1Multimodality treatment in hepatocellular carcinoma patients with tumor thrombi in portal vein显示文摘AIM To compare the therapeutic effect andsignificances of multimodality treatment forhepatocellular carcinoma (HCC) with tumorthrombi in portal vein (PVTT).METHODS HCC patients (n = 147) with tumortrombi in the main portal vein or the first branchof portal vein were divided into four groups bythe several therapeutic methods. There wereconservative treatment group in 18 out ofpatients (group A); and hepatic artery ligation(HAL) and/or hepatic artery infusion (HAl)group in 18 patients (group B), in whompostoberative chemoembolization was doneperiodically; group of removal of HCC with PVTTin 79 (group C) and group of transcatheterhepatic arterial chemoembolization (TACE) orHAl and/or portal vein infusion (PVI) afteroperation in 32 (group D).RESULTS The median survival period was 12months in our series and the 1-, 3-, and 5-yearsurvival rates were 44.3%, 24.5% and 15.2%,respectively. The median survival times were 2,5, 12 and 16 months in group A, B, C and D,respectively. The 1-, 3- and 5-year survival rateswere 5.6%, 0% and 0% in group A; 22.2%,5.6% and 0% in group B; 53.9%, 26.9% and16.6% in group C; 79.3%, 38.9% and 26.8% ingroup D, respectively. 4PD-1/PD-L1 pathway blockade works as an effective and practical therapy for cancer immunotherapy显示文摘Cancer immunotherapy has greatly advanced in recent years,and PD-1/PD-L1 blocking therapy has become a major pillar of immunotherapy.Successful clinical trials of PD-1/PD-L1 blocking therapies in cancer treatments have benefited many patients,which promoted the Food and Drug Administration(FDA)approval of PD-1/PD-L1 blocking drugs.In this review,we provide a detailed introduction of five PD-1/PD-L1 blocking drugs,with indications and studies,as a valuable reference for doctors and medical investigators.Moreover,the characteristics of PD-1/PD-L1 blocking therapies,including their universality and sustainability,are discussed in this review.Furthermore,we also discuss and predict the possibility of PD-L1 as an indication marker of PD-1/PD-L1 blocking therapy for pan-cancer treatment,and the current status of combination therapies.Long Jia Qi Zhang Rongxin Zhang 2018Cancer Biology & Medicine2018,15,2:18
5Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 6A kinetic study of the reaction of ozone with ethylene in a smog chamber under atmospheric conditions显示文摘Ozone is one of the key species in the processes of atmospheric chemistry, which can be taken as an indicator of oxidation capacity in the troposphere. The reaction of ozone with reactive gases is an important process in the troposphere. Experimental simulation equipment of smog chamber for atmospheric reactions is used to study the reaction of ozone with ethylene in real atmospheric environment with ozone concentrations of 100―200 ppb. The concentrations of ozone and ethylene were moni-tored during the reaction with the combination of Model 49C-O_3 Analyzer and GC-FID. A rate constant of 1.01×10^(-18) (cm^3·mol^(-1)·s-(~1)) was obtained at 286.5 K, under condition of which the half-life of ozone was 88 min. The results obtained from our experiments are in excellent agreement with those reported previously by other researchers under extremely low pressure in terms of matrixisolation technology. 10The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 11MPFFT:An Auto-Tuning FFT Library for OpenCL GPUs显示文摘Fourier methods have revolutionized many fields of science and engineering,such as astronomy,medical imaging,seismology and spectroscopy,and the fast Fourier transform(FFT) is a computationally efficient method of generating a Fourier transform.The emerging class of high performance computing architectures,such as GPU,seeks to achieve much higher performance and efficiency by exposing a hierarchy of distinct memories to software.However,the complexity of GPU programming poses a significant challenge to developers.In this paper,we propose an automatic performance tuning framework for FFT on various OpenCL GPUs,and implement a high performance library named MPFFT based on this framework.For power-of-two length FFTs,our library substantially outperforms the clAmdFft library on AMD GPUs and achieves comparable performance as the CUFFT library on NVIDIA GPUs.Furthermore,our library also supports non-power-of-two size.For 3D non-power-of-two FFTs,our library delivers 1.5x to 28x faster than FFTW with 4 threads and 20.01x average speedup over CUFFT 4.0 on Tesla C2050.Yan Li Yun-Quan Zhang Yi-Qun Liu Guo-Ping Long Hai-Peng Jia 2013Journal of Computer Science & Technology2013,28,1:10
