| 2 | Proteome analysis of alfalfa roots in response to water deficit stress显示文摘To evaluate the response of alfalfa to water deficit(WD) stress, WD-induced candidates were investigated through a proteomic approach. Alfalfa seedlings were exposed to WD stress for 12 and 15 days respectively, followed by 3 days re-watering. Water deficit increased H2O2 content, lipid peroxidation, DPPH(1,1-diphenyl-2-picrylhydrazyl)-radical scavenging activity, and the free proline level in alfalfa roots. Root proteins were extracted and separated by two-dimentional polyacrylamide gel electrophoresis(2-DE). A total of 49 WD-responsive proteins were identified in alfalfa roots; 25 proteins were reproducibly found to be up-regulated and 24 were down-regulated. Two proteins, namely cytosolic ascorbate peroxidase(APx2) and putative F-box protein were newly detected on 2-DE maps of WD-treated plants. We identified several proteins including agamous-like 65, albumin b-32, inward rectifying potassium channel, and auxin-independent growth promoter. The identified proteins are involved in a variety of cellular functions including calcium signaling, abacisic acid(ABA) biosynthesis, reactive oxygen species(ROS) regulation, transcription/ translation, antioxidant/detoxification/stress defense, energy metabolism, signal transduction, and storage. | 3 | Role of inflammasome regulation on immune modulators显示文摘Inflammatory responses are essential in eliminating harmful substrates from damaged tissue and inducing recovery.Several cytokines participate in and facilitate this response. Certain cytokines such as interleukin(IL)-1β and IL-18 are initially produced in precursor form in response to toll-like receptor(TLR) ligands and undergo maturation by inflammasomes, which are cytosolic multi-protein complexes containing nucleotide-binding oligomerization domain(NOD)-containing protein 2-like receptors(NLRs). Immune modulators targeting inflammasomes have been investigated to control inflammatory diseases such as metabolic syndrome. However, most immune modulators possessing anti-inflammasome properties attenuate production of other cytokines, which are essential for host defense. In this review, we analyzed the effect of anti-inflammasome agents on the production of cytokines which are not regulated by inflammasome and involving in initial immune responses. As a result, the infiammasome inhibitors are put into three categories: non-effector, stimulator, or inhibitor of cytokine production. Even the stimulator of cytokine production ameliorated symptoms resulting from inflammasome activation in mouse models. | 4 | Effects of temperature and Nickel content on magnetic properties of Ni-doped ZnO显示文摘Magnetic properties of diluted magnetic semiconductors (DMSs), Ni-doped ZnO materials, prepared by sol-gel method were investigated by measuring magnetization as functions of magnetic field. The Ni content affects the magnetic properties at low sintered temperature but it has few effects on the magnetic properties at high sintered temperature. | 5 | Inhibitory effects of rapamycin on the different stages of hepatic fibrosis显示文摘AIM:To investigate and compare the inhibitory effects of rapamycin in the different stages of liver fibrosis.METHODS:We performed bile duct ligation(BDL)in male Wistar rats(n=24).The experimental rats were classified into four groups:the BDL+/Rapa-group(untreated control,n=4),the BDL+/Rapa+group(treated14 d after BDL,n=8),the BDL+/Rapa++group(treated on the day after BDL,n=8),and the BDL-/Rapagroup(un-treated,sham-operated control,n=4).The BDL+/Rapa+and BDL+/Rapa++groups were administered rapamycin(2 mg/kg)for 28 d.The liver tissues were tested by immunohistochemical staining forα-smooth muscle actin(α-SMA)and cytokeratin.RESULTS:The liver mRNA levels of transforming growth factor(TGF)-β1 and platelet-derived growth factor(PDGF)were measured using the polymerase chain reaction.The protein levels of liver p70s6K and p-p70s6k were determined using Western blotting.α-SMA expression was lowest in the BDL+/Rapa++group.TGF-β1and PDGF expression levels in the rapamycin-treated group were lower than those in the un-treated group and higher than those in the control groups(TGF-β1:0.23±0.00 vs 0.34±0.01,0.23±0.0 vs 0.09±0.00,P<0.0001;PDGF:0.21±0.00 vs 0.34±0.01,0.21±0.0 vs 0.09±0.00,P<0.0001).The p70s6k and p-p70s6k levels decreased in the treated groups and were lowest in the BDL+/Rapa++group(p70s6k:1.05±0.17 vs 1.30±0.56,0.40±0.01 vs 1.30±0.56,P<0.0001;p-p70s6k:1.40±0.5 vs 1.67±0.12,0.70±0.01 vs 1.67±0.12,P<0.0001).CONCLUSION:The results of our study indicate that rapamycin has inhibitory effects on liver fibrosis,and the treatment is most effective in the early stages of fibrosis. | Yun Jeung Kim Eaum Seok Lee Seok Hyun Kim Heon Young Lee Seung Moo Noh Dae Young Kang Byung Seok Lee | 2014 | World Journal of Gastroenterology2014，20，23： | 3 |

| 7 | Reactivation of tuberculosis in hepatocellular carcinoma treated with transcatheter arterial chemoembolization: A report of 3 cases显示文摘Pulmonary tuberculosis is an opportunistic infection that can be reactivated in immunocompromised conditions, for example, in malignancy or after liver transplantation. Hepatocellular carcinoma (HCC) has a high mortality rate because it is frequently diagnosed at an advanced stage. Although surgical resection is the established curative measure for HCC, it is only feasible for earlystage HCC. Transcatheter arterial chemoembolization (TACE) is the most common treatment modality for patients with unresectable HCC. However, repeated TACE sessions and, occasionally, the tumor itself can further impair the reserve hepatic function and immunity. | 8 | Immune enhancement effect of an herb complex extract through the activation of natural killer cells and the regulation of cytokine levels in a cyclophosphamide-induced immunosuppression rat model显示文摘Objective: To investigate the effects of a herb complex extract(HCE) prepared from Cornus officinalis Sieb. Et Zucc., Eriobotrya japonica Lindley, and olive leaves on immune response of mouse spleen NK cells in vitro and in vivo analysis. Methods: The activity of natural killer(NK) cells was measured in splenocytes and YAC-1 cells. Mice were immunosuppressed using cyclophosphamide(5 mg/kg body weight). Three different doses of HCE(200, 400, and 800 mg/kg body weight) and red ginseng extract(800 mg/kg body weight) which was used as standard immunomodulatory herb were administered orally for 4 weeks. The body weight, dietary, water intake, organs(liver, thymus, and spleen) weight, completed blood count, and cytokines(tumor necrosis factor alpha, interferon gamma, and interleukin-2) production was measured. Results: At the maximum concentration of HCE, the activity of NK cells was increased by 48.5%. HCE increased liver, spleen, and thymus weights without altering numbers of white blood cells, lymphocytes, and neutrophils in a cyclophosphamide-induced immunosuppression rat model. However, HCE recovered the inhibited cytokine expression; HCE(800 mg/kg) increased cytokines levels. The results indicate the immune enhancement potential of this HCE. Conclusion: The HCE enhances immunity by increasing NK cell activity, regulating cytokine levels, and maintaining spleen weight.
