1ShRNA-mediated gene silencing of β-catenin inhibits growth of human colon cancer cells显示文摘AIM: To observe the gene silencing mediated by the specific shRNA targeted against β-catenin and its effect on cell proliferation and cycle distribution in the human colon cancer cell line Colo205. METHODS: Two shRNA plasmid vectors against β-catenin were constructed and transfected into Colo205 cells with LipofectamineTM2000. The down-regulations of β-catenin, c-myc and cyclinD1 expressions were detected by RT-PCR and western blot analysis. The cell proliferation inhibitions were determined by MTT assay and soft agar colony formation assay. The effect of these two β-catenin shRNAs on cell cycle distribution and apoptosis was examined by flow cytometry. RESULTS: These two shRNA vectors targeted against β-catenin efficiently suppressed the expression of β-catenin and its down stream genes, c-myc and cyclinD1. The expression inhibition rates were around 40%-50% either at the mRNA or at the protein level. The shRNA-mediated gene silencing of β-catenin resulted in significant inhibition of cell growth both on the culture plates and in the soft agar. Moreover, the cancer cells showed significant G0/G1 arrest and increased apoptosis at 72 h post transfection due to gene silencing. CONCLUSION: These specific shRNAs targeted against β-catenin could have a gene silencing effect and block the WNT signaling pathway. They could inhibit cell growth, increase apoptosis, and induce cell cycle arrest in Colo205 cells. 2A study of the efficacy of bacterial biofilm cleanout for gastrointestinal endoscopes显示文摘AIM:To compare the influence and clearance effect of enzymatic and non-enzymatic detergents against Escherichia coli (E. coli) biofilm on the inner surface of gastroscopes.METHODS:Teflon tubes were incubated in a mixture of different detergents and E. coli culture (106 CFU/mL) for 72 h at 15℃,and biofilms on the inner surface of the teflon tubes were analyzed by bacterial count and scanning electron microscopy. To evaluate the clear-ance effect of detergents,after biofilms were formed on the inner surface of Teflon tubes by 72 h lavage with E. coli culture,tubes were lavaged by enzymatic and non-enzymatic detergents at a speed of 250 mL/min,then biofilms on the inner surface were analyzed by bacterial count and scanning electron microscopy.RESULTS:Non-enzymatic detergent had a better inhi-bition function on biofilm formation than enzymatic de-tergent as it reduced bacterial burden by 2.4 log compared with the control samples (P = 0.00). Inhibition function of enzymatic detergent was not significantly different to that of control samples and reduced bac-terial burden by 0.2 log on average (P > 0.05). After lavaging at 250 mL/min for 3 min,no living bacteria were left in the tubes. Scanning electron microscopy observation showed biofi lms became very loose by the high shear force effect. CONCLUSION:Non-enzymatic detergent has a better inhibition effect on biofilm formation at room temperature. 4Medusoid Fossils from the Sinian Xingmincun Formation of Southern Liaoning Province显示文摘Abundant medusoid soft-bodied metazoan fossils have been found in the Sinian Xingmincun Formation(with an isotopic age of 650 Ma) at Qipanmo, Qidingshan and Yangtun of the Dalian-Jinzhou area, southern Liaoning Province; in addition, a number of macroscopic algal fossils have been discovered for the first time in the Getun Formation overlying the Xingmincun Formation. Medusoid fossils include 3 genera and 6 species(all of which are new). Judging from their biological characters, these medusoid fossils are considered to have an intimate affinity with the Ediacara fauna in the terminal Precambrian of Australia, as their characters are close to each other. The authors suggest that these medusoids and the Edicara fauna might be the products of the same period. To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://gffzzdbc7b6aaae734bddhqpv5oxqpkvkc6u0u.ffgz.tsg.suse.edu.cn/gsa or http://gffzz4eaf941a184140e3hqpv5oxqpkvkc6u0u.ffgz.tsg.suse.edu.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. 7Apatinib, a selective VEGFR2 inhibitor, improves the delivery of chemotherapeutic agents to tumors by normalizing tumor vessels in LoVo colon cancer xenograft mice显示文摘Tumor vascular normalization has been proposed as a therapeutic strategy for malignant neoplasms, which can also interpret the synergistic effect of anti-angiogenesis agents combined with chemotherapy. Apatinib (Apa), a highly selective VEGFR2 inhibitor, attracts much attentions due to its encouraging anticancer activity, especially in the clinical trials of combined treatment. In this study, we investigated whether Apa could promote vascular normalization in tumor in a certain time window. Mice bearing LoVo colon cancer xenograft were orally administrated Apa (150 mg kg^-1 per day) for 5, 7, 10, or 12 days. Apa significantly inhibited tumor growth and decreased the microvessel density. Using multi-photon microscopy and electron microscopy, we found that Apa improved tumor vessel morphology by pruning distorted vessel branches and decreased the gap between endothelial cells after a 7-day treatment. Furthermore, Apa decreased vessel leakage and in creased pericyte coverage on vascular en dothelial cells, suggesting that tumor vessels were more mature and integrated. The intratumoral distribution of adriamycin (ADR) in Apa group was improved from day 7 to 10 without change in plasma drug concen tration. Tumor blood perfusion was also in creased in this window, and the expression of hypoxia induced factor 1a was downregulated, suggesting the effect of Apa on alleviating tumor hypoxic micro-e nvironme nt. In conclusi on, Apa may improve the effective perfusi on of tumor vessels and in crease the intratumoral distribution of ADR in a certain time window via normalizing tumor vessels. 