HMGB1 expression vector, pc DNA3.1-HMGB1(P<0.01). 4Folk nomenclature of plants in Cistanche deserticola-associated community in South Gobi, Mongolia显示文摘Cistanche deserticola is an important medicinal plant in Mongolia.Despite its significant role in local healing systems,little traditional knowledge had been reported.The present study investigated folk names of C.deserticola and other species of the same community in Umnugobi Province,South Gobi region of Mongolia,based on ethnobotanical approaches.The high correspondence between folk names and scientific names of plant species occurring in Cistanche-associated community shows the scientific meaning of folk nomenclature and classification in Mongolia.The Mongolian and folk names of plants were formed on the basis of observations and understanding of wild plants including their morphology,phenology and traditional uses as well.Results from this study will support the conservation of C.deserticola itself,a rare and endangered plant species listed in the Monglian Red Data Book.Our documentation of folk nomenclature based on 96 plant species in the Cistanche community,as a part of traditional knowledge associated with biodiversity,will be very helpful for making strategy of plant biodiversity conservation in Mongolia.Urtnasan Mandakh Munkhjargal Battseren Danzanchadav Ganbat Turuutuvshin Ayanga Zolzaya Adiya Almaz Borjigidai Chunlin Long 2020Plant Diversity2020,42,6:1

6Fat deposition deficiency is critical for the high mortality of pre-weanling newborn piglets显示文摘Background: The high mortality of pre-weanling piglets is a dominant challenge which severely restricts the development of pig industry. A number of factors including nutrients imbalance and temperature variation during postnatal period of piglets have been reported to closely associated with the high mortality of postnatal piglets.This study aims to find out the relationship between fat deposition and survival of newborn piglets.Results: There were no differences in organ coefficient and bone density between the surviving and dead piglets(P > 0.05). The body weight and the fat deposition in the dead piglets were lower than the live individuals(P < 0.05).Consistently, the average sizes of white adipocytes in back and abdominal adipose tissues of dead piglets were smaller than the survivals(P < 0.05). The protein expression levels of adipocyte differentiation markers PPARγ and C/EBPα in the back and abdominal adipose tissues were lower in dead piglets compared to live piglets. The mRNA expressions of thermogenic markers PGC1α and PRDM16 in adipose tissues were decreased in the dead piglets(P < 0.05). The microarray of back fat samples from the surviving and dead piglets were conducted; two down-regulated genes namely AAMDC and CASTOR1 were identified from the dead piglets. According to quantitative real-time PCR(RT-PCR) analysis, the mRNA expression of AAMDC decreased, whereas CASTOR1 expression elevated in the dead piglets compared to the surviving piglets(P < 0.05).Conclusions: The fat deposition and adipocyte differentiation in the dead piglets are insufficient compared to the surviving piglets, which may attenuate the thermogenic ability of white adipose tissue(WAT).
