1The plant cell wall: Biosynthesis, construction, and functions显示文摘The plant cell wall is composed of multiple biopolymers, representing one of the most complex structural networks in nature. Hundreds of genes are involved in building such a natural masterpiece. However, the plant cell wall is the least understood cellular structure in plants. Due to great progress in plant functional genomics,manyachievementshavebeenmadein uncovering cell wall biosynthesis, assembly, and architecture, as well as cell wall regulation and signaling. Such information has significantly advanced our understanding of the roles of the cell wall in many biological and physiological processes and has enhanced our utilization of cell wall materials. The use of cutting-edge technologies such as single-molecule imaging,nuclear magnetic resonance spectroscopy, and atomic force microscopy has provided much insight into the plant cell wall as an intricate nanoscale network, opening up unprecedented possibilities for cell wall research. 2First-generation EGFR tyrosine kinase inhibitor therapy in 106 patients with compound EGFR-mutated lung cancer: a single institution’s clinical practice experience显示文摘Background:The antitumour efficacy of tyrosine kinase inhibitors(TKIs)in lung cancer patients with compound epidermal growth factor receptor(EGFR)mutations has not been resolved.Our study summarizes a single institutional experience of first-generation TKI therapy for lung cancers with compound EGFR mutations.Methods:A total of 106 consecutive patients with tumours bearing compound EGFR mutations were identified between January 2012 and May 2016;all patients received first-generation TKI therapy.Deletions in exon 19 and the L858R point mutation in exon 21 were considered common mutations;T790M was considered separately because of its association with TKIs resistances.Any other mutation was defined as a rare mutation.Patients were divided as follows:double common mutations(group A);common plus T790M mutations(group B);common plus rare muta-tions(group C);double rare mutations(group D);and rare plus T790M mutations(group E).A separate group of 115 consecutive patients with a single common mutation was created for comparative analysis(group F).Results:The frequency of patients with compound EGFR was 2.9%(114/3925)and their response rate to first-genera-tion TKIs was 50.9%,which was not significantly different from group F(67.0%,P=0.088).The progression-free survival(PFS)of the 106 patients receiving TKI therapy was worse than that of group F(median,9.1 vs.13.0 months,respec-tively;P<0.001).The PFS of the compound mutation group was shorter than that of the single common mutation group(median,10.1 months in group A,P=0.240;9.1 months in group B,P<0.001;9.6 months in group C,P=0.010;6.5 months in group D,P=0.048;5.4 months in group E,P=0.017).Patients with a co-occurring mutation in exon 20(excluding T790M)exhibited significantly worse PFS than the patients with other compound mutations or with a single common mutation(median,6.5 vs.9.1 vs.13.0 months,respectively,P=0.002).Conclusions:There was significant heterogeneity among the compound EGFR mutations and their response to first-generation TKIs.Individualized treatment in clinical practice should be considered for each case.Xiangyang Yu Xuewen Zhang Zichen Zhang Yongbin Lin Yingsheng Wen Yongqiang Chen Weidong Wang Lanjun Zhang 2018Cancer Communications2018,38,1:5

4Significance of combined detection of LunX mRNA and tumor markers in diagnosis of lung carcinoma显示文摘Objective:To evaluate the significance of combined detection of LunX mRNA,carcinoembryonic antigen(CEA),neuron-specific enolase(NSE),and cytokeratin 21-1 fragment(CYFRA21-1) in clinical diagnosis of lung carcinoma.Methods:Based on the quantitative RT-PCR and chemiluminescence immunoassay,the expression levels of LunX mRNA,CEA,NSE,and CYFRA21-1 in 113 patients with lung carcinoma(case group) and 30 healthy participants(control group) were detected.Meantime,the sensitivity,specificity,and accuracy of the combination detection were also explored.Results:The positive rates of LunX mRNA in peripheral blood and CEA,NSE,and CYFRA21-1 in serum were significantly higher in case group than those in control group(χ2=17.295,16.825,19.148,and 17.450;P<0.05).There was no statistical significance when positive rate of LunX mRNA was evaluated among different pathological types(χ2=0.047,P>0.05).The positive rate of LunX mRNA in stage Ⅰ+Ⅱ,Ⅲ,and Ⅳ had a significantly increasing tendency(χ2=10.565,32.462,P<0.05).The positive rate of CYFRA21-1 was highest in squamous carcinoma(78.5%),the positive rate of NSE was highest in small cell carcinoma(86.7%),and the positive rate of CEA wag highest in lung adenocarcinoma(80.4%).The sensitivity and accuracy of the combination detection were 91.1% and 88.1%,respectively.Conclusions:The combined detection of LunX mRNA and tumor markers(TMs) including CEA,NSE,and CYFRA21-1 in peripheral blood is helpful to increase the diagnostic accuracy of lung cancer.Also,it can inform the pathological typing of lung carcinoma.Hui Yu Xiuying Huang Zhihua Zhu Yi Hu Wei Ou Lanjun Zhang Ningning Zhou 2014Chinese Journal of Cancer Research2014,26,1:3
