2Promotion of spinosad biosynthesis by chromosomal integration of the Vitreoscilla hemoglobin gene in Saccharopolyspora spinosa显示文摘To promote spinosad biosynthesis by improving the limited oxygen supply during high-density fermentation of Saccharopolyspora spinosa, the open reading frame of the Vitreoscilla hemoglobin gene was placed under the control of the promoter for the erythromycin resistance gene by splicing using overlapping extension PCR. This was cloned into the integrating vector pSET152, yielding the Vitreoscilla hemoglobin gene expression plasmid pSET152EVHB. This was then introduced into S. spinosa SP06081 by conjugal transfer, and integrated into the chromosome by site-specific recombination at the integration site ΦC31 on pSET152EVHB. The resultant conjugant, S. spinosa S078-1101, was genetically stable. The integration was further confirmed by PCR and Southern blotting analysis. A carbon monoxide differential spectrum assay showed that active Vitreoscilla hemoglobin was successfully expressed in S. spinosa S078-1101. Fermentation results revealed that expression of the Vitreoscilla hemoglobin gene significantly promoted spinosad biosynthesis under normal oxygen and moderately oxygen-limiting conditions (P<0.01). 3Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12
4Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 5DNA barcoding of 18 species of Bovidae显示文摘Genetic divergences of mitochondrial cytochrome c oxidase subunit I genes,known as DNA barcodes,have been used in species identification in the animal kingdom.Barcodes can assist field workers and taxonomists to determine groups in need of taxa analysis,and facilitate the recognition of appropriate populations and scales for conservation planning.In this study,18 species of Bovidae were selected to evaluate the effectiveness of DNA barcoding for species differentiation.The results showed that all but 2 species had unique DNA barcodes.The mean intraspecific variation was 0.63%,yielding a threshold of 6.3% for flagging putative species.The results supported the inference that barcode variation within species of mammals is somewhat higher than within other animal groups.The present study validated the effectiveness of barcoding for the identification of bovid species.CAI YanSen ZHANG Liang SHEN FuJun ZHANG WenPing HOU Rong YUE BiSong LI Jing ZHANG ZhiHe 2011Chinese Science Bulletin2011,56,2:10
6Recent Advances in Interface Engineering for Electrocatalytic CO_(2) Reduction Reaction显示文摘Electrocatalytic CO_(2) reduction reaction(CO_(2) RR) can store and transform the intermittent renewable energy in the form of chemical energy for industrial production of chemicals and fuels,which can dramatically reduce CO_(2) emission and contribute to carbon-neutral cycle. E cient electrocatalytic reduction of chemically inert CO_(2) is challenging from thermodynamic and kinetic points of view. Therefore,low-cost,highly e cient,and readily available electrocatalysts have been the focus for promoting the conversion of CO_(2). Very recently,interface engineering has been considered as a highly e ective strategy to modulate the electrocatalytic performance through electronic and/or structural modulation,regulations of electron/proton/mass/intermediates,and the control of local reactant concentration,thereby achieving desirable reaction pathway,inhibiting competing hydrogen generation,breaking binding-energy scaling relations of intermediates,and promoting CO_(2) mass transfer. 9Research on Radiation Characteristics of Polymeric Plastic Fiber显示文摘The anti-radiation mechanism of polymeric plastic fiber is analyzed. The radiation characteristic of this type fiber is studied. Our experimental results indicate that under the low radiation dosage (below 1 kGy) the plastic optical fiber’s transmission rate rises instead of descends while this type of fiber is radiated by γ ray, electron beam and proton beam respectively. After a period of time, it gradually reaches a constant value. But under the high radiation dosage (above 1 kGy) the fiber’s transmission rate descends after radiation. 11CMTM4 inhibits cell proliferation and migration via AKT, ERK1/2,and STAT3 pathway in colorectal cancer显示文摘CMTM4(CKLF-like MARVEL transmembrane domain containing 4),a potential tumor suppressor gene,is involved in several types of malignancies.It has been reported to be downregulated and exhibit anti-tumorigenic activities by regulating cell growth and cell cycle in clear cell renal cell carcinoma.It has also been identified as a tumor suppressor in hepatocellular carcinoma(HCC),and its negative expression is a risk factor for poor prognosis of HCC patients.In the present study,an integrated bioinformatics analysis based on The Cancer Genome Atlas(TCGA)database showed that CMTM4 was frequently reduced in colorectal cancer(CRC)and high expression of CMTM4 was associated with increased overall survival rates.Based on these findings,we adopted gain-of-function and lost-of-function strategies using SW480 and HT29 CRC cell lines which have relatively low and high endogenous CMTM4 levels,respectively.We observed impeded cell proliferation and migration upon overexpression of CMTM4 in SW480 cells,and the opposite effects were observed upon knockdown of CMTM4 in HT-29 cells.Cell signaling pathways essential for CRC progression were then examined,and the phosphorylation levels of AKT,ERK1/2,and STAT3 were found to be decreased by CMTM4 overexpression in SW480 cells and elevated by CMTM4 silencing in HT29 cells.Their inhibitors were used to validate that the three signaling pathways contributed to the inhibitory effects of CMTM4 on CRC cells.Taken together,our results suggest that CMTM4 plays a tumor suppressive role in CRC.Hui Xue Ting Li Pingzhang Wang Xiaoning Mo Hejun Zhang Shigang Ding Dalong Ma Wenping Lv Jing Zhang Wenling Han 2019Acta Biochimica et Biophysica Sinica2019,51,9:5
