1Radiomics-based predictive risk score: A scoring system for preoperatively predicting risk of lymph node metastasis in patients with resectable non-small cell lung cancer显示文摘Objective: To develop and validate a radiomics-based predictive risk score(RPRS) for preoperative prediction of lymph node(LN) metastasis in patients with resectable non-small cell lung cancer(NSCLC).Methods: We retrospectively analyzed 717 who underwent surgical resection for primary NSCLC with systematic mediastinal lymphadenectomy from October 2007 to July 2016. By using the method of radiomics analysis, 591 computed tomography(CT)-based radiomics features were extracted, and the radiomics-based classifier was constructed. Then, using multivariable logistic regression analysis, a weighted score RPRS was derived to identify LN metastasis. Apparent prediction performance of RPRS was assessed with its calibration,discrimination, and clinical usefulness.Results: The radiomics-based classifier was constructed, which consisted of 13 selected radiomics features.Multivariate models demonstrated that radiomics-based classifier, age group, tumor diameter, tumor location, and CT-based LN status were independent predictors. When we assigned the corresponding score to each variable,patients with RPRSs of 0-3, 4-5, 6, 7-8, and 9 had distinctly very low(0%-20%), low(21%-40%), intermediate(41%-60%), high(61%-80%), and very high(81%-100%) risks of LN involvement, respectively. The developed RPRS showed good discrimination and satisfactory calibration (C-index: 0.785, 95% confidence interval(95% CI):0.780-0.790)Additionally, RPRS outperformed the clinicopathologic-based characteristics model with net reclassification index(NRI) of 0.711(95% CI: 0.555-0.867).Conclusions: The novel clinical scoring system developed as RPRS can serve as an easy-to-use tool to facilitate the preoperatively individualized prediction of LN metastasis in patients with resectable NSCLC. 2Warm temperature compromises JA-regulated basal resistance to enhance Magnaporthe oryzae infection in rice显示文摘Changes in global temperatures profoundly affect the occurrence of plant diseases.It is well known that rice blast can easily become epidemic in relatively warm weather.However,the molecular mechanism remains unclear.In this study,we show that enhanced blast development at a warm temperature(22C)compared with the normal growth temperature(28C)is rice plant-determined.Comparative transcriptome analysis revealed that jasmonic acid(JA)biosynthesis and signaling genes in rice could be effectively induced by Magnaporthe oryzae at 28C but not at 22C.Phenotypic analyses of the osaoc1 and osmyc2 mutants,OsCOI1 RNAi lines,and OsMYC2-OE plants further demonstrated that compromised M.oryzaeinduced JA biosynthesis and signaling lead to enhanced blast susceptibility at the warm temperature.Consistent with these results,we found that exogenous application of methyl jasmonate served as an effective strategy for improving blast resistance under the warm environmental conditions.Furthermore,decreased activation of JA signaling resulted in the downregulated expression of some key basal resistance genes at 22C when compared with 28C.Among these affected genes,OsCEBiP(chitin elicitorbinding protein precursor)was found to be directly regulated by OsMYB22 and its interacting protein OsMYC2,a key component of JA signaling,and this contributed to temperature-modulated blast resistance.Taken together,these results suggest that warm temperature compromises basal resistance in rice and enhances M.oryzae infection by reducing JA biosynthesis and signaling,providing potential new strategies for managing rice blast disease under warm climate conditions.Jiehua Qiu Junhui Xie Ya Chen Zhenan Shen Huanbin Shi Naweed I.Naqvi Qian Qian Yan Liang Yanjun Kou 2022Molecular Plant2022,15,4:5

