2Growth-associated protein 43 and neural cell adhesion molecule expression following bone marrow-derived mesenchymal stem cell transplantation in a rat model of ischemic brain injury显示文摘BACKGROUND:Transplantation of bone marrow-derived mesenchymal stem cells(BMSCs) improves motor functional recovery,but the mechanisms remain unclear.OBJECTIVE:To investigate expression of growth-associated protein 43(GAP-43) and neural cell adhesion molecule following BMSC transplantation to the lateral ventricle in rats with acute focal cerebral ischemic brain damage.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment using immunohistochemistry was performed at the laboratories of Department of Neurology,Renmin Hospital of Wuhan University and Doctoral Scientific Research Work Station of C-BONS PHARMA,Hubei Province,China,from January 2007 to December 2008.MATERIALS:Monoclonal mouse anti-rat 5-bromo-2-deoxyuridine and neural cell adhesion molecule antibodies were purchased from Sigma,USA;monoclonal mouse anti-rat GAP-43 antibody was purchased from Wuhan Boster,China.METHODS:Rat models of right middle cerebral artery occlusion were established using the thread method.At 1 day after middle cerebral artery occlusion,20 μL culture solution,containing 5 × 105 BMSCs,was transplanted to the left lateral ventricle using micro-injection.MAIN OUTCOME MEASURES:Scores of neurological impairment were measured to assess neural function.Expression of GAP-43 and neural cell adhesion molecule at the lesion areas was examined by immunohistochemistry.RESULTS:GAP-43 and neural cell adhesion molecule expression was low in brain tissues of the sham-operated group,but expression increased at the ischemic boundary(P < 0.05).Transplantation of BMSCs further enhanced expression of GAP-43 and neural cell adhesion molecule(P < 0.05) and remarkably improved neurological impairment of ischemic rats(P < 0.05).CONCLUSION:BMSC transplantation promoted neurological recovery in rats by upregulating expression of GAP-43 and neural cell adhesion molecule.Yu Peng Qimei Zhang Hui You Weihua Zhuang Ying Zhang Chengyan Li 2010Neural Regeneration Research2010,5,13:18
3Evaluating Conservation Effectiveness of Nature Reserves Established for Surrogate Species:Case of a Giant Panda Nature Reserve in Qinling Mountains,China显示文摘Many nature reserves are established to protect the habitat needs of particular endangered species of interest but their effectiveness for protecting other species is questionable.In this study,this effectiveness was evaluated in a nature reserve network located in the Qinling Mountains,Shaanxi Province,China.The network of reserves was established mainly for the conservation of the giant panda,a species considered as a surrogate for the conservation of many other endangered species in the region.The habitat suitability of nine protected species,including the giant panda,was modeled by using Maximum Entropy(MAXENT)and their spatial congruence was analyzed.Habitat suitability of these species was also overlapped with nature reserve boundaries and their management zones(i.e.,core,buffer and experimental zones).Results show that in general the habitat of the giant panda constitutes a reasonable surrogate of the habitat of other protected species,and giant panda reserves protect a relatively high proportion of the habitat of other protected species.Therefore,giant panda habitat conservation also allows the conservation of the habitat of other protected species in the region.However,a large area of suitable habitat was excluded from the nature reserve network.In addition,four species exhibited a low proportion of highly suitable habitat inside the core zones of nature reserves.It suggests that a high proportion of suitable habitat of protected species not targeted for conservation is located in the experimental and buffer zones,thus,is being affected by human activities.To increase their conservation effectiveness,nature reserves and their management zones need to be re-examined in order to include suitable habitat of more endangered species.The procedures described in this study can be easily implemented for the conservation of many endangered species not only in China but in many other parts of the world.XU Weihua Andrés VIA QI Zengxiang OUYANG Zhiyun LIU Jianguo LIU Wei WAN Hui 2014Chinese Geographical Science2014,24,1:12