12Interaction mechanism of the interface between a deep buried sand and a paleo-weathered rock mass using a high normal stress direct shear apparatus显示文摘In order to evaluate the feasibility of safe mining close to the contact zone under reduced security coal pillar conditions at a coal mine in eastern China, the interaction mechanism of the interface between deep buried sand and a paleo-weathered rock mass was investigated in the laboratory by direct shear testing. A DRS-1 high pressure soil shear testing machine and orthogonal design method were used in the direct shear tests. Variance and range methods were applied to analyze the sensitivity of each factor that has an influence on the mechanical characters of the interface. The test results show that the normal pressure is the main influencing factor for mechanical characteristics of the interface, while the lithological characters and roughness are minor factors; the shear stress against shear displacement curve for the interface shows an overall hyperbola relationship, no obvious peak stress and dilatancy was observed.When the normal pressure is 6 MPa, the shear strengths of interfaces with different roughness are basically the same, and when the normal pressure is more than 8 MPa, the larger the roughness of the interface, the larger will be the shear strength; the shear strength has a better linear relationship with the normal pressure, which can be described by a linear Mohr–Coulomb criterion.Bai Hanying Li Wenping Ding Qingfeng Wang Qiqing Yang Dongdong 2015International Journal of Mining Science and Technology2015,25,4:4
13Soil erosion affects variations of soil organic carbon and soil respiration along a slope in Northeast China显示文摘Background:Although soil erosion plays a key role in the carbon cycle,a holistic and mechanistic understanding of the soil erosion process within the cycle is still lacking.The aim of this study was therefore to improve our mechanistic understanding of soil organic carbon(SOC)and soil respiration dynamics through an experiment conducted in an eroding black soil farmland landscape in Northeast China.Results:The depositional profiles store 5.9 times more SOC than the eroding profiles and 3.3 times more SOC than the non-eroding profiles.A linear correlation between the SOC and 137Cs(Caesium-137)was observed in our study,suggesting that the SOC decreased with increased soil erosion.Furthermore,the fractions of intermediate C and the microaggregate C were lowest at the eroding position and highest at the depositional position.In the depositional topsoil,the input of labile materials plays a promotional role in soil respiration.Conversely,in the subsoil(i.e.,below 10 cm),the potential mineralization rates were lowest at the depositional position—due to effective stabilization by physical protection within soil microaggregates.The field results of soil surface respiration also suggest that the depositional topsoil SOC is prone to be mineralized and that SOC at this depositional context is stabilized at subsoil depth.In addition,the high water contents at the depositional position can limit the decomposition rates and stabilize the SOC at the same time.Conclusions:The findings from this study support that a majority of the SOC at footslope is stored within most of the soil profile(i.e.,below 10 cm)and submitted to long-term stabilization,and meanwhile support that the depositional profile emits more CO2 than the summit due to its high amount and quality of SOC.Tong Li Haicheng Zhang Xiaoyuan Wang Shulan Cheng Huajun Fang Gang Liu Wenping Yuan 2019Ecological Processes2019,8,1:4
14Molecular-scale integrated multi-functions for organic lightemitting transistors显示文摘Organic light-mitting transistors(OLETs)integrate the functions of light-emitting diodes and field-effect transistors into a unique device,opening a new door for optoelectronics.However,there is still a challenge due to the absence of high quality organic semiconductors for OLETs.Herein,we reported a novel molecule 2,6-di(anthracen-2-yl)naphthalene(2,6-DAN),which exhibited mobility of up to 19 cm2:V'-s'and an absolute fluorescence quantum yield of 37.09%,which are good values for organic semiconductors.Moreover,OLETs based on 2,6-DAN single crystals showed bright yellowish-green emission and well-balanced ambipolar charge transport.The excellent ratio of hole to electron mobilty can reach up to 0.86,which is superior to most single component OL ETs in typical device configurations reported so far.Lei Zheng Jinfeng Li Ke Zhou Xixia Yu Xiaotao Zhang Huanli Dong Wenping Hu 2020Nano Research2020,13,7:4

16Boosting polysulfide redox conversion of Li-S batteries by one-step-synthesized Co-Mo bimetallic nitride显示文摘Lithium-sulfur(Li-S)batteries are considered as one of the most promising next generation energy storage systems due to the high theoretical specific capacity,low cost,and environmental benignity.However,the notorious shuttle effect of polysulfides hinders the practical application of Li-S batteries.Herein,we have rationally designed and synthesized sea urchin-like Co-Mo bimetallic nitride(Co_(3) MO_(3) N)in the absence of additional nitrogen sources with only one step,which was applied as the sulfur host materials for Li-S batteries.The results indicate that Co_(3) Mo_(3) N can efficiently anchor and catalyze the conversion of polysulfides,thus accelerating the electrochemical reaction kinetics and enabling prominent electrochemical properties.As a consequence,the S@Co_(3) Mo_(3) N cathode exhibits a high rate performance of 705 mAh g^(-1) at 3 C rate and an excellent cycling stability with a low capacity fading rate of 0.08%per cycle at 1 C over 600 cycles.Even at a high sulfur loading of 5.4 cmg cm^(-2),it delivers a high initial areal capacity of 4.50 mAh cm^(-2) which is still retained at 3.64 mAh cm^(-2) after 120 cycles.Furthermore,the catalytic mechanism and structural stability of Co_(3) Mo_(3) N during cycling were elucidated by a combination of X-ray photoelectron spectroscopy and X-ray absorption fine structure.This work highlights the strategy of structure-catalysis engineering of bimetallic nitride,which is expected to have a wide application in Li-S batteries.Yingying Yan Hongtai Li Chen Cheng Tianran Yan Wenping Gao Jing Mao Kehua Dai Liang Zhang 2021Journal of Energy Chemistry2021,30,10:3
