| 1 | Mechanism of inhibiting type I interferon induction by hepatitis B virus X protein显示文摘Hepatitis B virus(HBV)is regarded as a stealth virus,invading and replicating efficiently in human liver undetected by host innate antiviral immunity.Here,we show that type I interferon(IFN)induction but not its downstream signaling is blocked by HBV replication in HepG2.2.15 cells.This effect may be partially due to HBV X protein(HBx),which impairs IFNβpromoter activation by both Sendai virus(SeV)and components implicated in signaling by viral sensors.As a deubiquitinating enzyme(DUB),HBx cleaves Lys63-linked polyubiquitin chains from many proteins except TANK-binding kinase 1(TBK1).It binds and deconjugates retinoic acid-inducible gene I(RIG I)and TNF receptor-associated factor 3(TRAF3),causing their dissociation from the downstream adaptor CARDIF or TBK1 kinase.In addition to RIG I and TRAF3,HBx also interacts with CARDIF,TRIF,NEMO,TBK1,inhibitor of kappa light polypeptide gene enhancer in B-cells,kinase epsilon(IKKi)and interferon regulatory factor 3(IRF3).Our data indicate that multiple points of signaling pathways can be targeted by HBx to negatively regulate production of type I IFN. | Junyi Jiang Hong Tang | 2010 | Protein & Cell2010,1,12: | 13 |
| 2 | Sox9 augments BMP2-induced chondrogenic differentiation by downregulating Smad7 in mesenchymal stem cells(MSCs)显示文摘Cartilage injuries caused by arthritis or trauma pose formidable challenges for effective clinical management due to the limited intrinsic proliferative capability of chondrocytes.Autologous stem cell-based therapies and transgene-enhanced cartilage tissue engineering may open new avenues for the treatment of cartilage injuries.Bone morphogenetic protein 2(BMP2)induces effective chondrogenesis of mesenchymal stem cells(MSCs)and can thus be explored as a potential therapeutic agent for cartilage defect repair.However,BMP2 also induces robust endochondral ossification.Although the precise mechanisms through which BMP2 governs the divergence of chondrogenesis and osteogenesis remain to be fully understood,blocking endochondral ossification during BMP2-induced cartilage formation may have practical significance for cartilage tissue engineering.Here,we investigate the role of Sox9-donwregulated Smad7 in BMP2-induced chondrogenic differentiation of MSCs.We find that overexpression of Sox9 leads to a decrease in BMP2-induced Smad7 expression in MSCs.Sox9 inhibits BMP2-induced expression of osteopontin while enhancing the expression of chondrogenic marker Col2a1 in MSCs.Forced expression of Sox9 in MSCs promotes BMP2-induced chondrogenesis and suppresses BMP2-induced endochondral ossification.Constitutive Smad7 expression inhibits BMP2-induced chondrogenesis in stem cell implantation assay.Mouse limb explant assay reveals that Sox9 expands BMP2-stimulated chondrocyte proliferating zone while Smad7 promotes BMP2-intitated hypertrophic zone of the growth plate.Cell cycle analysis indicates that Smad7 induces significant early apoptosis in BMP2-stimulated MSCs.Taken together,our results strongly suggest that Sox9 may facilitate BMP2-induced chondrogenesis by downregulating Smad7,which can be exploited for effective cartilage tissue engineering. | Chen Zhao Wei Jiang Nian Zhou Junyi Liao Mingming Yang Ning Hu Xi Liang Wei Xu Hong Chen Wei Liu Lewis L.Shi Leonardo Oliveira Jennifer Moriatis Wolf Sherwin Ho Aravind Athiviraham H.M.Tsai Tong-Chuan He Wei Huang | 2017 | Genes & Diseases2017,4,4: | 7 |
| 3 | Specific Properties of Air Flow Field Within the Grinding Zone显示文摘Air barrier of grinding means a boundary layer of air existing at the circumference of the rotating wheel,which hinders coolant from entry. This paper makes a research on air flow field of the grinding zone through experiments and numerical simulations,focusing on acquainting with the specific properties of the air flow field. Finite volume method is applied to analyze air flow field within grinding wheel in the course of numerical calculations. The test devices such as Hot-wire anemometer and Betz manometer are used during the experiments of testing the pressure and velocity within grinding zone. Results of experiments agree by and large with numerical results of calculations. The conclusions obtained in this paper,the distribution of wall pressure and the distribution of air flow velocity,are important and useful to navigate the delivery of coolant into