1Mechanisms of regulation of PFKFB expression in pancreatic and gastric cancer cells显示文摘Enzymes 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase-3 and-4(PFKFB-3 and PFKFB-4)play a significant role in the regulation of glycolysis in cancer cells as well as its proliferation and survival.The expression of these mRNAs is increased in malignant tumors and strongly induced in different cancer cell lines by hypoxia inducible factor(HIF)through active HIF binding sites in promoter region of PFKFB-4 and PFKFB-3 genes.Moreover,the expression and hypoxia responsibility of PFKFB-4 and PFKFB-3 was also shown for pancreatic(Panc1,PSN-1,and MIA PaCa-2)as well as gastric(MKN45 and NUGC3)cancer cells.At the same time,their basal expression level and hypoxia responsiveness vary in the different cells studied:the highest level of PFKFB-4 protein expression was found in NUGC3 gastric cancer cell line and lowest in Panc1 cells,with a stronger response to hypoxia in the pancreatic cancer cell line.Overexpression of different PFKFB in pancreatic and gastric cancer cells under hypoxic condition is correlated with enhanced expression of vascular endothelial growth factor(VEGF)and Glut1 mRNA as well as with increased level of HIF-1αprotein.Increased expression of different PFKFB genes was also demonstrated in gastric,lung,breast,and colon cancers as compared to corresponding nonmalignant tissue counterparts from the same patients,being more robust in the breast and lung tumors.Moreover,induction of PFKFB-4 mRNA expression in the breast and lung cancers is stronger than PFKFB-3mRNA.The levels of both PFKFB-4 and PFKFB-3 proteins in non-malignant gastric and colon tissues were more pronounced than in the non-malignant breast and lung tissues.It is interesting to note that Panc1and PSN-1 cells transfected with dominant/negative PFKFB-3(dnPFKFB-3)showed a lower level of endogenous PFKFB-3,PFKFB-4,and VEGF mRNA expressions as well as a decreased proliferation rate of these cells.Moreover,a similar effect had dnPFKFB-4.In conclusion,there is strong evidence that PFKFB-4 and PFKFB-3 isoenzymes are induced under hypoxia in pancreatic and other cancer cell lines,are overexpressed in gastric,colon,lung,and breast malignant tumors and undergo changes in their metabolism that contribute to the proliferation and survival of cancer cells.Thus,targeting these PFKFB may therefore present new therapeutic opportunities.Oleksandr H Minchenko Katsuya Tsuchihara Dmytro O Minchenko Andreas Bikfalvi Hiroyasu Esumi 2014World Journal of Gastroenterology2014,20,38:17
2A mathematical model of gas flow during coal outburst initiation显示文摘A proposed concept of outburst initiation examines the release of a large amount of gas from coal seams resulted from disintegrating thermodynamically unstable coal organic matter(COM).A coal microstructure is assumed to getting unstable due to shear component appearance triggered by mining operations and tectonic activities considered as the primary factor while COM disintegration under the impact of weak electric fields can be defined as a secondary one.The energy of elastic deformations stored in the coal microstructure activates chemical reactions to tilt the energy balance in a“coal–gas”system.Based on this concept a mathematical model of a gas flow in the coal where porosity and permeability are changed due to chemical reactions has been developed.Using this model we calculated gas pressure changes in the pores initiated by gas release near the working face till satisfying force and energy criteria of outburst.The simulation results demonstrated forming overpressure zone in the area of intensive gas release with enhanced porosity and permeability.The calculated outburst parameters are well combined with those evaluated by field measurements.Dmytro Rudakov Valeriy Sobolev 2019International Journal of Mining Science and Technology2019,29,5:8
3Fine equiaxed β grains and superior tensile property in Ti-6Al-4V alloy deposited by coaxial electron beam wire feeding additive manufacturing显示文摘Coarse columnar β grains result in anisotropic mechanical properties in Ti alloys deposited by additive manufacturing. This study reports that Ti-6Al-4V alloy fabricated by coaxial electron beam wire feeding additive manufacturing presents a weak anisotropy, high strength and ductility. The superior tensile property arises from a microstructure with fine equiaxed β grains(EGβ), discontinuous grain boundary α phase and short intragranular α lamellae. 6Engineering layered/spinel heterostructure via molybdenum doping towards highly stable Li-rich cathodes显示文摘Li-rich layered oxide(LLO),e.g.,Li_(1.12)[Mn_(0.56)Ni_(0.16)Co_(0.08)]O_(2)(LRMO),is considered as a promising cathode material due to its superior Li-storage capability.However,the poor cycling stability and large voltage decay,which are related to the phase transition,limit its industrialization process.Herein,a Mo-doped LRMO(Li_(1.12)[Mn_(0.56)Ni_(0.16)Co_(0.08)]_(0.98)Mo_(0.02)O_(2),LRMO-Mo2.0%)was successfully synthesized via a simple combination of co-precipitation with high-temperature calcination for solving the mentioned above-disadvantages.Compared with the pristine counterpart,the as-prepared LRMO-Mo2.0%shows more excellent electrochemical performance in terms of rate capability(reversible capacity of 118 mA·h·g^(−1) at 5 C),cyclic ability(94.3%capacity retention after 100 cycles at 0.2 C)and discharge midpoint voltage decay(0.11 V after 100 cycles).Systematic investigation of structural evolution and electrochemical kinetics elucidate that the synergic effect of robust oxygen framework and layered/spinel heterostructure is the key to its performance improvement.Such synergy helps to stabilize the layered structure by curbing the structural transformation and oxygen escaping during the electrochemical cycling.This work paved the way for the simple and efficient preparation of highly stable LLO cathode materials.Kun-Qi Geng Meng-Qian Yang Jun-Xia Meng Ling-Fei Zhou Yu-Qin Wang Sydorov Dmytro Qian Zhang Sheng-Wen Zhong Quan-Xin Ma 2022Tungsten2022,4,4:2

11A method for optimization of time intervals between rolling cuts on sorting humps显示文摘Sorting humps are the main technical means providing the breaking-and making-up of freight trains.Automation of sorting process by implementing microprocessor control systems is the main direction of increasing hump yard capacity, enhancement of safety in trains breaking-up, improvement of working conditions on hump yards. However, for wider introduction of such systems at the humps, it is necessary to reduce their cost.One of the main tasks of hump control systems is the determination of the optimal cars exit speeds from the retarders. Its solution is significantly complicated because of the uncertainty of cars rolling resistance coefficient,wind velocity and direction, retarder control errors. The paper is aimed at solving the problem of searching for such cars exit speeds from the retarders that provide the longest time intervals between them at the switches. Implementation of this method will make it possible to improve the quality of sorting process as a whole by improving the software of microprocessor control systems for automatic humping, rather than by increasing their technical facilities,which as a result will reduce the cost of their construction and operation.
