1miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. 2Effects of antisense CENP-B on the proliferation of HeLa (Tet-off) cells显示文摘In order to study the change of the expression of centromere protein B (CENP-B) caused by antisense transfection, proto-eukaryotically expressed fused protein GST-CENP-B (65 ku) was injected into mouse, and a peculiar anti-CENP-B serum MaCenpB was collected. A strain of transfected HeLa Tet-off cell HaCb, which contains antisense CENP-B expressing vector pBI-EGFP-as-CenpB, was prepared. Northern blot and Western blot were used to analyze the repression of internal CENP-B in transfected cells. According to the growth curve, the proliferation of HeLa (Tet-off) is repressed by antisense CENP-B, and the multiplication time is prolonged for 32.81 h. The analysis of flow cytometry revealed that, compared with HeLa (Tet-off), the G1 cell population of HaCb is increased (AG1=9%) while S fraction is decreased (AS=11%), but the G2/M phase is nearly unchanged (AG2/M=3%). In the meanwhile, the mi-totic index of HaCb declines greatly compared with that of HeLa (Tet-off). 3Two-dimensional self-healing hydrogen-bond-based supramolecula polymer film显示文摘Self-healing materials have been developed over the past decade with the recovery ability after damage.However,most researches focused on the self-healing process at three-dimension.Herein,we prepare monolayer self-healing hydrogen-bond-based supramolecular polymer film and explore the self-healing process at the two-dimensional limit.The healing process,which can be reversibly repeated for at least three times,is influenced by the temperature,the molecule-substrate interaction and the substrate roughness.In the application,the monolayer self-healing polymer film can be used to modify the SiO2 dielectric for copper phthalocyanine field effect transistor with improved mobility.This work will be valuable for developing two-dimensional functional self-healing materials in the future.Mengnan He Xiaosong Chen Donghua Liu Dacheng Wei 2019Chinese Chemical Letters2019,30,5:5
4A Fast and Memory-Efficient Approach to NDN Name Lookup显示文摘For name-based routing/switching in NDN, the key challenges are to manage large-scale forwarding Tables, to lookup long names of variable lengths, and to deal with frequent updates. Hashing associated with proper length-detecting is a straightforward yet efficient solution. Binary search strategy can reduce the number of required hash detecting in the worst case. However, to assure the searching path correct in such a schema, either backtrack searching or redundantly storing some prefixes is required, leading to performance or memory issues as a result. In this paper, we make a deep study on the binary search, and propose a novel mechanism to ensure correct searching path without neither additional backtrack costs nor redundant memory consumptions. Along any binary search path, a bloom filter is employed at each branching point to verify whether a said prefix is present, instead of storing that prefix here. By this means, we can gain significantly optimization on memory efficiency, at the cost of bloom checking before each detecting. 5Differential Expressing of Centromere Protein CenpG in Breast Cancer显示文摘Using indirect immunofluorescence (IIF), an anti-centromere protein CenpG-serum was verified. Western blot of the protein extracts of 31 samples of breast cancer tissues and their normal (not cancerous ) tissues a little far away from them in the same individuals showed that, in the majority of the tests (71%), centromere protein CenpG over expressed in breast cancer tissues. And moreover, a kind of protein component whose molecular weight is 43 kd, and which can be recognized by anti-CenpG serum was found in two of the cancer samples. 7China Antihypertensive Trial in Acute Ischemic Stroke II (CATIS-2): rationale and design显示文摘Backgrounds Increased blood pressure(BP)for patients who had an acute ischaemic stroke is associated with poor functional outcome,however the optimal timing of antihypertensive therapy is unknown.Aims We aim to compare early antihypertensive treatment to delayed antihypertensive treatment for reducing the risk of composite major disability and mortality at 3 months in acute ischaemic stroke.Design The China Antihypertensive Trial in Acute Ischemic Stroke II(CATIS-2)trial is a multicentre,randomised,open-label,blinded-endpoints trial that will be conducted in 100 hospitals in China.The primary outcome is the composite of death and major