1mTOR/miR-145-regulated exosomal GOLM1 promotes hepatocellular carcinoma through augmented GSK-3b/MMPs显示文摘Golgi membrane protein 1(GOLM1/GP73)is a serum marker of hepatocellular carcinoma(HCC).We have previously shown that mTOR promoted tumorigenesis of HCC through stimulating GOLM1 expression.In this study,we demonstrated that the mammalian target of rapamycin(mTOR)was a negative regulator of microRNA-145(miR-145)expression.miR-145 inhibited GOLM1 expression by targeting a coding sequence of GOLM1 gene.GOLM1 and miR-145 were inversely correlated in human HCC tissues.GOLM1-enriched exosomes activated the glycogen synthase kinase-3β/matrix metalloproteinases(GSK-3β/MMPs)signaling axis of recipient cells and accelerated cell proliferation and migration.In contrast,miR-145 suppressed tumorigenesis and metastasis.We suggest that mTOR/miR-145/GOLM1 signaling pathway should be targeted for HCC treatment.Xiaochen Gai Bufu Tang Fangming Liu Yuting Wu Fang Wang Yanling Jing Fuqiang Huang Di Jin Ling Wang Hongbing Zhang 2019Journal of Genetics and Genomics2019,46,5:13
2Internet of Things to network smart devices for ecosystem monitoring显示文摘Smart, real-time, low-cost, and distributed ecosystem monitoring is essential for understanding and managing rapidly changing ecosystems. However, new techniques in the big data era have rarely been introduced into operational ecosystem monitoring, particularly for fragile ecosystems in remote areas.We introduce the Internet of Things(IoT) techniques to establish a prototype ecosystem monitoring system by developing innovative smart devices and using IoT technologies for ecosystem monitoring in isolated environments. The developed smart devices include four categories: large-scale and nonintrusive instruments to measure evapotranspiration and soil moisture, in situ observing systems for CO2 and d13 C associated with soil respiration, portable and distributed devices for monitoring vegetation variables, and Bi-CMOS cameras and pressure trigger sensors for terrestrial vertebrate monitoring. These new devices outperform conventional devices and are connected to each other via wireless communication networks. The breakthroughs in the ecosystem monitoring IoT include new data loggers and longdistance wireless sensor network technology that supports the rapid transmission of data from devices to wireless networks. The applicability of this ecosystem monitoring IoT is verified in three fragile ecosystems, including a karst rocky desertification area, the National Park for Amur Tigers, and the oasis-desert ecotone in China. By integrating these devices and technologies with an ecosystem monitoring information system, a seamless data acquisition, transmission, processing, and application IoT is created. 3Lotus seed skin proanthocyanidin extract exhibits potent antioxidant property via activation of the Nrf2-ARE pathway显示文摘Lotus seed is well known as traditional food and medicine,but its skin is usually discarded.Recent studies have shown that lotus seed skin contains a high concentration of proanthocyanidins that have multi-functions,such as antioxidation,anti-inflammation,and anti-cancer effects.In the present study,we aimed to isolate and purify the proanthocyanidins from lotus seed skin by acetone extraction and rotary evaporation,identify their chemical structures by HPLC-MS-MS and NMR,and further investigate the antioxidant properties of the extract purified by macroporous resin (PMR) from lotus seed skin both in vitro and in vivo.The results showed that PMR mainly contained oligomeric proanthocyanidins,especially dimeric procyanidin B1 (PB1),procyanidin B2 and procyanidin B4.Although it had limited ability to directly scavenge radicals in vitro,PMR could significantly enhance the expressions of antioxidant proteins via activation of nuclear factory-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway in HepG2 cells.Molecular data revealed that PB1,a major component in PMR,stabilized Nrf2 by inhibiting the ubiquitination of Nrf2,which led to subsequent activation of the Nrf2-ARE pathway,including the enhancements of Nrf2 nuclear translocation,Nrf2-ARE binding and ARE transcriptional activity.Moreover,the in vivo results in high fat diet-induced mice further verified the powerful antioxidant property of PMR.These results revealed that lotus seed skin is a promising resource for functional food development.Tao Li Qili Li Weiguo Wu Yong Li De-xing Hou Hua Xu Baodong Zheng Shaoxiao Zeng Yang Shan Xiangyang Lu Fangming Deng Si Qin 2019Acta Biochimica et Biophysica Sinica2019,51,1:8
