| 1 | Synthesis, characterization and size control of zerovalent iron nanoparticles anchored on montmorillonite显示文摘Zerovalent iron nanoparticles have been successfully synthesized using sodium borohydride solution reduction of ferric trichloride hexahydrate in the presence of montmorillonite as an effective protective reagent and support as well. A combination of characterizations reveals that with high monodispersity these obtained iron nanoparticles are well dispersed on clay surface, virginal from boron related impurity, and oxidation resistant well with iron core-iron oxide shell structure. The shell thickness of 3 nm remains almost invariable under ambient conditions. | 2 | GCH1 plays a role in the high-altitude adaptation of Tibetans显示文摘Tibetans are well adapted to high-altitude hypoxia. Previous genome-wide scans have reported many candidate genes for this adaptation, but only a few have been studied. Here we report on a hypoxia gene(GCH1, GTP-cyclohydrolase I), involved in maintaining nitric oxide synthetase(NOS) function and normal blood pressure, that harbors many potentially adaptive variants in Tibetans. We resequenced an 80.8 kb fragment covering the entire gene region of GCH1 in 50 unrelated Tibetans. Combined with previously published data, we demonstrated many GCH1 variants showing deep divergence between highlander Tibetans and lowlander Han Chinese. Neutrality tests confirmed a signal of positive Darwinian selection on GCH1 in Tibetans. Moreover, association analysis indicated that the Tibetan version of GCH1 was significantly associated with multiple physiological traits in Tibetans, including blood nitric oxide concentration, blood oxygen saturation, and hemoglobin concentration. | 3 | Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. | 5 | Impact of the Built Environment on the Spatial Heterogeneity of Regional Innovation Productivity:Evidence from the Pearl River Delta,China显示文摘With the global economy increasingly dependent on innovation,urban discourse has shifted to consider what kinds of spatial designs may best nurture innovation.We examined the relationship between the built environment and the spatial heterogeneity of regional innovation productivity(RIP)using the example of China's Pearl River Delta(PRD).Based on a spatial database of 522546 patent data from 2017,this study proposed an innovation-based built environment framework with the following five aspects:healthy en-vironment,daily interaction,mixed land use,commuting convenience,and technology atmosphere.Combining negative binomial regression and Geodetector to examine the impact of the built environment on RIP,the results show that the spatial distribution of innovation productivity in the PRD region is extremely uneven.The negative binomial regression results show that the built environment has a significant impact on the spatial differentiation of RIP,and,specifically,that healthy environment,mixed land use,commuting convenience,and technology atmosphere all demonstrate significant positive impacts.Meanwhile,the Geodetector results show that the built environment factor impacts the spatial heterogeneity of RIP to varying degrees,with technology atmosphere demonstrating the greatest impact intensity.We conclude that as regional development discourse shifts focus to the knowledge and innovation economy,the innovation-oriented design and updating of built environments will become extremely important to policymakers. | WU Kangmin WANG Yang ZHANG Hong’ou LIU Yi YE Yuyao | 2021 | Chinese Geographical Science2021,31,3: | 5 |
| 6 | EP300 contributes to high-altitude adaptation in Tibetans by regulating nitric oxide production显示文摘The genetic adaptation of Tibetans to high altitude hypoxia likely involves a group of genes in the hypoxic pathway, as suggested by earlier studies. To test the adaptive role of the previously reported candidate gene EP300(histone acetyltransferase p300), we conducted resequencing of a 108.9 kb gene region of EP300 in 80 unrelated Tibetans. The allele-frequency and haplotype-based neutrality tests detected signals of positive Darwinian selection on EP300 in Tibetans, with a group of variants showing allelic divergence between Tibetans and lowland reference populations, including Han Chinese, Europeans, and Africans. Functional prediction suggested the involvement of multiple EP300 variants in gene expression regulation. More importantly, genetic association tests in 226 Tibetans indicated significant correlation of the adaptive EP300 variants with blood nitric oxide(NO) concentration. | 7 | Seismic vulnerability evaluation of axially loaded steel built-up laced members I:experimental results显示文摘An experimental program was initiated to investigate the seismic performance of built-up laced steel brace members. Quasi-static testing of twelve typical steel built-up laced member (BLM) specimens was conducted. These were designed to span a range of parameters typically encountered for such members based on findings from a survey of commonly used shapes and details that have been historically used. The specimens were subdivided into groups of three different cross-sectional shapes, namely built-up I-shape section, and built-up box shapes buckling about the x or the y axis. Within each group, global and local buckling slenderness ratios had either kl/r values of 60 or 120, and b/t ratios of 8 or 16. The specific inelastic cyclic behavior germane to each specimen, and general observations on overall member hysteretic behavior as a function of the considered parameters, are reported. | 9 | Seismic vulnerability evaluation of axially loaded steel built-up laced members II:evaluations显示文摘The test results described in Part 1 of this paper (Lee and Bruneau, 2008) on twelve steel built-up laced members (BLMs) subjected to quasi-static loading are analyzed to provide better knowledge on their seismic behavior. Strength capacity of the BLM specimens is correlated with the strength predicted by the AISC LRFD Specifications. Assessments of hysteretic properties such as ductility capacity, energy dissipation capacity, and strength degradation after buckling of the specimen are performed. The compressive strength of BLMs is found to be relatively well predicted by the AISC LRFD Specifications. BLMs with smaller kl/r were ductile but failed to reach the target ductility of 3.0 before starting to fracture, while those with larger kl/r could meet the ductility demand in most cases. The normalized energy dissipation ratio, EC/ET and the normalized compressive strength degradation, Cr'/Cr of BLMs typically decrease as normalized displacements δ/δb,exp increase, and the ratios for specimens with larger kl/r dropped more rapidly than for specimens with smaller kl/r; similar trends were observed for the monolithic braces. | 14 | Single-molecule imaging reveals the stoichiometry change of epidermal growth factor receptor during transactivation by β_2-adrenergic receptor显示文摘Stimulation of G protein-coupled receptors(GPCRs) can lead to the transactivation of the epidermal growth factor receptors(EGFR). The cross-communication between the two signaling pathways regulates several important physiological or pathological processes. However, the molecule mechanism underlying EGFR transactivation remains poorly understood. Here, we aim to study the GPCR-mediated EGFR transactivation process using the single-molecule fluorescence imaging and tracking approach.We found that although EGFR existed as monomers at the plasma membrane of resting cells, they became dimers and thus diffused slower following the activation of β2-adrenergic receptor(β2-AR) by isoproterenol(ISO). We further proved thatβ2-AR-mediated changes of EGFR in stoichiometry and dynamics were mediated by Src kinase.
