| 2 | Real-time PCR diagnosis of Schistosoma japonicum in low transmission areas of China显示文摘Background:Schistosomiasis in the People’s Republic of China(PRC)can be traced back to antiquity.In the past 60 years,the Chinese government has made great efforts to control this persistent disease with elimination slated by 2020 through the implementation of a comprehensive control strategy.This strategy aims to reduce the role of bovines and humans as sources of infection as a pre-requisite for elimination through transmission interruption.The goal of elimination will be achievable only by the implementation of a sustainable surveillance and control system,with sensitive diagnosis a key feature so that the true disease burden is not underestimated.Currently used diagnostics lack the necessary sensitivity to accurately determine the prevalence of Schistosoma japonicum infection in areas with low infection intensities.It is of critical importance to find and treat people and to identify animals with low-level infections if the National Control Programme for China is to achieve schistosomiasis elimination.Methods:We evaluated a real-time polymerase chain reaction(qPCR)assay using 633 human stool samples collected from five villages in Hunan,Anhui,Hubei,and Jiangxi provinces,and 182 bovine(70 cattle and 112 buffalo)stool samples obtained from four villages in Hunan,Anhui,and Jiangxi provinces in the PRC.All stool samples were subjected to the miracidium hatching test(MHT,a diagnostic procedure used in the National Schistosomiasis Control Programme)and the qPCR assay.Samples positive by MHT were subjected to either the Kato-Katz technique for humans,or the formalin-ethyl acetate sedimentation-digestion(FEA-SD)procedure for bovines,to determine infection intensities.Results:The qPCR assay exhibited a high level of sensitivity in the detection of S.japonicum infections.With both the human and bovine samples,a significantly higher prevalence was determined using the qPCR assay(11.06%humans,24.73%bovines)than with the MHT(0.93%humans,7.69%bovines).The animal contamination index(calculated using data obtained with the qPCR technique)for all positive bovines was 27618000 eggs per day,indicating a considerable amount of environmental egg contamination that would be underestimated using less sensitive diagnostic procedures.Conclusions:The qPCR assay we have evaluated will be applicable as a future field diagnostic and surveillance tool in low-transmission zones where schistosomiasis elimination is targeted and for monitoring post-intervention areas to verify that elimination has been maintained. | Pei He Catherine A.Gordon Gail M.Williams Yuesheng Li Yuanyuan Wang Junjian Hu Darren J.Gray Allen G.Ross Donald Harn Donald P.McManus | 2018 | Infectious Diseases of Poverty2018,7,1: | 7 |
| 3 | A hidden fault prediction model based on the belief rule base with power set and considering attribute reliability显示文摘Hidden faults are important characteristics of complex systems that cannot be observed directly.The hidden behavior of a system, such as the health state and safety level, has a direct correlation with these hidden faults. After the predicted hidden behavior reaches a fault boundary, certain measures must be taken to avoid fault occurrences. Thus, hidden faults can be predicted by the hidden behavior of a system.The belief rule base(BRB) has been used to predict hidden behaviors. However, two problems remain to be solved in engineering practice. First, when the observed information is absent, ignorance may exist in the output. If only global ignorance is considered, it may be unreasonable in certain cases, which can influence the prediction model. Second, the effects of disturbance factors such as noise and sensor quality may cause the reliability of the gathered information to decline, which indirectly leads to unreliability of the hidden behavior. Thus, to address the global ignorance and unreliable hidden behavior, a new hidden BRB model with a power set and considering attribute reliability(PHBRB-r) is proposed for hidden fault prediction. In the PHBRB-r model, the effects of disturbance factors on hidden behavior are considered using attribute reliability, and the discernment frame is a power set. | 4 | Differences in atheroma between Caucasian and Asian subjects with anterior stroke:A vessel wall MRI study显示文摘Background and purpose While extracranial carotid artery stenosis is more common among Caucasians and intracranial artery stenosis is more common among Asians,the differences in atherosclerotic plaque characteristics have not yet been extensively examined.We sought to investigate