2Celastrol induces ferroptosis in activated HSCs to ameliorate hepatic fibrosis via targeting peroxiredoxins and HO-1显示文摘Ferroptosis is a form of regulated cell death, characterized by excessive membrane lipid peroxidation in an iron-and ROS-dependent manner. Celastrol, a natural bioactive triterpenoid extracted from Tripterygium wilfordii, shows effective anti-fibrotic and anti-inflammatory activities in multiple hepatic diseases. However, the exact molecular mechanisms of action and the direct protein targets of celastrol in the treatment of liver fibrosis remain largely elusive. Here, we discover that celastrol exerts anti-fibrotic effects via promoting the production of reactive oxygen species(ROS) and inducing ferroptosis in activated hepatic stellate cells(HSCs). By using activity-based protein profiling(ABPP) in combination with bio-orthogonal click chemistry reaction and cellular thermal shift assay(CETSA), we show that celastrol directly binds to peroxiredoxins(PRDXs), including PRDX1, PRDX2, PRDX4 and PRDX6,through the active cysteine sites, and inhibits their anti-oxidant activities. Celastrol also targets to heme oxygenase 1(HO-1) and upregulates its expression in activated-HSCs. Knockdown of PRDX1, PRDX2,PRDX4, PRDX6 or HO-1 in HSCs, to varying extent, elevated cellular ROS levels and induced ferroptosis. 3A cyclodextrin-based nanoformulation achieves co-delivery of ginsenoside Rg3 and quercetin for chemo-immunotherapy in colorectal cancer显示文摘The immune checkpoint blockade therapy has profoundly revolutionized the field of cancer immunotherapy. However, despite great promise for a variety of cancers, the efficacy of immune checkpoint inhibitors is still low in colorectal cancer(CRC). This is mainly due to the immunosuppressive feature of the tumor microenvironment(TME). Emerging evidence reveals that certain chemotherapeutic drugs induce immunogenic cell death(ICD), demonstrating great potential for remodeling the immunosuppressive TME. In this study, the potential of ginsenoside Rg3(Rg3) as an ICD inducer against CRC cells was confirmed using in vitro and in vivo experimental approaches. The ICD efficacy of Rg3 could be significantly enhanced by quercetin(QTN) that elicited reactive oxygen species(ROS). To amelioratein vivo delivery barriers associated with chemotherapeutic drugs, a folate(FA)-targeted polyethylene glycol(PEG)-modified amphiphilic cyclodextrin nanoparticle(NP) was developed for co-encapsulation of Rg3 and QTN. The resultant nanoformulation(CD-PEG-FA.Rg3.QTN) significantly prolonged blood circulation and enhanced tumor targeting in an orthotopic CRC mouse model, resulting in the conversion of immunosuppressive TME. Furthermore, the CD-PEG-FA.Rg3.QTN achieved significantly longer survival of animals in combination with Anti-PD-L1. 4Spatial-temporal Characteristics and Factors Influencing Commuting Activities of Middle-class Residents in Guangzhou City, China显示文摘The middle class in metropolitan Chinese cities has become an important social group. With the rapid development of urbanization and constant advancement of suburbanization, the middle class has increasingly come to influence city traffic. Research into middle-class commuting activities thus has practical significance for improving traffic congestion and reducing the commuting burden in metropolitan cities. Based on a dataset formed by 816 completed surveys, this paper analyzes the commuting mode, time and distance of middle-class residents in Guangzhou City using the descriptive statistical method. The results indicate that private cars are the main commuting mode, followed by public transport. Meanwhile, middle-class residents mainly undertake medium-short time and medium-short distance commuting. The study subsequently uses multilevel logistic regression and multiple linear regression models to analyze the factors that influence commuting mode choice, time and distance. The gender, age, number of family cars, housing source and jobs-housing balance are the most important factors influencing commuting mode choice; housing, population density, jobs-housing balance and commuting mode significantly affect commuting time; and transport accessibility, jobs-housing balance and commuting mode are the notable factors affecting commuting distance. Finally, this paper analyzes what is affecting the commuting activities of middle-class residents and determines the differences in commuting activity characteristics and influence factors between middle-class and ordinary residents. 