4Heritability of growth traits in the Asian seabass (Lates calcarifer)显示文摘Growth is an economically important trait in aquaculture.To improve growth trait of the Asian seabass(Lates calcarifer)we have been carrying out,since 2004,a selective breeding program.This study focuses on growth traits in the F2 fish generation,comprised of offspring from 23 mass crosses from 383 F_(1) brooders.Using genotyping analysis for 10 microsatellites from both brood stock and progeny we have reconstructed the pedigree of each mass-cross.For F_(2) generation at 90 days post hatch(dph),we have recorded body weight(BW)for 12,117 individuals and total length(TL),standard length(SL)and condition factor(K_(tl) and K_(sl))for 3530 individuals;and all five traits for 2136 individuals at 270 dph.At 90 dph the average BW was 46.88±20.95 g.Combining pedigree information,recorded growth traits and Restricted Maximum Likelihood method,we have estimated that the narrow sense heritability(h^(2))in F_(1) fish for BW,TL,SL,K_(tl) and K_(sl) was at,90 dph,0.12±0.03,0.11±0.03,0.10±0.03,0.20±0.04 and 0.11±0.03,respectively and,at 270 dph,0.34±0.07,0.32±0.07,0.30±0.06,0.13±0.04 and 0.11±0.04,respectively.At 90 dph the realised heritability for BW was 0.13.Comparing with F_(1) generation,the growth performance of F_(2) fish was increased by 14.4%.Heritability of growth traits will be useful for future genetic improvement programmes of the Asian seabass.Baoqing Ye Ziyi Wan Le Wang Hongyan Pang Yanfei Wen Huiming Liu Bing Liang Huan Sein Lim Junhui Jiang Genhua Yue 2017Aquaculture and Fisheries2017,2,3:4
5A genome-wide association study uncovers a critical role of the RsPAP2 gene in red-skinned Raphanus sativus L.显示文摘Radish(Raphanus sativus L.)taproot contains high concentrations of flavonoids,including anthocyanins(ATCs),in redskinned genotypes.However,little information on the genetic regulation of ATC biosynthesis in radish is available.A genome-wide association study of radish red skin color was conducted using whole-genome sequencing data derived from 179 radish genotypes.The R2R3-MYB transcription factor production of anthocyanin pigment 2(PAP2)gene was found in the region associated with a leading SNP located on chromosome 2.The amino acid sequence encoded by the RsPAP2 gene was different from those of the other published RsMYB genes responsible for the red skin color of radish.The overexpression of the RsPAP2 gene resulted in ATC accumulation in Arabidopsis and radish,which was accompanied by the upregulation of several ATC-related structural genes.RsPAP2 was found to bind the RsUFGT and RsTT8 promoters,as shown by a dual-luciferase reporter system and a yeast one-hybrid assay.The promoter activities of the RsANS,RsCHI,RsPAL,and RsUFGT genes could be strongly activated by coinfiltration with RsPAP2 and RsTT8.These findings showed the effectiveness of GWAS in identifying candidate genes in radish and demonstrated that RsPAP2 could(either directly or together with its cofactor RsTT8)regulate the transcript levels of ATC-related genes to promote ATC biosynthesis,facilitating the genetic enhancement of ATC contents and other related traits in radish.Lianxue Fan YanWang Liang Xu Mingjia Tang Xiaoli Zhang Jiali Ying Cui Li Junhui Dong Liwang Liu 2020Horticulture Research2020,7,1:2

8Moso bamboo expansion decreased soil heterotrophic respiration but increased arbuscular mycorrhizal mycelial respiration in a subtropical broadleaved forest显示文摘Moso bamboo(Phyllostachys Pubescens)expansion into adjacent forests has been widely reported to affect plant diversity and its association with mycorrhizal fungi in subtropical China,which will likely have significant impacts on soil respiration.However,there is still limited information on how Moso bamboo expansion changes soil respiration components and their linkage with microbial community composition and activity.Based on a mesh exclusion method,soil respirations derived from roots,arbuscular mycorrhizal(AM)mycelium,and free-living microbes were investigated in a pure Moso bamboo forest(expanded),an adjacent broadleaved forest(nonexpanded),and a mixed bamboo-broadleaved forest(expanding).Our results showed that bamboo expansion decreased the cumulative CO_(2)effluxes from total soil respiration,root respiration and soil heterotrophic respiration(by 19.01%,30.34%,and 29.92%on average),whereas increased those from AM mycelium(by 78.67%in comparison with the broadleaved forests).Bamboo expansion significantly decreased soil organic carbon(C)content,bacterial and fungal abundances,and enzyme activities involved in C,N and P cycling whereas enhanced the interactive relationships among bacterial communities.In contrast,the ingrowth of AM mycelium increased the activities ofβ-glucosidase and N-acetyl-β-glucosaminidase and decreased the interactive relationships among bacterial communities.Changes in soil heterotrophic respiration and AM mycelium respiration had positive correlations with soil enzyme activities and fungal abundances.In summary,our findings suggest that bamboo expansion decreased soil heterotrophic respiration by decreasing soil microbial activity but increased the contribution of AM mycelial respiration to soil C efflux,which may potentially increase soil C loss from AM mycelial pathway.Wenhao Jin Jiaying Tu Qifeng Wu Liyuan Peng Jiajia Xing Chenfei Liang Shuai Shao Junhui Chen Qiufang Xu Hua Qin 2023Forest Ecosystems2023,10,3:1

