1Chemopreventive and Growth Inhibitory Effects of Selenium显示文摘There is very convincing evidence that a high dietary level of selenium substantially reduces the incidence of a wide variety of animal cancers. The human epidemiological evidence is less clear cut, but overall suggests that selenium may be protective: the evidence is strongest in men in relation to gastro-intestinal cancers. There is evidence that dietary selenium compounds reduce the formation of DNA adducts by carcinogens. Selenium compounds also inhibit growth in vitro and induce apoptosis. In general, there is a good correlation between the effectiveness of selenium compounds in chemoprevention and growth inhibition, implying that the mechanisms of growth inhibition and chemoprevention may be similar and that a major factor in the chemopreventive effects of selenium compounds in vivo is their ability to retard outgrowth of pre-malignant cells. Various hypotheses have been advanced as to how selenium compounds might prevent tumour cellgrowth. One is that they cause apoptosis by inducing oxidative stress. However, we have shown that the most potent selenium compound, selenodiglutathione (SDG), a natural metabolite of selenite, does not induce oxidative stress, at least not in the sarne way as other oxidants such as H2O2 and diamide. Firstly, a partially selenium-resistant variant cell line does not show increased resistance to H2O2. Moreover, SDG does not induce widespread tyrosine phosphorylation, including MAP and SAN kinases, like other oxidants such as H2O2 and diamide and its effects are not reversed by pretreatment with the tyrosine kinase inhibitor, herbimycin. 2A numerical study on fire endurance of wood beams exposed to three-side fire显示文摘To investigate the fire endurance of wood beams exposed to three-side fire, we conducted bearing capacity tests of two wood beams and experiments of five wood beams exposed to three-side fire. The finite element software ANSYS was also used to predict the fire endurance of those beams with the indirect order coupling method. It was found that the fire endurance decreases as the load level increases, and the reduction ratio tends to decrease. In the case of a certain load level, the fire endurance is improved if the section size is increased or covered by the fire protection coating. The central deformation increases as the fire duration increases, and the ratio of increase tends to rise. On another note, an increase in the density of wood leads to a rise in the fire endurance of a given beam. From the finite element method (FEM) calculation results, the fire endurance decreases as the load level increases, and the reduction ratio tends to decrease. When the load level is greater than 0.5, the fire endurance is significantly reduced, and it does not change significantly when the load level changes. 3Biocompatibility evaluation in vitro. Part I: Morphology expression and proliferation of human and rat osteoblasts on the biomaterials显示文摘The biocompatibility evaluation of calcium phosphate based biomaterials is performed by tissue culture in vitro model. Three kinds of bioceramic materials which are potential to deal with bone trauma and/or conduct tissue growth are recommodated. The biological research results show that human and animal osteoblast cells anchor the materials surface in two hours in culture. Confocal laser scanning microscopy (CLSM) demonstrated the normal cell distribution and proliferation on both of dense and porous biomaterials. Hydroxyapatite and tricalcium phosphate stimulate cell proliferation. However, DNA and protein synthesis were considerably limited and the apoptosis phenomenon would be present on the hydroxyapatite (HA) materials by adding Al, Mg elements. 4FT Ⅰ, a novel positive myeloid-lineage-specific transcription regulatory element within the mouse myeloperoxidase gene enhancer, En 1显示文摘FT Ⅰ (AAAAGGGGAAGCAGAG), a poly purine ele-ment within the myloid-lineage specific enhancer (En 1) of the mouse myeloperoxidase gene [1, 2] has been fur-ther characterised. 1, FT Ⅰ functions as a myeloid-lineage specific transcription regulatory element; 2, WEHI 3BD+ cells have higher binding activity to FT Ⅰ and express the proteins which could form the unique DNA-protein com-plex(es) of FT Ⅰ;. 3, The essential sequence for the specific DNA-protein interactions of FT Ⅰ is AAAAGGGGAAGC; 4, South-western analysis in conjunction with the compe-tition assay of the proteins binding to FT Ⅰ, has revealed a 28 kd protein in WEHI 3BD+ cells that displays the properties of the putative transcription factor which acts through FT Ⅰ. 5Removal characteristics of steroid estrogens in trickling filters显示文摘The discharge of steroid estroens from sewage treatment works (STW) is to be regulated by Environmental Quality Standard in the UK,thus requiring the understanding of removal characteristics of steroid estroens in trickling filters to benefit UK water industry with trickling filters used in 75% STWs. Two pilot-scale trickling filters were operated in parallel to treat synthetic sewage spiked with oestrone (E1),oestradiol (E2) and 17α-ethinyl oestradiol (EE2) at environmentally related concentrations. Control experiments show that biodegradation is the dominant removal mechanism although adsorption onto biofilm solids prior to biodegradation would be part of the overall mechanisms of estrogen removal. Approximately 44.7%-58.9% is removed by the pilot trickling filter normally operated,whilst the 1:1 recirculation increases 29.0%-32.2% estrogen removals by improved wetting rate and hydraulic retention time supported by tracer experiment with lithium chloride. 9Myeloidcell-lineage and premylocytic-stage-specific-expression of the mouse myeloperoxidase gene is controlled at initiation as well as elongation levels of transcription显示文摘The myeloperoxidase (MPO) is an important microbicidal protein present at high concentration in the primary granule of mature granulocyte and its expression is regulated in both myeloidcell-lineage and premyelocytic-stagespecific manners. A better understanding of the underlying control mechanisms should provide insights into the temporal and co-ordinate regulation of the gene expression during granulopoiesis. We have identified its promoter by mapping the start(s) of transcription using various molecular approaches together with demonstrating the promoter function of the relevant DNA segment in a transient transfection reporter assay. Besides the major start of transcription mapped at G residue, 11 nucleotide upstream of the 3’ end of exon 0, the usage of that is specific to the MPO expressing cell lines, we have shown that irrespective of the MPO-expression status of the hematopoietic cells, transcription occurs broadly within a two kb region upstream of the 5’ proximity of the gene, and is largely terminated in nitron 2.
