2Cholesterol homeostasis regulated by ABCA1 is critical for retinal ganglion cell survival显示文摘Genome-wide association studies have suggested a link between primary open-angle glaucoma and the function of ABCA1.ABCA1 is a key regulator of cholesterol efflux and the biogenesis of high-density lipoprotein(HDL) particles. Here, we showed that the POAG risk allele near ABCA1 attenuated ABCA1 expression in cultured cells. Consistently, POAG patients exhibited lower ABCA1 expression, reduced HDL, and higher cholesterol in white blood cells. Ablation of Abca1 in mice failed to form HDL, leading to elevated cholesterol levels in the retina. Counting retinal ganglion cells(RGCs) by using an artificial intelligence(AI) program revealed that Abca1-deficient mice progressively lost RGCs with age. Single-cell RNA sequencing(scRNA-seq) revealed aberrant oxidative phosphorylation in the Abca1-/-retina, as well as activation of the mTORC1 signaling pathway and suppression of autophagy. Treatment of Abca1-/-mice using atorvastatin reduced the cholesterol level in the retina,thereby improving metabolism and protecting RGCs from death. Collectively, we show that lower ABCA1 expression and lower HDL are risk factors for POAG. Accumulated cholesterol in the Abca1-/-retina causes profound aberrant metabolism, leading to a POAG-like phenotype that can be prevented by atorvastatin. 3Reactive oxygen species and nitric oxide induce senescence of rudimentary leaves and the expression profiles of the related genes in Litchi chinensis显示文摘Litchi is one of the most important subtropical evergreen fruit trees in southern Asia.Previous studies indicated that high-temperature conditions encourage growth of rudimentary leaves in panicles and suppress flowering.We have demonstrated that methyl viologen dichloride hydrate(MV)and sodium nitroprusside(SNP)promoted flowering in litchi partially by inhibiting the growth of rudimentary leaves via reactive oxygen species(ROS)and nitric oxide(NO).In the present study,we examined the microstructure and ultrastructure,programmed cell death(PCD)ratio,nuclei morphology of the rudimentary leaves,and the expression of senescence-related genes after the treatment with ROS or NO.The results showed that chromatins of the ROS-or NO-treated cells in the rudimentary leaves were condensed.Fusion of the cytoplasm-digesting vesicles with the vacuole and degradation of cytoplasm forming scattered debris were found in those of the treated cells.Treatment with ROS or NO increased the cell PCD ratio.Morphology of the nuclei stained by propidium iodide(PI)showed that nuclei shape became irregular after the ROS or NO treatment.Further,the expression levels of LcRboh,LcMC-1-like,and LcPirin were higher in the ROS-and NO-treated rudimentary leaves than those in the control ones,suggesting that these genes may be involved in the ROS and NO-induced senescence and abscission of the rudimentary leaves in litchi.Our results suggested that ROS and NO play an important role in inducing the senescence of the rudimentary leaves,and ROS-and NO-induced PCD may be involved in the regulation of the rudimentary leaf growth in litchi.Haifang Yang Hye-Ji Kim Houbin Chen Yong Lu Xingyu Lu Congcong Wang Biyan Zhou 2018Horticulture Research2018,5,1:2
4Clinical Features and Differential Diagnosis of Acute Idiopathic Blind Spot Enlargement Syndrome显示文摘Purpose:To study the clinical manifestations and the diagnostic and differential diagnostic characteristics of acute idiopathic blind spot enlargement syndrome(AIBSES).Methods:.Six patients diagnosed with AIBSES underwent complete eye examinations including fundus photography,fundus fluorescein angiography(FFA), indocyanine green angiography(ICGA), optical coherence tomography(OCT),electroretinogram(ERG), and visual field examinations.Results: All patients had enlarged blind spots of variable sizes and densities..Three eyes had mild swelling of the optic disc and one eye had peripapillary scarring that corresponded to the permanent field defect. Two patients who underwent FFA had fluorescein leakage of blood vessels around the optic disc and ICGA highlighted diffuse, small hypofluorescent spots scattering throughout the posterior pole..OCT showed that the inner and outer segment(IS / OS).line were absent in five patients and the middle cone outer segment tip line was absent in the nasal macular area in one eye.Conclusion: AIBSES is a rare outer retinopathy. Visual field examination and OCT are the most important means of detection. 5Deep Sc-RNA sequencing decoding the molecular dynamic architecture of the human retina显示文摘The human retina serves as a light detector and signals transmission tissue.Advanced insights into retinal disease mechanisms and therapeutic strategies require a deep understanding of healthy retina molecular events.Here,we sequenced the m RNA of over 0.6 million single cells from human retinas across six regions at nine different ages.Sixty cell sub-types have been identified from the human mature retinas with unique markers.We revealed regional and age differences of gene expression profiles within the human retina.Cell-cell interaction analysis indicated a rich synaptic connection within the retinal cells.Gene expression regulon analysis revealed the specific expression of transcription factors and their regulated genes in human retina cell types.Some of the gene’s expression,such as DKK3,are elevated in aged retinas.A further functional investigation suggested that over expression of DKK3 could impact mitochondrial stability.Overall,decoding the molecular dynamic architecture of the human retina improves our understanding of the vision system.Lulin Huang Runze Li Lin Ye Shanshan Zhang Huaping Tian Mingyan Du Chao Qu Shujin Li Jie Li Mu Yang Biao Wu Ran Chen Guo Huang Ling Zhong Hongjie Yang Man Yu Yi Shi Changguan Wang Houbin Zhang Wei Chen Zhenglin Yang 2023Science China(Life Sciences)2023,66,3:0
