2From Golden Rice to aSTARice:Bioengineering Astaxanthin Biosynthesis in Rice Endosperm显示文摘Carotenoids are important phytonutrients with antioxidant properties,and are widely used in foods and feedstuffs as Supplements.Astaxanthin,a red-colored ketocarotenoid,has strong antioxidant activity and thus can benefit human health.However,astaxanthin is not produced in most higher plants.Here we report the bioengineering of astaxanthin biosynthesis in rice endosperm by introducing four synthetic genes,sZmPSY1,sPaCrtl,sCrBKT,and sHpBHY,which encode the enzymes phytoene synthase,phytoene desaturase,β-carotene ketolase,and β-carotene hydroxylase,respectively.Transgneic overexpression of two (sZmPSY1 and sPaCrtl),three (sZmPSY1,sPaCrtl and sCrBKT),and all these four genes driven by rice endosperm-specific promoters established the Carotenoid/ketocarotenoid/astaxanthin biosynthetic pathways in the endosperm and thus resulted in various types of germplasm,from the yellow-grained β-caro- tene-enriched Golden Rice to orange-red-grained Canthaxanthin Rice and Astaxanthin Rice,respectively. 3Weighted Markov chains for forecasting and analysis in Incidence of infectious diseases in jiangsu Province,China显示文摘This paper first applies the sequential cluster method to set up the classification standard of infectious disease incidence state based on the fact that there are many uncertainty characteristics in the incidence course.Then the paper presents a weighted Markov chain,a method which is used to predict the future incidence state.This method assumes the standardized self-coefficients as weights based on the special characteristics of infectious disease incidence being a dependent stochastic variable.It also analyzes the characteristics of infectious diseases incidence via the Markov chain Monte Carlo method to make the long-term benefit of decision optimal.Our method is successfully validated using existing incidents data of infectious diseases in Jiangsu Province.In summation,this paper proposes ways to improve the accuracy of the weighted Markov chain,specifically in the field of infection epidemiology.Zhihang Peng Changjun Bao Yang Zhao Honggang Yi Letian Xia Hao Yu Hongbing Shen Feng Chen 2010The Journal of Biomedical Research2010,24,3:10

6Hd1,Ghd7,and DTH8 synergistically determine the rice heading date and yield-related agronomic traits显示文摘Heading date determines the seasonal and regional adaptation of rice(Oryza sativa L.)varieties and is mainly controlled by photoperiod sensitivity(PS).The core heading date genes Hd1,Ghd7,DTH8,and PRR37 act synergistically in regulating the PS.In this study,we systematically analyze the heading date,PS,and agronomic traits of eight homozygous lines with various combinations of Hd1,Ghd7,and DTH8 alleles in the prr37 background under long-day(LD)and short-day(SD)conditions,respectively.We find that Hd1 alone promotes heading,regardless of the day length.However,under LDs,Hd1 suppresses flowering,in coordination with functional Ghd7 or with Ghd7 and DTH8.These loci cooperate to negatively regulate the Ehd1-Hd3 a/RFT1 pathway and delay heading.Under SDs,Hd1 competes with various heading suppressors to promote heading.Therefore,the dual function of Hd1 is vital for PS.The lines carrying Hd1 alone show reduced plant height with fewer primary and secondary branches in panicles.Lines carrying Ghd7 and DTH8(with hd1)show delayed heading and improve agronomic traits.Overall,our results reveal the regulation of rice PS flowering by the core heading date genes and their effects on agronomic traits,providing valuable information for the selection of rice varieties for adaptation to different light and temperature conditions.Kangli Sun Minghui Huang Wubei Zong Dongdong Xiao Chen Lei Yanqiu Luo Yingang Song Shengting Li Yu Hao Wanni Luo Bingqun Xu Xiaotong Guo Guangliang Wei Letian Chen Yao-Guang Liu Jingxin Guo 2022Journal of Genetics and Genomics2022,49,5:4
7A flexible,multifunctional,optoelectronic anticounterfeiting device from high-performance organic light-emitting paper显示文摘As a primary anticounterfeiting technology,most paper anticounterfeiting devices take advantage of photoresponsive behaviors of certain security materials or structures,thus featuring low-security threshold,which has been a critical global issue.To incorporate optoelectronic devices into existing anticounterfeiting technology suggests a feasible avenue to address this challenge.Here we report a high-performance organic light-emitting paper-based flexible anticounterfeiting(FAC)device with multiple stimuli-responsiveness,including light,electricity,and their combination.Without sacrificing the preexisted security information on the paper,we fabricate FAC device in a facile,low-cost yet high-fidelity fashion by integrating patterned electro-responsive and photo-responsive organic emitters onto paper substrates.By introducing optical microcavities,the FAC device shows considerable color shift upon different viewing angle and applied voltage,which is easily discernible by naked eyes.Notably,the FAC device is bendable,unclonable,and durable(a half-lifetime over 4000 hours at 100 cd m^(−2)).Teng Pan Shihao Liu Letian Zhang Wenfa Xie Cunjiang Yu 2022Light(Science & Applications)2022,11,3:2