4Size,morphology,and composition of lunar samples returned by Chang'E-5 mission显示文摘This study focuses on the physical and chemical properties of surficial lunar regolith(LR)samples returned from the Moon by the Chang’E-5(CE-5)mission.Insights regarding the effect of a new sampling geological site on the surficial lunar sample CE5 C0400 were illustrated using nondestructive techniques such as laser diffractometry coupled with image analysis,X-ray computed tomography,and field emission scanning electron microscopy equipped with energy dispersive spectroscopy,and Xray diffraction combined with Rietveld refinement.From the characterization analyses,the CE-5 sampling site in the northeastern Oceanus Procellarum on the Moon yields a unique collection of relatively regular-shaped and fine basalt-dominated particles.The median grain size D_(50) is(55.24±0.96)μm,falling within the relatively low end of the range of the Apollo lunar returned samples.The coefficient of uniformity C_(u)of 15.1 and the coefficient of curvature C_(c)of 1.7 could classify CE5 C0400 to be well-graded.The minerals in CE5 C0400 comprise approximately 44.5%pyroxene,30.4%plagioclase,3.6%olivine,and6.0%ilmenite.There is a relatively low content of approximately 15.5%glass phase in the CE-5 lunar sample.From the results,we deduce that the CE-5 LR structure could have mainly resulted from micrometeoroid impacts to achieve such a high level of maturity.Hui Zhang Xian Zhang Guang Zhang Keqi Dong Xiangjin Deng Xiaosong Gao Yaodong Yang Yuan Xiao Xiao Bai Kaixin Liang Yiwei Liu Wenbin Ma Shaofan Zhao Ce Zhang Xiaojing Zhang Jian Song Wei Yao Hong Chen Weihua Wang Zhigang Zou Mengfei Yang 2022Science China(Physics,Mechanics & Astronomy)2022,65,2:4
5Chronic intermittent hypoxia promotes the development of experimental non-alcoholic steatohepatitis by modulating Treg/Th17 differentiation显示文摘The present study aims to characterize the effect of chronic intermittent hypoxia and HIF1α on the non-alcoholic steatohepatitis (NASH)process in mice,and to explore the role of the Treg/Th17 balance in the formation of NASH inflammation and fibrosis.To achieve this purpose,simple steatosis was induced in mice by high-fat diet administration.Subsequently,chronic intermittent hypoxia was simulated by intraperitoneally injecting sodium nitrite.The changes of inflammation, fibrosis,and Treg/Th17 balance in the liver were.quantified under chronic intermittent hypoxia condition and after tail vein injection of HIF1α-siRNA.In addition,T cells were cultured in vitro, and HIF1α expression was either blocked or overexpressed under chronic intermittent hypoxia or normal conditions.Then,the changes of Treg/Th17 balance,inflammatory factors,and cell pathways were measured in each group.Our results demonstrated that chronic intermittent hypoxia accelerates the NASH process,while tail vein injection of HIF1α-siRNA improves liver histology and function.Chronic intermittent hypoxia alters the ratio of Th17 and Treg cells through HIF1α and roTOR signaling,and increases the expressions of NF-κB,IL-6,and IL-17,but decreases IL-10 expression.Inhibition of the mTOR-HIF1α-TLR4-IL-6 pathway increases the ratio of Treg/Th17. 6Concise and efficient direct-view generation of arbitrary cylindrical vector beams by a vortex half-wave plate显示文摘A concise, efficient, and practical direct-view scheme is presented to generate arbitrary cylindrical vector(CV)beams, including CV beams, vortex beams, and cylindrical vector vortex(CVV) beams, by a vortex half-wave plate(VHP). Six kinds of first-order and other high-order CV beams, such as azimuthally polarized(AP) beams, antivortex radial polarization mode beams, and three-order AP beams, are formed by simply rotating a half-wave plate. The Stokes parameters and double-slit interference of multitype CV beams are investigated in detail.The polarization parameters, including degree of polarization, polarization azimuth, and ellipticity, are obtained,which demonstrates the efficient generation of CV beams. In addition, the double-slit interference experiment is introduced in the setup, and fringe misplacement and tilt appear for CVV beams, in which the misplacement number M is 2P+1 for P ≤ 2 and 2P-1 for P ≥ 3, where P is the polarization order number, and the fringe tilt offset is positively related to the topological charge number l of CVV beams. In addition, new types of VHPs can be formed by cascading two or more VHPs when the types of available VHPs are limited, assisting in more flexible generation of multitype CV beams. 7A composite insulation structure for silicon-based planar neuroprobes显示文摘Silicon-based planar neuroprobes are composed of silicon substrate,conducting layer,and insulation layers of SiO 2 or SiN membrane.The insulation layer is very important because it affects many key parameters of neuprobes,like impedance,SNR(signal noise ratio),reliability,etc.Monolayer membrane of SiO 2 or SiN are not good choices for insulation layer,since defects and residual stress in these layers can induce bad passivation.In this paper a composite insulation structure is studied,with thermal SiO 2 as the lower insulation layer and with multilayer membrane composed of PECVD SiO 2 and SiN as the upper insulation layer.This structure not only solves the problem of residual stress but also ensures a good probe passivation.So it's a good choice for insulation layer of neuroprobes.ZHAO Hui PEI WeiHua CHEN SanYuan GUI Qiang TANG RongYu GUO Kai CHEN HongDa 2012Science China(Technological Sciences)2012,55,9:2
