1Regulatory effects of antitumor agent matrine on FOXO and PI3K-AKT pathway in castration-resistant prostate cancer cells显示文摘We previously demonstrated that matrine could inhibit the proliferating, migrating, as well as invading processes of both PC-3 and DU145 cells. However, the underlying molecular mechanisms have not yet been clearly defined. In this study, using various techniques such as high throughput sequencing technology, bioinformatics, quantitative real-time PCR, and immunoblot analysis,we aimed to understand whether matrine serves as a novel regulator of FOXO and PI3K-AKT signaling pathway. DU145 and PC-3 cell lines were cultured for 24 h in vitro. Cells were treated with either matrine or control serum for 48 h, followed by extraction of total RNA. The RNA was sequenced using HiSeq 2500 high-throughput sequencing platform (Illumina). A gene library was established and quality analysis of read data carried out. Integrated database from the website DAVID was used to analyze Gene Ontology (GO), and Kyoto encyclopedia of genes and genomes (KEGG) pathway of differential genes was used for pathway analysis, screening for fold differences of more than two times. The FOXO and PI3K-AKT signaling pathways were screened, and expression levels of mRNA and core protein detected by real-time PCR and immunoblotting, respectively. High throughput sequencing and GO analysis revealed that differentially expressed genes before and after treatment played an important role in cell metabolic process, growth process, anatomical structure formation, cellular component organization, and biological regulation. KEGG signal pathway analysis revealed that FOXO and PI3K-AKT signal pathways had a significant difference between before and after matrine-treated androgen-independent prostate cancer cells PC-3 and DU145. Real-time PCR showed that matrine treatment led to a significant increase in the expression levels of FOXO1A, FOXO3A, FOXO4, and FOXO6 in DU145 and PC-3 cells (P<0.01 or P<0.05), whereas the PI3K expression levels decreased (P<0.01). Similarly, immunoblotting revealed a significant increase (P<0.05) in the expression levels of FOXO1A FOXO3A, FOXO4, and FOXO6 in both PC-3 and DU145 cells, whereas PI3K expression levels decreased (P<0.05). Matrine had a broad regulating effect on the mRNA expression profiles of both PC-3 and DU145 cells. Matrine may inhibit cell proliferation, migration, as well as invasion, and induce apoptosis in both PC-3 and DU145 cells through FOXO and PI3K-AKT signaling pathways. 2Retrieving crop leaf area index by assimilation of MODIS data into a crop growth model显示文摘Leaf area index (LAI) is an important parameter in monitoring crop growth. One of the methods for retrieving LAI from remotely sensed observations is through inversion of canopy reflectance models. Many model inversion methods fail to account for variable LAI values at different crop growth stages. In this research, we use the crop growth model to describe the LAI changes with crop growth, and consider a priori LAI values at different crop growth stages as constraint information. The key approach of this research is to assimilate multiple canopy reflectance values observed at different growth stages and a priori LAI values into a coupled crop growth and radiative transfer model sequentially using a variational data assimilation algorithm. Adjoint method is used to minimize the cost function. Any other information source can be easily incorporated into the inversion cost function. The validation results show that the time series of MODIS canopy reflectance can greatly reduce the uncertainty of the inverted LAI values. 3Integrated Chinese-western therapy versus western therapy alone on survival rate in patients with non-small-cell lung cancer at middlelate stage显示文摘OBJECTIVE: To compare the effects of integrated Chinese-Western therapy versus Western therapy alone on the survival rate of patients with non-small-cell lung cancer (NSCLC) at middle-late stage and to evaluate prognostic factors. METHODS: We selected 98 inpatients with middle-late stage NSCLC diagnosed from March 2009 to March 2011 and randomly divided them into two groups, with 49 cases in each group, and the clinical data were analyzed retrospectively.The control group was treated by the combined methods of Western Medicine, including chemotherapy, supportive treatment and symptomatic treatment. The observation group was treated by injection and prescriptions of Chinese medicine based on Traditional Chinese Medicine syndrome differentiation and by the same combined methods of western treatment used in the control group. After treatment, the survival rates of the patients were compared by the stage of cancer and evaluation of 24 prognostic factors analyzed by a Cox regressionmodel, and the clinical data were statistically analyzed. RESULTS: The survival rates of all patients were over 90.0% at 1 and 3 months after treatment with no significant differences between the two groups (P>0.05); In the observation group the survival rates at 6 months and 1 year were 93.4% and 42.8%, respectively, being superior to 85.6% and 18.3% in the control group (P<0.05). The median survival time in the observation group was superior to the control group (P<0.05); The effects of 24 prognostic factors were significantly better in the observation group than in the control group (P<0.05). 4Aerodynamic characteristics research on wide-speed range waverider configuration显示文摘Waverider generated from a given flow field has a high lift-to-drag ratio because of attached bow shock on leading edge. However, leading edge blunt and off-design condition can make bow shock off leading edge and have unfavorable influence on aerodynamic characteristics. So these two problems have always been concerned as important engineering science issues by aeronautical engineering scientists. In this paper, through respectively using low speed and high speed waverider design principles, a wide-speed rang vehicle is designed, which can level takeoff and accelerate to hypersonic speed for cruise. In addition, sharp leading edge is blunted to alleviated aeroheating. 