| 1 | Intestinal lysozyme liberates Nod1 ligands from microbes to direct insulin trafficking in pancreatic beta cells显示文摘Long-range communication between intestinal symbiotic bacteria and extra-intestinal organs can occur through circulating bacterial signal molecules, through neural circuits, or through cytokines or hormones from host cells. Here we report that Nod1 ligands derived from intestinal bacteria act as signal molecules and directly modulate insulin trafficking in pancreatic beta cells. The cytosolic peptidoglycan receptor Nod1 and its downstream adapter Rip2 are required for insulin trafficking in beta cells in a cell-autonomous manner. Mechanistically, upon recognizing cognate ligands, Nod1 and Rip2 localize to insulin vesicles, recruiting Rab1a to direct insulin trafficking through the cytoplasm. Importantly, intestinal lysozyme liberates Nod1 ligands into the circulation, thus enabling long-range communication between intestinal microbes and islets. The intestine-islet crosstalk bridged by Nod1 ligands modulates host glucose tolerance. | 4 | The EGFR-P38 MAPK axis up-regulates PD-L1 through miR-675-5p and down-regulates HLA-ABC via hexokinase-2 in hepatocellular carcinoma cells显示文摘Background:Immunotherapy has been shown to be a promising strategy against human cancers.A better understanding of the immune regulation in hepatocellular carcinoma(HCC)could help the development of immunotherapy against HCC.The epidermal growth factor receptor(EGFR)signaling is frequently activated in HCC and plays important roles in tumorigenesis.However,its role in HCC immunity is still largely unknown.This study aimed to investigate the impact of EGFR signaling on programmed death-ligand 1(PD-L1)and human leukocyte antigen class-I(HLA-I)expression in HCC cells and its underlying mechanisms.Methods:The expression of phosphorylated EGFR(p-EGFR),PD-L1,and HLAI(HLA-ABC)in HCC specimens was detected by immunohistochemistry,and their correlations were analyzed.PD-L1 and HLA-ABC expression in EGFRactivated HCC cells were detected by quantitative real-time PCR,Western blotting,and flow cytometry,and T cell-mediated lysis was performed to test the immunosuppressive effects of PD-L1 and HLA-ABC alterations in HCC cells.Furthermore,the underlying mechanisms of EGFR activation-induced PD-L1 up-regulation and HLA-ABC down-regulation were explored by animal experiments,luciferase reporter assay,and gene gain-and loss-of-function studies.Results:p-EGFR was positively correlated with PD-L1 and negatively correlated with HLA-ABC expression in HCCs.EGFR activation by its ligand EGF up-regulated PD-L1 and down-regulated HLA-ABC in HCC cells,which was functionally important and could be abolished by the EGFR inhibitor,gefitinib,both in vitro and in vivo.Mechanistically,enhanced P38 mitogenactivated protein kinase(MAPK)activation down-regulated microRNA-675-5p(miR-675-5p)and up-regulated glycolysis-related enzyme hexokinase 2(HK2);miR-675-5p down-regulation enhanced the stability of PD-L1 mRNA probably via the 3’-untranslated region(3’-UTR)of PD-L1 and thereby caused PD-L1 accumulation,and HK2 up-regulation enhanced aerobic glycolysis and mediated a decrease in HLA-ABC.Conclusions:The EGFR-P38 MAPK axis could up-regulate PD-L1 through miR-675-5p and down-regulate HLA-ABC via HK2 in HCC cells.Our study reveals a novel signaling network that may cause immune suppression in HCC and suggests that EGFR signaling can be targeted for HCC immunotherapy. | Zongcai Liu Fen Ning Yanna Cai Huiying Sheng Ruidan Zheng Xi Yin Zhikun Lu Ling Su Xiaodan Chen Chunhua Zeng Haifang Wang Li Liu | 2021 | Cancer Communications2021,41,1: | 5 |
| 5 | Link Prediction in Brain Networks Based on a Hierarchical Random Graph Model显示文摘Link prediction attempts to estimate the likelihood of the existence of links between nodes based on available brain network information, such as node attributes and observed links. In response to the problem of the poor efficiency of general link prediction methods applied to brain networks, this paper proposes a hierarchical random graph model based on maximum likelihood estimation. This algorithm uses brain network data to create a hierarchical random graph model. Then, it samples the space of all possible dendrograms using a Markov-chain Monte Carlo algorithm. Finally, it calculates the average connection probability. It also employs an evaluation index.Comparing link prediction in a brain network with link prediction in three different networks(Treponemapallidum metabolic network, terrorist networks, and grassland species food webs) using the hierarchical random graph model, experimental results show that the algorithm applied to the brain network has the highest prediction accuracy in terms of AUC scores. With the increase of network scale, AUC scores of the brain network reach 0.8 before gradually leveling off. In addition, the results show AUC scores of various algorithms computed in networks of eight different scales in 28 normal people. They show that the HRG algorithm is far better than random prediction and the ACT global index, and slightly inferior to local indexes CN and LP. | 6 | An on-chip intestine-liver model for multiple drugs absorption and metabolism behavior simulation显示文摘A double-layer microfluidic chip integrated with a hollow fiber(HF)was developed to reconstitute the intestine-liver functionality for studying the absorption and metabolism of combination drugs.Caco-2 cells were inoculated in the HF cavity at the top of the serpentine channel to simulate the intestinal