| 1 | The first offshore natural gas hydrate production test in South China Sea显示文摘Natural gas hydrates (NGH)is one of key future clean energy resources.Its industrialized development will help remit the huge demand of global natural gas,relieve the increasing pressure of the environment, and play a vital role in the green sustainable growth of human societies.Based on nearly two decades' studying on the reservoir characteristics in the South China Sea (SCS)and the knowledge of reservoir system,the China Geological Survey (CGS)conducted the first production test on an optimal target selected in Shenhu area SCS in 2017.Guided by the 'three-phase control'exploitation theory which focused on formation stabilization,technologies such as formation fluid extraction,well drilling and completing, reservoir stimulating,sand controlling,environmental monitoring,monitoring and preventing of secondary formation of hydrates were applied.The test lasted for 60 days from May 10^th when starting to pump, drop pressure and ignite to well killing on July 9^th,with gas production of 3.09×10^5 m^3 in total,which is a world record with the longest continuous duration of gas production and maximal gas yield.This successful test brings a significant breakthrough on safety control of NGH production. | Jin-fa Li Jian-liang Ye Xu-wen Qin Hai-jun Qiu Neng-you Wu Hai-long Lu Wen-wei Xie Jing-an Lu Fei Peng Zhen-qiang Xu Cheng Lu Zeng-gui Kuang Jian-gong Wei Qian-yong Liang Hong-feng Lu Bei-bei Kou | 2018 | China Geology2018,1,1: | 117 |
| 2 | The second natural gas hydrate production test in the South China Sea显示文摘Clayey silt reservoirs bearing natural gas hydrates(NGH)are considered to be the hydrate-bearing reservoirs that boast the highest reserves but tend to be the most difficult to exploit.They are proved to be exploitable by the first NGH production test conducted in the South China Sea in 2017.Based on the understanding of the first production test,the China Geological Survey determined the optimal target NGH reservoirs for production test and conducted a detailed assessment,numerical and experimental simulation,and onshore testing of the reservoirs.After that,it conducted the second offshore NGH production test in 1225 m deep Shenhu Area,South China Sea(also referred to as the second production test)from October 2019 to April 2020.During the second production test,a series of technical challenges of drilling horizontal wells in shallow soft strata in deep sea were met,including wellhead stability,directional drilling of a horizontal well,reservoir stimulation and sand control,and accurate depressurization.As a result,30 days of continuous gas production was achieved,with a cumulative gas production of 86.14×104 m3.Thus,the average daily gas production is 2.87×10^4 m^3,which is 5.57 times as much as that obtained in the first production test.Therefore,both the cumulative gas production and the daily gas production were highly improved compared to the first production test.As indicated by the monitoring results of the second production test,there was no anomaly in methane content in the seafloor,seawater,and atmosphere throughout the whole production test.This successful production test further indicates that safe and effective NGH exploitation is feasible in clayey silt NGH reservoirs.The industrialization of hydrates consists of five stages in general,namely theoretical research and simulation experiments,exploratory production test,experimental production test,productive production test,and commercial production.The second production test serves as an important step from the exploratory production test to experimental production test. | Jian-liang Ye Xu-wen Qin Wen-wei Xie Hai-long Lu Bao-jin Ma Hai-jun Qiu Jin-qiang Liang Jing-an Lu Zeng-gui Kuang Cheng Lu Qian-yong Liang Shi-peng Wei Yan-jiang Yu Chunsheng Liu Bin Li Kai-xiang Shen Hao-xian Shi Qiu-ping Lu Jing Li Bei-bei Kou Gang Song Bo Li He-en Zhang Hong-feng Lu Chao Ma Yi-fei Dong Hang Bian | 2020 | China Geology2020,3,2: | 22 |

| 6 | Promotion of spinosad biosynthesis by chromosomal integration of the Vitreoscilla hemoglobin gene in Saccharopolyspora spinosa显示文摘To promote spinosad biosynthesis by improving the limited oxygen supply during high-density fermentation of Saccharopolyspora spinosa, the open reading frame of the Vitreoscilla hemoglobin gene was placed under the control of the promoter for the erythromycin resistance gene by splicing using overlapping extension PCR. This was cloned into the integrating vector pSET152, yielding the Vitreoscilla hemoglobin gene expression plasmid pSET152EVHB. This was then introduced into S. spinosa SP06081 by conjugal transfer, and integrated into the chromosome by site-specific recombination at the integration site ΦC31 on pSET152EVHB. The resultant conjugant, S. spinosa S078-1101, was genetically stable. The integration was further confirmed by PCR and Southern blotting analysis. A carbon monoxide differential spectrum assay showed that active Vitreoscilla hemoglobin was successfully expressed in S. spinosa S078-1101. Fermentation results revealed that expression of the Vitreoscilla hemoglobin gene significantly promoted spinosad biosynthesis under normal oxygen and moderately oxygen-limiting conditions (P<0.01). | 7 | Genesis of the Wulong gold deposit,Liaoning Province,NE China:Constrains