1Chemical comparison of dried rehmannia root and prepared rehmannia root by UPLC-TOF MS and HPLC-ELSD with multivariate statistical analysis显示文摘To identify the chemical differences which lead to the different therapeutic effects of dried rehmannia root(DRR)and prepared rehmannia root(PRR),we compared the chemical composition of decoctions of randomly purchased DRR and PRR using ultra performance liquid chromatography(UPLC)coupled with time-of-fight mass spectrometry and high performance liquid chromatography(HPLC)coupled with evaporative light scattering detection(ELSD)with the aid of multivariate statistical analysis.Both approaches clearly revealed compositional and quantitative differences between DRR and PRR.UPLC-MS data indicated stachyose,rehmaiono-side A(or rehmaionoside B),acteoside(or forsythiaside,or isoacteoside),6-O-coumaroylajugol(or 6-O-E-feruloylajugol,or 6-O-Z-feruloylajugol)as important discriminators between DRR and PRR decoctions.HPLC-ELSD analysis showed that the content of fructose in the decoctions of PRR was about four times greater than that of DRR(P<10^(-5)),while sucrose content in the decoctions of PRR was only about one seventh of that in DRR(P<0.01).Our results suggest that some compounds,such as fructose,stachyose and rehmaionoside,may be responsible for the differing therapeutic effects of DRR and PRR.Furthermore,improvements in quality control for PRR,which is currently lacking in the Chinese Pharmacopoeia,are recommended.Qiande Liang Jing Ma Zengchun Ma Yuguang Wang Hongling Tan Chengrong Xiao Ming Liu Beibei Lu Boli Zhang Yue Gao 2013Acta Pharmaceutica Sinica B2013,3,1:8
2Spaceflight and simulated microgravity suppresses macrophage development via altered RAS/ERK/NFκB and metabolic pathways显示文摘Spaceflight-associated immune system weakening ultimately limits the ability of humans to expand their presence beyond the earth's orbit. A mechanistic study of microgravity-regulated immune cell function is necessary to overcome this challenge. Here, we demonstrate that both spaceflight (real) and simulated microgravity significantly reduce macrophage differentiation, decrease macrophage quantity and functional polarization, and lead to metabolic reprogramming, as demonstrated by changes in gene expression profiles. Moreover, we identified RAS/ERK/NFκB as a major microgravity-regulated pathway. Exogenous ERK and NFκB activators significantly counteracted the effect of microgravity on macrophage differentiation. In addition, microgravity also affects the p53 pathway, which we verified by RT-qPCR and Western blot. 3Recent Advancements on Photothermal Conversion and Antibacterial Applications over MXenes-Based Materials显示文摘The pernicious bacterial proliferation and emergence of super-resistant bacteria have already posed a great threat to public health,which drives researchers to develop antibiotic-free strategies to eradicate these fierce microbes.Although enormous achievements have already been achieved,it remains an arduous challenge to realize efficient sterilization to cut off the drug resistance generation.Recently,photothermal therapy(PTT)has emerged as a promising solution to efficiently damage the integrity of pathogenic bacteria based on hyperthermia beyond their tolerance.Until now,numerous photothermal agents have been studied for antimicrobial PTT.Among them,MXenes(a type of two-dimensional transition metal carbides or nitrides)are extensively investigated as one of the most promising candidates due to their high aspect ratio,atomic-thin thickness,excellent photothermal performance,low cytotoxicity,and ultrahigh dispersibility in aqueous systems.Besides,the enormous application scenarios using their antibacterial properties can be tailored via elaborated designs of MXenes-based materials.In this review,the synthetic approaches and textural properties of MXenes have been systematically presented first,and then the photothermal properties and sterilization mechanisms using MXenes-based materials are documented.Subsequently,recent progress in diverse fields making use of the photothermal and antibacterial performances of MXenes-based materials are well summarized to reveal the potential applications of these materials for various purposes,including in vitro and in vivo sterilization,solar water evaporation and purification,and flexible antibacterial fabrics.Last but not least,the current challenges and future perspectives are discussed to provide theoretical guidance for the fabrication of efficient antimicrobial systems using MXenes.Shuyan Hao Hecheng Han Zhengyi Yang Mengting Chen Yanyan Jiang Guixia Lu Lun Dong Hongling Wen Hui Li Jiurong Liu Lili Wu Zhou Wang Fenglong Wang 2022Nano-Micro Letters2022,14,11:4