5Identification of maize seed varieties based on near infrared reflectance spectroscopy and chemometrics显示文摘False seeds can often be seen in the maize seed market,leading to a serious decline in maize yield.Those existing variety identification methods are expensive,time consuming,and destructive to seeds.The aim of this study is to develop a cheap,fast and non-destructive method which can robustly identify large amounts of maize seed varieties based on near-infrared reflectance spectroscopy(NIRS)and chemometrics.Because it is difficult to establish models for every variety in the market,this study mainly investigated the performance of models based on a large number of samples(more than 40 major varieties in the market).The reflectance spectra of maize seeds were collected by two modes(bulk kernels mode and single kernel mode).Both collection modes can be applied to identification,but only the single kernel mode can be applied to purity sorting.The spectra were pretreated with smoothing,the first derivative and vector normalization;and then principal component analysis(PCA),linear discriminant analysis(LDA)and biomimetic pattern recognition(BPR)were applied to establish identification models.The environmental factors such as producing areas and years have a significant influence on the performance of the models.Therefore,the method to improve the robustness of the models was investigated in this study.New indexes(correct acceptance degree(CAD),correct rejection degree(CRD)and correct degree(CD))were defined to analyze the performance of the models more accurately.Finally,the models obtained a mean correct discrimination rate of over 90%,and exhibited robust properties for samples harvested from different areas and years.The results showed that NIR technology combined with chemometrics methods such as PCA,LDA,and BPR could be a suitable and alternative technique to identify the authenticity of maize seed varieties.Yongjin Cui Lanjun Xu Dong An Zhe Liu Jiancheng Gu Shaoming Li Xiaodong Zhang Dehai Zhu 2018International Journal of Agricultural and Biological Engineering2018,11,2:3

10The role of human ADA2a in the regulation of p53 acetylation and stability显示文摘The tumor suppressor protein p53 is a well-known transcription factor that functions as a critical component of the genotoxic stress response via regulating the expression of effector proteins that control cellular fate following DNA damage. The human p300/CBP-associated factor(PCAF)-containing histone acetyltransferase(HAT) complex is important for the stability and activity of p53 via its acetylation. The human homolog of yeast alteration/deficiency in activation 2a(ADA2a) is a stable component of the human PCAF-containing HAT complex. In this study,we demonstrated that p53 and hADA2a physically interact with each other in human HEK 293T cells. Using overexpression and small interfering RNA-mediated knockdown,we demonstrated that hADA2a stabilizes p53 via promoting its acetylation at lysine 320 - a PCAF-dependent acetylation site. Furthermore,hADA2a can potentiate the transcriptional activity of p53 at the BAX and p21 promoters to induce cell apoptosis and cell cycle arrest. 12Experimental replacement of thoracic esophageal segment with a biomaterial artificial esophagus in dogs显示文摘Objective:To implant 80 mm-long artificial esophagi constructed of biomateriai in dogs,observe the perioperative survival rates and the incidence of postoperative complications,and study the mechanisms of postoperative healing.Methods:Specimens of the implanted esophagus,the'neo-esophagi',were taken for histopathologic study 1,3,6,12,24 months after operation.Results:The incidence of anastomotic leakage after the artificial esophagus implantation was 3.33%.The perioperative survival rate was 96.67%.The incidence of postoperative stenosis in the'neo-esophagi'was 81.48%;the stenoses were treated by expanding with esophagoscopy and implanting a stent.Epithelization of the mucosa in the'neo-esophagi'was completed in 3 to 6 months