the grinding zone. | 6 | Structural Asymmetry of Dorsolateral Prefrontal Cortex Correlates with Depressive Symptoms: Evidence from Healthy Individuals and Patients with Major Depressive Disorder显示文摘In this study,we investigated the role of structural asymmetry of the dorsolateral prefrontal cortex(DLPFC) in the continuum of depression from healthy individuals to patients.Structural magnetic resonance imaging was performed in 70 patients with major depressive disorder(MDD),49 matched controls,and 349 healthy university students to calculate structural asymmetry indexes of the DLPFC.First-episode,treatment-naive MDD patients showed a relatively lower asymmetry index than healthy controls,and their asymmetry index was negatively correlated with the depressive symptoms.This abnormality was normalized by antidepressants in medicated MDD patients.Furthermore,the asymmetry index was negatively correlated with the depressive symptoms in university students;this was replicated at two time points in a subgroup of students,suggesting good test–retest reliability.Our findings are consistent with previous studiesthat support the imbalance hypothesis of MDD and suggest a potential structural basis underlying the functional asymmetry of the DLPFC in depression.In future,the structural index of the DLPFC may become a potential biomarker to evaluate individuals' risk for the onset of MDD. | Wei Liu Yu Mao Dongtao Wei Junyi Yang Xue Du Peng Xie Jiang Qiu | 2016 | Neuroscience Bulletin2016,32,3: | 4 |
| 7 | Distr ibution, risk and bioavailability of metals in sediments of Lake Yamdrok Basin on the Tibetan Plateau, China显示文摘Total contents of metals in soil and sediments on the Tibetan Plateau of China have been widely analyzed,but existing information is insufficient to effectively evaluate metal ecological risk because of a lack of metal bioavailability data.In this study,distribution,potential risk,mobility and bioavailability of metals in sediments of Lake Yamdrok Basin in Tibet of China were explored by combined use of total digestion,sequential extraction and the diffusive gradient in thin-films(DGT).Average concentrations of Cr,Ni,Cu,Zn,As,Cd and Pb in surface sediments were 31.25,30.31,22.00,45.04,31.32,0.13 and 13.39 mg/kg,respectively.Higher levels of metals were found near the inflowing rivers.Residual form was dominant in Cr,Ni,Zn,Cd and Pb,and reducible form was dominant in As and Cd.Metals in surface sediments showed a low enrichment degree overall,but Cd and As had higher ecological risk levels than the other metals.Furthermore,there was a larger average proportion of exchangeable form of As(20.4%)and Cd(9.0%)than the other metals(1.7%-3.3%),implying their higher mobility and release risk.Average DGT-labile concentrations of Cr,Ni,Cu,Zn,As,Cd and Pb were 0.5,4.5,0.7,25.1,60.0,0.22 and 1.0μg/L,respectively.The DGT-labile As was significantly correlated with extractable As forms(p<0.01),suggesting that extractable As in sediments acts as a'mobile pool'for bioavailable As.These results suggest potential risks of As and Cd,especially As,deserve further attention in Lake Yamdrok Basin. | Feifei Che Junyi Chen Bo Zhang Xia Jiang Shuhang Wang | 2020 | Journal of Environmental Sciences2020,32,11: | 4 |
| 8 | Tenofovir vs.entecavir on recurrence of hepatitis B virus-related hepatocellular carcinoma beyond Milan criteria after hepatectomy显示文摘Background:Hepatectomy for hepatocellular carcinoma(HCC)beyond the Milan criteria is shown to be beneficial.However,a high rate of post-operative HCC recurrence hinders the long-term survival of the patients.This study aimed to investigate and compare the impacts of tenofovir(TDF)and entecavir(ETV)on the recurrence of hepatitis B viral(HBV)-related HCC beyond the Milan criteria.Methods:Data pertaining to 1532 patients who underwent hepatectomy and received antiviral therapy between January 2014 and January 2019 were collected from five centers.Recurrence-free survival(RFS)analysis was performed using the Kaplan-Meier method.Cox proportional hazards regression analysis was performed to determine prognostic factors for HCC recurrence.Results:The analysis incorporates 595 HBV-related HCC patients.The overall 5-year RFS was 21.3%.Among them,533 and 62 patients received ETV and TDF treatment,respectively.The 1-,3-,and 5-year RFS rates were 46.3%,27.4%,and 19.6%,respectively,in the ETV group compared with 65.1%,41.8%,and 37.2%,respectively,in the TDF