1A systematic investigation of high harmonic generation using mid-infrared driving laser pulses显示文摘We report on a systematic investigation of the influences of gas pressure,focal position and focusing geometry on high harmonic generation by use of mid-infrared femtosecond laser pulses. We also discuss the spatial characteristics of harmonics under different focusing conditions. 2Real-time emergency route generating algorithm in tunnel显示文摘As part of the digital mine system, a real time emergency route generating algorithm for a large scale metal mine is studied. The tunnel is abstracted and mathematically described by the center line model. A series of definitions are made and the center lines are regularized. In order to improve the quality of the final routes, a center line dataset preprocessing is done according to the factors including the slope threshold and the tunnel closed state information, etc. While in preprocessing, the mineshaft and shaft with the steep slope are excluded implicitly. The interface is preserved for point-like objects (e.g. blowers), directions (e.g. wind direction of blowers) and so on. The final path finding algorithm is optimized by the filter operation that the calculation is only performed at the endpoints and hub points, which can further reduce computing data amount. In our experiment, 3 368 nodes out of 22 401 nodes are selected as key nodes, therefore, the data processing amount of the algorithm is reduced to 1/7 and the routes can be found in real time. 4A Custom-Tailored Multi-TW Optical Electric Field for Gigawatt Soft-X-Ray Isolated Attosecond Pulses显示文摘Since the first isolated attosecond pulse was demonstrated through high-order harmonics generation(HHG)in 2001,researchers’interest in the ultrashort time region has expanded.However,one realizes a limitation for related research such as attosecond spectroscopy.The bottleneck is concluded to be the lack of a high-peak-power isolated attosecond pulse source.Therefore,currently,generating an intense attosecond pulse would be one of the highest priority goals.In this paper,we review our recent work of a TW-class parallel three-channel waveform synthesizer for generating a gigawatt-scale soft-X-ray isolated attosecond pulse(IAP)using HHG.By employing several stabilization methods,we have achieved a stable 50 mJ three-channel opticalwaveform synthesizer with a peak power at the multi-TW level.This optical-waveform synthesizer is capable of creating a stable intense optical field for generating an intense continuum harmonic beam thanks to the successful stabilization of all the parameters.Furthermore,the precision control of shot-to-shot reproducible synthesized waveforms is achieved.Through the HHG process employing a loose-focusing geometry,an intense shot-to-shot stable supercontinuum(50–70 eV)is generated in an argon gas cell.This continuum spectrum supports an IAP with a transform-limited duration of 170 as and a submicrojoule pulse energy,which allows the generation of a GW-scale IAP.Another supercontinuum in the soft-X-ray region with higher photon energy of approximately 100–130 eV is also generated in neon gas from the synthesizer.The transform-limited pulse duration is 106 as.Thus,the enhancement of HHG output through optimized waveform synthesis is experimentally proved.Bing Xue Yuuki Tamaru Yuxi Fu Hua Yuan Pengfei Lan Oliver D.Mücke Akira Suda Katsumi Midorikawa Eiji J.Takahashi 2021Ultrafast Science2021,,1:1
5Parental and transgenerational impairments of thyroid endocrine system in zebrafish by 2,4,6-tribromophenol显示文摘Many environmental contaminants could be transmitted fromparents and generate impairments to their progeny.The 2,4,6-tribromophenol(TBP),a novel brominated flame retardant which has been frequently detected in various organisms,was supposed to be bioaccumulated and intergenerational transmitted in human beings.Previous studies revealed that TBP could disrupt thyroid endocrine system in zebrafish larvae.However,there is no available data regarding the parental and transgenerational toxicity of this contaminant.Thus,in this study adult zebrafish were exposed to environmental contaminated levels of TBP for 60 days to investigate the parental and transgenerational impairments on thyroid endocrine system.Chemical analysis verified the bioaccumulation of TBP in tested organs of parents(concentration:liver>gonads>brain)and its transmission into eggs.For adults,increased thyroid hormones,disturbed transcriptions of related genes and histopathological changes in thyroid follicles indicate obvious thyroid endocrine disruptions.Transgenerational effects are indicated by the increased thyroid hormones both in eggs(maternal source)and in developed larvae(newly synthesized),aswell as disrupted transcriptional profiles of key genes in HPT axis.The overall results suggest that the accumulated TBP could be transmitted from parent to offspring and generate thyroid endocrine disruptions in both generations.Yuxi Zhou Juanjuan Fu Min Wang Yongyong Guo Lihua Yang Jian Han Bingsheng Zhou 2023Journal of Environmental Sciences2023,,2:1