12Development and case study of a science-based software platform to support policy making on air quality显示文摘This article describes the development and implementations of a novel software platform that supports real-time, science-based policy making on air quality through a user-friendly interface. The software, RSM-VAT, uses a response surface modeling(RSM) methodology and serves as a visualization and analysis tool(VAT) for three-dimensional air quality data obtained by atmospheric models. The software features a number of powerful and intuitive data visualization functions for illustrating the complex nonlinear relationship between emission reductions and air quality benefits. 13Pre-impact trajectory planning for minimizing base attitude disturbance in space manipulator systems for a capture task显示文摘Aimed at capture task for a free-floating space manipulator, a scheme of pre-impact trajectory planning for minimizing base attitude disturbance caused by impact is proposed in this paper.Firstly, base attitude disturbance is established as a function of joint angles, collision direction and relative velocity between robotic hand and the target.Secondly, on the premise of keeping correct capture pose, a novel optimization factor in null space is designed to minimize base attitude disturbance and ensure that the joint angles do not exceed their limits simultaneously.After reaching the balance state, a desired configuration is achieved at the contact point.Thereafter, particle swarm optimization(PSO) algorithm is employed to solve the pre-impact trajectory planning from its initial configuration to the desired configuration to achieve the minimized base attitude disturbance caused by impact and the correct capture pose simultaneously.Finally, the proposed method is applied to a 7-dof free-floating space manipulator and the simulation results verify the effectiveness.Zhang Long Jia Qingxuan Chen Gang Sun Hanxu 2015Chinese Journal of Aeronautics2015,28,4:9
14Effect of spinal cord extracts after spinal cord injury on proliferation of rat embryonic neural stem cells and Notch signal pathway in vitro显示文摘Objective:To investigate the effect of the spinal cord extracts(SCE)after spinal cord injuries(SCIs)on the proliferation of rat embryonic neural stem cells(NSCs)and the expressions of mRNA of Notch1 as well as of Hes1 in this process in vitro.Methods:The experiment was conducted in 4 different mediums:NSCs+PBS(Group A-blank control group),NSCs+SCE with healthy SD rats(Croup B-normal control group),NSCs+SCE with SD rats receiving sham-operation treatment(Croup C-sham-operation group)and NSCs+SCE with SCIs rats(Group D-paraplegic group).Proliferative abilities of 4 different groups were analyzed by MTT chromatometry after co-culture for 1,2,3,4 and 5 d,respectively.The expressions of Notch 1 and Hes1 mRNA were also detected with RT-PCR after co-culture for 24 and 48 h,respectively.Results:After co-culture for 1,2,3,4 and 5 d respectively,the MTT values of group D were significantly higher than those of group A,group B and group C(P<0.05).However,there were no significantly differences regarding MTT values between group A,group B and group C after co-culture for 1,2,3,4 and 5 d,respectively(P>0.05).Both the expressions of Notch1 and Hes1 mRNA of group D were significantly higher than those of other 3 groups after co-culture for 24 h and 48 h as well(P<0.05).But there was no difference oin expressions of Notch1 and Hes1 mRNA among group A,group B and group C after co-culture for 24 h and 48 h(P>0.05).There was no difference in expressions of Notch1and Hes1 mRNA between 24 h and 48 h treatment in group D.Conclusions:SCE could promote the proliferation of NSCs.It is demonstrated that the microenvironment of SCI may promote the proliferation of NSCs.Besides,SCE could increase the expression of Notch1 and Hes1 mRNA of NSC.It can be concluded that the Notch signaling pathway activation is one of the mechanisms that locally injured microenvironment contributes to the proliferation of ENSC after SCIs.This process may be performed by up-regulating the expressions of Notch1 and Hes1 gene.Qing-Zhong Zhou Ge Zhang Hai-Bo Long Fei Lei Fei Ye Xu-Feng Jia Yun-Long Zhou Jian-Ping Kang Da-Xiong Feng 2014Asian Pacific Journal of Tropical Medicine2014,7,7:9