from noble gases,radiogenic and stable isotope studies显示文摘TheWulong lode deposit contains over 80 tonnes of gold with an average grade of 5.35 g/t.It is one of the largest deposits in Dandong City,Liaoning Province in northeast China.Previous studies on the deposit focused on its geological characteristics,geochemistry,fluid inclusions,and the timing of gold mineralization.However,controversy remains regarding the origin of the ore-forming fluids and metals,and the genesis of the gold deposit.This paper presents zircon UePb and pyrite RbeSr ages and S,Pb,He,and Ar isotopic results along with quartz H and O isotopic data for all litho-units associated with the deposit.Laser ablation inductively coupled mass spectrometry measurements yielded zircon UePb dates for samples of pre-mineralization rocks like granite porphyry dike,the Sanguliu granodiorite,fine-grained diorite,and syn-mineralization diorite,as well as post-mineralization dolerite,and lamprophyre;their emplacement ages are 1261 Ma,1241 Ma,1231 Ma,1201 Ma,1192 Ma,and 1152 Ma,respectively.The pyrite RbeSr isochron age is 1191 Ma,indicating that both magmatism and mineralization occurred during the Early Cretaceous.The d18OH2O values of ore-forming hydrothermal fluids from the quartzepolymetallic sulfide vein stage vary from 4.8&to 6.5&,and the dDV-SMOW values are between67.7&and75.9&,indicating that the ore-forming fluids were primarily magmatic.The noble gas isotope compositions of fluid inclusions hosted in pyrite suggest that the ore-forming fluids were dominantly derived from crustal sources with minor mantle input.Sulfur isotopic values of pyrite vary between 0.2&and 3.5&,suggesting that S was derived from a homogeneous magmatic source or possibly from fluids derived from the crust.The Pb isotopic compositions of sulfides(207Pb/204Pb?15.51 e15.71,206Pb/204Pb?17.35e18.75,208Pb/204Pb?38.27e40.03)indicate that the Pb of the Wulong gold deposit is a mixture of crust and mantle components.Geochronological and geochemical data,together with the regional geological history,indicate that Early Cretaceous magmatism and mineralization of the Wulong gold deposit occurred during the rollback of the subducting Paleo-Pacific Plate,which resulted in lithospheric thinning and the destruction of the North China Craton(NCC),which indicates that the deposit is of magmaticehydrothermal origin. | Peng Zhang Linlin Kou Yan Zhao Zhongwei Bi Deming Sha Renping Han Zhumin Li | 2020 | Geoscience Frontiers2020,11,2: | 13 |
| 8 | Network pharmacology-based prediction and verification of the molecular targets and pathways for schisandrin against cerebrovascular disease显示文摘AIM: To illuminate the molecular targets for schisandrin against cerebrovascular disease based on the combined methods of network pharmacology prediction and experimental verification. METHOD: A protein database was established through constructing the drug-protein network from literature mining data. The protein-protein network was built through an in-depth exploration of the relationships between the proteins. The computational platform was implemented to predict and extract the sensitive sub-network with significant P-values from the protein-protein network. Then the key targets and pathways were identified from the sensitive sub-network. The most related targets and pathways were also confirmed in hydrogen peroxide(H2O2)-induced PC12 cells by Western blotting. RESULTS: Twelve differentially expressed proteins(gene names: NFKB1, RELA, TNFSF10, MAPK1, CHUK, CASP8, PIGS2, MAPK14, CREB1, IFNG, APP, and BCL2) were confirmed as the central nodes of the interaction network(45 nodes, 93 edges). The NF-κB signaling pathway was suggested as the most related pathway of schisandrin for cerebrovascular disease. Furthermore, schisandrin was found to suppress the expression and phosphorylation of IKKα, as well as p50 and p65 induced by H2O2 in PC12 cells by Western blotting. | 11 | Resistance reduction by bionic coupling of the earthworm lubrication function显示文摘Based on the biological coupling theory, the resistance reduction characteristic of the surface morphology and surface wettability of the earthworm were studied in this paper. The parameters of surface dorsal pore and corrugation were extracted. According to these parameters, the lubrication mechanism of the earthworm surface was analyzed. The distribution of the pores and surface morphology were designed and the bionic coupling samples were prepared. The positive pressure, lubricant flow rate and advancing velocity were selected as the experiment factors while the soil friction resistance as observed object. According to the obtained data of bionic coupling samples from the testing system of biologic signal for tiny soil adhesion test, the optimal samples from the bionic coupling resistance reduction tests were selected through the range analysis. Compared to the normal ones, the soil resistance of bionic coupling samples was reduced by 76.8%. | 16 | Autologous transplantation with fewer fibers repairs large peripheral nerve defects显示文摘Peripheral nerve injury is a serious disease and its repair is challenging. A cable-style autologous graft is the gold standard for repairing long peripheral nerve defects; however, ensuring that the minimum number of transplanted nerve attains maximum therapeutic effect remains