12Hepatic transcriptome signatures in mice and humans with nonalcoholic fatty liver disease显示文摘Background:Nonalcoholic fatty liver disease(NAFLD)is the main reason for cirrhosis and hepatocellular carcinoma.As a starting point for NAFLD,the treatment of nonalcoholic fatty liver(NAFL)is receiving increasing attention.Mice fed a high-fat diet(HFD)and hereditary leptin deficiency(ob/ob)mice are important NAFL animal models.However,the comparison of these mouse models with human NAFL is still unclear.Methods:In this study,HFD-fed mice and ob/ob mice were used as NAFL animal models.Liver histopathological characteristics were compared,and liver transcriptome from both mouse models was performed using RNA sequencing(RNA-seq).RNAseq data obtained from the livers of NAFL patients was downloaded from the GEO database.Global gene expression profiles in the livers were further analyzed using functional enrichment analysis and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway.Results:Our results showed that the biochemical parameters of both mouse models and human NAFL were similar.Compared with HFD-fed mice,ob/ob mice were more similar in histologic appearance to NAFL patients.The liver transcriptome characteristics partly overlapped in mice and humans.Furthermore,in the NAFL pathway,most genes showed similar trends in mice and humans,thus demonstrating that both types of mice can be used as models for basic research on NAFL,considering the differences.Conclusion:Our findings show that HFD-fed mice and ob/ob mice can mimic human NAFL partly in pathophysiological process.The comparative analysis of liver transcriptome profile in mouse models and human NAFL presented here provides insights into the molecular characteristics across these NAFL models.Yiming Ding Xulei Dai Miaoye Bao Yuanming Xing Junhui Liu Sihai Zhao Enqi Liu Zuyi Yuan Liang Bai 2023Animal Models and Experimental Medicine2023,6,4:0
13C19orf66 Inhibits Japanese Encephalitis Virus Replication by Targeting-1 PRF and the NS3 Protein显示文摘The Japanese encephalitis serogroup of the neurogenic Flavivirus has a specific feature that expresses a non-structural protein NS1'produced through a programmed-1 ribosomal frameshifting(-1 PRF).Herein,C19orf66,a novel member of interferon-stimulated gene(ISG)products,exhibited significant activity of antagonizing Japanese encephalitis virus(JEV)infection.Overexpression of C19orf66 in 293T cells significantly inhibited JEV replication,while knock-down of endogenous C19orf66 in HeLa cells and A549 cells significantly increased virus replication.Notably,C19orf66 had an inhibitory effect on frameshift production of JEV NS1'.The inhibition was more significant when C19orf66 and JEV NS1-NS2A were co-expressed in the 293T cells.Both C19orf66-209 and C19orf66-Zinc^(mut) did not significantly change the NS1'to NS1 ratio and had weaker antiviral effects than C19orf66.Similarly,C19orf66-209 and C19orf66-Zinc^(mut) had no significant effect on the expression of the JEV NS3 protein,whose expression was down-regulated by C19orf66 via the lysosome-dependent pathway.These findings suggest that C19orf66 may possess at least two different mechanisms of antagonizing JEV infection.This study identified C19orf66 as a novel interferon-stimulated gene product that can inhibit JEV replication by targeting-1 PRF and the NS3 protein.The study provides baseline information for the future development of broad-spectrum antiviral agents against JEV.Du Yu Yundi Zhao Junhui Pan Xingmiao Yang Zhenjie Liang Shengda Xie Ruibing Cao 2021Virologica Sinica2021,36,6:0
14Japanese Encephalitis Virus NS2B-3 Protein Complex Promotes Cell Apoptosis and Viral Particle Release by Down-Regulating the Expression of AXL显示文摘Japanese encephalitis virus(JEV) is a flavivirus transmitted by mosquitoes that causes severe encephalitis in humans and animals. It has been suggested that AXL, a transmembrane protein, can promote the replication of various flaviviruses,such as dengue(DENV), Zika(ZIKV), and West Nile(WNV) viruses. However, the effect of AXL on JEV infection has not yet been determined. In the present study, we demonstrate that AXL is down-regulated after JEV infection in the late stage. JEV NS2B-3 protein specifically interacted with AXL, and