| 8 | Decreased Average Albumin-to-Globulin Ratio During Radiotherapy Worsens the Prognosis of Esophageal Squamous Cell Carcinoma:Implications for Nutritional Management显示文摘Objective Malnutrition and cachexia are common in esophageal squamous cell carcinoma(ESCC)patients undergoing radiotherapy.This study evaluated how malnutrition-and cachexia-related indicators,including the albumin-to-globulin ratio(AGR),and their changes during radiotherapy predict the treatment outcomes.Methods We reviewed a total of 172 ESCC patients receiving radiotherapy(as a primary cohort)and performed a Cox regression analysis on potential prognostic factors,including the AGR,as well as the TNM stage and concomitant chemotherapy.A subsequent receiver operating characteristics(ROC)curve and Kaplan Meier survival analysis was performed for ESCC patients stratified by the average AGR cut-off point.Results In addition to the well-documented factors(i.e.TNM stage and concomitant chemotherapy),the average AGR was a significant prognostic factor for ESCC patients receiving radiotherapy.By plotting the ROC curve of the average AGR with regard to the ESCC prognosis,we obtained cut-off points for the overall patients(cut-off point:1.5,AUC:0.636),for patients with stageⅡ/Ⅲdisease(cut-off point:1.5,AUC:0.611),and for patients with stageⅣdisease(cut-off point:1.84,AUC:0.900).The ESCC patients with higher average AGR had a significantly more favorable OS compared with those with a lower average AGR.Notably,ESCC patients who had an increasingΔAGR during radiotherapy had a considerably more favorable OS compared with those with a decreasingΔAGR.All of these findings were reproducible in the validation cohort.Conclusion A lower average AGR is indicative of a poorer prognosis for ESCC patients following radiotherapy.Improving the nutritional status and preventing or ameliorating cachexia might contribute to improving the prognosis of ESCC patients. | Fen Cai Xiu Lian Zhang Xiao Ling Wu Ting Ting Zhuang Lei Huang Yi Fei Tu Yuan Yuan Lu Min Xia Jiang Qing Ying Zhang Yan Lin Chang Chun Ma | 2021 | Journal of Nutritional Oncology2021,6,2: | 8 |

| 10 | Enhancement of innate immune system in monocot rice by transferring the dicotyledonous elongation factor Tu receptor EFR显示文摘The elongation factor Tu(EF-Tu) receptor(EFR) in cruciferous plants specifically recognizes the N-terminal acetylated elf18 region of bacterial EF-Tu and thereby activates plant immunity.It has been demonstrated that Arabidopsis EFR confers broad-spectrum bacterial resistance in the EFR transgenic solanaceous plants.Here,the transgenic rice plants(Oryza sativa L.ssp.japonica cv.Zhonghua 17) and cell cultures with constitutive expression of AtEFR were developed to investigate whether AtEFR senses EF-Tu and thus enhances bacterial resistance in the monocot plants.We demonstrated that the Xanthomonas oryzae-derived elf18 peptide induced oxidative burst and mitogen-activated protein kinase activation in the AtEFR transgenic rice cells and plants,respectively.Pathogenesis-related genes,such as OsPBZ1,were upregulated dramatically in transgenic rice plant and cell lines in response to elf18 stimulation.Importantly,pretreatment with elf18 triggered strong resistance to X.oryzae pv.oryzae in the transgenic plants,which was largely dependent on the AtEFR expression level.These plants also exhibited enhanced resistance to rice bacterial brown stripe,but not to rice fungal blast.Collectively,the results indicate that the rice plants with heterologous expression of AtEFR recognize bacterial EF-Tu and exhibit enhanced broad-spectrum bacterial disease resistance and that pattern recognition receptor-mediated immunity may be manipulated across the two plant classes,dicots and monocots. | Fen Lu Huiqin Wang Shanzhi Wang Wendi Jiang Changlin Shan Bin Li Jun Yang Shiyong Zhang Wenxian Sun | 2015 | Journal of Integrative Plant Biology2015,57,7: | 6 |
| 11 | The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy. | Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu | 2021 | Cancer Communications2021,41,1: | 5 |
| 12 | Polysaccharides extracted from the roots of Bupleurum chinense DC modulates macrophage functions显示文摘The present study aimed to investigate the effects of polysaccharides extracted from Bupleurum chinense DC(BCPs) on macrophage functions. In the in vivo experiment, 1 m L of 5% sodium thioglycollate was injected into the abdomen of the mice on Day 0 and macrophages were harvested on Day 4. The macrophages were cultured in plates and treated with different concentrations of BCPs and stimulus. Effects of BCPs on macrophage functions were assessed by chemotaxis assay, phagocytosis assay and Enzyme-Linked Immunosorbent Assay(ELISA). Our results showed the enhanced chemotaxis, phagocytosis and secretion of nitric oxide(NO) and inflammatory cytokines by macrophages when treated with BCPs. However, when chemotaxis and phagocytosis were up-regulated by complement components or opsonized particles, BCPs inhibited these effects. Also, the NO production induced by lipopolysaccharides(LPS) was suppressed by BCPs mildly. Moreover, BCPs had an inhibitory effect on the [Ca^(2+)]i elevation of macrophages. | 15 | Variability of microphysical characteristics in the “21·7” Henan extremely heavy rainfall event显示文摘In this study, significant rainfall microphysical variability is revealed for the extremely heavy rainfall event over Henan Province in July 2021(the “21·7” Henan EHR event) using a dense network of disdrometers and two polarimetric radars.The broad distributions of specific drop size distribution(DSD) parameters are identified in heavy rainfall from the disdrometer observations, indicating obvious microphysical variability on the surface. A K-means clustering algorithm is adopted to objectively classify the disdrometer datasets into separate groups, and distinct DSD characteristics are found among these heavy rainfall groups. Combined with the supporting microphysical structures obtained through radar observations, comprehensive microphysical features of the DSD groups are derived. An extreme rainfall group is dominantly formed in the deep convection over the plain regions, where the high number of concentrations and large mean sizes of surface raindrops are underpinned by both active ice-phase processes and efficient warm-rain collision-coalescence processes in the vertical direction. Convection located near orographic regions is characterized by restricted ice-phase processes and high coalescence efficiency of liquid hydrometeors, causing the dominant DSD group to comprise negligible large raindrops. | 19 | Analysis of the cause of crack in cold rolling of QSn6.5-0.1 strip显示文摘In order to know the cause of cracks in cold rolling of QSn6.5 0.1 copper alloy strip, a lot of experiments and analysis were done. The microstructure changes of QSn6.5 0.1 were investigated by means of metallurgical microscope. The morphology of cracks and surface defects were examined using scanning electron microscope. Macroscopic residual stresses produced in every process during manufacturing in the QSn6.5 0.1 strip were measured by X ray diffraction method and hole drilling method. The results show that the cracks in the QSn6.5 0.1 cold rolling strip were caused due to the derivation of metallurgical defects, such as SnO 2, S, fine looses,the inverse segregation unable to clear up when milling, and the accumulation of all kinds of resi dual stresses. When the accumulation of the residual stress reaches the material′s breaking strength, the cracks will be generated.