4A disintegrin and metalloproteinase with thrombospondin motif 1(ADAMTS1) expression increases in acute aortic dissection显示文摘Acute aortic dissection(AAD) is a life-threatening cardiovascular disease caused by progressive medial degeneration of the aortic wall. A disintegrin and metalloproteinase with thrombospondin motifs 1(ADAMTS1) is a recently identified extracellular metalloproteinase participating in the development of vascular disease, such as atherosclerosis. In the present study, we found that ADAMTS1 was significantly elevated in blood samples from AAD patients compared with patients with acute myocardial infarction and healthy volunteers. Based on these findings, we established an AAD model by infusing angiotensin II in older mice. AAD was successfully developed in aorta tissues, with an incidence of 42% after 14 days in the angiotensin II group. Macrophage and neutrophil infiltration was observed in the media of the aorta, and ADAMTS1 overexpression was found in the aorta by Western blot and immunohistochemistry. Double immunofluorescence staining showed the expression of ADAMTS1 in macrophages and neutrophils. Consistent with the upregulation of ADAMTS1 in aortic dissection tissues, versican(a proteoglycan substrate of ADAMTS1) was degraded significantly more in these tissues than in control aortic tissues. 5Image formation of brain function in patients suffering from knee osteoarthritis treated with moxibustion显示文摘OBJECTIVES:Functional magnetic resonance imaging(fMRI) technology was used to study changes to the resting state blood flow in the brains of patients with knee osteoarthritis(KOA) before and after treatment with moxibustion at the acupoint of the left Dubi(ST 35) and to probe the cerebral mechanism underlying the effect of moxibustion.METHODS:The resting state brain function of 30 patients with left KOA was scanned with fMRI before and after treatment with moxibustion.The analytic methods of fractional amplitude of low frequency fluctuation(fALFF) and regional homogeneity(ReHo) were used to observe changes in resting state brain function.RESULTS:The fALFF values of the right cerebrum,extra-nucleus,left cerebellum,left cerebrum and white matter of patients after moxibustion treatment were higher than before treatment,and the fALFF values of the precentral gyrus,frontal lobe and occipital lobe were lower than before treatment(P<0.05,K≥85).The ReHo values of the thalamus,extra-nucleus and parietal lobe of patients were much higher than those before moxibustion treatment,and the ReHo values of the right cerebrum,left cerebrum and frontal lobe were lower than before treatment(P<0.05,K≥85).CONCLUSION:The influence of moxibustion on obvious changes in brain regions basically conforms to the way that pain and warmth is transmitted in the body,and the activation of sensitive systems in the body may be objective evidence of channel transmission.The regulation of brain function by moxibustion is not in a single brain region but rather in a network of many brain regions.Hongwu Xie Fangming Xu Rixin Chen Tianyou Luo Mingren Chen Weidong Fang Fajin Lü Fei Wu Yune Song Jun Xiong 2013Journal of Traditional Chinese Medicine2013,33,2:7