6Heptanuclear brucite disk with cyanide bridges in a cocrystal and tracking its pyrolysis to an efficient oxygen evolution electrode显示文摘The development of efficient oxygen evolution reaction(OER)catalysts is still lacking in exploration of the mechanism of controlled pyrolysis of precursors among new material platforms.Here,a novel Co-based coordination molecular cluster has been first introduced as precursor to obtain metallic cobalt core shelled by N-doped carbon(Co@NC)structure which operates as an oxygen evolution electrode.Specifically,a new cocrystal compound,[Co7II(l3-CN)6(mmimp)6][CoIICl3N(CN)2]á3CH3OH(Co7+1,mmimp=2-methoxy-6-((methylimino)-methyl)phenol),was isolated consisting of Brucite disks of cobalt where the usual bridging l3-OH is replaced by l3-CN produced by the in-situ decomposition of dicyanamide(NC-N-CNà).The cobalt atoms are bonded through the nitrogen atom of the cyanide.Remarkably,time dependent thermogravimetric-mass spectrometry(TG-MS)analysis was utilized to track its pyrolysis process.It allowed us to propose a possible formation process of the Co@NC structure from Co7+1.Interestingly,an extremely superior OER electrode is optimized for Co@NC-600 having the lowest overpotential of257 m V at 10 m A/cm2in 1 mol/L KOH solution.The present study pins down the importance of clusters of transition metals on realizing distinct nanostructures operating as highly efficient OER electrocatalyst.Jian-Qiang Zhao Dandan Cai Jun Dai Mohamedally Kurmoo Xu Peng Ming-Hua Zeng 2019Science Bulletin2019,64,22:4
7Retina-Inspired Organic Heterojunction-Based Optoelectronic Synapses for Artificial Visual Systems显示文摘For the realization of retina-inspired neuromorphic visual systems which simulate basic functions of human visual systems,optoelectronic synapses capable of combining perceiving,processing,and memorizing in a single device have attracted immense interests.Here,optoelectronic synaptic transistors based on tris(2-phenylpyridine)iridium(Ir(ppy)_(3))and poly(3,3-didodecylquarterthiophene)(PQT-12)heterojunction structure are presented.The organic heterojunction serves as a basis for distinctive synaptic characteristics under different wavelengths of light.Furthermore,synaptic transistor arrays are fabricated to demonstrate their optical perception efficiency and color recognition capability under multiple illuminating conditions.The wavelength-tunability of synaptic behaviors further enables the mimicry of mood-modulated visual learning and memorizing processes of humans.More significantly,the computational dynamics of neurons of synaptic outputs including associated learning and optical logic functions can be successfully demonstrated on the presented devices.This work may locate the stage for future studies on optoelectronic synaptic devices toward the implementation of artificial visual systems.Junyao Zhang Yang Lu Shilei Dai Ruizhi Wang Dandan Hao Shiqi Zhang Lize Xiong Jia Huang 2021Research2021,,1:3
8Performance Analysis of Full-Duplex Based Two-Way Relaying显示文摘Cooperative communication is regarded as a promising technique for improving the reliability of wireless communication links and enhancing the radio coverage simultaneously. Unlike the conventional half-duplex(HD) mode relaying techniques,the full-duplex based two-way relaying(FD-TWR) enables data exchanges between two nodes to be completed within a single time-slot,thus resulting in a significant improvement in the spectrum efficiency. In this paper,the channel model of the FD-TWR is first given out,followed by deriving the critical performance metrics,including the received signal-to-interference-plus-noise ratio(SINR),the upper bound of the ergodic capacity and the closedform solution of the proposed FD-TWR under amplify-and-forward(AF) mode. Furthermore,taking the limit of sum-transmit-power into account,we formulate the objective function of the optimal power allocation of FD-TWR as an extreme-value problem by deriving the optimal transmit power for both the source nodes and the relay node. As long as the self-interference(SI) signal in the FD-TWR nodes can be sufficiently suppressed,the proposed scheme is shown to outperform the conventional HD mode in terms of both the ergodic capacity and the outage probability. 