tissue for drug absorption and transport studied,and Hep G2 cells,seeded in the bottom chamber,were used to mimic the liver for metabolism-related studies.Genistein and dacarbazine were selected for combination drug therapy and its effects on cell viability,hepatotoxicity,and cell cycle arrest under drug-conditioned culture were investigated.The results suggested that the combined concentration below-100μg/m L had no significant inhibitory effect on Hep G2 cell viability,and therefore Hep G2 cells maintained their drug metabolism ability.When the drug concentration was increased above 250μg/m L,Hep G2 cells underwent apoptosis.Detection of metabolites by mass spectrometry proved the effective metabolism in the microchip model.This dynamic,co-culture microchip successfully provided a podium for long-term observation of absorption,transport,and metabolism of combination drugs,and could be an effective in vitro simulation model for further clinical research. | Mingsha Jie Haifeng Lin Ziyi He Hanyang Liu Haifang Li Jin-Ming Lin | 2018 | Science China Chemistry2018,61,2: | 3 |
| 7 | SciFinder-guided rational design of fluorescent carbon dots for ratiometric monitoring intracellular pH fluctuations under heat shock显示文摘Intracellular pH plays a significant role in various biological processes, including cell proliferation,apoptosis, metabolism, enzyme activity and homeostasis. In this work, a novel design strategy for the preparation of pH responsive carbon dots(CDs-pH) for ratiometric intracellular imaging was reported. By using SciFinder database, fluorescent CDs-pH with the required p Kavalue of 6.84 were rationally designed, which is vital important for precise sensing of intracellular pH. As a result, the synthesized CDspH demonstrated robust ability to test pH fluctuations within the physiological range of 5.4-7.4. The CDspH was further utilized for fluorescent ratiometric imaging of pH in living HeLa cells, effectively avoided the influence of autofluorescence from native cellular species. Moreover, real-time monitoring of intracellular pH fluctuation under heat shock was successfully realized. | 9 | Forest soil CO_2 fluxes as a function of understory removal and N-fixing species addition显示文摘We report on the effects of forest management practices of understory removal and N-fixing species(Cassia alata) addition on soil CO2 fluxes in an Eucalyptus urophylla plantation(EUp),Acacia crassicarpa plantation(ACp),10-species-mixed plantation(Tp),and 30-species-mixed plantation(THp) using the static chamber method in southern China.Four forest management treatments,including(1) understory removal(UR);(2) C.alata addition(CA);(3) understory removal and replacement with C.alata(UR+CA);and(4) control without any disturbances(CK),were applied in the above four forest plantations with three replications for each treatment.The results showed that soil CO2 fluxes rates remained at a high level during the rainy season(from April to September),followed by a rapid decrease after October reaching a minimum in February.Soil CO2 fluxes were significantly higher(P < 0.01) in EUp(132.6 mg/(m2.hr)) and ACp(139.8 mg/(m2.hr)) than in Tp(94.0 mg/(m2.hr)) and THp(102.9 mg/(m2.hr)).Soil CO2 fluxes in UR and CA were significantly higher(P < 0.01) among the four treatments,with values of 105.7,120.4,133.6 and 112.2 mg/(m2.hr) for UR+CA,UR,CA and CK,respectively.Soil CO2 fluxes were positively correlated with soil temperature(P < 0.01),soil moisture(P < 0.01),NO3?-N(P < 0.05),and litterfall(P < 0.01),indicating that all these factors might be important controlling variables for soil CO2 fluxes.This study sheds some light on our understanding of soil CO2 flux dynamics in forest plantations under various management practices. | Haifang Li Shenglei Fu Hongting Zhao Hanping Xia | 2011 | Journal of Environmental Sciences2011,23,6: | 3 |

Following a brief introduction about history and development of multi-channel microchip and MS combination, we will elaborate the key issues of constructing chip-MS platform interface. Then exciting progresses made in this field should be reviewed with well exemplified works, including chip-MS technology for cell introduction, pretreatment of cell secretions and cell metabolite analysis. We will also describe the development of integrated total analysis systems proposed by our group. | 11 | Novel Model Using Kernel Function and Local Intensity Information for Noise Image Segmentation显示文摘It remains a challenging task to segment images that are distorted by noise and intensity inhomogeneity.To overcome these problems, in this paper, we present a novel region-based active contour model based on local intensity information and a kernel metric. By introducing intensity information about the local region, the proposed model can accurately segment images with intensity inhomogeneity. To enhance the model's robustness to noise and outliers, we introduce a kernel metric as its objective functional. To more accurately detect boundaries, we apply convex optimization to this new model, which uses a weighted total-variation norm given by an edge indicator function. Lastly, we use the split Bregman iteration method to obtain the numerical solution. | 13 | Analysis of cDNA sequence,protein structure and expression of parotid secretory protein in pig显示文摘Parotid secretory protein (PSP) secreted abundantly in saliva, whose function is related with the anti-bacterial effect. The PSP cDNA has been isolated from pig parotid glands by 3′ and 5′ rapid amplification of cDNA end (RACE), based