| 2 | Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. | 4 | Analysis of genetic architecture and favorable allele usage of agronomic traits in a large collection of Chinese rice accessions显示文摘Genotyping and phenotyping large natural populations provide opportunities for population genomic analysis and genome-wide association studies(GWAS). Several rice populations have been re-sequenced in the past decade;however, many major Chinese rice cultivars were not included in these studies. Here, we report large-scale genomic and phenotypic datasets for a collection mainly comprised of 1,275 rice accessions of widely planted cultivars and parental hybrid rice lines from China. The population was divided into three indica/Xian and three japonica/Geng phylogenetic subgroups that correlate strongly with their geographic or breeding origins. We acquired a total of 146 phenotypic datasets for 29 agronomic traits under multi-environments for different subpopulations. With GWAS, we identified a total of 143 significant association loci, including three newly identified candidate genes or alleles that control heading date or amylose content. Our genotypic analysis of agronomically important genes in the population revealed that many favorable alleles are underused in elite accessions, suggesting they may be used to provide improvements in future breeding efforts. | 5 | Effect of Ginkgo biloba leaf extract on cerebral cortex amino acid levels in cerebral ischemia model rats显示文摘OBJECTIVE: To investigate the effect of Ginkgo biloba leaf extract on amino acid levels in the cerebral cortex of cerebral ischemia model rats induced by middle cerebral artery occlusion(MCAO).METHODS: A rat model of cerebral ischemia was established by MCAO. Male rats were divided into a negative control group(Control), a sham-operated group(Sham), an ischemic group(MCAO), and an ischemic group treated with Ginkgo biloba leaf extract(MCAO_D). All groups were divided into two subgroups with occlusion times of 12 and 24 h, respectively. The levels of 18 endogenous amino acids in the cerebral cortex were quantified by triple quadrupole-liquid chromatography-mass spectrometry.RESULTS: Compared with the MCAO group, behavioral performance, neurological deficit score, and cerebral infarct volume were significantly improved in the MCAO_D group(P < 0.05, P < 0.01). Compared with the sham group, the levels of 17 amino acids in the cerebral cortex were markedly changed in the MCAO group. The levels of Alanine(Ala), Isoleucine(Ile), Glutamic acid(Glu), Serine(Ser), Valine(Val), Phenylalanine(Phe), Proline(Pro),Threonine(Thr), Lysine(Lys), Tyrosine(Tyr), Hydroxyproline(Hyp), Arginine(Arg), Leucine(Leu),Tryptophan(Trp), and Glycine(Gly) were increased(P < 0.001, P < 0.05), while levels of Gln and Tau were decreased(P < 0.001, P < 0.05). Compared with the MCAO group, Ginkgo biloba extract treatment in the MCAO_D group significantly down-regulated the levels of 11 amino acids, especially those of Arg, Thr, and Ser in 12 or 24 h.CONCLUSION: Injection of Ginkgo biloba leaf extract has a therapeutic effect on model rats with MCAO-induced cerebral ischemia by acting on amino acids in the cerebral cortex. | 6 | The study on seasonal characteristics of water masses in the western East China Sea shelf area显示文摘On the basis of the CTD data and the modeling results in the winter and summer of 2009, the seasonal characteristics of the water masses in the western East China Sea shelf area were analyzed using a cluster analysis method. The results show that the distributions and temperature-salinity characteristics of the water masses in the study area are of distinct seasonal difference. In the western East China Sea shelf area, there are three water masses during winter, i.e., continental coastal water(CCW), Taiwan Warm Current surface water(TWCSW) and Yellow Sea mixing water(YSMW), but four ones during summer, i.e., the CCW, the TWCSW, Taiwan Warm Current deep water(TWCDW) and the YSMW. Of all, the CCW, the TWCSW and the TWCDW are all dominant water masses. The CCW, primarily characterized by a low salinity, has lower temperature, higher salinity and smaller spatial extent in winter than in summer. The TWCSW is warmer, fresher and smaller in summer than in winter, and it originates mostly from the Kuroshio surface water(KSW) northeast of Taiwan, China and less from the Taiwan Strait water during winter, but it consists of the strait water and the KSW during summer. | 7 | Activity of cold vortex in northeastern China and its connection with the characteristics of precipitation and circulation during 1960-2012显示文摘Based on the daily reanalysis data released by NCEP/NCAR and the daily precipitation of 753 Chinese stations from May to August during the period of 1960 to 2012, the statistical characteristics of the cold vortex in northeastern China were analyzed. In addition, the strength index, which described the characteristics of the vortex consistently and frequently, and the geographical distribution were given by continuous anomalies of circulation. Based on this index, the activity routines of the cold vortex, characteristics of atmospheric circulation, and their effects on precipitation in northeastern China were analyzed. The results show that: the activities of the cold vortex exhibit remarkable features of annual and interdecadal oscillation, and the vortex high frequency and its characteristics of atmospheric circulation are