4Spatial and temporal patterns of the inter-annual oscillations of glacier mass over Central Asia inferred from Gravity Recovery and Climate Experiment(GRACE) data显示文摘Monitoring glacier mass balance is crucial to managing water resources and also to understanding climate change for the arid and semi-arid regions of Central Asia. This study extracted the inter-annual oscillations of glacier mass over Central Asia from the first ten principal components(S-PCs) of filtered variability via multichannel singular spectral analysis(MSSA), based on gridded data of glacier mass inferred from Gravity Recovery and Climate Experiment(GRACE) data obtained from July 2002 to March 2015. Two significant cycles of glacier mass balance oscillations were identified. The first cycle with a period of 6.1-year accounted for 54.5% of the total variance and the second with a period of 2.3-year accounted for 4.3%. The 6.1-year oscillation exhibited a stronger variability compared with the 2.3-year oscillation. For the 6.1-year oscillation, the results from lagged cross-correlation function suggested that there were significant correlations between glacier mass balances and precipitation variations with the precipitation variations leading the response of glacier mass balances by 9–16 months.ZHU Chuandong LU Yang SHI Hongling ZHANG Zizhan 2017Journal of Arid Land2017,9,1:2
5Identification of Ubiquitinated Proteins from Human Multiple Myeloma U266 Cells by Proteomics显示文摘Objective To identify ubiquitinated proteins from complex human multiple myeloma (MM) U266 cells,a malignant disorder of differentiated human B cells.Methods Employing a globally proteomic strategy combining of immunoprecipitation,LC-MS/MS and SCX-LC-MS analysis to identified ubiquitination sites,which were identified by detecting signature peptides containing a GG-tag (114.1 Da) and an LRGG-tag (383.2 Da).Results In total,52 ubiquitinated proteins containing 73 ubiquitination sites of which 14 and 59 sites contained LRGG-tag and GG-tag were identified,respectively.Conclusion Classification analysis by of the proteins identified in the study based on the PANTHER showed that they were associated with multiple functional groups.This suggested the involvement of many endogenous proteins in the ubiquitination in MM.JIA HongLing LIU ChaoWu GE Feng XIAO ChuanLe LU ChunHua WANG Tong HE QingYu 2011Biomedical and Environmental Sciences2011,24,4:2

10Aldosterone may induce visfatin expression via the GR-ERK1/2 signal pathway in 3T3-L1 adipocytes显示文摘The main function of aldosterone is to regulate blood pressure and electrolytic balance;however,recent studies have also focused on the effect of aldosterone on glucose metabolism especially in adipose tissue.Aldosterone was found to inhibit insulin-induced glucose uptake in 3T3-L1 adipocytes [1] and aldosterone deficiency was found to prevent high-fat-feeding-induced hyperglycemia and adipocyte dysfunction in mice [2].Visfatin,an adipokine discovered by the team of Fukuhara,is mainly released by adipose tissue [3].Treatment of 3T3-L1 adipocytes and L6 myocytes with visfatin increased basal glucose uptake,and glucose release was suppressed in H4IIEC3 hepatocytes after visfatin treatment [3].It has also been suggested that visfatin may act as a proinflammatory cytokine [4] and produce chronic inflammation in adipocytes [5].Eyileten et al.[6] reported that treatment with angiotensin-converting enzyme inhibitor,which can reduce plasma aldosterone concentration,decreased circulating visfatin level in diabetic nephropathy patients.These findings suggest a possible interaction between the renin–angiotensin–aldosterone system(RAAS)and visfatin.Considering the important roles of aldosterone and visfatin and the association between RAAS and visfatin,we hypothesize that there may be a direct regulatory interaction between aldosterone and visfatin.To test this hypothesis,3T3-L1 adipocytes were used as the cell model and the expression of visfatin was measured by qRT-PCR and western blot analysis.Junhua Ma Xuemei Zhao Jichen Zhang Fei Sun Jiancan Lu Hongling Zhu Mingjun Gu Jingnan Wang Xiangqi Li 2019Acta Biochimica et Biophysica Sinica2019,51,3:1

13Differences in Phosphorus Absorption and Utilization Efficiency of Soybean in Mature Period under Phosphorus Stress显示文摘In this study,four phosphorus-inefficient soybean genotypes( D03,D05,D17 and D18) and four phosphorus-efficient soybean genotypes( D31,D34,D37 and D38) were selected as experimental materials for soil culture experiment under high and low phosphorus treatments,to investigate the grain yield,phosphorus content,phosphorus uptake and the relationship between phosphorus utilization efficiency and phosphorus efficiency of soybean genotypes with different phosphorus efficiency in mature period. According to the experimental results,under low phosphorus treatment,four phosphorus-efficient soybean genotypes exhibited significantly high phosphorus uptake in mature period,especially for D31 and D37; however,three phosphorus-efficient genotypes showed no advantages in adaptability of phosphorus utilization efficiency,while only D31 exhibited high phosphorus utilization and absorption capacity. Correlation analysis and path analysis showed that phosphorus deficiency of soybean was mainly determined by phosphorus absorption capacity,and phosphorus deficiency under(- P) treatment was significantly higher than( + P) treatment. Phosphorus uptake and phosphorus utilization efficiency posed great direct effects on phosphorus efficiency,and phosphorus uptake exhibited a greater contribution; in addition,these two factors both posed small indirect effects. In mature period at reproductive growth stage,phosphorus absorption efficiency( phosphorus uptake) was the main variation source of phosphorus efficiency of various soybean genotypes in mature period. 