10Single cell analysis reveals inhibition of angiogenesis attenuates the progression of heterotopic ossification in Mkx^(−/−)mice显示文摘Tendon heterotopic ossification(HO)is characterized by bone formation inside tendon tissue,which severely debilitates people in their daily life.Current therapies fail to promote functional tissue repair largely due to our limited understanding of HO pathogenesis.Here,we investigate the pathological mechanism and propose a potential treatment method for HO.Immunofluorescence assays showed that the Mohawk(MKX)expression level was decreased in human tendon HO tissue,coinciding with spontaneous HO and the upregulated expression of osteochondrogenic and angiogenic genes in the tendons of Mkx^(−/−)mice.Single-cell RNA sequencing analyses of wild-type and Mkx^(−/−)tendons identified three cell types and revealed the excessive activation of osteochondrogenic genes during the tenogenesis of Mkx^(−/−)tendon cells.Single-cell analysis revealed that the gene expression program of angiogenesis,which is strongly associated with bone formation,was activated in all cell types during HO.Moreover,inhibition of angiogenesis by the small-molecule inhibitor BIBF1120 attenuated bone formation and angiogenesis in the Achilles tendons of both Mkx mutant mice and a rat traumatic model of HO.These findings provide new insights into the cellular mechanisms of tendon HO and highlight the inhibition of angiogenesis with BIBF1120 as a potential treatment strategy for HO.Junxin Lin Yuwei Yang Wenyan Zhou Chao Dai Xiao Chen Yuanhao Xie Shan Han Huanhuan Liu Yejun Hu Chenqi Tang Varitsara Bunpetch Dandan Zhang Yishan Chen Xiaohui Zou Di Chen Wanlu Liu Hongwei Ouyang 2022Bone Research2022,10,4:1
11Observation of CO2^++ dication in the dayside Martian upper atmosphere显示文摘Doubly charged positive ions(dications)are an important component of planetary ionospheres because of the large energy required for their formation.Observations of these ions are exceptionally difficult due to their low abundances;until now,only atomic dications have been detected.The Neutral Gas and Ion Mass Spectrometer(NGIMS)measurements made on board the recent Mars Atmosphere and Volatile Evolution mission provide the first opportunity for decisive detection of molecular dications,CO2^++in this case,in a planetary upper atmosphere.The NGIMS data reveal a dayside averaged CO2^++distribution declining steadily from 5.6 cm^−3 at 160 km to below 1 cm^−3 above 200 km.The dominant CO2^++production mechanisms are double photoionization of CO2 below 190 km and single photoionization of CO2^+at higher altitudes;CO2++destruction is dominated by natural dissociation,but reactions with atmospheric CO2 and O become important below 160 km.Simplified photochemical model calculations are carried out and reasonably reproduce the data at low altitudes within a factor of 2 but underestimate the data at high altitudes by a factor of 4.Finally,we report a much stronger solar control of the CO2^++density than of the CO2+density.Hao Gu Jun Cui DanDan Niu LongKang Dai JianPing Huang XiaoShu Wu YongQiang Hao Yong Wei 2020Earth and Planetary Physics2020,4,4:1

18Effect of electroacupuncture on inflammatory signal expression in local tissues of rats with chronic pelvic pain syndrome based on purinergic 2X7 receptor/NOD-like receptor pyrin domaincontaining 3 signal pathway显示文摘OBJECTIVES: To study the expression of inflammatory signal in local prostate tissue of chronic pelvic pain syndrome(CPPS) rats by electroacupuncture(EA) of Guanyuan(CV4), Zhongji(CV3), Huiyang(BL35) and Sanyinjiao(SP6), and to explore the possible mechanism of anti-inflammatory and analgesic effects of EA. METHODS : A total of 36 Sprague-Dawley male rats were randomly divided into three groups: control, model and EA(n=12 rats/group). The CPPS model was made by injection of CFA into ventral lobes of the prostate(0.1 m L). Electric acupuncture apparatus was applied to stimulate Guanyuan(CV4), Zhongji(CV3), bilateral Huiyang(BL35) and Sanyinjiao(SP6) acupoints in EA group. The general condition of rats was observed and the prostate index(PI) was