described more accurately by the strength index of the cold vortex, which shows a high correspondence with the vortex precipitation during early summer and midsummer in the northeast. In strong(weak) vortex years, the general circulation in the middle and high latitudes of Eurasia is to the advantage(disadvantage) of the formation, development and maintenance of the cold vortex, thus it is easy(difficult) to form the circulation which is beneficial to transmit vapor from south to north during the period of July to August. Blocking over the Ural Mountains prevails(does not prevail) in early summer, and blocking over the Sea of Okhotsk prevails(does not prevail) in midsummer. | 9 | Fe-based metallic glass:An efficient and energy-saving electrode material for electrocatalytic degradation of water contaminants显示文摘Excessive consumption of electrical energy has hampered the widespread application of electrochemical technology for degradation of various contaminants. In this paper, a Fe-based metallic glass(MG) was demonstrated as a new type of electrocatalyst to effectively and economically degrade an azo dye. In comparison to other typical electrodes, Fe-based MG electrodes exhibit a minimized degradation time, and the specific energy is 4-6 orders of magnitude lower than that of dimensionally stable anode(DSA), metal-like boron-doped diamond(BDD) and other electrodes. As sacrificial electrode materials, Fe-based MGs have less specific electrode mass consumption than iron electrodes. | 10 | A novel cancer vaccine with the ability to simultaneously produce anti-PD-1 antibody and GM-CSF in cancer cells and enhance Th1-biased antitumor immunity显示文摘Tumor escape from immune-mediated destruction has been associated with immunosuppressive mechanisms that inhibit T-cell activation.A promising strategy for cancer immunotherapy is to disrupt key pathways regulating immune tolerance,such as program death-1(PD-1/PD-L1)pathway in the tumor environment.However,the determinants of response to anti-PD-1 monoclonal antibodies(mAbs)treatment remain incompletely understood.In murine models,PD-1 blockade alone fails to induce effective immune responses to poorly immunogenic tumors,but is successful when combined with additional interventions,such as cancer vaccines.Novel cancer vaccines combined with antibody may offer promising control of cancer development and progression.In this investigation,we generated a novel tumor cell vaccine simultaneously expressing anti-PD-1 mAbs and granulocyte-macrophage colony stimulating factor(GM-CSF)in CT26 colon cancer and B16-F10 melanoma.The antitumor effect of the vaccine was verified by therapeutic and adoptive animal experiments in vivo.The antitumor mechanism was analyzed using Flow cytometry,Elispot and in vivo intervention approaches.The results showed that tumor cell vaccine secreting PD-1 neutralizing antibodies and GM-CSF induced remarkable antitumor immune effects and prolonged the survival of tumor-bearing animals compared with animals treated with either PD-1 mAbs or GM-CSF alone.Antitumor effects and prolonged survival correlated with strong antigen-specific T-cell responses by analyzing CD11c^(+)CD86^(+)DC,CD11b^(+)F4/80^(+)MΦcells,increased ratio of Teff/Treg in the tumor microenvironment,and higher secretion levels of Th1 proinflammatory cytokines in serum.Furthermore,the results of ELISPOT and in vivo blocking strategies further confirmed that the antitumor immune response is acquired by CD4 and CD8 T immune responses,primarily dependent on CD4 Th1 immune response,not NK innate immune response.The combination of PD-1 blockade with GM-CSF secretion potency creates a novel tumor cell vaccine immunotherapy,affording significantly improved antitumor responses by releasing the state of immunosuppressive microenvironment and augmenting the tumor-reactive T-cell responses. | Hongwei Tian Gang Shi Qin Wang Yiming Li Qianmei Yang Chunlei Li Guoyou Yang Min Wu Qian Xie Shuang Zhang Yang Yang Rong Xiang Dechao Yu Yuquan Wei Hongxin Deng | 2016 | Signal Transduction and Targeted Therapy2016,1,1: | 3 |

| 14 | Mechanism and kinetics of treatment of acid orange II by aged Fe-Si-B metallic glass powders显示文摘The mechanism and kinetics of acid orange II(AOII) treated by aged gas-atomized Fe-Si-B metallic glass(MG) powders were investigated in this study. The decolorization reaction is shown to obey the pseudofirst-order kinetic model, and the treatment processes could be divided into two stages: a slow step followed by a rapid one. This observation is in accordance with the following results, the azo dye is simply adsorbed onto the Fe-based MG powders in the initial stage, because the oxide layer coated on the powder surface depresses the degradation reaction by covering the activity sites, and then the degradation occurs with the desquamation of the powders. The AOII could be degraded with a rapid reaction rate when the Fe-based MG powders are applied to the treatment process again, because of the consumption of the oxide layer and the unchanged core of the Fe-based MGs. Experimental results show that the resistances for PrFeO_3 and NdFeO_3 in CO_2 gas are larger than those in air and the responses for PrFeO_3and NdFeO_3 sensors increase with an increase in room-temperature relative humidity. When exposed to1000 ppm CO_2, the response of PrFeO_3 thick film based on nano-powders annealed at 700℃can reach8.44 at 160℃for the background of wet air with 58%of