| 1 | Effects of dietary arginine levels on growth performance,body composition, serum biochemical indices and resistance ability against ammonia-nitrogen stress in juvenile yellow catfish(Pelteobagrus fulvidraco)显示文摘This experiment was conducted to investigate the effects of dietary arginine levels on growth performance, body composition, serum biochemical indices and resistance ability against ammonia-nitrogen stress in juvenile yellow catfish(Pelteobagrus fulvidraco). Five isonitrogenous and isolipidic diets(42%protein and 9% lipid) were formulated to contain graded levels of arginine(2.44%, 2.64%, 2.81%, 3.01% and3.23% of diet), by supplementing L-Arginine HCI. Seven hundred juvenile yellow catfish with an initial average body weight of 1.13 ± 0.02 g were randomly divided into 5 groups with 4 replicates of 35 fish each and each group was fed one of the diets. After 56 d feeding, fish were exposed to 100 mg/L of ammonia-nitrogen for 72 h. The results showed that weight gain(WG) and specific growth rate(SGR) in2.64% and 2.81% groups were significantly higher than those in 3.23% group(P < 0.05). The feed conversation ratio(FCR) in 2,64%, 2,81% and 3.01% groups was significantly decreased when compared with3,23% group. The protein efficiency ratio(PER) in 2.64% group was significantly higher than that in 2.44%and 3.23% groups(P < 0.05). The condition factor(CF) of fish was significantly higher in 2.81% group than that in 2,44% group(P < 0.05). Dietary arginine levels had no significant effect on hepatosomatic index(HSI), viscerosomatic index(VSI), and whole-body dry matter, crude protein, crude lipid, ash contents, as well as serum total protein(TP), triglyceride(TG), glucose(GLU), urea nitrogen(UN) contents and aspartate aminotransferase(AST), alanine aminotransferase(ALT) activities(P> 0.05). After the fish were challenged to ammonia-nitrogen for 72 h, their cumulative mortality rate in 2.81% group was significantly lower than that in 2.44% group(P < 0.05). The results suggested that dietary arginine level at 2.81%could optimize anti-ammonia-nitrogen stress ability of juvenile yellow catfish and a level of 3.23%arginine seemed to depress the growth performance of fish and decreased their tolerance to the ammonia-nitrogen stress under current study. | 3 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 4 | Modeling and boundary control of a flexible marine riser coupled with internal fluid dynamics显示文摘In this paper, vibration reduction of a flexible marine riser with time-varying internal fluid is studied by using boundary control method and Lyapunov's direct method. To achieve more accurate and practical riser's dynamic behavior, the model of marine riser with time-varying internal fluid is modeled by a distributed parameter system (DPS) with partial differential equations (PDEs) and ordinary differential equations (ODEs) involving functions of space and time. The dynamic responses of riser are completely different if the time-varying internal fluid is considered. Boundary control is designed at the top boundary of the riser based on original infinite dimensionality PDEs model and Lyapunov's direct method to reduce the riser's vibrations. The uniform boundedness and closed-loop stability are proved based on the proposed boundary control. | 5 | Effects of elevated CO_2 on the nutrient compositions and enzymes activities of Nilaparvata lugens nymphs fed on rice plants显示文摘Elevated CO 2 may reduce the tolerance of Nilaparvata lugen (N. lugens) to adverse environmental factors through the biological and physiological degeneration of N. lugens. In an artificial climate box, under 375 and 750 μL L 1 CO 2 levels, the rice stems nutrient content, the nutrient content and enzyme activities of N. lugens nymph fed on rice seedlings exposed to ambient and elevated CO 2 were studied. The results showed that rice stems had significantly higher protein and total amino acid levels under ambient than elevated CO 2 levels. Nymphs had significantly higher protein levels in the ambient CO 2 treatment, while their glucose levels were significantly lower under ambient CO 2 conditions. Significantly higher trypsin activity was observed in nymphs grown in elevated CO 2 . Significantly lower activities of the protective enzymes total superoxide dismutase and catalase were observed in the nymphs under ambient CO 2 . Meanwhile, the activity of the detoxification enzyme glutathione S-transferase was significantly higher in the ambient CO 2 treatment. | 6 | First observation of Karlodinium venef icum from the East China Sea and the coastal waters of Germany显示文摘Harmful algal blooms often occurred in the East China Sea (ECS) and the German coastal waters of the North Sea but Karlodinium veneficum had not been taxonomically reported. Two strains of Karlodinium (LAMB090611 and LAMB010601) were isolated from the two areas in 2009. The morphological characteristics and molecular phylogeny of two strains are compared on the basis of observation of a light microscope, a scanning electron microscope (SEM), a laser scanning micro-scope (LSM) and an internal transcribed spacer (ITS) sequence data. The mean cell length of strain LAMB090611 is (14.2 ± 1.8) μm (range 11.1-18.7 μm) and mean width is (10.8 ± 1.5) μm (range 8.2- 14.7 μm). The mean cell length of strain LAMB010601 is (15.1 ± 1.2) μm (range 12.7-17.9 μm) and the mean width is (11.4 ± 1.1) μm (range 9.1-14.7 μm), respectively. The two strains are similar in morphological characteristics, including a straight apical groove, distinct ventral pore, sulcal extension, cingulum displacement, two or four irregular shaped chloroplasts within the cell and almost equal sized epicone and hypocone. The large and round nucleus is located at the center or at the hypocone of the cell. The sequence length of the ECS strain LAMB090611 and the German strain LAMB010601 is 640 and 646 bp, respectively. The GC content is 49%. The