1Development of a core set of SNP markers for the identifi cation of upland cotton cultivars in China显示文摘Considering the advantages of single nucleotide polymorphisms(SNP) in genotyping and variety identification, the first set public SNP markers at Cotton Marker Database(http://gffzz743c1d093f394620ho9xx66fw9kwp666w.ffgz.tsg.suse.edu.cn/) were validated and screened across standard varieties of cotton distinctness, uniformity and stability(DUS) test, aiming to obtain an appropriate set of core SNP markers suitable for upland cotton cultivars in China. A total of 399 out of 1 005 SNPs from 270 loci including 170 insertions-deletions(In Dels) were evaluated for their polymorphisms among 30 standard varieties using Sanger sequencing. As a result, 147 loci were sequenced successfully, 377 SNPs and 49 In Dels markers were obtained. Among the 377 SNP markers, 333 markers(88.3%) were polymorphic between Gossypium hirsutum and G. barbadense, while 164 markers(43.5%) were polymorphic within upland cotton. As for In Del markers, the polymorphic rate is relatively lower than that of SNP both between species and within species. The homozygous DNA locus ratio of 121 SNPs was higher than 86.2% while that of other 43 SNPs was less than 70%. Only 64 SNPs displayed completely homozygous genotypes among all of the detected upland cotton varieties with 100% homozygous DNA locus ratio. At last, a set of 23 pairs of core SNPs were achieved in view of avoidance of linkage, with polymorphism information content(PIC) values varying from 0.21 to 0.38 with an average of 0.28. Genotype characteristics and genetic diversity were analyzed based on the set of core markers, while 40 pairs of core simple-sequence repeats(SSR) primers comprised of 10 sets of four multiplex PCR combinations were also used for analysis based on fluorescence detection system. Comparison results indicated that the genetic diversity level was almost equal, while various varieties were significantly different from each other. Genetic relationship revealed by SSR markers is related to geographic source to a certain extent. 2Inhibitory effect of Salicornia europaea on the marine alga Skeletonema costatum显示文摘Exploiting the negative biochemical interference between plants and algal species has been suggested as a method to control harmful algal blooms.In this work,we investigated the inhibitory effect of the salt marsh halophyte Salicornia europaea against the marine alga Skeletonema costatum.S.europaea suppressed the growth of S.costatum in a nutrient-sufficient co-culture system,indicating that the inhibition of algal growth was because of the phytotoxic effect of S.europaea,rather than nutrient competition.We tested aqueous and organic extracts from S.europaea roots against S.costatum.The organic extracts inhibited growth and affected the cell size and chlorophyll a content of S.costatum in a dose-dependent manner.Among the three tested organic extracts,the methanol extract had the greatest effects on S.costatum,followed by butanol extract,and then the chloroform extract.Two flavonoids,rutin and quercetin-3-D-glucoside,were identified in the methanol extract by high performance liquid chromatography.The concentration of rutin was much higher than that of quercetin-3-D-glucoside.In an algal bioassay,rutin inhibited the growth of S.costatum and the inhibitory effect increased with increasing rutin concentration and with decreasing initial algal density.Therefore,we concluded that S.europaea negatively affects the growth of S.costatum,and that rutin,a metabolite of S.europaea,may play a role in this inhibitory effect.JIANG Dan HUANG LingFeng LIN YongQing NIE LingLing LV SuLian KUANG TingYun LI YinXin 2012Science China(Life Sciences)2012,55,6:8

4CGE model-based analysis of the neutralized hybrid carbon policy and its decomposed effects on economic growth,carbon reduction,and energy utilization costs显示文摘The hybrid policy is a flexible policy tool that combines features of carbon trading and carbon taxation.Its economic and environmental effects under China's background are still not studied in detail.Given the exogenous carbon reduction targets,carbon prices,and carbon tax-rates,by computable general equilibrium modeling methods and factor decomposition methods,this article investigates direct and cascaded effects of the hybrid policy on economic growth,energy utilization,and carbon emission on the national level and the sector