8Recruited CD68^(+)CD206^(+)macrophages orchestrate graft immune tolerance to prompt xenogeneic-dentin matrix-based tooth root regeneration显示文摘Successful regenerative medicine strategies of xenogeneic extracellular matrix need a synergistic balance among inflammation,fibrosis,and remodeling process.Adaptive macrophage subsets have been identified to modulate inflammation and orchestrate the repair of neighboring parenchymal tissues.This study fabricated PPARγ-primed CD68+CD206+M2 phenotype(M2γ),and firstly verified their anti-inflammatory and tissue-regenerating roles in xenogeneic bioengineered organ regeneration.Our results showed that Th1-type CD3^(+)CD8^(+)T cell response to xenogeneic-dentin matrix-based bioengineered root complex(xeno-complex)was significantly inhibited by M2γmacrophage in vitro.PPARγactivation also timely recruited CD68^(+)CD206^(+)tissue macrophage polarization to xeno-complex in vivo.These subsets alleviated proinflammatory cytokines(TNF-α,IFN-γ)at the inflammation site and decreased CD3^(+)CD8^(+)T lymphocytes in the periphery system.When translated to an orthotopic nonhuman primate model,PPARγ-primed M2 macrophages immunosuppressed IL-1β,IL-6,TNF-α,MMPs to enable xeno-complex to effectively escape immune-mediated rejection and initiate graft-host synergistic integrity.These collective activities promoted the differentiation of odontoblast-like and periodontal-like cells to guide pulp-dentin and cementum-PDLs-bone regeneration and rescued partially injured odontogenesis such as DSPP and periostin expression.Finally,the regenerated root showed structure-biomechanical and functional equivalency to the native tooth.The timely conversion of M1-to-M2 macrophage mainly orchestrated odontogenesis,fibrogenesis,and osteogenesis,which represents a potential modulator for intact parenchymal-stromal tissue regeneration of targeted organs.Hui Li Jingjing Sun Hefeng Yang Xue Han Xiangyou Luo LiJun Liao Bo Yang Tian Zhu Fangjun Huo Weihua Guo Weidong Tian 2021Bioactive Materials2021,6,4:2

11Blood flow activation in rat somatosensory cortex under sciatic nerve stimulation revealed by laser speckle imaging显示文摘In many functional neuroimaging research the change of local cerebral blood flow (CBF) induced by sensory stimulation is regarded as an indicator of the change in cortical neuronal activity although a precise and full spatio-temporal description of local CBF response coupled to neural activity has still not been laid out. Using the laser speckle imaging technique a relatively large exposed area in somatosensory cortex of rat was imaged for the observation of the variations of CBF during sciatic nerve stimulation. The results showed that cerebral blood flow activation was spatially localized and discretely distributed in the targeted microvasculature. Individual arteries, veins and capillaries in different diameters were activated with the time going. 19A novel algorithm for transmission image reconstruction of tomographic gamma scaners显示文摘It is difficult to develop image reconstruction algorithms for tomographic gamma scanning based on drummed radioactive residues or wastes.In this paper,a novel reconstruction algorithm of transmission image for tomographic gamma scanning is proposed.It is based on the conventional transmission equation and equivalent gamma-ray track length modified by a Monte Carlo method.The algorithm is implemented by simulating the samples on the established platform.For the verification experiments of the algorithm,several cubic voxel samples were designed and manufactured.Experimental tests were conducted.The tomographic gamma scanning of transmission images is compared with the linear attenuation coefficients by the simulated values and experimental data with the algorithm and the reference values.The results show that the absolute relative errors of the reconstructed images are less than 5%.ZHANG Quanhu HUI Weihua WANG Dong HE Bin GU Zhongmao LI Ze QIAN Shaojun 2010Nuclear Science and Techniques2010,21,3:1