6Thick Miocene eolian deposits on the Huajialing Mountains:The geomorphic evolution of the western Loess Plateau显示文摘The geomorphic evolution of northwestern China during the Cenozoic has been a subject of much geological interest because of its link with the uplift of the Himalayan-Tibetan complex.Much information about these changes is recoverable from the sedimentary sequences of the region.We report here on the thick eolian deposits mantling the Huajialing Mountains,a relatively flat mountain range in the western Loess Plateau.Correlation of magnetic susceptibility stratigraphy with the QA-I Miocene eolian sequence dates a 134.7 m section(NL-VII) for the interval from 18.7 to 11.8 Ma,as confirmed by micro-mammalian fossils.These eolian deposits demonstrate a much wider distribution of the Miocene eolian deposits,and also indicate that the topography contrasts in the western Loess Plateau,including the uplifts of the Huajialing Mountains and the bedrock highlands in the Qinan region,were formed by the early Miocene.The near-continuous Miocene eolian sequence from 18.7 to 11.8 Ma indicates that the substratum of Huajialing had not experienced any intense tectonic changes during this time interval,which suggests further,the relative tectonic stability of the nearby Tibetan Plateau.ZHAN Tao GUO ZhengTang WU HaiBin GE JunYi ZHOU Xin WU ChunLin ZENG FangMing 2011Science China Earth Sciences2011,54,2:7
7Coagulation factors VII, IX and X are effective antibacterial proteins against drug-resistant Gram-negative bacteria显示文摘Infections caused by drug-resistant“superbugs”pose an urgent public health threat due to the lack of effective drugs;however, certain mammalian proteins with intrinsic antibacterial activity might be underappreciated. Here, we reveal an antibacterial property against Gram-negative bacteria for factors VII, IX and X, three proteins with well-established roles in initiation of the coagulation cascade. These factors exert antibacterial function via their light chains (LCs). Unlike many antibacterial agents that target cell metabolism or the cytoplasmic membrane, the LCs act by hydrolyzing the major components of bacterial outer membrane, lipopolysaccharides, which are crucial for the survival of Gram-negative bacteria. The LC of factor VII exhibits in vitro efficacy towards all Gram-negative bacteria tested, including extensively drug-resistant (XDR) pathogens, at nanomolar concentrations. It is also highly effective in combating XDR Pseudomonas aeruginosa and Acinetobacter baumannii infections in vivo. Through decoding a unique mechanism whereby factors VII, IX and X behave as antimicrobial proteins, this study advances our understanding of the coagulation system in host defense, and suggests that these factors may participate in the pathogenesis of coagulation disorder-related diseases such as sepsis via their dual functions in blood coagulation and resistance to infection. 9Prevalence, risk factors, and survival associated with pulmonary hypertension and heart failure among patients with underlying coronary artery disease: a national prospective, multicenter registry study in China显示文摘Background: Coronary artery disease (CAD) is the commonest cause of heart failure (HF), whereas pulmonary hypertension (PH) has not been established or reported in this patient population. Therefore, we assessed the prevalence, risk factors, and survival in CAD-associated HF (CAD-HF) complicated with PH.Methods: Symptomatic CAD-HF patients were continuously enrolled in this prospective, multicenter registry study. Echocardiography, coronary arteriography, left and right heart catheterization (RHC), and other baseline clinical data were recorded. Patients were followed up and their survival was recorded.Results: One hundred and eighty-two CAD-HF patients were enrolled, including 142 with HF with a preserved ejection fraction (heart failure with preserved ejection fraction [HFpEF];left ventricular ejection fraction [LVEF] ≥50%) and 40 with a reduced ejection fraction (heart failure with reduced ejection fraction [HFrEF];LVEF < 50%). PH was diagnosed with RHC in 77.5% of patients. Patients with PH showed worse hemodynamic parameters and higher mortality. HFrEF-PH patients had worse survival than HFpEF-PH patients. CAD-HF patients with an enlarged left ventricular end-diastolic diameter and reduced hemoglobin were at higher risk of PH. Nitrate treatment reduced the risk of PH. Elevated creatinine and mean pulmonary arterial pressure (mPAP), diastolic pressure gradient (DPG) ≥7 mmHg, and previous myocardial infarction (MI) entailed a higher risk of mortality in CAD-HF patients with PH.Conclusions: PH is common in CAD-HF and worsens the hemodynamics and survival in these patients. Left ventricle enlargement and anemia increase the risk of PH in CAD-HF. Patients may benefit from nitrate medications. 