5Discussions on real-world acupuncture treatments for chroni clow-back pain in older adults显示文摘Chronic low-back pain(CLBP) is one of the most common pain conditions. Current clinical guidelines for low-back pain recommend acupuncture for CLBP. However, there are very few high-quality acupuncture studies on CLBP in older adults. Clinical acupuncture experts in the American Traditional Chinese Medicine Association(ATCMA) were interested in the recent grant on CLBP research announced by the National Center for Complementary and Integrative Health. The ATCMA experts held an online discussion on the subject of real-world acupuncture treatments for CLBP in older adults. Seven participants, each with more than 20 years of acupuncture practice, discussed their own unique clinical experience while another participant talked about the potential mechanism of acupuncture in pain management. As a result of the discussion, a picture of a similar treatment strategy emerged across the participants for CLBP in older adults. 6lonization and dissociation of acetonitrile by intense femtosecond laser pulse显示文摘Photoionization and photodissociation of CH3CN were studied by a linear time of flight mass spectrometer coupled with 800 nm, 50 fs laser pulses at intensities of 6.3×1013-1.2×1014 W/cm2. The laser power dependences for principal ions CH3CN+, CH2CN+, CHCN+ and CCN+ were measured, which are consistent with the numbers of photons required to produce the ions via multiphoton ionization and dissociation. 11SUSPENDED SEDIMENT TRANSPORT CAPACITY OF OPEN CHANNEL FLOW显示文摘A turbulent bursting-based formulation for suspended sediment transport capacity is verified with data from Brownlie (1981). Previous formulations are examined using the results of data analysis. Satisfactory performance of the turbulent bursting-based formulation is demonstrated, in addition to its strong physical background. Einstein’s formula is found to over-or under-estimate sediment transport capacity in some cases. Celik and Rodi’s correlation is shown to behave poorly. The WUHEE curve does not fit the measured data well. Further, the transport capacity is exhibited to increase nonlinearly with the nondimensional quantity u3 /ghw. This behavior upsets many existing formulas, in which the transport capacity is represented as linear functions of U3 /ghw. 16Effectiveness of two different acupuncture strategies in patients with vulvodynia Study protocol for a pilot pragmatic controlled trial显示文摘Background: Vulvodynia, or vulvar pain, is a common condition in women; however, there are few evidence-based clinical trials evaluating nonpharmacological therapies for this condition. Acupuncture is one complementary and integrative medicine therapy used by some patients with vulvodynia. This study evaluates two different acupuncture strategies for the treatment of vulvodynia and aims to evaluate whether either of the acupuncture protocols reduces vulvar pain, pain duration or pain with intercourse. The study also examines how long the effect of acupuncture lasts in women with vulvodynia.Methods/design: The study is designed as a randomized controlled trial, focused on two acupuncture protocols. Fifty-one patients who have had vulvodynia for more than 3 months will be recruited.Among them, 34 patients will be randomized into Groups 1 a and 1 b; those who are unwilling to receive acupuncture will be recruited into the standard care group(Group 2). Patients in Group 1 a will have acupuncture focused on the points in the pudendal nerve distribution area, while patients in Group 1 b will receive acupuncture focused on traditional(distal) meridian points. Patients in Group 2 will receive routine conventional treatments, such as using pain medications, local injections and physical therapies or other nonsurgical procedures. Acupuncture will last 45 min per session, once or twice a week for6 weeks. The primary outcome measurement will be objective pain intensity, using the cotton swab test.The secondary outcome measurement will be subjective patient self-reported pain intensity, which will be conducted before cotton swab test. Pain intensities will be measured by an 11-point Numeric Pain Rating Scale. Pain duration and pain score during intercourse are recorded. Local muscle tension, tenderness and trigger points(Ashi points) are also recorded. 17Superior performance of a 2 kHz pulse Nd:YAG laser based on a gradient-doped crystal显示文摘Herein,we report a homemade new Nd:YAG crystal rod that contains a gradient dopant of 0.39–0.80 at.%Nd3+ from end to end,achieving superior performance of a 2 kHz Nd:YAG pulse laser at 1064 nm. The optical-tooptical conversion efficiency reached 53.8%,and the maximum output power of the laser was 24.2 W,enhanced by 35.9% compared with a uniform crystal rod with the same total concentration of Nd3+. Significantly,our experiments revealed that the gradient concentration crystal produced a relatively even pumping distribution along the rod axis,greatly reducing the temperature gradient as well as having a smaller thermal effect. 18Spectroscopic analysis of high electric field enhanced ionization in laser filaments in air for corona guiding显示文摘We report on a systematic experimental study on the fluorescence spectra produced from a femtosecond laser filament in air under a high electric field. The electric field alone was strong enough to create corona discharge(CD). Fluorescence spectra from neutral and ionic air molecules were measured and compared with pure high-voltage CD and pure laser filamentation(FIL). Among them, high electric field assisted laser FIL produced nitrogen fluorescence more efficiently than either pure CD or pure FIL processes. The nonlinear enhancement of fluorescence from the interaction of the laser filament and corona discharging electric field resulted in a more efficient ionization along the laser filament zone, which was confirmed by the spectroscopic measurement of both ionization-induced fluorescence and plasma-scattered 800 nm laser pulses.