plaque location and characteristics within extracranial carotid and intracranial arteries in symptomatic Caucasians and Chinese using vessel wall MRI.Methods Subjects with recent anterior circulation ischaemic stroke were recruited and imaged at two sites in the USA and China using similar protocols.Both extracranial carotid and intracranial arteries were reviewed to determine plaque location and characteristics.Results The prevalence of extracranial carotid plaque in Caucasians and Chinese was 73.1%and 49.1%,respectively(p=0.055).Prevalence of intracranial plaque was 38.5%and 69.1%in Caucasians and Chinese,respectively(p=0.02).Furthermore,42% of Caucasians and 16%of Chinese had high-risk plaque(HRP)features(intraplaque haemorrhage,luminal surface disruption)in the extracranial carotid artery(p=0.03).The prevalence of HRP features in intracranial arteries was not significantly different between the two cohorts(4%vs 11%;p=0.42).Conclusions Differences in the location and characteristics of cerebrovascular atherosclerosis were identified by vessel wall MRI in US Caucasian and Chinese subjects with recent anterior circulation ischaemic stroke.Extracranial carotid plaques with HRP features were more common in Caucasians.Intracranial plaques were more common in Chinese subjects,but no significant difference between the two cohorts in intracranial HRP prevalence was found.Larger studies using vessel wall imaging to investigate racial differences in cerebrovascular disease may inform underlying mechanisms of HRP development and may ultimately help guide appropriate therapy. | Hiroko Watase Mi Shen Binbin Sui Peiyi Gao Dong Zhang Jie Sun Niranjan Balu Daniel S Hippe Gail P Jarvik Xihai Zhao Rui Li Shuo Chen Chun Yuan Thomas S Hatsukami | 2021 | Stroke & Vascular Neurology2021,6,1: | 4 |
| 5 | Estimating the prevalence of Echinococcus in domestic dogs in highly endemic for echinococcosis显示文摘Background:Cystic echinococcosis(CE)and alveolar echinococcosis(AE)are highly endemic in Xiji County of Ningxia Hui Autonomous Region(NHAR)in China where the control campaign based on dog de-worming with praziquantel has been undertaken over preceding decades.This study is to determine the current prevalence of Echinococcus granulosus and E.multilocularis in domestic dogs and monitor the echinococcosis transmission dynamics.Methods:Study villages were selected using landscape patterns(Geographic Information System,GIS)for Echinococcus transmission“hot spots”,combined with hospital records identifying risk areas for AE and CE.A survey of 750 domestic dogs,including copro-sampling and owner questionnaires,from 25 selected villages,was undertaken in 2012.A copro-multiplex PCR assay was used for the specific diagnosis of E.granulosus and E.multilocularis in the dogs.Data analysis,using IBM SPSS Statistics,was undertaken,to compare the prevalence of the two Echinococcus spp.in dogs between four geographical areas of Xiji by theχ^(2)test.Univariate analysis of the combinations of outcomes from the questionnaire and copro-PCR assay data was carried out to determine the significant risk factors for dog infection.Results:The highest de-worming rate of 84.0%was found in the northwest area of Xiji County,and significant differences(P<0.05)in the de-worming rates among dogs from the four geographical areas of Xiji were detected.The highest prevalence(19.7%,59/300)of E.multilocularis occurred in northwest Xiji,though the highest prevalence(18.1%,38/210)of E.granulosus occurred in southwest Xiji.There was no significant difference(P>0.05)in the prevalence of E.granulosus in dogs from the northwest,southwest,northeast,and southeast of Xiji,but there were significant differences(P<0.05)between dogs infected with E.multilocularis from the four areas.None of the other independent variables was statistically significant.Conclusions:The results from this study indicate a high prevalence of both E.granulosus and E.muiltilocularis in dogs in Xiji County,NHAR.Transmission of E.multilocularis was more impacted by geographical risk-factors in Xiji County than that of E.granulosus.Dogs have the potential to maintain the transmission of both species of Echinococcus within local Xiji communities,and the current praziquantel dosing of dogs appears to be ineffective or poorly implemented in this area. | Cong-Nuan Liu Yang-Yang Xu Angela M.Cadavid-Restrepo Zhong-Zi Lou Hong-Bin Yan Li Li Bao-Quan Fu Darren J.Gray Archie A.Clements Tamsin S.Barnes Gail M.Williams Wan-Zhong Jia Donald P.McManus Yu-Rong Yang | 2018 | Infectious Diseases of Poverty2018,7,1: | 3 |