10Morphological and physiological analysis of narrow and striped leaf 1(nsl1) mutant of rice(Oryza sativa L.) and the gene mapping显示文摘The shape and color of rice leaves are important agronomic traits that directly influence the proportion of sunlight energy utilization and ultimately affect the yield and quality.A new mutant exhibiting stable inheritance was identified as derived from ethyl methane sulfonate(EMS)-treated restorer Jinhui 10,tentatively named as narrow and striped leaf 1(nsl1).The nsl1 displayed pale white leaves at the seeding stage and then white striped leaves in parallel to the main vein at the jointing stage.Meanwhile,its leaf blades are significantly narrower than the control group of Jinhui 10.The chloroplast structures of cells in the white striped area of the nsl1 mutant break down,and the photosynthetic pigments are significantly lower than that of the wild type.Moreover,fluorescence parameters,such as F0,F v/F m,U psII,qp,and ETR,in the nsl1 mutant are significantly lower than those of the wild type,and the photosynthetic efficiency is also significantly decreased.These changes in leaf color and shape,together with physiological changes in the nsl1,result in smaller plant height and a decrease in the most important agronomic traits,such as the number of grains per panicle,grain weight,etc.Genetic analysis shows that the narrow and striped traits of the nsl1 mutant are controlled by a single recessive nuclear gene,which is located between InDel 16and InDel 12 in chromosome 3.The physical distance is204 kb.So far,no similar genes of such leaf color and shape in this area have been reported.This study has laid a solid foundation for the gene cloning and function analysis of NSL1.Fangming Zhao Yong Zhang Yanru Wu Tongming Wang Ling Ma Zhenglin Yang Xia Wei Xianchun Sang Yinghua Ling Nan Wang Changwei Zhang Guanghua He 2014Chinese Science Bulletin2014,59,9:3
11Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors.Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang 2022Animal Models and Experimental Medicine2022,5,1:3
12Delphinidin-induced autophagy protects pancreatic β cells against apoptosis resulting from high-glucose stress via AMPK signaling pathway显示文摘Hyperglycemia,a diagnostic characteristic of diabetes mellitus,is detrimental to pancreatic 0 cells.Delphinidin,a member of the anthocyanin family,inhibits glucose absorption,increases glucagonlike peptide(GLP-1)secretion,and improves insulin secretion in diabetes.However,whether delphinidin plays a protective role in pancreatic p-cell mass and function is not clear.In this study,delphinidin was found to decrease the high-glucose-induced apoptosis of RIN-m5F pancreatic β cells.In addition,delphinidin induced autophagy in RIN-m5F cells under the normal and highglucose conditions,while 3-methyladenine(3-MA)inhibition of autophagy significantly diminished the protective role of delphinidin against high-glucose-induced apoptosis of pancreatic β cells.Delphinidin also decreased the level of cleaved caspase 3 and increased the phosphorylation level of AMP-activated protein kinase a(AMPKa)Thr172.Compound C,an AMPK inhibitor,was found to decrease the ratio of LC3-II/LC3-I,and the apoptotic rate of high-glucose-injured cells was in creased after treatme nt with delphinidin,in dicating that delphinidi n attenuated the n egative effects of high-glucose stress to cells.In con elusion,our data dem on strate that delphi nidin protects pancreatic B cells against high-glucose-induced injury by autophagy regulation via the AMPK signaling pathway.These findings might shed light on the underlying mechanisms of diabetes and help improve the prevention and therapy of this common disease.Dengni Lai Mingyong Huang Lingyan Zhao Yan Tian Yong Li Dongpo Liu Yanyang Wu Fangming Deng 2019Acta Biochimica et Biophysica Sinica2019,51,12:3