nucleotide similarity of the two strains is 98.1%. The sequence divergence is 0.003. Both strains are confirmed as Karlo-dinium veneficum (D. Ballantine) J. Larsen and this is the first taxonomic report from China and Germany coastal waters. | 7 | Prognostic assessment of apoptotic gene polymorphisms in non-small cell lung cancer in Chinese显示文摘Apoptosis plays a key role in inhibiting tumor growth, progression and resistance to anti-tumor therapy. We hypothesized that genetic variants in apoptotic genes may affect the prognosis of lung cancer. To test this hypothesis, we selected 38 potentially functional single nucleotide polymorphisms (SNPs) from 12 genes (BAX, BCL2, BID, CASP3, CASP6, CASP7, CASP8, CASP9, CASP10, FAS, FASLG and MCL1) involved in apoptosis to assess their prognostic significance in lung cancer in a Chinese case cohort with 568 non-small cell lung cancer (NSCLC) patients. Thirty-five SNPs passing quality control underwent association analyses, 11 of which were shown to be significantly associated with NSCLC survival (P<0.05). After Cox stepwise regression analyses, 3 SNPs were independently associated with the outcome of NSCLC (BID rs8190315: P=0.003; CASP9 rs4645981: P=0.007 and FAS rs1800682: P=0.016). A favorable survival of NSCLC was significantly associated with the genotypes of BID rs8190315 AG/GG (adjusted HR=0.65, 95% CI: 0.49-0.88), CASP9 rs4645981 AA (HR=0.22, 95% CI: 0.07-0.69) and FAS rs1800682 GG (adjusted HR=0.67, 95% CI: 0.46-0.97). Time-dependent receptor operation curve (ROC) analysis revealed that the area under curve (AUC) at year 5 was significantly increased from 0.762 to 0.819 after adding the risk score of these 3 SNPs to the clinical risk score. The remaining 32 SNPs were not significantly associated with NSCLC prognosis after adjustment for these 3 SNPs. | 8 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |

While changing the content of polyvinylpyrrolidone, we found that carbon content was adjustable on the surface of lithium vanadium phosphate. By optimizing the carbon content among the composites, the electrochemical performance can be enhanced significantly. The results of electrochemical performance tests suggested that the samples exhibited good cycle performance and high discharge capability in the voltages between 3.0-4.8 V. | 14 | All-solid-state flexible asymmetric supercapacitors with high energy and power densities based on NiCo_2S_4@MnS and active carbon显示文摘Electrode material based on a novel core–shell structure consisting of NiCo_2S_4(NCS) solid fiber core and Mn S(MS) sheet shell(NCS@MS) in situ grown on carbon cloth(CC) has been successfully prepared by a simple sulfurization-assisted hydrothermal method for high performance supercapacitor. The synthesized NiCo_2S_4@Mn S/CC electrode shows high capacitance of 1908.3 F g^(-1) at a current density of 0.5 A g^(-1) which is higher than those of NiCo_2S_4 and Mn S at the same current density. A flexible all-solid-state asymmetric supercapacitor(ASC) is constructed by using NiCo_2S_4@Mn S/CC as positive electrode, active carbon/CC as negative electrode and KOH/poly(vinyl alcohol)(PVA) as electrolyte. The optimized ASC shows a maximum energy density of 23.3 Wh kg^(-1) at 1 A g^(-1), a maximum power density of about7.5 kw kg^(-1) at 10 A g^(-1) and remarkable cycling stability. After 9000 cycles, the ASC still exhibited67.8% retention rate and largely unchanged charge/discharge curves. The excellent electrochemical properties are resulted from the novel core–shell structure of the NiCo_2S_4@Mn S/CC electrode, which possesses both high surface area for Faraday redox reaction and superior kinetics of charge transport. | 19 | Association of assisted reproductive technology, germline de novo mutations and congenital heart defects in a prospective birth cohort study显示文摘Emerging evidence suggests that children conceived through assisted reproductive technology(ART)have a higher risk of congenital heart defects(CHDs)even when there is no family history.De novo mutation(DNM)is a well-known cause of sporadic congenital diseases;however,whether ART procedures increase the number of germline DNM(gDNM)has not yet been well studied.Here,we performed whole-genome sequencing of 1137 individuals from 160 families conceived through ART and 205 families conceived spontaneously.Children conceived via ART carried 4.59 more gDNMs than children conceived spontaneously,including 332 paternal and 1.26 maternal DNMs,after correcting for parental age at conception,cigarette smoking,alcohol drinking,and exercise behaviors.Paternal DNMs in offspring conceived via ART are characterized by C>T substitutions at CpG sites,which potentially affect protein-coding genes and are significantly associated with the increased risk of CHD.In addition,the accumulation of non-coding functional mutations was independently associated with CHD and 87.9% of the mutations were originated from the father.Among ART offspring,infertility of the father was associated with elevated paternal DNMs;usage of both recombinant and urinary follicle-stimulating hormone and high-dosage human chorionic gonadotropin trigger was associated with an increase of maternal DNMs.In sum,the increased gDNMs in offspring conceived by ART were primarily originated from fathers,indicating that ART itself may not be a major reason for the accumulation of gDNMs.Our findings emphasize the importance of evaluating the germline status of the fathers in families with the use of ART. | Cheng Wang Hong Lv Xiufeng Ling Hong Li Feiyang Diao Juncheng Dai Jiangbo Du Ting Chen Qi Xi Yang Zhao Kun Zhou Bo Xu Xiumei Han Xiaoyu Liu Meijuan Peng Congcong Chen Shiyao Tao Lei Huang Cong Liu Mingyang Wen Yangqian Jiang Tao Jiang Chuncheng Lu Wei Wu Di Wu Minjian Chen Yuan Lin Xuejiang Guo Ran Huo Jiayin Liu Hongxia Ma Guangfu Jin Yankai Xia Jiahao Sha Hongbing Shen Zhibin Hu | 2021 | Cell Research2021,31,8: | 2 |