promoted AXL degradation through the ubiquitin–proteasome pathway. AXL-degradation increased cell apoptosis by disrupting phosphatidylinositol 3-kinase(PI3 K)/Akt signal transduction. In addition, the degradation of AXL promoted JEV release to supernatant, whereas the virus in the cell lysates decreased. The supplementation of AXL ligand Gas6 inhibited the JEV-mediated degradation of AXL. 15Therapeutic sponge prevents postoperative breast cancer recurrence by sustainably dissociating into CD44-targeted nanoplatform显示文摘Locoregional recurrence and distant metastasis of breast cancer still pose a significant risk for patients’survival.To address the clinical challenge,functional absorbable sponges(HA-SH/PP-Dox/Lap/COL I(HCNPs))were constructed by biomimetic extracellular matrix of collagen I/hyaluronic acid complex conjugated with doxorubicin/lapatinib(Dox/Lap)-loaded nanoparticles.The HCNPs sponge exhibited excellent clotting ability and blood absorption rate.Worthily,Dox/Lap-loaded nanoparticles were synchronously endowed with a large number of oligo hyaluronic acid segments after degradation,which thus enhanced the ability of targeting into CD44-overexpressed tumor cells.The implantable HCNPs sponge in resected cavity of postoperative 4T1 models inhibited the spread of scattered tumor cells by absorbing the inevitable bleeding.More importantly,CD44 targeted nanoparticle with suitable Dox/Lap proportion continuously released from sponge to kill tumor cells of surrounding HCNPs and those remaining at surgical margin,thus prevented local recurrence as well as distant metastasis.Therefore,the functional HCNPs sponge might provide a safer and more effective strategy for postoperative treatment of cancer.Junhui Sui Mingda Zhao Zhihao Guo Jiafeng Li Jie Chen Hongli Chen Jie Liang Yong Sun Xingdong Zhang Yujiang Fan 2024Nano Research2024,17,3:0
16Melatonin-induced DNA demethylation of metal transporters and antioxidant genes alleviates lead stress in radish plants显示文摘Melatonin(MT)is a tryptophan-derived natural product that plays a vital role in plant response to abiotic stresses,including heavy metals(HMs).However,it remains elusive how exogenous MT mediates lead(Pb)accumulation and detoxification at the methylation and transcriptional levels in radish.In this study,decreased Pb accumulation and increased antioxidant enzyme activity were detected under MT treatment in radish.Single-base resolution maps of DNA methylation under Pb stress(Pb200)and Pb plus MT treatment(Pb_50MT)were first generated.The genome-wide methylation level was increased under Pb stress,while an overall loss of DNA methylation was observed under MT treatment.The differentially methylated region(DMR)-associated genes between Pb_50MT and Pb200 were uniquely enriched in ion binding terms,including cation binding,iron ion binding,and transition metal ion binding.Hyper-DMRs between Pb200 and Control exhibited a decreasing trend of methylation under Pb_50MT treatment.A few critical upregulated antioxidant genes(e.g.,RsAPX2,RsPOD52 and RsGST)exhibited decreased methylation levels under MT treatment,which enabled the radish plants to scavenge lead-induced reactive oxygen species(ROS)and decrease oxidative stress.Notably,several MT-induced HM transporter genes with low methylation(e.g.,RsABCF5,RsYSL7 and RsHMT)and transcription factors(e.g.,RsWRKY41 and RsMYB2)were involved in reducing Pb accumulation in radish roots.These findings could facilitate comprehensive elucidation of the molecular mechanism underlying MT-mediated Pb accumulation and detoxification in radish and other root vegetable crops.Mingjia Tang Liang Xu Yan Wang Junhui Dong Xiaoli Zhang Kai Wang Jiali Ying Cui Li Liwang Liu 2021Horticulture Research2021,8,1:0
17Perovskite/silicon-based heterojunction tandem solar cells with 14.8%conversion efficiency via adopting ultrathin Au contact显示文摘A rising candidate for upgrading the performance of an established narrow-bandgap solar technology without adding much cost is to construct the tandem solar cells from a crystalline silicon bottom cell and a high open-circuit voltage top cell.Here,we present a four-terminal tandem solar cell architecture consisting of a selffiltered planar architecture perovskite top cell and a silicon heterojunction bottom cell.A transparent ultrathin gold electrode has been used in perovskite solar cells to achieve a semi-transparent device.The transparent ultrathin gold contact could provide a better electrical conductivity and optical