15Combination of mild therapeutic hypothermia and adipose-derived stem cells for ischemic brain injury显示文摘Mild therapeutic hypothermia has been shown to mitigate cerebral ischemia, reduce cerebral edema, and improve the prognosis of patients with cerebral ischemia. Adipose-derived stem cell-based therapy can decrease neuronal death and infiltration of inflammatory cells, exerting a neuroprotective effect. We hypothesized that the combination of mild therapeutic hypothermia and adipose-derived stem cells would be neuroprotective for treatment of stroke. A rat model of transient middle cerebral artery occlusion was established using the nylon monofilament method. Mild therapeutic hypothermia(33°C) was induced after 2 hours of ischemia. Adipose-derived stem cells were administered through the femoral vein during reperfusion. The severity of neurological dysfunction was measured by a modified Neurological Severity Score Scaling System. The area of the infarct lesion was determined by 2,3,5-triphenyltetrazolium chloride staining. Apoptotic neurons were detected by terminal deoxynucleotidyl transferase-mediated d UTP-biotin nick end labeling(TUNEL) staining. The regeneration of microvessels and changes in the glial scar were detected by immunofluorescence staining. The inflammatory responses after ischemic brain injury were evaluated by in situ staining using markers of inflammatory cells. The expression of inflammatory cytokines was measured by reverse transcription-polymerase chain reaction. Compared with mild therapeutic hypothermia or adipose-derived stem cell treatment alone, their combination substantially improved neurological deficits and decreased infarct size. They synergistically reduced the number of TUNEL-positive cells and glial fibrillary acidic protein expression, increased vascular endothelial growth factor levels, effectively reduced inflammatory cell infiltration and down-regulated the m RNA expression of the proinflammatory cytokines interleukin-1β, tumor necrosis factor-α and interleukin-6. 16Cystathionine-γ-lyase ameliorates the histone demethylase JMJD3-mediated autoimmune response in rheumatoid arthritis显示文摘Cystathionine-γ-lyase(CSE),an enzyme associated with hydrogen sulfide(H2S)production,is an important endogenous regulator of inflammation.Jumonji domain-containing protein 3(JMJD3)is implicated in the immune response and inflammation.Here,we investigated the potential contribution of JMJD3 to endogenous CSE-mediated inflammation in rheumatoid arthritis(RA).Upregulated CSE and JMJD3 were identified in synovial fibroblasts(SFs)from RA patients as well as in the joints of arthritic mice.Knocking down CSE augmented inflammation in IL-1β-induced SFs by increasing JMJD3 expression.In addition,CSE−/−mice with collagen-induced arthritis(CIA)developed severe joint inflammation and bone erosion.Conversely,overexpressing CSE inhibited JMJD3 expression by the transcription factor Sp-1 and was accompanied by reduced inflammation in IL-1β-treated SFs.Furthermore,JMJD3 silencing or the administration of the JMJD3 inhibitor GSK-J4 significantly decreased the inflammatory response in IL-1β-treated SFs,mainly by controlling the methylation status of H3K27me3 at the promoter of its target genes.GSK-J4 markedly attenuated the severity of arthritis in CIA mice.In conclusion,suppressing JMJD3 expression by the transcription factor Sp-1 is likely responsible for the ability of CSE to negatively modulate the inflammatory response and reduce the progression of RA.Weijun Wu Ming Qin Wanwan Jia Zheng Huang Zhongzheng Li Di Yang Mengwei Huang Chenxi Xiao Fen Long Jianchun Mao Philip K.Moore Xinhua Liu Yi Zhun Zhu 2019Cellular & Molecular Immunology2019,16,8:9
17Celastrol induces lipophagy via the LXRα/ABCA1 pathway in clear cell renal cell carcinoma显示文摘Celastrol is a triterpene derived from the traditional Chinese medicine Tripterygium wilfordii Hook f,which displays potential anticancer activity.In the present study,we investigated the anticancer effects of celastrol against clear cell renal cell carcinoma(ccRCC)and the underlying mechanisms.Using Cancer Genome Atlas(TCGA)database and genotype-tissue expression(GTEx)database we conducted a bioinformatics analysis,which showed that the mRNA levels of liver-X receptorsα(LXRα)and ATP-binding cassette transporter A1(ABCA1)in ccRCC tissues were significantly lower than those in adjacent normal tissues.This result was confirmed by immunoblotting analysis of 4 ccRCC clinical specimens,which showed that the protein expression of LXRαand ABCA1 was downregulated.Similar results were obtained in a panel of ccRCC cell lines(786-O,A498,SN12C,and OS-RC-2).In 786-O and SN12C cells,treatment with celastrol(0.25–2.0μM)concentration-dependently inhibited the cell proliferation,migration,and invasion as well as the epithelial-mesenchymal transition(EMT)process.Furthermore,we demonstrated that celastrol inhibited the invasion of 786-O cells through reducing