8Bexarotene improves motor function after spinal cord injury in mice显示文摘Spinal cord injury is a challenge in orthopedics because it causes irreversible damage to the central nervous system.Therefore,early treatment to prevent lesion expansion is crucial for the management of patients with spinal cord injury.Bexarotene,a type of retinoid,exerts therapeutic effects on patients with cutaneous T-cell lymphoma and Parkinson's disease.Bexarotene has been proven to promote autophagy,but it has not been used in the treatment of spinal cord injury.To investigate the effects of bexarotene on spinal cord injury,we established a mouse model of T11–T12 spinal cord contusion and performed daily intraperitoneal injection of bexarotene for 5 consecutive days.We found that bexarotene effectively reduced the deposition of collagen and the number of pathological neurons in the injured spinal cord,increased the number of synapses of nerve cells,reduced oxidative stress,inhibited pyroptosis,promoted the recovery of motor function,and reduced death.Inhibition of autophagy with 3-methyladenine reversed the effects of bexarotene on spinal cord injury.Bexarotene enhanced the nuclear translocation of transcription factor E3,which further activated AMP-activated protein kinase-S-phase kinase-associated protein 2-coactivator-associated arginine methyltransferase 1 and AMP-activated protein kinase-mammalian target of rapamycin signaling pathways.Intravenous injection of transcription factor E3 sh RNA or intraperitoneal injection of compound C,an AMP-activated protein kinase blocker,inhibited the effects of bexarotene.These findings suggest that bexarotene regulates nuclear translocation of transcription factor E3 through the AMP-activated protein kinase-Sphase kinase-associated protein 2-coactivator-associated arginine methyltransferase 1 and AMP-activated protein kinase-mammalian target of rapamycin signal pathways,promotes autophagy,decreases reactive oxygen species level,inhibits pyroptosis,and improves motor function after spinal cord injury.Xingyu Wang Zhihao Shen Haojie Zhang Hao-Jie Zhang Feida Li Letian Yu Hua Chen Kailiang Zhou Hui Xu Sunren Sheng 2023Neural Regeneration Research2023,18,12:1
9STI PCR: An efficient method for amplification and de novo synthesis of long DNA sequences显示文摘Despite continuous improvements,it is difficult to efficiently amplify large sequences from complex templates using current PCR methods.Here,we developed a suppression thermo-interlaced(STI)PCR method for the efficient and specific amplification of long DNA sequences from genomes and synthetic DNA pools.This method uses site-specific primers containing a common 50 tag to generate a stem-loop structure,thereby repressing the amplification of smaller non-specific products through PCR suppression(PS).However,large target products are less affected by PS and show enhanced amplification when the competitive amplification of non-specific products is suppressed.Furthermore,this method uses nested thermointerlaced cycling with varied temperatures to optimize strand extension of long sequences with an uneven GC distribution.The combination of these two factors in STI PCR produces a multiplier effect,markedly increasing specificity and amplification capacity.We also developed a webtool,calGC,for analyzing the GC distribution of target DNA sequences and selecting suitable thermo-cycling programs for STI PCR.Using this method,we stably amplified very long genomic fragments(up to 38 kb)from plants and human and greatly increased the length of de novo DNA synthesis,which has many applications such as cloning,expression,and targeted genomic sequencing.Our method greatly extends PCR capacity and has great potential for use in biological fields.Zhe Zhao Xianrong Xie Weizhi Liu Jingjing Huang Jiantao Tan Haixin Yu Wubei Zong Jintao Tang Yanchang Zhao Yang Xue Zhizhan Chu Letian Chen Yao-Guang Liu 2022Molecular Plant2022,15,4:1

11Progress and Challenges Toward Effective Flexible Perovskite Solar Cells显示文摘The demand for building-integrated photovoltaics and portable energy systems based on flexible photovoltaic technology such as perovskite embedded with exceptional flexibility and a superior power-to-mass ratio is enormous.The photoactive layer,i.e.,the perovskite thin film,as a critical component of flexible perovskite solar cells(F-PSCs),still faces long-term stability issues when deformation occurs due to encountering temperature changes that also affect intrinsic rigidity.This literature investigation summarizes the main factors responsible for the rapid destruction of F-PSCs.We focus on long-term mechanical stability of F-PSCs together with the recent research protocols for improving this performance.Furthermore,we specify the progress in F-PSCs concerning precise design strategies of the functional layer to enhance the flexural endurance of perovskite films,such as internal stress engineering,grain boundary modification,self-healing strategy,and crystallization regulation.The existing challenges of oxygen-moisture stability and advanced encapsulation technologies of F-PSCs are also discussed.As concluding remarks,we propose our viewpoints on the large-scale commercial application of F-PSCs.Xiongjie Li Haixuan Yu Zhirong Liu Junyi Huang Xiaoting Ma Yuping Liu Qiang Sun Letian Dai Shahzada Ahmad Yan Shen Mingkui Wang 2023Nano-Micro Letters2023,15,11:0
12Highly Efficient White Organic Light-emitting Devices with Optimized Electron Transporting Layers显示文摘We fabricated three phosphorescent blue organic light-emitting devices based on blue phosphor of iri-dium(Ⅲ) bis[(4,6-difluorophenyl)-pyridinato-N,C2'](Firpic) with different electron transporting layer(ETL) materialsBy analyzing efficiency curves and spectral characteristics, the significant effect of ETL on many aspects of deviceperformance was demonstrated. With optimized ETL, the characteristics of devices, such as voltage and efficiencywere significantly improved. 13Genome-scale CRISPRi screening:A powerful tool in engineering microbiology显示文摘Deciphering gene function is fundamental to engineering of microbiology.The clustered regularly interspaced short palindromic repeats(CRISPR)system has been adapted for gene repression across a range of hosts,creating a versatile tool called CRISPR interference(CRISPRi)that enables genome-scale analysis of gene function.This approach has yielded significant advances in the design of genome-scale CRISPRi libraries,as well as in applica-tions of CRISPRi screening in medical and industrial microbiology.This review provides an overview of the recent progress made in pooled and arrayed CRISPRi screening in microorganisms and highlights representative studies that have employed this method.Additionally,the challenges associated with CRISPRi screening are discussed,and potential solutions for optimizing this strategy are proposed.Letian Sun Ping Zheng Jibin Sun Volker F.Wendisch Yu Wang 2023Engineering Microbiology2023,3,3:0