level,with China's national input-output data-set.Stepwisely,policy scenarios with irrational estimated results are selectively excluded based on comprehensive evaluation among economic,carbon reduction and other policy targets.As a result,against national economic conditions in 2007,the hybrid policy,with a carbon reduction target of -10%,a carbon tax-rate of around $10,and a ceiling carbon price of $40,is highly recommended,because of its significant lower economic loss,lower energy utilization cost,and practical robustness against fluctuation of energy market and carbon market.Furthermore,by decomposition analysis,carbon reduction-related costs are decomposed into a direct part that includes carbon allowance price and carbon tax,and an indirect part as the energy price incremental induced by direct carbon costs.Gross carbon reduction may be decomposed into three parts such as energy intensity,economic scale,and technical progress.And,carbon taxation is the main policy tool that stimulates to improve the energy efficiency.Rui Sun Dan Kuang 2015Chinese Journal of Population,Resources and Environment2015,13,1:7
5Effect analysis of carbon trading on Economy-Energy-Environment system and calculation of reasonable carbon price intervals显示文摘This paper constructs a 4-lier computable general equilibrium model which includes such modules as modeling carbon emission constraints and carbon trading(CT),and incorporates the cost of carbon emissions into constant elasticity of substitute production function.Under scenario settings under different carbon abatement targets,based on 2007 national social accounting matrix and related statistical data about energy consumption and carbon emission,effects on economic outputs,energy consumption,and carbon abatement are estimated and analyzed at both macro and sector level.By calculating selected novel indicators that compromise between macroeconomic opportunity cost and achievable carbon abatement,reasonable carbon price intervals are given for enhancing the robustness and liquidity of carbon market.Further,by decomposition and share-weighted methods,expected carbon abatement and energy price are measured and analyzed in details.Some results are meaningful for fundamental design of the future carbon market.Given constant energy utilization and carbon abatement technologies at the macro level,the higher the carbon price the more actual carbon abatement;the more gross domestic product loss,the less energy consumption.Accwding to the overall situation estimated for 2007 in China,the advice given is to introduce a carbon abatement target rate(R_c)of-10%,which is helpful to make carbon market stable against unexpected carbon price shocks between[6.9,35]/tC with less economic loss.According to Kaya decomposition,after introduction of carbon pricing,carbon abatement is mainly contributed by the effects of energy intensity(EI)and technical progress.Further,CT may help reduce energy consumption and induce transformation to a low-carbon energy structure.At the sector level,the introduction of CT could induce economic recession in all sectors,especially energy.However,the overall economic structure remains unchanged to some extent.CT will help reduce energy consumption in all sectors,especially energy.Overall utilization costs of the energy composite can be divided in two,market price and carbonrelated costs.Carbon-related costs mainly contribute to variation in the utilization cos of the energy composite;carbon pricing may help non-energy sectors achieve sufficient carbon abatement by pushing up energy utilization cost.However,despite achievable carbon abatement by the energy sector being relatively high,induced by carbon pricing,there is still significant potential for other incentive policies to stimulate further abatement,such as energy resources taxation and transportation fuel taxation,especially in the sectors of coal and transportation.Finally,some advice is proposed in regard to policy decisions and further research.Rui Sun Dan Kuang Dongqin Chang 2015Chinese Journal of Population,Resources and Environment2015,13,2:5