after surgery Structures such as submucosal muscle layers,mucous glands,nerve fibers,capillaries,etc.were regenerated after 12 months,and then reconstruction of the fibrous connective tissue layer was completed.Conclusion:Implanting a biomaterial artificial esophagus accomplishes safe reconstruction of defects in the esophagus.Advanced cellular structure of'neo-esophagus'can be regenerated after 1 year.Postoperative stenosis,which is related to hyperplasia and retraction of scar tissue,is still the most common complications which limiting the clinical application of the artificial esophagus.Zhang Lanjun Rong Tiehua Su Xiaodong Wu Qiuliang Xu Guoliang Lin Peng Long Hao FU Jianhua 2008Journal of Medical Colleges of PLA(China)2008,23,1:1
13中国人非小细胞肺癌表皮生长因子受体EGFR基因突变与血浆纤维蛋白原水平之间的相关性(英文)显示文摘Objective:The plasma fibrinogen levels had not only been used as an independent prognostic parameter for the patients with non-small cell lung cancer (NSCLC), but also as a promising biomarker for evaluating the efficacy of chemotherapy. This study aimed to investigate the correlation between the plasma fibrinogen levels and epidermal growth factor receptor (EGFR) gene mutation and clinical-pathological characteristics of Chinese patients with NSCLC. Methods:In this retrospective study, NSCLC specimens collected from 352 patients between November 2009 and November 2011 were selected to detect EGFR gene mutation with real-time polymerase chain reaction (RT-PCR). In these specimens, 308 ones were also detected EGFR gene copy number with fluorescence in situ hybridization (FISH). Coagulation makers were examined prior to the operations. The association between the plasma fibrinogen levels and EGFR gene mutation and clinical-pathological characteristics were analyzed using SPSS 16.0 software. Results:The median pre-operation plasma fibrinogen level was 3.55 g/L (109/352) patients with higher plasma fibrinogen level (> 4.0 g/L). The lower plasma fibrinogen levels correlated significantly with EGFR gene mutations (P < 0.001), the similar result was seen in platelet counts (P = 0.026). A linear correlation was found between the plasma fibrinogen levels and the platelet counts in NSCLC patients (R 2 = 0.209, P < 0.001). Pre-operation plasma fibrinogen levels correlated with gender (P < 0.001), smoking status (P < 0.001), and histology (P < 0.001). There were significant link between the above clinical-pathological characteristics and EGFR gene mutations. In addition, EGFR gene mutation was correlated with FISH-positive status (P < 0.001). Moreover, both plasma fibrinogen level (P = 0.024) and the EGFR gene copy number (P = 0.040) had significant relationships with the pathological TNM stage. Conclusion:This study showed that a significant relevance between plasma fibrinogen levels and EGFR gene mutations. The plasma fibrinogen level might be as a clinical decision parameter for evaluating the efficacy of anti-EGFR tyrosine kinase inhibitors (TKIs) in NSCLC. The patients of NSCLC had higher indicate have poor benefits from anti-EFGR TKIs. 14SUMOylation-triggered ALIX activation modulates extracellular vesicles circTLCD4-RWDD3 to promote lymphatic metastasis of non-small cell lung cancer显示文摘Lymph node(LN)metastasis is one of the predominant metastatic routes of non-small cell lung cancer(NSCLC)and is considered as a leading cause for the unsatisfactory prognosis of patients.Although lymphangiogenesis is well-recognized as a crucial process in mediating LN metastasis,the regulatory mechanism involving lymphangiogenesis and LN metastasis in NSCLC remains unclear.In this study,we employed high-throughput sequencing to identify a novel circular RNA(circRNA),circTLCD4-RWDD3,which was significantly upregulated in extracellular vesicles(EVs)from LN metastatic NSCLC and was positively associated with deteriorated OS and DFS of patients with NSCLC from multicenter clinical cohort.Downregulating the expression of EV-packaged circTLCD4-RWDD3 inhibited lymphangiogenesis and LN metastasis of NSCLC both in vitro and in vivo.Mechanically,circTLCD4-RWDD3 physically interacted with hnRNPA2B1 and mediated the SUMO2 modification at K108 residue of hnRNPA2B1 by upregulating UBC9.Subsequently,circTLCD4-RWDD3-induced SUMOylated hnRNPA2B1 was recognized by the SUMO interaction motif(SIM)of ALIX and activated ALIX to recruit ESCRT-III,thereby facilitating the sorting of circTLCD4-RWDD3 into NSCLC cell-derived EVs.Moreover,EV-packaged circTLCD4-RWDD3 was internalized by lymphatic endothelial cells to activate the transcription of PROX1,resulting in the lymphangiogenesis and LN metastasis of