reflectance-scattering to maintain the performance of the top cell compared with the traditional metal oxide contact.The four-terminal tandem solar cell yields an efficiency of 14.8%,with contributions of the top(8.98%)and the bottom cell(5.82%),respectively.We also point out that in terms of optical losses,the intermediate contact of self-filtered tandem architecture is the uppermost problem,which has been addressed in this communication,and the results show that reducing the parasitic light absorption and improving the long wavelength range transmittance without scarifying the electrical properties of the intermediate hole contact layer are the key issues towards further improving the efficiency of this architecture device.Lin Fan Fengyou Wang Junhui Liang Xin Yao Jia Fang Dekun Zhang Changchun Wei Ying Zhao Xiaodan Zhang 2017Journal of Semiconductors2017,38,1:0

19Back contact interfacial modification mechanism in highly-efficient antimony selenide thin-film solar cells显示文摘Antimony selenide(Sb_(2)Se_(3))is a potential photovoltaic(PV)material for next-generation solar cells and has achieved great development in the last several years.The properties of Sb_(2)Se_(3)absorber and back contact influence the PV performances of Sb_(2)Se_(3)solar cells.Hence,optimization of back contact characteristics and absorber orientation are crucial steps in raising the power conversion efficiency(PCE)of Sb_(2)Se_(3)solar cells.In this work,MoO2was introduced as an intermediate layer(IL)in Sb_(2)Se_(3)solar cells,and comparative investigations were conducted.The growth of(211)-oriented Sb_(2)Se_(3)with large grains was facilitated by introducing the MoO2IL with suitable thickness.The MoO2IL substantially lowered the back contact barrier and prevented the formation of voids at the back contact,which reduced the thickness of the MoSe2interface layer,inhibited carrier recombination,and minimized bulk and interfacial defects in devices.Subsequently,significant optimization enhanced the open-circuit voltage(VOC)of solar cells from 0.481 V to 0.487 V,short-circuit current density(JSC)from 23.81 m A/cm^(2)to 29.29 m A/cm^(2),and fill factor from 50.28%to 57.10%,which boosted the PCE from 5.75%to 8.14%.Junhui Lin Guojie Chen Nafees Ahmad Muhammad Ishaq Shuo Chen Zhenghua Su Ping Fan Xianghua Zhang Yi Zhang Guangxing Liang 2023Journal of Energy Chemistry2023,,5:0
20Spatial variation of lipophilic marine algal toxins and its relationship with physicochemical parameters in spring in Laizhou Bay,China显示文摘Lipophilic marine algal toxins(LMATs)are produced by some toxigenic microalgae,which pose a serious threat to marine ecosystem and even human health.The occurrence and environmental control factors of LMATs in the surface seawater and phytoplankton in spring in Laizhou Bay in which Huanghe(Yellow)River estuary is included,in Shandong,East China were investigated.Okadaic acid(OA),pectenotoxin-2(PTX2),dinophysistoxin-1(DTX1),pectenotoxin-2 seco acid(PTX2 SA),DTX2,7-epi-PTX2 SA,PTX11,and 13-desmethyl spirolide C(SPX1)were detected from the surface seawater samples,and PTX2,7-epi-PTX2 SA,OA,DTX2,DTX1,PTX2 SA,and PTX11 were discovered in the phytoplankton samples showed a decreasing trend.The concentrations of∑LMATs in the seawater and phytoplankton ranged 2.03-74.38 ng/L on average of 13.72 ng/L and 0.98-479.27 pg/L on average of 50.20 pg/L,respectively.The joint influence of terrigenous input and internal circulation could promote the growth,toxin production,and toxin release of toxin-producing algae,leading to a higher content of LMATs in the bay nearby the Huanghe River estuary in both seawater and phytoplankton.The concentration of LMATs in spring was higher than that in summer,showing obvious seasonal variation.In addition,no significant correlation between most of the physiochemical parameters and LMAT contents in seawater was revealed by correlation analysis except for the positive correlation between DTX2 and chlorophyll a,OA and NH4-N.However,the increase of dissolved inorganic nitrogen content in seawater could enhance the production of OA,DTX1,and DTX2 in phytoplankton due probably to that the inorganic N input could benefit the growth and stimulate toxin production of toxin-producing algae.The result also proved that some PTX2 may be originated from Procentrum spp.and OA,DTXs and part of PTX2 may be originated from Dinophysis spp.Xiuping HE Junhui CHEN Jiuming WANG Xia SUN Ming XIN Baodong WANG Shengkang LIANG 2022Journal of Oceanology and Limnology2022,40,6:0