poorly understood. In this study, a rat model of common peroneal nerve defect was established by resecting a 10-mm long right common peroneal nerve. Rats receiving transplantation of the common peroneal nerve in situ were designated as the in situ graft group. Ipsilateral sural nerves(10–30 mm long) were resected to establish the one sural nerve graft group, two sural nerves cable-style nerve graft group and three sural nerves cable-style nerve graft group. Each bundle of the peroneal nerve was 10 mm long. To reduce the barrier effect due to invasion by surrounding tissue and connective-tissue overgrowth between neural stumps, small gap sleeve suture was used in both proximal and distal terminals to allow repair of the injured common peroneal nerve. At three months postoperatively, recovery of nerve function and morphology was observed using osmium tetroxide staining and functional detection. The results showed that the number of regenerated nerve fibers, common peroneal nerve function index, motor nerve conduction velocity, recovery of myodynamia, and wet weight ratios of tibialis anterior muscle were not significantly different among the one sural nerve graft group, two sural nerves cable-style nerve graft group, and three sural nerves cable-style nerve graft group. These data suggest that the repair effect achieved using one sural nerve graft with a lower number of nerve fibers is the same as that achieved using the two sural nerves cable-style nerve graft and three sural nerves cable-style nerve graft. We evaluated the recovery from lung damage and femoral head necrosis in an observational cohort study of SARS patients using pulmonary CT scans, hip joint MRI examinations, pulmonary function tests and hip joint function questionnaires. Eighty medical staff contracted SARS in 2003. Two patients died of SARS, and78 were enrolled in this study from August 2003 to March 2018. Seventy-one patients completed the 15-year follow-up. The percentage of pulmonary lesions on CT scans diminished from 2003(9.40 ± 7.83)% to 2004(3.20 ± 4.78)%(P < 0.001) and remained stable thereafter until 2018(4.60 ± 6.37)%. Between 2006 and 2018, the proportion of patients with interstitial changes who had improved pulmonary function was lower than that of patients without lesions, as demonstrated by the one-second ratio(FEV1/FVC%, t = 2.21, P = 0.04) and mid-flow of maximum expiration(FEF25%–75%, t = 2.76, P = 0.01). The volume of femoral head necrosis decreased significantly from 2003(38.83 ± 21.01)% to 2005(30.38 ± 20.23)%(P = 0.000 2), then declined slowly from 2005 to 2013(28.99 ± 20.59)% and plateaued until 2018(25.52 ± 15.51)%. Pulmonary interstitial damage and functional decline caused by SARS mostly recovered, with a greater extent of recovery within 2 years after rehabilitation. As a critical problem in cooperative air combat,outcome of WTA directly influences the battle. Along with the cost of weapons rising rapidly, it is indispensable to design a target assignment model that can ensure minimizing targets survivability and weapons consumption simultaneously. Afterwards an algorithm named as improved artificial fish swarm algorithm-improved harmony search algorithm(IAFSA-IHS) is proposed to solve the problem. | 20 | Molecular cloning, sequence analysis, and cadmium stress-rated expression changes of BTG1 in freshwater pearl mussel(Hyriopsis schlegelii)显示文摘The B cells translocation gene 1(BTG1) is a member of the BTG/TOB family of anti-proliferative genes,which have recently emerged as important regulators of cell growth and differentiation among vertebrates.Here,for the first time we cloned the full-length cDNA sequence of Hyriopsis schlegelii(Hs-BTG1),an economically important freshwater shellfish and potential indicator of environmental heavy metal pollution,for the first time.Using rapid amplification of cDNA ends(RACE) together with splicing the EST sequence from a haemocyte cDNA library,we found that Hs-BTG1 contains a 525 bp open reading frame(ORF) encoding a 174 amino-acid polypeptide,a 306 bp 5' untranslated region(5' UTR),and a 571 bp 3' UTR with a Poly(A) tail as well as a transcription termination signal(AATAAA).Homologue searching against GenBank revealed that Hs-BTG1 was closest to Crassostrea gigas BTG1,sharing 50.57% of protein identities.Hs-BTG1 also shares some typical features of the BTG/TOB family,possessing two well-conserved A and B boxes.Clustering analysis of Hs-BTG1 and other known BTGs showed that Hs-BTG1 was also closely related to BTG1 of C.gigas from the invertebrate BTG1 clade.Function prediction via homology modeling showed that both Hs-BTG1 and C.gigas BTG1 share a similar three-dimensional structure with Homo sapiens BTG1.Tissue-specific expression analysis of the Hs-BTG1 via real-time PCR showed that the transcripts were constitutively expressed,with the highest levels in the hepatopancreas and gills,and the lowest in both haemocyte and muscle tissue.Expression levels of Hs-BTG1 in hepatopancreas(2.03-fold),mantle(2.07-fold),kidney(2.2-fold) and haemocyte(2.5-fold) were enhanced by cadmium(Cd2+) stress,suggesting that Hs-BTG1 may have played a significant role in H.schlegelii adaptation to adverse environmental conditions. | Kou PENG Cheng-Yuan WANG Jun-Hua WANG Jun-Qing SHENG Jian-Wu SHI Jian LI Yi-Jiang HONG | 2014 | Zoological Research2014,35,5: | 7 |