NSCLC.Importantly,blocking EV-mediated transmission of circTLCD4-RWDD3 via mutating SIM in ALIX or K108 residue of hnRNPA2B1 inhibited the lymphangiogenesis and LN metastasis of NSCLC in vivo.Our findings reveal a precise mechanism underlying SUMOylated hnRNPA2B1-induced EV packaging of circTLCD4-RWDD3 in facilitating LN metastasis of NSCLC,suggesting that EV-packaged circTLCD4-RWDD3 could be a potential therapeutic target against LN metastatic NSCLC.Xiayao Diao Chao Guo Hanhao Zheng Ke Zhao Yuming Luo Mingjie An Yan Lin Jiancheng Chen Yuanlong Li Yuting Li Xuehan Gao Jiaqi Zhang Mengxin Zhou Wenliang Bai Lei Liu Guige Wang Lanjun Zhang Xiaotian He Rusi Zhang Zhihua Li Changhao Chen Shanqing Li 2023Signal Transduction and Targeted Therapy2023,8,12:0
15A solid-state nanopore-based single-molecule approach for label-free characterization of plant polysaccharides显示文摘Polysaccharides are important biomacromolecules existing in all plants,most of which are integrated into a fibrillar structure called the cell wall.In the absence of an effective methodology for polysaccharide analysis that arises from compositional heterogeneity and structural flexibility,our knowledge of cell wall architecture and function is greatly constrained.Here,we develop a single-molecule approach for identifying plant polysaccharides with acetylated modification levels.We designed a solid-state nanopore sensor supported by a free-standing SiNx membrane in fluidic cells.This device was able to detect cell wall polysaccharide xylans at concentrations as low as 5 ng/mL and discriminate xylans with hyperacetylated and unacetylated modifications.We further demonstrated the capability of this method in distinguishing arabinoxylan and glucuronoxylan in monocot and dicot plants.Combining the data for categorizing polysaccharide mixtures,our study establishes a single-molecule platformfor polysaccharide analysis,opening a new avenue for understanding cell wall structures,and expanding polysaccharide applications.Yao Cai Baocai Zhang Liyuan Liang Sen Wang Lanjun Zhang Liang Wang Hong-Liang Cui Yihua Zhou Deqiang Wang 2021Plant Communications2021,2,2:0

18A Study of Molecular Resection Margins for Esophageal Squamous Cell Carcinoma Using Large Pathologic Sections显示文摘OBJECTIVE It has been shown that application of molecular biological techniques to surgical margins of some cancers could predict risk of local recurrence. However, the optimal length of surgical resection with tumor-free surgical margins for esophageal squamous cell carcinoma (ES-CC) is unknown. This study was conducted to evaluate the optimal length of surgical resection for ESCC with molecularly tumor-free surgical margins marked by p53 and Ki67. METHODS Surgical specimens from 70 patients with ESCC were collected for study. The lengths of the upper margin, tumor, and lower margin of every specimen were measured during the operation. Each specimen was divided into three large pathologic sections, stained with H&E and immunohistochemically for p53 and Ki67, and examined microscopically. The lengths of the upper and lower resection ends were measured for p53 and Ki67 positive expression. The actual surgical lengths were calculated by the principle of rational shrinkage. RESULTS All surgical margins were histologically tumor-free, while the positive rates of p53 and Ki67 were 66% and 54%. The positive rates of p53 and Ki67 in the upper resection end were 17% and 20%. The mean lengths of the upper resection end showing p53 and Ki67 positive expression were 1.08±1.12 cm and 1.64±1.01 cm, and the maximum lengths were 3.73 cm and 3.26 cm. The positive rates of p53 and Ki67 in the lower resection end were 20% and 23%. The mean lengths of the lower resection end of p53 and Ki67 with positive expression were 1.11±1.15 cm and 1.34±0.94 cm, and the maximum lengths were 3.73 cm and 3.61 cm. 19Influence of impact energy on work hardening ability of austenitic manganese steel and its mechanism显示文摘To further understand the hardening mechanism of austenitic manganese steel under actual working conditions, the work hardening ability was studied and the microstructures of austenitic manganese steel were observed under different impact energies. The work hardening mechanism was also analyzed. The results show that the best strain hardening effect could be received only when the impact energy reaches or exceeds the critical impact energy. The microstructural observations reveal that dislocations, stacking faults and twins increase with raising impact energy of the tested specimens. The hardening mechanism changes at different hardening degrees.