15Regulation of jasmonate signaling by reversible acetylation of TOPLESS in Arabidopsis显示文摘The plant hormone jasmonate(JA)regulates plant immunity and adaptive growth by orchestrating a genome-wide transcriptional program.Key regulators of JA-responsive gene expression include the master transcription factor MYc2,which is repressed by the conserved Groucho/Tup1-like corepressor TOPLESS(TPL)in the resting state.However,the mechanisms underlying TPL-mediated transcriptional repression of MYc2 activity and hormone-dependent switching between repression and de-repression remainenigmatic.Here,we report the regulation of TPLactivity and JAsignaling byreversibleacetylation of TPL.We found that the histone acetyltransferase GCN5 could mediate TPL acetylation,which enhances its interaction with the NOVEL-INTERACTOR-OF-JAZ(NINJA)adaptor and promotes its recruitment to MYc2 target promoters,facilitating transcriptional repression.Conversely,TPL deacetylation by the histonedeacetylase HDA6 weakens TPL-NINJA interaction and inhibitsTPL recruitmentto MYC2 target promoters,facilitating transcriptional activation.In the resting state,the opposing activities of GCN5 and HDA6 maintain TPL acetylation homeostasis,promoting transcriptional repression activity of TPL.In response to JA elicitation,HDA6 expression is transiently induced,resulted in decreased TPL acetylation and repressor activity,thereby transcriptional activation of MYC2 target genes.Thus,the GCN5-TPL-HDA6 module main tains the homeostasis of acetylated TPL,thereby determining the transcriptional state of JA-responsive genes.Our findings uncovered a mechanism by which the TPL corepressor activity in JA signaling is activelytuned inarapidandreversiblemanner.Chunpeng An Lei Deng Huawei Zhai Yanrong You Fangming Wu Qingzhe Zhai Alain Goossens Chuanyou Li 2022Molecular Plant2022,15,8:2
16The establishment of polypeptide PSMA-targeted chimericantigen receptor-engineered natural killer cells forcastration-resistant prostate cancer and the induction of ferroptosis-related cell death显示文摘Background:The mortality of castration-resistant prostate cancer(CRPC)is high due to lack of an effective treatment.Chimeric antigen receptor(CAR)-based therapy is a promising immunotherapeutic strategy.Here,we aimed to design a novel CAR-natural killer(NK)cells with a clinically significant tumoricidal effect on CRPC.Methods:We constructed novel CAR-NK92MI cells with a CD244-based recombinant lentiviral vector.Different intracellular segments(CD244,NKG2D,or CD3ζ)were screened to identify the best candidate according to cell lysis assay and CD107a expression levels.To enhance the affinity of the CAR to the tumor antigen,we compared an antibody specific for prostate-specific membrane antigen(anti-PSMA)with PSMA-targeted polypeptide(p-PSMA),which was screened by phage display combinatorial library.Then,CAR-NK92MI cells with both a high affinity for PSMA and a strong tumoricidal capacity were generated.In addition,we verified their tumor-killing effect in vitro and in vivo.The release of cytokine by NK92MI cells was compared with that by CAR-NK92MI cells through flow cytometry and enzyme-linked immunosorbent assay.Moreover,ferroptosis-related cell death was explored as a possible underlying mechanism.Results:Three different CAR intracellular regions CAR1(CD244),CAR2(CD244,NKG2D)and CAR3(CD244,NKG2D,and CD3ζ)were constructed.CAR2 was chosen to confer a stronger tumoricidal ability on CAR-NK92MI cells.Compared with anti-PSMA,p-PSMA exhibited enhanced affinity for the tumor antigen.Thus,p-PSMA-CAR-NK92MI cells,which expressed CAR with a polypeptide-based antigen-binding region,an intracellular CD244 and a NKG2D costimulatory domain,were generated.They could selectively and successfully kill PSMA+target cells and exhibited specific lysis rate of 73.19%for PSMA-positive C4-2 cells and 33.04%for PSMA-negative PC3 cells.Additionally,p-PSMA-CAR-NK92MI cells had significantly higher concentrations of IFN-γ,TNF-αand granzyme B than NK92MI cells.In a CRPC cancer xenograft model,p-PSMA-CAR-NK92MI cells significantly inhibited tumor growth and exerted a more consistent killing effect than NK92MI cells.Moreover,ferroptosis is a potential mechanism through which CAR-NK92MI cells attack cancer cells,and is triggered by IFN-γ.Conclusions:p-PSMA-CAR-NK92MI cells can effectively kill CRPCPSMA+cells in vitro and in vivo.This strategy may provide additional treatment options for patients with CRPC.Liyuan Wu Fei Liu Le Yin Fangming Wang Hui Shi Qinxin Zhao Feiya Yang Dong Chen Xiying Dong Yuchun Gu Nianzeng Xing 2022Cancer Communications2022,42,8:2
