| 1 | Hedgehog signaling in gastrointestinal carcinogenesis and the gastrointestinal tumor microenvironment显示文摘The Hedgehog(HH) signaling pathway plays important roles in gastrointestinal carcinogenesis and the gastrointestinal tumor microenvironment(TME). Aberrant HH signaling activation may accelerate the growth of gastrointestinal tumors and lead to tumor immune tolerance and drug resistance.The interaction between HH signaling and the TME is intimately involved in these processes, for example, tumor growth, tumor immune tolerance, inflammation, and drug resistance. Evidence indicates that inflammatory factors in the TME, such as interleukin 6(IL-6) and interferon-g(IFN-g), macrophages, and T cell-dependent immune responses, play a vital role in tumor growth by affecting the HH signaling pathway. Moreover, inhibition of proliferating cancer-associated fibroblasts(CAFs) and inflammatory factors can normalize the TME by suppressing HH signaling. Furthermore, aberrant HH signaling activation is favorable to both the proliferation of cancer stem cells(CSCs) and the drug resistance of gastrointestinal tumors. | 4 | Predictably synthesizing a library of white-light-emitting perovskites显示文摘The low-dimensional organic-inorganic halide perovskites with self-trapped exciton emission have promising prospects for single-phase white-light emitters. However, so far, these broadband white-light-emitting(BWLE) perovskites were synthesized by trial-and-error testing spacing molecules. Here, we developed a steric hindrance regulation strategy to predictably synthesize BWLE perovskites. The molecules containing C–C(–NH_(2))–C groups were introduced into low-dimensional perovskites, which brings a large steric hindrance in-plane orientation. The bigger C–C(–NH_(2))–C bond angle would induce larger structural distortion in perovskites, which leads to the higher rate of self-trapping of excitons and the deeper self-trapping depth. The photoluminescence spectra of the synthesized perovskites can cover the cool-to-warm white light region. Overall, we fabricated a material library consisting of 40 kinds of BWLE compounds according to this strategy. | 12 | Nitrate transporter NRT1.1 and anion channel SLAH3 form a functional unit to regulate nitrate-dependent alleviation of ammonium toxicity显示文摘Ammonium(NH_(4)^(+))and nitrate(NO_(3)^(-))are major inorganic nitrogen(N)sources for plants.When serving as the sole or dominant N supply,NH_(4)^(+)often causes root inhibition and shoot chlorosis in plants,known as ammonium toxicity.NO_(3)^(-) usually causes no toxicity and can mitigate ammonium toxicity even at low concentrations,referred to as nitrate-dependent alleviation of ammonium toxicity.Our previous studies indicated a NO_(3)^(-) efflux channel SLAH3 is involved in this process.However,whether additional components contribute to NO_(3)^(-)-mediated NH_(4)^(+)detoxification is unknown.Previously,mutations in NO_(3)^(-) transporter NRT1.1 were shown to cause enhanced resistance to high concentrations of NH_(4)^(+).Whereas,in this study,we found when the high-NH_(4)^(+) medium was supplemented with low concentrations of NO_(3)^(-),nrt1.1 mutant plants showed hyper-sensitive phenotype instead.Furthermore,mutation in NRT1.1 caused enhanced medium acidification under high-NH_(4)^(+)/Iow-NO_(3)^(-) condition,suggesting NRT1.1 regulates ammonium toxicity by facilitating H+uptake.Moreover,NRT1.1 was shown to interact with SLAH3 to form a transporter-channel complex.Interestingly,SLAH3 appeared to affect NO_(3)^(-) influx while NRT1.1 influenced NO_(3)^(-) efflux,suggesting NRT1.1 and SLAH3 regulate each other at protein and/or gene expression levels.Our study thus revealed NRT1.1 and SLAH3 form a functional unit to regulate nitrate-dependent alleviation of ammonium toxicity through regulating NO_(3)^(-) transport and balancing rhizosphere acidification. | Chengbin Xiao Doudou Sun Beibei Liu Xianming Fang Pengcheng Li Yao Jiang Mingming He Jia Li Sheng Luan Kai He | 2022 | Journal of Integrative Plant Biology2022,64,4: | 0 |

| 14 | Mevalonate improves anti-PD-1/PD-L1 efficacy by stabilizing CD274 mRNA显示文摘Mevalonate metabolism plays an important role in regulating tumor growth and progression;however,its role in immune evasion and immune checkpoint modulation remains unclear.Here,we found that non-small cell lung cancer(NSCLC)patients with higher plasma mevalonate response better to antiPD-(L)1 therapy,as indicated by prolonged progression-free survival and overall survival.Plasma mevalonate levels were positively correlated with programmed death ligand-1(PD-L1)expression in tumor tissues.In NSCLC cell lines and patient-derived cells,supplementation of mevalonate significantly upregulated the expression of PD-L1,whereas deprivation of mevalonate reduced PD-L1 expression.Mevalonate increased CD274 mRNA level but did not affect CD274 transcription.Further,we confirmed that mevalonate improved CD274 mRNA stability.Mevalonate promoted the affinity of the AU-rich elementbinding protein HuR to the 3'-UTR regions of CD274 mRNA and thereby stabilized CD274 mRNA.By in vivo study,we further confirmed that mevalonate addition enhanced the anti-tumor effect of anti-PD-L1,increased the infiltration of CD8^(+)T cells,and improved cytotoxic function of T cells.Collectively,our findings discovered plasma mevalonate levels positively correlated with the therapeutic efficacy of anti-PD-(L)1 antibody,and provided the evidence that mevalonate supplementation could be an immunosensitizer in NSCLC. | Wenxin Zhang Xiaohui Pan Yanjun Xu Hongjie Guo Mingming Zheng Xi Chen Honghai Wu Fengming Luan Qiaojun He Ling Ding Bo Yang | 2023 | Acta Pharmaceutica Sinica B2023,13,6: | 0 |