lipid accumulation;celastrol concentration-dependently promoted autophagy to reduce lipid storage.Moreover,we revealed that celastrol dramatically activated LXRαsignaling,and degraded lipid droplets by inducing lipophagy in 786-O cells.Finally,celastrol promoted cholesterol efflux from 786-O cells via ABCA1.In high-fat diet-promoted ccRCC cell line 786-O xenograft model,administration of celastrol(0.25,0.5,1.0 mg·kg^(−1)·d^(−1),for 4 weeks,i.p.)dose-dependently inhibited the tumor growth with upregulated LXRαand ABCA1 protein in tumor tissue.In conclusion,this study reveals that celastrol triggers lipophagy in ccRCC by activating LXRα,promotes ABCA1-mediated cholesterol efflux,suppresses EMT progress,and ultimately inhibits cell proliferation,migration,and invasion as well as tumor growth.Thus,our study provides evidence that celastrol can be used as a lipid metabolism-based anticancer therapeutic approach.Chan-juan Zhang Neng Zhu Jia Long Hong-tao Wu Yu-xiang Wang Bi-yuan Liu Duan-fang Liao Li Qin 2021Acta Pharmacologica Sinica2021,42,9:8
18The role of FGF21 in the pathogenesis of cardiovascular disease显示文摘The morbidity and mortality of cardiovascular diseases(CVDs)are increasing worldwide and seriously threaten human life and health.Fibroblast growth factor 21(FGF21),a metabolic regulator,regulates glucose and lipid metabolism and may exert beneficial effects on the cardiovascular system.In recent years,FGF21 has been found to act directly on the cardiovascular system and may be used as an early biomarker of CVDs.The present review highlights the recent progress in understanding the relationship between FGF21 and CVDs including coronary heart disease,myocardial ischemia,cardiomyopathy,and heart failure and also explores the related mechanism of the cardioprotective effect of FGF21.FGF21 plays an important role in the prediction,treatment,and improvement of prognosis in CVDs.This cardioprotective effect of FGF21 may be achieved by preventing endothelial dysfunction and lipid accumulating,inhibiting cardiomyocyte apoptosis and regulating the associated oxidative stress,inflammation and autophagy.In conclusion,FGF21 is a promising target for the treatment of CVDs,however,its clinical application requires further clarification of the precise role of FGF21 in CVDs.Ying Zhang Dan Liu Xiao-Xue Long Qi-Chen Fang Wei-Ping Jia Hua-Ting Li 2021Chinese Medical Journal2021,,24:8
19Large-area straight,regular periodic surface structures produced on fused silica by the interference of two femtosecond laser beams through cylindrical lens显示文摘Inhomogeneity and low efficiency are two important factors that limit the application of laser-induced periodic surface structures(LIPSSs),especially on glass surfaces.In this study,two-beam interference(TBI)of femtosecond lasers was used to produce large-area straight LIPSSs on fused silica using cylindrical lenses.Compared with those produced us-ing a single circular or cylindrical lens,the LIPSSs produced by TBI are much straighter and more regular.Depending on the laser fluence and scanning velocity,LIPSSs with grating-like or spaced LIPSSs are produced on the fused silica sur-face.Their structural colors are blue,green,and red,and only green and red,respectively.Grating-like LIPSS patterns oriented in different directions are obtained and exhibit bright and vivid colors,indicating potential applications in surface coloring and anti-counterfeiting logos.Long Chen Kaiqiang Cao Yanli Li Jukun Liu Shian Zhang Donghai Feng Zhenrong Sun Tianqing Jia 2021Opto-Electronic Advances2021,4,12:7
20Smog chamber studies of ozone formation potentials for isopentane显示文摘The incremental reactivity and ozone formation potential of isopentane have been studied with chamber experiments and computer simulations. The chemical mechanism used in the computer simulations is an isopentane sub-mechanism from the Master Chemical Mechanism (MCM). The results from the chamber experiments suggest that the MCM can well simulate i-C5H12-NOx chamber experiments. The heterogeneous reaction of NO2 and water is an important source for OH radicals in the chamber experiments. The photolysis of HONO is responsible for the initiation of isopentane in photochemical reactions. The reaction rate constant for NO2 → HONO was determined to be 3.9×10-4―5.9×10-3 min-1 by conducting 3 sets of CO-NOx-air irradiations. 5 sets of isopentane-NOx irradiations under different conditions were performed in our chamber. Compared with the experiment with a low relative humidity (RH), an increase in RH can increase the reaction rate of NO2 with H2O, so that the peak ozone occurs earlier.