17Mediator tail module subunits MED16 and MED25 differentially regulate abscisic acid signaling in Arabidopsis显示文摘MED25 has been implicated as a negative regulator of the abscisic acid(ABA)signaling pathway.However,it is unclear whether other Mediator subunits could associate with MED25 to participate in the ABA response.Here,we used affinity purification followed by mass spectrometry to uncover Mediator subunits that associate with MED25 in transgenic plants.We found that at least26 Mediator subunits,belonging to the head,middle,tail,and CDK8 kinase modules,were copurified with MED25 in vivo.Interestingly,the tail module subunit MED16 was identified to associate with MED25 under both mock and ABA treatments.We further showed that the disruption of MED16 led to reduced ABA sensitivity compared to the wild type.Transcriptomic analysis revealedthattheexpressionofseveral ABA-responsive genes was significantly lower in med16 than those in wild type.Furthermore,we discovered that MED16 may possibly compete with MED25 to interact with the key transcription factor ABA INSENSITIVE 5(ABI5)to positively regulate ABA signaling.Consistently,med16 and med25 mutants displayed opposite phenotypes in ABA response,cuticle permeability,and differential ABI5-mediated EM1 and EM6 expression.Together,our data indicate that MED16 and MED25 differentially regulate ABA signaling byantagonisticallyaffectingABI5-mediated transcription in Arabidopsis.Pengcheng Guo Leelyn Chong Fangming Wu Chuan-Chih Hsu Chuanyou Li Jian-Kang Zhu Yingfang Zhu 2021Journal of Integrative Plant Biology2021,63,4:1
18Challenges and opportunities in remote sensing-based crop monitoring:a review显示文摘Building a more resilient food system for sustainable development and reducing uncertainty in global food markets both require concurrent and near-real-time and reliable crop information for decision making.Satellite-driven crop monitoring has become a main method to derive crop information at local,regional,and global scales by revealing the spatial and temporal dimensions of crop growth status and production.However,there is a lack of quantitative,objective,and robust methods to ensure the reliability of crop information,which reduces the applicability of crop monitoring and leads to uncertain and undesirable consequences.In this paper,we review recent progress in crop monitoring and identify the challenges and opportunities in future efforts.We find that satellite-derived metrics do not fully capture determinants of crop production and do not quantitatively interpret crop growth status;the later can be advanced by integrating effective satellite-derived metrics and new onboard sensors.We have identified that ground data accessibility and the negative effects of knowledge-based analyses are two essential issues in crop monitoring that reduce the applicability of crop monitoring for decisions on food security.Crowdsourcing is one solution to overcome the restrictions of ground-truth data accessibility.We argue that user participation in the complete process of crop monitoring could improve the reliability of crop information.Encouraging users to obtain crop information from multiple sources could prevent unconscious biases.Finally,there is a need to avoid conflicts of interest in publishing publicly available crop information.Bingfang Wu Miao Zhang Hongwei Zeng Fuyou Tian Andries B Potgieter Xingli Qin Nana Yan Sheng Chang Yan Zhao Qinghan Dong Vijendra Boken Dmitry Plotnikov Huadong Guo Fangming Wu Hang Zhao Bart Deronde Laurent Tits Evgeny Loupian 2023National Science Review2023,10,4:1