room-temperature relative humidity (RH),which is much larger than the corresponding value (3.03) in wet air with 25%RH. The sensing response S of NdFeO_3 thick-film sensor based on nano-powders annealed at 600℃to 3000 ppm CO_2 at the operating temperature 200℃can reach 2.36 for the background of wet air with 72%RH, which is larger than the corresponding value (1.83) in the air with 25%RH. | 16 | Yield photosynthesis and leaf anatomy of maize in inter-and mono-cropping systems at varying plant densities显示文摘Increasing plant density can increase cereal crop yields. However, the physiological and anatomical mechanisms of grain yield increase at high plant densities in maize-based intercropping systems are not well understood. A two-year field experiment was conducted in 2018 and 2019 to investigate grain yield, photosynthetic characteristics, stomatal traits, and leaf anatomy of maize plants in an intercropping system with high plant densities. Two cropping patterns(monocropping and intercropping) and three plant densities(D1, 78,000 plants ha^(-1);D2, 103,500 plants ha^(-1);D3, 129,000 plants ha;were arranged in a randomized block design. Increasing maize plant density significantly increased maize yield, and intercropping gave a significant yield advantage over monocropping under the same plant density. Intercropping combined with high plant density increased the leaf area and SPAD value of maize,increasing the photosynthesis rates after the harvest of pea. At the twelfth leaf stage, the stomatal density and stomatal area of intercrops combined with medium plant density increased by respectively 10.5%and 18.4% relative to their values for the corresponding density of monocrops. Although leaf thickness of maize was reduced by increasing plant density, the chloroplast number and grana lamella number were higher in intercropping than in monocropping under different plant densities. | 17 | Dasatinib inhibits proliferation of liver cancer cells,but activation of Akt/mTOR compromises dasatinib as a cancer drug显示文摘Dasatinib is a multi-target protein tyrosine kinase inhibitor.Due to its potent inhibition of Src,Abl,the platelet-derived growth factor receptor(PDGFR)family kinases,and other oncogenic kinases,it has been investigated as a targeted therapy for a broad spectrum of cancer types.However,its efficacy has not been significantly extended beyond leukemia.The mechanism of resistance to dasatinib in a wide array of cancers is not clear.In the present study,we investigated the effect of dasatinib on hepatocellular carcinoma cell growth and explored the underlying mechanisms.Our results showed that dasatinib potently inhibited the proliferation of SNU-449 cells,but not that of other cell lines,such as SK-Hep-1,even though it inhibited the phosphorylation of Src on both negative and positive regulation sites in all these cells.Dasatinib activated the phosphoinositide-dependent protein kinase1(PDK1)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR)signaling pathway in SK-Hep-1 cells,but not in SNU-449 cells.Blocking the Akt/mTOR signaling pathway strongly promoted the efficacy of dasatinib in SK-Hep-1 cells.In SNU-449 cells,dasatinib promoted apoptosis and the cleavage of caspase-3 and caspase-7,induced cell cycle arrest in the G1 phase,and inhibited the expression of Cyclin-dependent kinase(CDK4)/6/CyclinD1 complex.These findings demonstrate that dasatinib exerts its anti-proliferative effect on hepatocellular cell proliferation by blocking the Src family kinases;however,it causes Akt activation,which compromises dasatinib as an anti-cancer drug. | Chang Liu Xiaoxia Zhu Yuqi Jia Fenqing Chi Keru Qin Jinhong Pei Chan Zhang Xiuli Mu Hongwei Zhang Xiushan Dong Jun Xu Baofeng Yu | 2021 | Acta Biochimica et Biophysica Sinica2021,53,7: | 2 |
| 18 | Construction of Highly Emissive Pt(Ⅱ) Metallacycles upon Irradiation显示文摘Photoswitchable or photoactivatable fluorescent species have been found wide applications within supramolecular chemistry and materials science.In this study,we successfully constructed two highly emissive Pt(Ⅱ)metallacycles from the diarylethene ligands via coordination-driven self-assembly.Different from the most known fluorescent metallacycles,the obtained metallacycles have displayed'turn-on'fluorescence switching.They are non-fluorescent in solution,but they emit highly yellow or orange fluorescence under ultraviolet irradiation.The metallacycles were well characterized by 1H NMR,31P NMR and ESI-TOF-MS.The photochromic properties of the resultant metallacycles were investigated by 1H NMR,31P NMR,UV/Vis spectrum and fluorescence spectrum.Notably,NMR studies revealed that these two metallacycles featured excellent cyclization efficiency(90% conversion efficiency).Moreover,the closed-ring isomers of the metallacycles displayed relatively high quantum yield(ФF=0.5).DFT simulations demonstrated that the antiparallel configuration of the diarylethene ligand had an angle closed to 120°,which was more stable in energy compared to the parallel configuration,thus allowing for the facile construction of highly emissive metallacycles.We believe that such highly emissive metallacycles which are in-situ prepared upon irradiation can be used as new fluorescence materials for sensing and bioimaging in the future. | Yi Qin Ying Zhang Guangqiang Yin Yuxuan Wang Changwei Zhang Lijun Chen Hongwei Tan Xiaopeng Li Lin Xu Haibo Yang | 2019 | Chinese Journal of Chemistry2019,37,4: | 2 |