group(P<0.001).Multivariate analysis showed that TDF treatment(hazard ratio[HR]:0.604,P=0.005),cirrhosis(HR:1.557,P=0.004),tumor size(HR:1.037,P=0.008),microvascular invasion(MVI)(HR:1.403,P=0.002),portal vein tumor thrombus(PVTT)(HR:1.358,P=0.012),capsular invasion(HR:1.228,P=0.040),and creatinine levels(CREA)(HR:0.993,P=0.031)were statistically significant prognostic factors associated with RFS.Conclusions:Patients with HCC beyond the Milan criteria exhibited a high rate of HCC recurrence after hepatectomy.Compared to the ETV therapy,TDF administration significantly lowered the risk of HCC recurrence. | Junyi Shen Weili Qi Junlong Dai Shusheng Leng Kangyi Jiang Yu Zhang Shun Ran Chuan Li Tianfu Wen | 2022 | Chinese Medical Journal2022,,3: | 3 |

In this scheme, multiple antennas are employed to strengthen the robustness in transmission, and to get more diversity gain. The bit error rate(BER) of the MIMO-CDSK is studied analytically in AWGN channel model and multipath fading channel model. The theory and simulation results show that, the performance gain can be obtained with multiple antennas allocated in the transmitter and receiver. | 16 | Differential SW16.1 allelic effects and genetic backgrounds contributed to increased seed weight after soybean domestication显示文摘Although seed weight has increased following domestication from wild soybean(Glycine soja) to cultivated soybean(Glycine max), the genetic basis underlying this change is unclear. Using mapping populations derived from chromosome segment substitution lines of wild soybean, we identified SW16.1 as the causative gene underlying a major quantitative trait locus controlling seed weight.SW16.1 encodes a nucleus-localized LIM domaincontaining protein. Importantly, the GsSW16.1 allele from wild soybean accession N24852 had a negative effect on seed weight, whereas the GmSW16.1 allele from cultivar NN1138-2 had a positive effect. Gene expression network analysis,reverse-transcription quantitative polymerase chain reaction, and promoter-luciferase reporter transient expression assays suggested that SW16.1 regulates the transcription of MT4, a positive regulator of seed weight. The natural variations in SW16.1 and other known seed weight genes were analyzed in soybean germplasm. The SW16.1 polymorphism was associated with seed weight in 247 soybean accessions, showing much higher frequency of positive-effect alleles in cultivated soybean than in wild soybean. Interestingly,gene allele matrix analysis of the known seed weight genes revealed that G. max has lost 38.5%of the G. soja alleles and that most of the lost alleles had negative effects on seed weight. | 20 | Measuring Mental Workload Using ERPs Based on FIR,ICA,and MARA显示文摘Mental workload is considered to be strongly linked to human performance,and the ability to measure it accurately is key for balancing human health and work.In this study,brain signals were elicited by mental arithmetic tasks of varying difficulty to stimulate different levels of mental workload.In addition,a finite impulse response(FIR)filter,independent component analysis(ICA),and multiple artifact rejection algorithms(MARAs)were used to filter event-related potentials(ERPs).Then,the data consisting of ERPs,subjective ratings of mental workload,and task performance,were analyzed through the use of variance and Spearman’s correlation during a simulated computer task.We found that participants responded faster and performed better in the easy task condition,followed by the medium and high-difficulty conditions,which verifies the validity of the ERP filtering.Moreover,larger P2 and P3 waveforms were evoked as the task difficulty increased,and a higher task difficulty elicited a more enhanced N300.Correlation analysis revealed a negative relationship between the amplitude of P3 and the subjective ratings,and a positive relationship between the P3 amplitude and accuracy.The results presented in this paper demonstrate that a combination of FIR,ICA,and MARA methods can filter ERPs in the non-invasive real-time measurement of workload.Additionally,frontocentral P2,N3,and parietal P3 components showed differences between genders.The proposed measurement of mental workload can be useful for real-time identification of mental states and can be applied to human-computer interaction in the future. | Yu Sun Yi Ding Junyi Jiang Vincent G.Duffy | 2022 | Computer Systems Science & Engineering2022,41,5: | 1 |