6Neuroprotective mechanism of TMP269, a selective class ⅡA histone deacetylase inhibitor, after cerebral ischemia/reperfusion injury显示文摘TMP269 is a selective class ⅡA histone deacetylase inhibitor that has a protective effect on the central nervous system, whose specific mechanism of action is unclear. We aimed to reveal the optimal concentration of TMP269 for protecting against cerebral ischemia/reperfusion injury and its neuroprotective mechanism. Male Sprague-Dawley rats were randomly divided into sham, ischemia/reperfusion, and 1, 4, 10 and 16 mg/kg TMP269 groups. Cerebral ischemia/reperfusion injury was induced by middle cerebral artery occlusion. TMP269 was intraperitoneally administered at different doses 0.5 hours before ischemia induction. Western blot assay and immunohistochemistry were used to detect effects of TMP269 on histone 2 acetylation. The results showed that the level of histone 2 acetylation was increased 24 hours after TMP269 injection. 2,3,5-Triphenyltetrazolium chloride staining was utilized to examine effect of TMP269 on infarct volume. The results found that different doses of TMP269 could reduce the infarct volume. Western blot assay, immunohistochemistry and Evans blue staining were employed to measure the effect of TMP269 on blood-brain barrier. The results showed that TMP269 counteracted the abnormal endothelial cell permeability changes caused by cerebral ischemia/reperfusion. Western blot assay and immunohistochemistry were used to determine the effect of TMP269 on tissue kallikrein. The results found that TMP269 up-regulated the expression of tissue kallikrein. Western blot assay further determined the optimal concentration to be 4 mg/kg. In conclusion, TMP269 plays a neuroprotective role by up-regulating the level of histone 2 acetylation, alleviating endothelial cell injury after cerebral ischemia/reperfusion, and up-regulating the expression of tissue kallikrein. 7Prostate-specific membrane antigen expression in hepatocellular carcinoma,cholangiocarcinoma,and liver cirrhosis显示文摘BACKGROUND Primary liver cancer includes three subtypes:Hepatocellular carcinoma(HCC),intrahepatic cholangiocarcinoma(CCA),and combined hepatocellular carcinoma.Patients with primary liver cancer experienced poor prognosis and high mortality,so early detection of liver cancer and improved management of metastases are both key strategies to reduce the death toll from liver cancer.Prostate-specific membrane antigen(PSMA)expression in the tumor-associated neovasculature of nonprostate malignancies including liver cancer has been reported recently,but conclusive evidence of PSMA expression based on the pathological type of liver cancer remains limited.AIM To study the expression of PSMA in HCC,CCA,and liver cirrhosis.METHODS A total of 446 formalin-fixed paraffin-embedded(FFPE)liver tumor and liver cirrhosis tissue samples were obtained retrospectively from the Pathology Department of Tongji Hospital.Immunohistochemistry was used to detect PSMA expression in these 446 FFPE liver biopsy specimens(213 HCC,203 CCA,and 30 liver cirrhosis).The tumor compartment and the associated neovascular endothelium were separately analyzed.PSMA expression was examined by two certified pathologists,and the final results were presented in a 4-point scoring system(0-3 points).Correlation between PSMA expression and clinicopathological information was also assessed.RESULTS PSMA was expressed primarily in the neovascular endothelium associated with tumors.The positive rate of PSMA staining in HCC was significantly higher than that in CCA(86.8%vs 79.3%;P=0.001)but was only 6.6%in liver cirrhosis(P=0.000).HCC cases had more 3-score PSMA staining than CCA had(89/213,41.8%vs 35/203,17.2%;P=0.001).PSMA expression correlated positively with the stage and grade of HCC and CCA.In both liver cancer subtypes,there were more PSMA+cases in stages III–V diseases than in stages I and II.High staining intensity of PSMA was more frequently observed in liver cancers at high grade and advanced stage.There was no significant association of PSMA expression with sex,age,region,α-fetoprotein,hepatitis B surface antigen,or tumor size in both tumor subtypes.CONCLUSION Neovascular PSMA may be a promising marker to differentiate HCC from liver cirrhosis and a prognostic marker for anti-tumor angiogenesis therapy for HCC.Li-Xing Chen Si-Juan Zou Dan Li Jian-Yuan Zhou Zhao-Ting Cheng Jun Zhao Yuan-Li Zhu Dong Kuang Xiao-Hua Zhu 2020World Journal of Gastroenterology2020,26,48:3