8Mechanisms and pharmacokinetic/pharmacodynamic profiles underlying the low nephrotoxicity and ototoxicity of etimicin显示文摘Etimicin(ETM),a fourth-generation aminoglycosides(AGs),is now widely clinically used in China due to its high efficacy and low toxicity.However,the mechanisms underlying its low nephrotoxicity and ototoxicity remain unclear.In the present study we compared the antibacterial and toxicity profiles of etimicin,gentamicin(GM,a second-generation AG),and amikacin(AMK,a third-generation AG),and investigated their pharmacokinetic properties in the toxicity target organs(kidney and inner ear)and subcellular compartments.We first demonstrated that ETM exhibited superior antibacterial activities against clinical isolates to GM and AMK,and it exerted minimal nephrotoxicity and ototoxicity in rats following multi-dose administration.Then,we conducted pharmacokinetic studies in rats,showed that the three AGs accumulated in the kidney and inner ear with ETM being distributed to a lesser degree in the two toxicity target organs as compared with GM and AMK high-dose groups.Furthermore,we conducted in vitro experiments in NRK-52E rat renal tubular epithelial cells and HEI-OC1 cochlear hair cells,and revealed that all the three AGs were distributed predominantly in the mitochondria with ETM showing minimal accumulation;they not only directly inhibited the activity of mitochondrial complexesⅣandⅤbut also inhibited mitochondrial function and its related PGC-1α-NRF1-TFAM pathway;ETM caused minimal damage to the mitochondrial complex and mitochondrial biogenesis.Our results demonstrate that the minimal otonephrotoxicity of ETM results from its lesser accumulation in mitochondria of target cells and subsequently lesser inhibition of mitochondrial function.These results provide a new strategy for discovering novel AGs with high efficacy and low toxicity.Lan Yao Jing-wei Zhang Bin Chen Ming-min Cai Dong Feng Qi-zhi Wang Xin-yu Wang Jian-guo Sun Yi-wen Zheng Guang-ji Wang Fang Zhou 2020Acta Pharmacologica Sinica2020,41,6:10

12Development of high-temperature superconducting filters operating at temperatures above 90 K显示文摘This paper represents the development of a high temperature superconducting (HTS) filter, the highest operating temperature of which is up to 93 K. The filter is designed for S band with 4% fractional bandwidth and fabricated using thallium-barium-calcium-copper oxide (Tl2Ba2CaCu2O8) thin films. At 93 K, the measurements of the filter show that the insertion loss in the passband is less than 0.22 dB, the return loss is better than 20 dB, and the out-of-band rejection is more than 80 dB. The analysis on the characteristics of the filter operating at different temperatures shows that the filter can work well at temperatures around 90 K. The temperature of 93 K is the highest among the previous reports for HTS filters. 13Targeting redox-altered plasticity to reactivate synaptic function: A novel therapeutic strategy for cognitive disorder显示文摘Redox-altered plasticity refers to redox-dependent reversible changes in synaptic plasticity via altering functions of key proteins, such as N-methyl-D-aspartate receptor(NMDAR). Age-related cognitive disorders includes Alzheimer’s disease(AD), vascular dementia(VD), and age-associated memory impairment(AAMI). Based on the critical role of NMDAR-dependent long-term potentiation(LTP) in memory, the increase of reactive oxygen species in cognitive disorders, and the sensitivity of NMDAR to the redox status, converging lines have suggested the redox-altered NMDAR-dependent plasticity might underlie the synaptic dysfunctions associated with cognitive disorders. In this review, we summarize the involvement of redox-altered plasticity in cognitive disorders by presenting the available evidence. According to reports from our laboratory and other groups, this 'redox-altered plasticity' is Hydrogen sulfidemore similar to functional changes rather than organic injuries, and strategies targeting redox-altered plasticity using pharmacological agents might reverse synaptic dysfunctions and memory abnormalities in the early stage of cognitive disorders. 