14Characterizing three-dimensional features of Antarctic subglacial lakes from the inversion of hydraulic potential——Lake Vostok as a case study显示文摘To estimate basal water storage beneath the Antarctic ice sheet, it is essential to have data on the three-dimensional characteristics of subglacial lakes. We present a method to estimate the water depth and surface area of Antarctic subglacial lakes from the inversion of hydraulic potential method. Lake Vostok is chosen as a case study because of the diverse and comprehensive measurements that have been obtained over and around the lake. The average depth of Lake Vostok is around 345±4 m. We estimated the surface area of Lake Vostok beneath the ice sheet to be about 13300±594 km^2. The lake consists of two sub-basins separated by a ridge at water depths of about 200–300 m. The surface area of the northern sub-basin is estimated to be about half of that of the southern basin. The maximum depths of the northern and southern sub-basins are estimated to be about 450 and 850 m, respectively. Total water volume is estimated to be about 4658±204 km^3. These estimates are compared with previous estimates obtained from seismic data and inversion of aerogravity data. 15Influences of the tank liquid lateral sloshing and mass time-varying on high clearance self-propelled sprayer ride comfort显示文摘To explore the influence of the lateral sloshing and the time-varying mass of the liquid in the tank on the ride comfort of the high-clearance sprayer,a spring-mass-damping equivalent mechanics that can describe the lateral sloshing of the liquid under different filling ratios was constructed based on the equivalent criterion.The Fluent was used to simulate the moment acting on the wall of the tank by the lateral sloshing of the liquid,and then the parameters of the equivalent mechanical model are obtained by fitting and solving.Comparative analysis of Fluent simulation and bench test on lateral sloshing of tank liquid under different filling ratios.The results show that the lateral sloshing trend of the tank liquid level obtained from the Fluent simulation and the bench test was consistent,which proved the accuracy of the Fluent fluid simulation process and the correctness of the required equivalent mechanical model parameters.Incorporating a liquid sloshing equivalent model,a four-degree-of-freedom vertical dynamic model of the sprayer half-car was established.Subsequently,the performance of the sprayer was systematically analyzed and compared under the excitation of a bump road and a random E-level road.This investigation took into account varying liquid filling ratios of 10%,50%,and 90%.The focus lay on evaluating the vertical acceleration of the sprayer body,dynamic deflection of the suspension,and dynamic load on the tires in response to these road conditions.This analysis is conducted independently of the liquid sloshing factor.The results show that the lateral sloshing of the liquid medicine significantly reduces the ride smoothness of the machine,and makes the vibration response of the machine produce a certain hysteresis effect.With the reduction of the quality of the liquid medicine in the spray tank,the vibration amplitude of the sprayer body gradually decreases,the hysteresis effect is also gradually weakened.The results presented in this study offer a theoretical foundation for the analysis of ride comfort and the optimization of chassis structure in high-clearance sprayers.Liquan Lu Junchang Zhang Cui Cui Jun Chen Yu Chen Hongling Jing Shuo Zhang Sugirbay Adilet Chenwei Hu Jiayu Cao 2024International Journal of Agricultural and Biological Engineering2024,17,1:0

17Growth suppressor lingerer regulates bantam microRNA to restrict organ size显示文摘The evolutionarily conserved Hippo signaling pathway plays an important role in organ size control by regulating cell proliferation and apoptosis.Here,we identify Lingerer(Lig)as a growth suppressor using RNAi modifying screen in Drosophila melanogaster.Loss of lig increases organ size and upregulates bantam(ban)and the expression of the Hippo pathway target genes,while overexpression of lig results in diminished ban expression and organ size reduction.We demonstrate that Lig C-terminal exhibits dominant-negative function on growth and ban expression,and thus plays an important role in organ size control and ban regulation.In addition,we provide evidence that both Yki and Mad are essential for Lig-induced ban expression.We also show that Lig regulates the expression of the Hippo pathway target genes partially via Yorkie.Moreover,we find that Lig physically interacts with and requires Salvador to restrict cell growth.Taken together,we demonstrate that Lig functions as a critical growth suppressor to control organ size via ban and Hippo signaling.Liang Dong Jinhui Li Hongling Huang Meng-Xin Yin Jinjin Xu Peixue Li Yi Lu Wenqing Wu Hang Yang Yun Zhao Lei Zhang 2015Journal of Molecular Cell Biology2015,7,5:0