on the conserved signal peptide region among the known mammalian PSP. The result of homologous comparison shows that pig PSP and human PSP shares the high identity at the level of the primary, secondary and tertiary protein structure. A search for functionally significant protein motifs revealed a unique amino acid sequence pattern consisting of the residues Leu-X(6)-Leu-X(6)-Leu- X(7)-Leu-X(6)-Leu-X(6)-Leu near the amino-terminal portion of the protein, which is important to its function. | 14 | On-chip solid phase extraction coupled with electrophoresis using modified magnetic microspheres as stationary phase显示文摘A poly(dimethylsiloxane)(PDMS)/glass hybrid microchip for on-line solid phase extraction (SPE) and electrophoresis separation has been developed and evaluated. The SPE microchannel was crossed to the electrophoresis microchannel. All the microfluidic channels were etched on the glass substrate. The magnetic microspheres were coated with hydroxyl-terminated poly-dimethylsiloxane (PDMS-OH) serving as extraction phase,which could be conveniently immobilized into the sample pretreatment channel by magnetic field. The PDMS-OH microspheres were mobilized into and out of the pretreatment channel by injection flow. The 0.1 μmol/L solution of fluorescence isothiocyanate (FITC)-labeled phenylalanine (Phe) was electrically injected into the SPE channel and extracted onto the PDMS-OH microspheres bed. The enriched FITC-labeled Phe was electrically eluted by 9 mmol/L sodium acetate containing 10% acetonitrile and electrically driven into the electrophoresis channel and then separated. The preconcentration factor could reach 87.5 after sufficient extraction. A linear preconcentration curve was obtained with the initial FITC-labeled Phe concentration ranging from 6 nmol/L to 300 nmol/L (R2=0.9922) with 200 s loading time. | 15 | Combination of dynamic hollow fiber liquid-phase microextraction with HPLC analysis for the determination of UV filters in cosmetic products显示文摘A simple method based on hollow fiber liquid-phase microextraction (HF-LPME) followed by high performance liquid chro-matography (HPLC) analysis was successfully developed for the determination of UV filters in cosmetic products. A canular extractor was assembled by mounting a hollow fiber inside an external tube with a tee-connector. The organic solvent was immobilized into the fiber to form a liquid membrane as the acceptor phase. The sample was continuously injected into the extractor and the UV filters were extracted from the aqueous sample into organic acceptor phase. The main parameters affecting HF-LPME including extraction solvent, sample volume, sample flow rate, pH values and ionic strength were investigated. Toluene has been verified to be suitable as the acceptor phase. Under the optimized HF-LPME conditions, the enrichment factors of five UV filters varying from 24 to 57 were achieved. The limits of detection for the five UV filters were in the range of 1-100 μg L-1 .The relative standard deviations (RSDs) of HF-LPME and HPLC analysis were lower than 5.2%. | 16 | Construction of on-line supercritical fluid extraction with reverse phase liquid chromatography–tandem mass spectrometry for the determination of capsaicin显示文摘A novel on-line system composed of supercritical fluid extraction(SFE), dilution line and reverse phase liquid chromatography/mass spectrometry(RPLC/MS) was constructed for on-line extraction and reverse phase separation of some fat-soluble components in foods. Three columns including a trap column,concentration column and analytical column were used for trapping the fat-soluble components, on-line enrichment and reverse phase separation, respectively. Capsaicinoids were on-line extracted by a CO_2 supercritical fluid, then concentrated and separated by using the C_(18) columns, finally detected by mass spectrometry(MS). Capsaicin eluted at 10.1 min and limit of detection(LOD, S/N=3) for the standard solution is 0.55pg. The linearity was calculated with a value of coefficient of determination(R^2)≥0.998 in the range of 1.1–8.5 ng. | 19 | Combining Residual Attention Mechanisms and Generative Adversarial Networks for Hippocampus Segmentation显示文摘This research discussed a deep learning method based on an improved generative adversarial network to segment the hippocampus.Different convolutional configurations were proposed to capture information obtained by a segmentation network.In addition,a generative adversarial network based on Pixel2Pixel was proposed.The generator was a codec structure combining a residual network and an attention mechanism to capture detailed information.The discriminator used a convolutional neural network to discriminate the segmentation results of the generated model and that of the expert.Through the continuously transmitted losses of the generator and discriminator,the generator reached the optimal state of hippocampus segmentation.T1-weighted magnetic resonance imaging scans and related hippocampus labels of 130 healthy subjects from the Alzheimer’s disease Neuroimaging Initiative dataset were used as training and test data;similarity coefficient,sensitivity,and positive predictive value were used as evaluation indicators.Results showed that the network model could achieve an efficient automatic segmentation of the hippocampus and thus has practical relevance for the correct diagnosis of diseases,such as Alzheimer’s disease. | Hongxia Deng Yuefang Zhang Ran Li Chunxiang Hu Zijian Feng Haifang Li | 2022 | Tsinghua Science and Technology2022,27,1: | 1 |