| 6 | Development of an educational cartoon to prevent worm infections in Chinese schoolchildren显示文摘Background:With more than two billion people infected worldwide,soil-transmitted helminths(STH)are the most widespread infections.To date,STH control efforts rely predominantly on recurrent mass drug administration(MDA),which does not prevent reinfection.Additional public health measures including novel health educational tools are required for more sustained integrated control of STH.We describe the development of an educational cartoon video(The Magic Glasses)targeting STH infections in Chinese schoolchildren and its pilot testing in China.We applied an extensive community-based mixed methods approach involving input from the target group of 9–10 year old schoolchildren and key informants,such as teachers,doctors and parents,in order to identify potential STH infection risks in the study area and to formulate key messages for the cartoon.The development of the educational cartoon included three major steps:formative research,production,and pilot testing and revision.Results:We found that most adults and approximately 50%of the schoolchildren were aware of roundworm(Ascaris)infection,but knowledge of transmission,prevention and treatment of STH was poor.Observations in the study area showed that unhygienic food practices,such as eating raw and unwashed fruit or playing in vegetable gardens previously fertilised with human faeces,posed major STH infection risks.Conclusions:It was crucial to assess the intellectual,emotional,social and cultural background of the target population prior to video production in order to integrate the key messages of the cartoon into everyday situations.Overall,our strategy for the development of the cartoon and its incorporation into a health education package proved successful,and we provide a summary of recommendations for the development of future educational videos based on our experiences in China. | Franziska A Bieri Li-Ping Yuan Yue-Sheng Li Yong-Kang He Andrew Bedford Robert S Li Feng-Ying Guo Sheng-Ming Li Gail M Williams Donald P McManus Giovanna Raso Darren J Gray | 2013 | Infectious Diseases of Poverty2013,2,1: | 2 |

| 12 | Osteosarcoma Segmentation in MRI Based on Zernike Moment and SVM显示文摘Osteosarcoma is primary malignant neoplasms derived from cells of mesenchymal origin, and often has distinct phenotypes at different stages. The location of tumor and reaction zone can be identified by an expert in magnetic resonance imaging (MRI), with MRI being one of the choices for evaluating the extent of osteosarcoma. However, it is still a challenge to automatically extract tumor from its surrounding tissues because of their low intensity differences in MRI. We investigated an approach based on Zernike moment and support vector machine (SVM) for osteosarcoma segmentation in T1-weighted image (TIWI). Firstly, the different order moments around each pixel are calculated in small windows. Secondly, the grayscale and the module values of different order moments are used as a texture feature vector which is then used as the training set for SVM. Finally, an SVM classifier is trained based on this set of features to identify the osteosarcoma, and the segmented tumor tissue is rendered in 3D by the ray casting algorithm based on graphics processing unit (GPU). | 14 | Histopathological Analysis of Liver During Pregnancy Toxemia in Small-tailed Han Sheep显示文摘The present work aimed to investigate the microscopic and ultramicroscopic structure of the liver to assess the level of hepatic impairment during pregnancy toxemia. Seven pregnant small-tailed Han sheep of negative urine ketone bodies were assessed in this study. Toxemia was induced by limiting food and movement late in pregnancy. Three sheep developed obvious clinical symptoms with motor weakness, depression, anorexia, locomotion disturbances, blindness and languishment. We harvested their liver tissues as pathological material, and used routine histological and electronic microscopy methods to observe the histopathological changes in small-tailed Han sheep induced by pregnancy toxemia. Autopsy of the livers of the sheep revealed deep yellow coloration, intumescence and hemorrhage on the surface. Microstructural features indicated fatty degeneration, which is a main characteristic of fatty liver. Hepatocellular ultrastructural changes were observed under an electronic microscope. The characteristic findings were nucleolus concentration, vacuolation of mitochondria and excessive glycogen granules in the cytoplasm. Via this experimental protocol, pregnancy toxemia of sheep was successfully induced, providing a pathological model for the study of this disease.