10Selenium-enriched Cardamine violifolia protects against sepsis-induced intestinal injury by regulating mitochondrial fusion in weaned pigs显示文摘Sepsis is a life-threatening organ dysfunction caused by the dysregulated response of the host to an infection, and treatments are limited. Recently, a novel selenium source, selenium-enriched Cardamine violifolia(SEC) has attracted much attention due to its anti-inflammatory and antioxidant properties, but little is known about its role in the treatment of sepsis. Here, we found that SEC alleviated LPS-induced intestinal damage, as indicated by improved intestinal morphology, and increased disaccharidase activity and tight junction protein expression. Moreover, SEC ameliorated the LPS-induced release of pro-inflammatory cytokines, as indicated by decreased IL-6 level in the plasma and jejunum. Moreover, SEC improved intestinal antioxidant functions by regulating oxidative stress indicators and selenoproteins. In vitro, TNF-α-challenged IPEC-1 cells were examined and showed that selenium-enriched peptides, which are the main functional components extracted from Cardamine violifolia(CSP), increased cell viability, decreased lactate dehydrogenase activity and improved cell barrier function. Mechanistically, SEC ameliorated LPS/TNF-α-induced perturbations in mitochondrial dynamics in the jejunum and IPEC-1 cells. Moreover, CSPmediated cell barrier function is primarily dependent on the mitochondrial fusion protein MFN2 but not MFN1. 11Isochlorogenic acid A attenuates progression of liver fibrosis through regulating HMGB1/TLR4/NF-κB signaling pathways显示文摘OBJECTIVE Liver fibrosis is a chronic damage process related to the further progression of hepatic cirrhosis and has yet no truly effective treatment is available.This study aimed to investigate the effects of isochlorogenic acid A(ICQA)on liver fibrosis induced by carbon tetrachloride(CCl4)and clarify the underlying mechanism.METHODS Rats were treated with CCl4 for eight weeks in order to induce liver fibrosis and simultaneously orally administered with ICQA(10,20 and 40 mg·kg-1).RESULTS ICQA had significant protective effect on liver injury,inflammation,and fibrosis in rats.Meanwhile,ICQA prevented the activation of hepatic stellate cells(HSC)as indicated by inhibiting the overexpres⁃sion of a-smooth muscle actin(a-SMA).In addition,reduced fibrosis was found to be associated with decreased protein expression of high-mobility group box 1(HMGB1)and toll like receptor(TLR)4.Moreover,ICQA supressed the cytoplasmic translocation of HMGB1 in rat liver.Further investigations indicated that ICQA treatment significantly attenuated nuclear translocation of the nuclear factor-κB(NF-κB)p65 and inhibited degradation of IkBa expression in the liver of rats with liver fibrosis.CONCLUSION ICQA has hepatoprotective and anti-fibrotic effects in rats with liver fibrosis through modulating the HMGB1/TLR4/NF-κB signaling pathways.LIU Xin KUANG Kai MEI Dan ZHANG Bo 2019中国药理学与毒理学杂志2019,33,9:1

13Moxibustion modulates working memory in patients with amnestic mild cognitive impairment:a functional magnetic resonance imaging study显示文摘OBJECTIVE:To investigate the correlation between changes in brain activity associated with working memory and assessment scales of memory scores in amnestic mild cognitive impairment(aMCI)before and after moxibustion therapy.METHODS:aMCI patients were randomized into the moxibustion treatment(MT)group and the placebo moxibustion(PM)group.Each group received either moxibustion therapy or a placebo moxibustion for eight weeks.Neuropsychological performance and functional brain responses to a working memory task were assessed at baseline and at the end of treatment.Memory function was evaluated individually by the Rivermead behavioral memory test(RBMT),and working memory was assessed by the N-back task.RESULTS:Compared with the PM group,RBMT score changes were significant(P<0.05).In the MT group,the accuracy of the N-back texts increased compared with those before the intervention.After moxibustion intervention,the right insula,postcentral gyrus,precentral gyrus,superior temporal gyrus,thalamus,lingual gyrus,calcarine sulcus,posterior cingulate gyrus,middle frontal gyrus and anterior frontal gyrus were significantly activated(P=0.01,Cluster-level Family-Wise Error=0.05).Pearson correlation analysis showed that the insula,lingual gyrus and posterior cingulate gyrus were associated with changes in N-back score.Spearman correlation analysis showed that the postcentral gyrus,superior temporal gyrus,thalamus,lingual gyrus,and posterior cingulate gyrus were correlated with RBMT score changes.CONCLUSION:Moxibustion treatment improved memory in aMCI patients and was associated with the activation of the brain region of the insula,lingual gyrus,posterior cingulate gyrus,postcentral gyrus,superior temporal gyrus,and thalamus,which may be an important mechanism by which moxibustion improves the memory function.LI Xingjie LIU Qiqi XIA Rui LIU Jun WANG Dan SHI Jiao KUANG Yuxing DAI Yalan HUANG Haoyu TANG Wei CHEN Shangjie 2023Journal of Traditional Chinese Medicine2023,43,4:1

