1Development of composite PLGA microspheres containing exenatide-encapsulated lecithin nanoparticles for sustained drug release显示文摘This study aimed to prepare poly(D, L-lactic-co-glycolic acid) microspheres(PLGA-Ms)by a modified solid-in-oil-in-water(S/O/W) multi-emulsion technique in order to achieve sustained release with reduced initial burst and maintain efficient drug concentration for a prolonged period of time. Composite PLGA microspheres containing exenatideencapsulated lecithin nanoparticles(Ex-NPs-PLGA-Ms) were obtained by initial fabrication of exenatide-loaded lecithin nanoparticles(Ex-NPs) via the alcohol injection method,followed by encapsulation of Ex-NPs into PLGA microspheres. Compared to Ms prepared by the conventional water-in-oil-in-water(W/O/W) technique(Ex-PLGA-Ms), Ex-NPs-PLGAMs showed a more uniform particle size distribution, reduced initial burst release, and sustained release for over 60 d in vitro. Cytotoxicity studies showed that Ms prepared by both techniques had superior biocompatibility without causing any detectable cytotoxicity.In pharmacokinetic studies, the effective drug concentration was maintained for over 30 d following a single subcutaneous injection of two types of Ms formulation in rats, potentially prolonging the therapeutic action of Ex. In addition, administration of Ex-NPs-PLGA-Ms resulted in a more smooth plasma concentration-time profile with a higher area under the curve(AUC) compared to that of Ex-PLGA-Ms. Overall, Ex-NPs-PLGA-Ms prepared by the novel S/O/W method could be a promising sustained drug release system with reduced initial burst release and prolonged therapeutic efficacy.Ni Dong Chune Zhu Junhuang Jiang Di Huang Xing Li Guilan Quan Yang Liu Wen Tan Xin Pan Chuanbin Wu 2020Asian Journal of Pharmaceutical Sciences2020,15,3:7
2Study on Acute Toxicity of Purified Protein HAS1 Expression in KM Mice显示文摘[Objectives]The antifungal protein HAS1 newly obtained was evaluated for acute toxicity safety in KM mice according to relevant national regulations,so as to eliminate people s concerns about the safety of transgenic plants.[Methods]The acute toxicity of the purified protein HAS1 was observed by intragastric administration of mice,and the poisoning symptoms,poisoning degree,recovery and death were observed.[Results]No abnormal toxicity symptoms were observed in the test group,the vehicle control group and the blank control group.The main tissues and organs were not abnormal in gross anatomy.The average body weight of each group showed an increasing trend compared with before administration at 1,3,7,11 and 14 d after administration.It was found that after giving the purified protein HAS1 to KM mice at a cumulative dose of 64 mg/kg a day,no obvious toxicity was observed in the acute toxicity test,indicating that the test substance was non-toxic by oral administration.[Conclusions]This study provides a basis for further use of the protein and its coding genes.Guoru XIONG Gengfeng ZHAO Jungang WANG Guilan XING Shuzhen ZHANG 2019Agricultural Biotechnology2019,8,4:1

4Genomic insights on the contribution of introgressions from Xian/Indica to the genetic improvement of Geng/Japonica rice cultivars显示文摘Hybridization between Xian/indica(XI)and Geng/japonica(GJ)rice combined with utilization of plant ideotypes has greatly contributed to yield improvements in modern GJ rice in China over the past 50 years.To explore the genomic basis of improved yield and disease resistance in GJ rice,we conducted a large-scale genomic landscape analysis of 816 elite GJ cultivars representing multiple eras of germplasm from China.We detected consistently increasing introgressions from three XI subpopulations into GJ cultivars since the 1980s and found that the XI genome introgressions significantly increased the grain number per panicle(GN)and decreased the panicle number per plant.This contributed to the improvement of plant type during modern breeding,changing multi-tiller plants tomoderate tiller plants with a large panicle size and increasing the blast resistance.Notably,we found that key gene haplotypes controlling plant architecture,yield components,and pest and disease resistance,including IPA1,SMG1,DEP3,Pib,Pi-d2,and Bph3,were introduced from XI rice by introgression.By GWAS analysis,we detected a GN-related gene Gnd5,which had been consistently introgressed from XI into GJ cultivars since the 1980s.Gnd5 is a GRAS transcription factor gene,and Gnd5 knockout mutants showed a significant reduction in GN.The estimated genetic effects of genes varied among different breeding locations,which explained the distinct introgression levels of XI gene haplotypes,including Gnd5,DEP3,etc.,to these GJ breeding pedigrees.These findings reveal the genomic contributions of introgressions from XI to the trait improvements of GJ rice cultivars and provide new insights for future rice genomic breeding.Di Cui Han Zhou Xiaoding Ma Zechuan Lin Linhua Sun Bing Han Maomao Li Jianchang Sun Jin Liu Guixiu Jin Xianju Wang Guilan Cao Xing Wang Deng Hang He Longzhi Han 2022Plant Communications2022,3,3:0
5Skin Anaphylaxis Test of Purified Protein HAS1 in Cavies显示文摘The antifungal protein HAS1 is a new antifungal protein isolated from the genome of Bacillus subtilis HAS,which has a good inhibitory effect on various pathogenic fungi in sugarcane.This study aimed at evaluating the immunological transfer reaction of the test sample through repeated skin contact by observing whether the cavy skin repeatedly exposed to purified protein HAS1 suffers from allergic reaction and how strong is the allergic reaction.The results showed that the test group and the vehicle control group exhibited no allergic reaction in the skin immediately and at 24,48 and 72 h and had an mean reaction score and a sensitization rate both of 0,so the results of the naked eye observation were both nonallergenic to cavy skin;and in the positive control group,the cavies were observed to be highly sensitized immediately after removing the drug,mildly sensitized at 24 h,and not sensitized at 48 and 72 h,which meant the skin allergy was alleviated with the observation time.It is suggested that the purified protein HAS1 is negative for cavy skin anaphylaxis test (nonallergenic to cavy skin),which provides an experimental basis for further utilization of the protein and its coding gene.Guoru XIONG Gengfeng ZHAO Jungang WANG Guilan XING Shuzhen ZHANG 2019Agricultural Biotechnology2019,8,5:0