14Pulsed radiofrequency inhibits expression of P2X3 receptors and alleviates neuropathic pain induced by chronic constriction injury in rats显示文摘Background:Pulsed radiofrequency (PRF) is a minimally invasive interventional technique that provides a novel and effective treatment strategy for neuropathic pain (NP).PRF is advantageous because it does not damage nerves and avoids sensory loss after treatment.At present,animal studies have demonstrated that PRF is safe and effective for relieving the NP associated with sciatic nerve damage in rats with chronic constriction injury (CCI).However,the mechanism through which this effect occurs is unknown.An increasing body of evidence shows that the expression of the P2X ligand-gated ion channel 3 (P2X3) receptor is closely related to NP;this study was to investigate whether the expression of this receptor is involved in NP relief due to PRF.Methods:A total of 36 healthy adult male Sprague-Dawley (SD) rats were randomly divided into three groups:Sham group,CCI group,and PRF group.The right sciatic nerve was ligated in CCI group and PRF group to establish a CCI model;the right sciatic nerve was separated but not ligated in Sham group.On day 14 after the operation,PRF was administered to the ligated sciatic nerve in PRF group (42℃,45 V,2 min).A non-live electrode was placed at the exposed sciatic nerve for the rats in Sham and CCI groups.The hindpaw withdrawal threshold (HWT) and thermal withdrawal latency (TWL) were measured at the right hindpaw at different time points before and after PRF or sham therapy.On day 28 after treatment,the dorsal root ganglion (DRG) and spinal dorsal horn of the right L4-6 were harvested from each group to determine the mRNA and protein levels of the P2X3 receptor.Results:On day 28 after PRF treatment,the HWT (8.33 ± 0.67 g vs.3.62 ± 0.48 g) and TWL (25.42 ± 1.90 s vs.15.10 ± 1.71 s) were significantly higher in PRF group as compared to CCI group (P < 0.05).The mRNA expression of the P2X3 receptor in the DRG in PRF group was 23.7% lower than that in CCI group (P < 0.05),in the spinal dorsal horns in PRF group was 22.7% lower than that in CCI group (P < 0.05).The protein expression of the P2X3 receptor in the DRG in PRF group was 27.8 % lower than that in CCI group (P < 0.05),in the spinal dorsal horns in PRF group was 35.6% lower than that in CCI group (P < 0.05).Conclusion:PRF possibly reduces NP in CCI rats by inhibiting the expression of the P2X3 receptor in the L4-6 DRG and spinal dorsal horns.Miao Fu Lan Meng Hao Ren Fang Luo 2019Chinese Medical Journal2019,,14:8

16Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 18Establishment of in Vitro Regeneration System of Bitter Melon (Momordica Charantia L. )显示文摘Explants of bitter melon can produce three types of callus: green, yellow green and fragile yellow callus. During the period of inducing callus, the explants are not sensitive to the kinds and proportions of phytohormone. The rate of callus inductivity is about 90.0%. However, in the process of callus differentiating adventitious bud, the kind, proportion and quantity of phytohormone and the type of callus made different result. The adventitious buds were induced successfully on MS medium+6-BA (N6-benzyladenine) 4.0 mg l-1+KT(kinetin) 2.0 mg l-1) by yellow green callus. The frequency was about 66.7 %. Yellow callus did not differentiated adventitious bud. The frequency of green callus differentiation was very low (<15.0 %) even under the most suitable conditions. The MS + ZT (zeatin) 5.0 mgl-1 +KT 0.5 mgl-1 medium was suitable for the proliferation of bud and three weeks later, the coefficient of proliferation was about 5-6. The 1/2 MS + ZT 0.02 mgl-1 or 1/2 MS media were suitable for in vitro rooting of shoot, the shoot on them could produce 6-7 new roots in three weeks. 19A novel thioredoxin reductase inhibitor inhibits cell growth and induces apoptosis in HL-60 and K562 cells显示文摘Human thioredoxin reductase (TrxR) system is associated with cancer cell growth and anti-apoptosis process. Effects of 1,2-bis(1,2-benzisoselenazolone-3(2H)-ketone)ethane (BBSKE),a novel TrxR inhibitor,were investigated on human leu-kemia cell lines HL-60 and K562. BBSKE treatment induced cell growth inhibition and apoptosis in both cell lines. Apoptosis induced by BBSKE is through Bcl-2/Bax and caspase-3 pathways. Ehrlich's ascites carcinoma-bearing mice were used to inves-tigate the anti-tumor effect of BBSKE in vivo. Tumor-bearing mice treated with BBSKE showed an increase of life span with a comparable effect to cyclophosphamide (CTX). 20Determination of Indoxacarb Residue in Foodstuffs of Plant and Animal Origin by GC-ECD and LC-MS/MS显示文摘An effective method for the trace analysis of indoxacarb residue in foodstuffs of plant and animal origin [grapefruit, ginger, fresh soybean, bamboo shoot, qing-gen-cai (cruciferous vegetable), chicken, fish, and pork] was developed using gas chromatography (GC-ECD) and liquid chromatography tandem mass spectrometry (LC-MS/MS). Samples were extracted using acetone and n-hexane mixed solvent (1:2, v/v) and then purified using solid-phase extraction (SPE) columns. The extracts were analyzed using GC-ECD and LC-MS/MS. The multiple reaction monitoring (MRM) scheme used involved transitions of the precursor ions to selected two product ions in which one pair for identification was m/z 529 → 293 and another pair for quantification was m/z 529→249. The detection limits (LODs) of the method were 0.0015 and 0.0006 mg kg-1, and the quantification limits (LOQs) were 0.005 and 0.002 mg kg-1 for GC-ECD and LC-MS/MS, respectively. The relative standard deviations (RSDs) of recovery for indoxacarb were lower than 15% in 10 types of agro-products. Ten repetitive determinations of recovery achieved good reproducibility for indoxacarb and the recovery ranged from 72.08 to 113.74%.