16Both IDO1 and TDO contribute to the malignancy of gliomas via the Kyn–AhR–AQP4 signaling pathway显示文摘Indoleamine 2,3-dioxygenase 1(IDO1),indoleamine 2,3-dioxygenase 2(IDO2),and tryptophan 2,3-dioxygenase(TDO)initiate the first step of the kynurenine pathway(KP),leading to the transformation of L-tryptophan(Trp)into L-kynurenine(Kyn)and other downstream metabolites.Kyn is known as an endogenous ligand of the aryl hydrocarbon receptor(AhR).Activation of AhR through TDO-derived Kyn is a novel mechanism to support tumor growth in gliomas.However,the role of IDO1 and IDO2 in this mechanism is still unknown.Herein,by using clinical samples,we found that the expression and activity of IDO1 and/or TDO(IDO1/TDO)rather than IDO2 were positively correlated with the pathologic grades of gliomas.The expression of IDO1/TDO rather than IDO2 was positively correlated with the Ki67 index and overall survival.The expression of IDO1/TDO was positively correlated with the expression of aquaporin 4(AQP4),implying the potential involvement of IDO1/TDO in glioma cell motility.Mechanistically,we found that IDO1/TDO accounted for the release of Kyn,which activated AhR to promote cell motility via the Kyn–AhR–AQP4 signaling pathway in U87MG glioma cells.RY103,an IDO1/TDO dual inhibitor,could block the IDO1/TDO–Kyn–AhR–AQP4 signaling pathway and exert anti-glioma effects in GL261 orthotopic glioma mice.Together,our results showed that the IDO1/TDO–Kyn–AhR–AQP4 signaling pathway is a new mechanism underlying the malignancy of gliomas,and suggest that both IDO1 and TDO might be valuable therapeutic targets for gliomas.Lisha Du Zikang Xing Bangbao Tao Tianqi Li Dan Yang Weirui Li Yuanting Zheng Chunxiang Kuang Qing Yang 2020Signal Transduction and Targeted Therapy2020,5,1:0
17Serum erythropoietin and transferrin in children with idiopathic nephrotic syndrome显示文摘Idiopathic nephrotic syndrome(INS)is char-acterized by marked urinary excretion of albumin and other intermediate-sized plasma proteins,such as trans-ferrin and vitamin D-binding protein.Some cases even develop anemia.The aim of this study was to investigate the changes in serum iron,transferrin,and erythropoietin,and the relationships between serum and urine transferrin and erythropoietin.Thirty-seven children with INS and 35 age-and sex-matched healthy children were investi-gated.The indexes related to iron metabolism,including serum iron,ferritin,transferrin,total iron-binding capa-city(TIBC),transferrin saturation,and hematological parameters[hemoglobin(Hb),mean corpuscular volume(MCV),mean corpuscular hemoglobin(MCH)],and urin-ary transferrin and erythropoietin were measured in 37 children with INS before treatment and at the remission stage.Thirty-five age-and sex-matched healthy children served as controls.Serum iron levels(18.8±3.8)mmol/L in INS patients before treatment were significantly lower than those of the healthy controls(22.2±3.8)mmol/L and those measured at the remission stage(21.0±3.5)mmol/L(all P<0.01).Serum transferrin levels in INS patients before therapy(1.9±0.3)g/L also decreased compared with the healthy controls(3.1±0.5)g/L and the measures at the remission stage(2.9±0.6)g/L(all P<0.01).In contrast,serum TIBC and transferrin saturation were sig-nificantly higher in INS patients before treatment than in the healthy controls[TIBC(56.4±9.2)mmol/L vs(50.7±6.8)mmol/L,P<0.01;transferrin saturation(55.7±9.2)%vs(46.4±8.2)%,P<0.01]and they were also higher than the measures at remission stage[(51.9±7.7)mmol/L and(47.4±13.3)mmol/L](all P<0.01).Serum transferrin was positively correlated with serum albumin(r= 50.609,P<0.01)and negatively correlated with urinary transferrin(r= 520.550,P<0.01)in INS patients before treatment.We conclude that serum iron,transferrin and erythropoietin levels are markedly decreased in INS patients,which may be partially related to the urinary loss of these indexes.Hongzhu LU Lin WANG Qihong FAN Dan LIU Wanming ZHANG Yuesha YUAN Hongyan KUANG 2008Frontiers of Medicine2008,2,3:0
