| 4 | Efficient editing DNA regions with high sequence identity in actinomycetal genomes by a CRISPR-Cas9 system显示文摘Actinobacteria able to produce varieties of bioactive natural products have been long appreciated by the field of drug discovery and development.Recently,a few of CRISPR/Cas9 systems bearing different types of replicons(pSG5 and pIJ101)were developed to efficiently edit their genomes.Despite wide application in gene editing,their utility in editing challenging DNA regions e.g.high sequence identity has not been compared.In this study,we confirmed that the widely used temperature-sensitive pSG5 replicon is indeed not suitable for editing modular polyketide synthase(PKS)genes due to causing unpredicted gene recombination.This problem can be addressed by replacing the pSG5 with the segregationally unstable pIJ101 replicon.By introducing a counterselection marker CodA,convenient cloning sites in the single guide RNAs(sgRNAs)and homologous template scaffolds,we developed a new CRISPR-Cas9 system pMWCas9.This system was successfully used to delete/replace erythromycin PKS and other biosynthetic genes in Saccharopolyspora erythraea and Streptomyces sp.AL2110.By swapping the promoters of antB and antC with ermE and kasOp,we achieved a deacyl-antimycin hyper producer which produces a 9-fold higher yield than the original Streptomyces sp.AL2110 strain.Our results provide a robust and useful Cas9 tool for genetic studies in Actinobacteria. | Jingjun Mo Shuwen Wang Wan Zhang Chunyu Li Zixin Deng Lixin Zhang Xudong Qu | 2019 | Synthetic and Systems Biotechnology2019,4,2: | 2 |
| 5 | Residential Appliance Detection Using Attentionbased Deep Convolutional Neural Network显示文摘Improving energy efficiency management has become an important task for current electricity market participating entities,and monitoring consumption of pivotal appliances plays an important role in many applications.This paper focuses on detecting whether a residence possesses a certain type of appliance based on their electricity consumption and the problem of class imbalance within deep learning model training for large power appliances with the state‘ON’.We propose a datadriven deep learning approach with attention mechanism to detect residential appliances from low-resolution aggregate energy consumption data.Firstly,the historical consumption profile of each user is divided into a specific length and labeled with the status of an appliance to generate training and test samples.Then,a deep convolutional neural network model with attention mechanism is trained,and the trained model is utilized to classify the test samples.Meanwhile,we obtain appliance status in a residence based on classification of multiple samples.Finally,we propose a novel approach of data generation for class imbalance of appliance detection using generative adversarial networks.In order to guarantee the quality,we devise a mechanism of self-validation to ensure generated data approximating real distribution of minor class samples.Experiments are conducted on a low-frequency smart meter data set sampled once every 30 minutes,and the results show that the proposed model performs better than hidden Markov model based algorithms and has good application prospects. | Chunyu Deng Kehe Wu Binbin Wang | 2022 | CSEE Journal of Power and Energy Systems2022,8,2: | 2 |

| 7 | Controlling the morphology, size and phase of Nb_2O_5 crystals for high electrochemical performance显示文摘The morphology, size and phase of the material play a crucial role in its electrochemical performance.Herein, the nano-sized niobium pentoxide(Nb_2O_5) with different morphologies and phase structures are synthesized through a very simple thermal treatment method, including the pseudo hexagonal Nb_2O_5 nanosheets and pseudo hexagonal Nb_2O_5 nanoparticles, orthorhombic Nb_2O_5 nanoparticles. The synthesized pseudohexagonal Nb2O5 nanosheets and orthorhombic Nb_2O_5 nanoparticles exhibit better cycling and rate performance than the pseudohexagonal Nb2 O5 nanoparticles due to the different morphologies and phase structures. The T-Nb_2O_5-700 nanoparticles show the higher capacity(175 mAh/g) than that of TT-Nb_2O_5-500 nanosheets(127 mAh/g) and TT-Nb_2O_5-600 nanoparticles(39 mAh/g) at a current density of 50 mA/g and good rate performance with a capacity of 140 mAh/g at 1.0 A/g. The excellent rate capability and cycling stability of orthorhombic T-Nb_2O_5 may be ascribed to the dominant contribution of pseudocapacitive effect. | 10 | Overcoming Perovskite Corrosion and De-Doping Through Chemical Binding of Halogen Bonds Toward Efficient and Stable Perovskite Solar Cells显示文摘4-tert-butylpyridine(TBP)is an indispensable additive for the hole transport layer in highly efficient perovskite solar cells(PSCs),while it can induce corrosion decomposition of perovskites and de-doping effect of spiro-OMeTAD,which present huge challenge for the stability of PSCs.Herein,halogen bonds provided by 1,4-diiodotetrafluorobenzene(1,4-DITFB)are employed to bond with TBP,simultaneously preventing perovskite decomposition and eliminating de-doping effect of oxidized spiro-OMeTAD.Various characterizations have proved strong chemical interaction forms between 1,4-DITFB and TBP.With the incorporation of halogen bonds,perovskite film can maintain initial morphology,crystal structure,and light absorbance;meanwhile,the spiro-OMeTAD film shows a relatively stable conductivity with good charge transport property.Accordingly,the device with TBP complex exhibits significantly enhanced stability in N_(2) atmosphere or humidity environment.Furthermore,a champion power conversion efficiency of 23.03%is obtained since perovskite is no longer damaged by TBP during device preparation.This strategy overcomes the shortcomings of TBP in n-i-p PSCs community and enhances the application potential of spiro-OMeTAD in fabricating efficient and stable PSCs. | Guanhua Ren Wenbin Han Qiang Zhang Zhuowei Li Yanyu Deng Chunyu Liu Wenbin Guo | 2022 | Nano-Micro Letters2022,14,11: | 0 |