6Peptide-drug conjugates(PDCs):a novel trend of research and development on targeted therapy,hype or hope?显示文摘Peptide-drug conjugates(PDCs)are the next generation of targeted therapeutics drug after antibody-drug conjugates(ADCs),with the core benefits of enhanced cellular permeability and improved drug selectivity.Two drugs are now approved for market by US Food and Drug Administration(FDA),and in the last two years,the pharmaceutical companies have been developing PDCs as targeted therapeutic candidates for cancer,coronavirus disease 2019(COVID-19),metabolic diseases,and so on.The therapeutic benefits of PDCs are significant,but poor stability,low bioactivity,long research and development time,and slow clinical development process as therapeutic agents of PDC,how can we design PDCs more effectively and what is the future direction of PDCs?This review summarises the components and functions of PDCs for therapeutic,from drug target screening and PDC design improvement strategies to clinical applications to improve the permeability,targeting,and stability of the various components of PDCs.This holds great promise for the future of PDCs,such as bicyclic peptide-toxin coupling or supramolecular nanostructures for peptide-conjugated drugs.The mode of drug delivery is determined according to the PDC design and current clinical trials are summarised.The way is shown for future PDC development.Chen Fu Lifeng Yu Yuxi Miao Xinli Liu Zhaojin Yu Minjie Wei 2023Acta Pharmaceutica Sinica B2023,13,2:1
7Inhibition of fibroblast activation protein ameliorates cartilage matrix degradation and osteoarthritis progression显示文摘Fibroblast activation protein(Fap)is a serine protease that degrades denatured type I collagen,α2-antiplasmin and FGF21.Fap is highly expressed in bone marrow stromal cells and functions as an osteogenic suppressor and can be inhibited by the bone growth factor Osteolectin(Oln).Fap is also expressed in synovial fibroblasts and positively correlated with the severity of rheumatoid arthritis(RA).However,whether Fap plays a critical role in osteoarthritis(OA)remains poorly understood.Here,we found that Fap is significantly elevated in osteoarthritic synovium,while the genetic deletion or pharmacological inhibition of Fap significantly ameliorated posttraumatic OA in mice.Mechanistically,we found that Fap degrades denatured type II collagen(Col II)and Mmp13-cleaved native Col II.Intra-articular injection of r Fap significantly accelerated Col II degradation and OA progression.In contrast,Oln is expressed in the superficial layer of articular cartilage and is significantly downregulated in OA.Genetic deletion of Oln significantly exacerbated OA progression,which was partially rescued by Fap deletion or inhibition.Intra-articular injection of r Oln significantly ameliorated OA progression.Taken together,these findings identify Fap as a critical pathogenic factor in OA that could be targeted by both synthetic and endogenous inhibitors to ameliorate articular cartilage degradation.Aoyuan Fan Genbin Wu Jianfang Wang Laiya Lu Jingyi Wang Hanjing Wei Yuxi Sun Yanhua Xu Chunyang Mo Xiaoying Zhang Zhiying Pang Zhangyi Pan Yiming Wang Liangyu Lu Guojian Fu Mengqiu Ma Qiaoling Zhu Dandan Cao Jiachen Qin Feng Yin Rui Yue 2023Bone Research2023,11,1:0
8Therapeutic potential of an anti-HER2 single chain antibody-DM1 conjugates for the treatment of HER2-positive cancer显示文摘Antibody–drug conjugates(ADCs)take the advantage of monoclonal antibodies to selectively deliver highly potent cytotoxic drugs to tumor cells,which have become a powerful measure for cancer treatment in recent years.To develop a more effective therapy for human epidermal growth factor receptor 2(HER2)-positive cancer,we explored a novel ADCs composed of anti-HER2 scFv–HSA fusion antibodies conjugates with a potent cytotoxic drug DM1.The resulting ADCs,T-SA1–DM1 and T-SA2–DM1(drug-to-antibody ratio in the range of 3.2–3.5)displayed efficient inhibition in the growth of HER2-positive tumor cell lines and the half-maximal inhibitory concentration on SKBR-3 and SKOV3 cells were both at the nanomolar levels in vitro.In HER2-positive human ovarian cancer xenograft models,T-SA1–DM1 and T-SA2–DM1 also showed remarkable antitumor activity.Importantly,three out of six mice exhibited complete remission without regrowth in the high-dose group of T-SA1–DM1.On the basis of the analysis of luminescence imaging,anti-HER2 scFv–HSA fusion antibodies,especially T-SA1,showed strong and rapid tumor tissue penetrability and distribution compared with trastuzumab.Collectively,the novel type of ADCs is effective and selective targeting to HER2-positive cancer,and may be a promising antitumor drug candidate for further studies.Hang Zhang Yuxi Wang Yangping Wu Xiaohua Jiang Yiran Tao Yuqin Yao Yujia Peng Xiangzheng Chen Yuyin Fu Lin Yu Ruixue Wang Qinhuai Lai Weirong Lai Wenting Li Yuhuan Kang Shuli Yi Ying Lu Lantu Gou Min Wu Jinliang Yang 2017Signal Transduction and Targeted Therapy2017,2,1:0
9Bisimulation Lattice of Asymmetric Chi Calculus with Mismatch显示文摘This paper carries out a systematic investigation into the bisimulation lattice of asymmetric chi calculus with a mismatch combinator. It is shown that all the sixty three L bisimilarities collapse to twelve distinct relations and they form a bisimulation lattice with respect to set inclusion. 10How faithfully can π be interpreted in SA?显示文摘The π calculus and the safe ambient calculus are two of the widely studied variants of process calculi in the field of concurrency theory.The former is the most classic model for mobile processes and the latter is well known for its nested structure.However,the relationship between these two models deserves further investigation.In this paper,we reinvestigate this problem thoroughly.We first give the strengthened encoding criteria.Then we propose the encoding of the synchronous π-calculus into the safe ambients calculus.The encoding scheme is a generalization and improvement of Levi and Sangiorgi's work when moving from the asynchronous π-calculus to the synchronous π-calculus.We show the merits of the encoding by checking the mentioned criteria.LONG Huan FU YuXi 2013Science China(Information Sciences)2013,56,3:0
