| 1 | Chemical synthesis of Ub-AMC via ligation of peptide hydrazides显示文摘The C-terminal conjugate of ubiquitin with 7-amino-4-methylcoumarin (Ub-AMC) is an important probe for fluorescencebased analysis of deubiquitinating enzyme (DUB) activity. It is important to develop more efficient methods for the preparation of Ub-AMC because the currently available technology is still expensive for scaled-up production. In the present work we report an efficient strategy for total chemical synthesis of Ub-AMC through ligation of peptide hydrazides. Three peptide segments are assembled via N-to-C sequential ligation and the resulting product is converted to Ub-AMC via TCEP-mediated desulfurization. | 9 | Field Efficacy Test of 22% Fipronil FS against Sugarcane Stem Borers and Thrips显示文摘[Objective] The paper was to explore the field efficacy of 22% fipronil FS against sugarcane stem borer and thrips( Baliothrips serratus Kobus).[Method]The field trial was conducted by foliar spraying of 22% fipronil FS and mixed broadcasting with fertilizer in 2014. [Result]Foliar spraying of 22%fipronil FS at the dose of 50 m L /667 m^2 had the best control effect against sugarcane stem borer,and the control effect at 90 d post administration was 70. 70%,significantly higher than that of control agent(61. 86%). Medium and high dose of 22% fipronil FS had better control effects against sugarcane thrips. | 11 | Fructooligosaccharides enhance the therapeutic effect of xiao-zhi-fang on non-alcoholic fatty liver disease via regulating intestinal flora显示文摘Background:Non-alcoholic fatty liver disease(NAFLD)is a frequently occurring clinical disease,which could be treated by traditional Chinese medicine.However,the effects and mechanisms of xiao-zhi-fang(XZF)alone or in combination with fructooligosaccharides(FOS)on NAFLD remains unclear.Methods:This study established the NAFLD mouse model by feeding with a high-fat diet and by administering with XZF and/or FOS.Moreover,the fat content in the liver tissues was evaluated by oil red O and hematoxylin and eosin staining,and the liver function indices were examined using a biochemical analyzer.Changes in the intestinal flora were monitored using 16S rRNA gene sequencing.The results of the current study showed that the alanine aminotransferase,aspartate aminotransferase,total triglyc-eride,total cholesterol levels,and the amount of lipid accumulation were markedly elevated in NAFLD mice.Results:XZF alone or in combination with FOS significantly reduced lipid accumulation in the liver and blood lipid levels in NAFLD mice.Moreover,XZF alone or in combination with FOS notably altered the intestinal flora structure in NAFLD mice.Consequently,17 operational taxonomic units were decreased and nine were increased in the XZF and/or FOS groups.Conclusions:FOS may accelerate the intestinal microecological balance in NAFLD mice after treatment with XZF by promoting intestinal flora growth,thereby enhancing the therapeutic effect of XZF on NAFLD. | Jiongshan Zhang Yue Li Gemin Xiao Yongwei Li Heping Xie Yan Chen | 2021 | Liver Research2021,5,4: | 0 |
| 12 | A secreted phospholipase A_(2) (BmsPLA_(2)) regulates melanization of immunity through BmDDC in the silkworm Bombyx mori显示文摘Insect immune-associated phospholipase A_(2) (PLA_(2)) is an important target of pathogen invasion. Melanization, an effective defense response, has significant correlations with other immune responses to coordinate immune attack against invaders. However, the effect of PLA_(2) on melanization has not yet been reported in insects or other arthropods. In this work, we cloned a PLA_(2) gene (BmsPLA_(2)), and its protein had characteristic features of secreted PLA_(2) (sPLA_(2)). After injection of bacteria, BmsPLA_(2) expression and sPLA_(2) activity in hemolymph significantly increased. BmsPLA_(2) fluorescence was transferred from the cytoplasm to the cell membranes of circulating hemocytes. These results indicated that BmsPLA_(2) was related to hemolymph immunity in silkworms. Interestingly, reducing BmsPLA_(2) by RNA interference decreased melanosis (melanistic hemocytes) levels in vivo and in vitro, while BmsPLA_(2) overexpression had the opposite effect. The larval survival and melanization rate in the hemocoel both slowed depending on the PLA_(2) inhibitor dosage. These results demonstrated that BmsPLA_(2) plays a role in melanization during the immune process of silkworms. Surprisingly, the level of BmDDC matched the degree of melanization in various observations. BmDDC expression showed a significant increase, with the peak occurring later than that of BmsPLA_(2) after injection of bacteria, implying that BmsPLA_(2) was activated prior to BmDDC. Moreover, the alteration of BmsPLA_(2) by RNA interference or overexpression led to altered BmDDC levels. These results suggested that BmsPLA_(2) regulates the melanization response in silkworms through BmDDC. | 14 | SBC(Sanhuang Xiexin Tang combined with Baihu Tang plus Cangzhu)alleviates NAFLD by enhancing mitochondrial biogenesis and ameliorating inflammation in obese patients and mice显示文摘In the context of non-alcoholic fatty liver disease (NAFLD), characterized by dysregulated lipid metabolism in hepatocytes, the quest for safe and effective therapeutics targeting lipid metabolism has gained paramount importance. Sanhuang Xiexin Tang (SXT) and Baihu Tang (BHT) have emerged as prominent candidates for treating metabolic disorders. SXT combined with BHT plus Cangzhu (SBC) has been used clinically for Weihuochisheng obese patients. This retrospective analysis focused on assessing the anti-obesity effects of SBC in Weihuochisheng obese patients. We observed significant reductions in body weight and hepatic lipid content among obese patients following SBC treatment. To gain further insights, we investigated the effects and underlying mechanisms of SBC in HFD-fed mice. The results demonstrated that SBC treatment mitigated body weight gain and hepatic lipid accumulation in HFD-fed mice. Pharmacological network analysis suggested that SBC may affect lipid metabolism, mitochondria, inflammation, and apoptosis—a hypothesis supported by the hepatic transcriptomic analysis in HFD-fed mice treated with SBC. Notably, SBC treatment was associated with enhanced hepatic mitochondrial biogenesis and the inhibition of the c-Jun N-terminal kinase (JNK)/nuclear factor-kappa B (NF-κB) and extracellular signal-regulated kinase (ERK)/NF-κB pathways. In conclusion, SBC treatment alleviates NAFLD in both obese patients and mouse models by improving lipid metabolism, potentially through enhancing mitochondrial biogenesis. | 16 | Effect of Different Additive Combinations on Production Performance of Layer显示文摘To explore the effect of different additive combinations on double-low rapeseed cake( meal) utilization and layer production performance,we randomly divided 756 Roman brown shell layers around 50 weeks age into seven groups to detect production performance,of which group 1 was control group supplied with basic diet( added with 9% double-low rapeseed cake),while groups 2- 7 were experimental groups added with Chinese herbal medicine,enzyme preparation,probiotics and peptide antibiotics in basic diet. The results showed that group 4 added with Chinese herbal medicine + enzyme preparation + peptide antibiotics received the best effect,resulting in significantly increased laying rate( 70. 50%,P < 0. 05),significantly reduced feed-egg ratio( 2. 46,P < 0. 05),and extremely increased egg Haugh unit( P < 0. 01).