The first strategy is based on the construction of an intermediate pseudo[1]rotaxane followed by the desired bis-[l]rotaxane. The other one is based on the construction of the desired bis-[1]rotaxane directly via a condensation reaction through host-guest interactions between a mono-functionalized pillar[5]arene and the axle. | 14 | Effects of seagrass leaf litter decomposition on sediment organic carbon composition and the key transformation processes显示文摘Seagrass leaf litters are an important source of sediment organic carbon(SOC). However, the mechanisms of seagrass leaf litter decomposition influencing SOC composition and the key transformation processes remain unknown. We performed a laboratory chamber experiment to compare the labile organic carbon(OC) composition and the enzyme activities governing SOC transformation between the seagrass group(seagrass leaf litter addition) and the control group. The results showed that the seagrass leaf litter decomposition significantly elevated the salt-extractable carbon(SEC) content and the SEC/SOC. Additionally,the levels of invertase, polyphenol oxidase, and cellulase in the seagrass leaf litters addition group were generally higher than in the control group, which could elevate recalcitrant OC decomposition. Following 24 days incubation, addition of seagrass leaf litter increased the amount of CO_2 released, but decreased the SOC content. | 16 | Sequence structure, sedimentary evolution and their controlling factors of the Jurassic Lianggaoshan Formation in the East Sichuan Basin, SW China显示文摘Based on the data of outcrops, seismic sections, thin sections, heavy mineral assemblages and detrital zircon U-Pb dating, the sedimentary characteristics, lake level fluctuation and provenance characteristics of the Middle Jurassic Lianggaoshan Formation(J_(2)l) in eastern Sichuan Basin, SW China, were investigated to reveal the control of tectonic movements of the surrounding orogenic belts on the sedimentary systems. The J_(2)lmainly developed a delta–lake sedimentary system, which contained a complete third-order sequence that was subdivided into four lake level up-down cycles(fourth-order sequence).The lake basins of cycles Ⅰ and Ⅱ were mainly distributed in eastern Sichuan Basin, while the lake basins of cycles Ⅲ and Ⅳ migrated to central Sichuan Basin, resulting in the significant difference in sedimentary characteristics between the north and the south of eastern Sichuan Basin. The provenance analysis shows that there were three types of provenances for J_(2)l. Specifically, the parent rocks of Type Ⅰ were mainly acidic igneous rocks and from the proximal northern margin of the Yangtze Plate;the parent rocks of Type Ⅱ were intermediate-acid igneous rocks and metamorphic rocks and from the central parts of the southern and northern Qinling orogenic belts;the parent rocks of Type Ⅲ were mainly metamorphic rocks followed by intermediate–acid igneous rocks, and from the North Daba Mountain area. It is recognized from the changes of sedimentary system and provenance characteristics that the sedimentary evolution of J_(2)lin eastern Sichuan Basin was controlled by the tectonic compression of the Qinling orogenic belt. In the early stage, the lake basin was restricted to the east of the study area, and Type Ⅰ provenance was dominant. With the intensifying north-south compression of the Qinling orogenic belt, the lake basin expanded rapidly and migrated northward, and the supply of Type Ⅱ provenance increased. | 17 | In-Orbit Calibration and Data Analysis of China's First Grazing Incidence Focusing X-Ray Pulsar Telescope显示文摘The grazing incidence focusing X-ray pulsar telescope(iFXPT), as the main payload of the X-ray Pulsar Navigation Test Satellite(XPNAV-1), will have great significance on China's space scientific exploration and X-ray pulsar navigation. With PSR B0531+21(Crab Pulsar) as the observation target, the pulsar profile has been recovered based on the data obtained by iFXPT, realizing the main objective of 'observing' PSR B0531+21 for the first time in China. This payload mainly consists of the Wolter-I X-ray optics, silicon drift detector, magnetic deflector, electronics, high-energy particle shield and high-stability structures. Currently, the iFXPT, with its good in-orbit performance, has obtained a considerable observation data. The effective area, sensitivity and energy response have been calibrated both on ground and in-orbit, demonstrating a high degree of consistency. Meanwhile, the in-orbit observation data and information for pulsar navigation has also been analyzed simultaneously.