disability(modified Rankin Scale score≥3)at 3 months of randomisation.Antihypertensive treatment will be received immediately after randomisation in the early treatment group,aimed at average systolic BP by 10%-20%reduction within the first 24 hours,and achieving an average BP level of<140/90 mm Hg within 5 days.Patients in the delayed treatment group will discontinue any antihypertension medications for the first 7 days of randomisation,and will receive antihypertensive therapy achieving a BP goal of<140/90 mm Hg after 7 days.Conclusion The CATIS-2 trial will be testing the hypotheses that early BP lowering leads to improved functional outcome without any other harms,and developing clinical guidelines of the BP management for patients who had an acute ischaemic stroke.Liping Liu Yilong Wang Xuewei Xie Dacheng Liu Aili Wang Penglian Wang Suwen Shen Chongke Zhong Yufei Wei Tan Xu Yuesong Pan Yong Jiang Xia Meng David Wang Yonghong Zhang Jiang He Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:3
8SecIngress:An API Gateway Framework to Secure Cloud Applications Based on N-Variant System显示文摘Based on the diversified technology and the cross-validation mechanism,the N-variant system provides a secure service architecture for cloud providers to protect the cloud applications from attacks by executing multiple variants of a single software in parallel and then checking their behaviors’consistency.However,it is complex to upgrade current Software as a Service(SaaS)applications to adapt N-variant system architecture.Challenges arise from the inability of tenants to adjust the application architecture in the cloud environment,and the difficulty for cloud service providers to implement N-variant systems using existing API gateways.This paper proposes SecIngress,an API gateway framework,to overcome the challenge that it is hard in the cloud environment to upgrade the applications based on N-variants system.We design a two-stage timeout processing method to lessen the service latency and an Analytic Hierarchy Process Voting under the Metadata mechanism(AHPVM)to enhance voting accuracy.We implement a prototype in a testbed environment and analyze the security and performance metrics before and after deploying the prototype to show the effectiveness of SecIngress.The results reveal that SecIngress enhances the reliability of cloud applications with acceptable performance degradation.Dacheng Zhou Hongchang Chen Guozhen Cheng Weizhen He Lingshu Li 2021China Communications2021,18,8:2

14An electrical-coupling-suppressing MEMS gyroscope with feed-forward coupling compensation and scalable fuzzy control显示文摘This paper proposes a novel electrical coupling suppressing and drive closed loop control method for a MEMS gyroscope with feed-forward coupling compensation(FCC) and scalable fuzzy control. Theoretical analysis of the novel method is described in detail, and it is very simple to realize. Experimental results demonstrate that the electrical anti-resonant peaks located at the amplitude-frequency and phase-frequency responses are both eliminated by FCC control, and the height of the amplitude resonant peak increases more than 24 d B over 1800 Hz span. In addition, the overshoot of the transient response with scalable fuzzy control is smaller than 5%, and the settling time is less than 15 ms. The stabilities of the resonant amplitude and phase of the drive-mode velocity with scalable fuzzy control are about 15 ppm and 11 ppm, respectively. The scale factor of the gyroscope is measured to be 33.98 m V/deg/s with nonlinearity about 0.08%. 17A novel robust design method for the sense mode of a MEMS vibratory gyroscope based on fuzzy reliability and Taguchi design显示文摘This paper proposes a novel robust design method for the sense mode of a MEMS vibratory gyroscope based on fuzzy reliability and Taguchi design. The principles of fuzzy reliability and Taguchi design are both introduced and described in detail. Experimental results demonstrate that the signal to noise ratio of the robust design scheme is better than those of the other experimental schemes.Over the full temperature range from -40 to 80°C, the temperature sensitivities of phase margin, gain margin, sensitivity margin,the maximum amplitude of open loop system, bandwidth of closed loop system, and the performance function of the robust design system are all smaller than those of the original design system. Meanwhile, the temperature sensitivity of the bandwidth of the robust design system is improved to 126 from 1075 ppm/°C.