calculated. The thermal pain threshold was collected after each therapeutic course. Histopathological changes of the prostate tissue were examined by hematoxylin-eosin staining method. The expression levels of tumor necrosis factor α(TNF-α), interleukin-1β(IL-1β) and prostaglandin E2(PGE2) in prostatic homogenates were measured by enzyme linked immunosorbent assay(ELISA). Moreover, the expression levels of purinergic 2X7 receptor(P2X7R), NOD-like receptor pyrin domain-containing 3(NLRP3), caspase-1 and interleukin-18(IL-18) m RNA were quantified by quantitative real-time polymerase chain reaction. RESULTS: Compared with control group, the PI of rats increased, and the thermal pain threshold decreased significantly in model group. The morphological structure of prostate tissues of rats in model group was severely damaged with a large number of inflammatory cells infiltration. Additionally, the levels of TNF-α, IL-1β and PGE2 were higher, and the expressions of P2X7R, NLRP3, caspase-1 and IL-18 m RNA were higher than those in control group. After EA treatment, the PI was significantly decreased, the thermal pain threshold was significantly increased, and the tissue damage was significantly improved. The expressions of inflammatory cytokines were lower in EA group, and expression of P2X7R/NLRP3 pathway was down-regulated. CONCLUSION: The effect of EA at Guanyuan(CV4), Zhongji(CV3), Huiyang(BL35) and Sanyinjiao(SP6) can improve inflammation and pain symptoms of CPPS rats induced by Complete Freund’s adjuvant(CFA). 19Data analysis of dose map verification for IMRT显示文摘In this paper we analyze the results of dose map verifications for patient’s IMRT (Intensity-modulated radiation therapy) plans and study the factors that may influence the accuracy of verification. MapCHECK, a two-dimensional diode array, was used to measure the dose maps for 1242 plans (14540 fields) from May 2004 to August 2008. The plans were designed with Pinnacle3 planning system. The passing rate of beams was determined when the acceptance criterion was 2%/2 mm, 3%/3 mm and 4%/4 mm. And the data with 3%/3 mm criteria was analyzed in more detail. The considered factors were beam modeling, optimization mode and treatment site. The median passing rate of total fields was 93.5%, 98.8%, and 100% when the acceptance criterion was 2%/2 mm, 3%/3 mm and 4%/4 mm, and the interquartile range were 11.1%, 3.8%, and 1.3%, respectively. The results of direct machine parameter optimization (DMPO) planning mode was better than those of multiple-step mode and beam modeling of new accelerators was better than that of old accelerators. These indicate that beam modeling is the key point of improving passing rate of IMRT verification and the influence of treatment site was little. 20Is Solar Wind electron precipitation a source of neutral heating in the nightside Martian upper atmosphere?显示文摘Solar Wind(SW)electron precipitation is able to deposit a substantial amount of energy in the nightside Martian upper atmosphere,potentially exerting an influence on its thermal structure.This study serves as the first investigation of such an issue,with the aid of the simultaneous measurements of both neutral density and energetic electron intensity made on board the recent Mars Atmosphere and Volatile Evolution(MAVEN)spacecraft.We report that,from a statistical point of view,the existing measurements do not support a scenario of noticeable neutral heating via SW electron precipitation.However,during 3%−4%of the MAVEN orbits for which data are available,strong correlation between nightside temperature and electron intensity is observed,manifested as collocated enhancements in both parameters,as compared to the surrounding regions.In addition,our analysis also indicates that neutral heating via SW electron precipitation tends to be more effective at altitudes below 160 km for integrated electron intensities above 0.01 ergs·cm^−2·s^−1 over the energy range of 3−450 eV.The results reported here highlight the necessity of incorporating SW electron precipitation as a heat source in the nightside Martian upper atmosphere under extreme circumstances such as during interplanetary coronal mass ejections.LongKang Dai Jun Cui DanDan Niu Hao Gu YuTian Cao XiaoShu Wu HaiRong Lai 2021Earth and Planetary Physics2021,5,1:0
