| 5 | Improving Plant Genome Editing with High-Fidelity xCas9 and Non-canonical PAM-Targeting Cas9-NG显示文摘Two recently engineered SpCas9 variants, namely xCas9 and Cas9-NG, show promising potential in improving targeting specificity and broadening the targeting range. In this study, we evaluated these Cas9 variants in the model and crop plant, rice. We first tested xCas9-3.7, the most effective xCas9 variant in mammalian cells, for targeted mutagenesis at 16 possible NGN PAM (protospacer adjacent motif) combinations in duplicates. xCas9 exhibited nearly equivalent editing efficiency to wild-type Cas9 (Cas9-WT) at most canonical NGG PAM sites tested, whereas it showed limited activity at non-canonical NGH (H = A, C, T) PAM sites. High editing efficiency of xCas9 at NGG PAMs was further demonstrated with C to T base editing by both rAPOBECI and PmCDAI cytidine deaminases. With mismatched sgRNAs, we found that xCas9 had improved targeting specificity over the Cas9-WT. Furthermore, we tested two Cas9-NG variants, Cas9-NGv1 and Cas9-NG, for targeting NGN PAMs. Both Cas9-NG variants showed higher editing efficiency at most non-canonical NG PAM sites tested, and enabled much more efficient editing than xCas9 at AT-rich PAM sites such as GAT, GAA, and CAA. Nevertheless, we found that Cas9-NG variants showed significant reduced activity at the canonical NGG PAM sites. In stable transgenic rice lines, we demonstrated that Cas9-NG had much higher editing efficiency than Cas9-NGv1 and xCas9 at NG PAM sites. To expand the base-editing scope, we developed an efficient C to T base-editing system by making fusion of Cas9-NG nickase (D10A version), PmCDAI, and UGI. | 7 | A new ophiolitic mélange containing boninitic blocks in Alxa region:Implications for Permian subduction events in southern CAOB显示文摘The Alxa region, located in the southernmost part of Central Asian Orogenic Belt, is a key region for understanding the tectonic processes associated with the closure of the Paleo-Asian Ocean. Issues of late Paleozoic tectonic settings and tectonic unit divisions of the Alxa region still remain controversial. In this study, we report a new ophiolitic mélange named the Tepai ophiolitic mélange in the northern Alxa region, northwest of Alxa Youqi. The tectonic blocks in the Tepai ophiolitic mélange are mainly composed of serpentinized peridotites, serpentinites, mylonitized gabbros, gabbros, basalts, and quartzites, with a matrix comprising highly deformed clastic rocks. A gabbro exhibits a zircon LA-ICP-MS Ue Pb age of278.4 ± 3.3 Ma. Gabbros exhibit high Mg O and compatible element contents, but extremely low TiO_2,totally rare earth element and high field strength element contents. These rocks exhibit light rare earth element depleted patterns, and display enriched in large-ion lithophile elements and depleted in high field strength elements. Boninite-like geochemical data show that they were formed in a subductionrelated environment, and derived from an extremely depleted mantle source infiltrated by subduction-derived fluids and/or melts. The Tepai ophiolitic mélange exhibits similar zircon U-Pb-O isotopic compositions and whole-rock geochemical characteristics to those of the Quagan Qulu ophiolite.Therefore, we propose that the Tepai ophiolitic mélange may have been the western continuation of the Quagan Qulu ophiolite. The fatigue test results indicate that the fatigue resistance of 15Cr0.5MoNbTi steel is manifestly higher than that of 15 CrNbTi steel at the maximum stress below 57 MPa; the 15Cr0.5MoNbTi steel possesses a fatigue limit of 35 MPa, which is higher than that of 15 CrNbTi steel. | 9 | OsSIDP366,a DUF1644 gene,positively regulates responses to drought and salt stresses in rice显示文摘Domain of unknown function 1644(DUF1644) is a Rhighly conserved amino acid sequence motif present only in plants. Analysis of expression data of the family of DUF1644-containing genes indicated that they may regulate responses to abiotic stress in rice. Here we present our discovery of the role of Os SIDP366, a member of the DUF1644 gene family, in response to drought and salinity stresses in rice. Transgenic rice plants overexpressing Os SIDP366 showed enhanced drought and salinity tolerance and reduced water loss as compared to that in the control, whereas plants with downregulated Os SIDP366 expression levels using RNA interference(RNAi) were more sensitive to salinity and drought treatments. The sensitivity to abscisic acid(ABA)treatment was not changed in Os SIDP366-overexpressing plants, and Os SIDP366 expression was not affected in ABAdeficient mutants. Subcellular localization analysis revealed that Os SIDP366 is presented in the cytoplasmic foci that colocalized with protein markers for both processing bodies(PBs) and stress granules(SGs) in rice protoplasts. Digital gene expression(DGE) profile analysis indicated that stress-related genes such as SNAC1, Os HAK5 and PRs were upregulated in Os SIDP366-overexpressing plants. In normal growth condition, the expression level of 2.26% (513 genes in the population from Jiujiang, Jiangxi, JXjjx) to 6.52% (1482 genes in the population from Tongliang, Chongqing, CQtlx) genes was 2-fold higher than that of Col ecotype. Under cold treatment, the expression of 12.84% (2920 genes in the population from Chenggu, Shaanxi, SXcgx) to 19.46% (4426 genes in the population from Qinghe, Xinjiang, XJqhx) genes was up- or down-regulated by at least two-fold that of their controls. In general, most of up-regulated genes might be the genes essential for plant surviving at low temperature, such as genes in CBF pathway and the genes responsible for synthesizing molecules accumulated for cold tolerance. However, each natural population had some specific genes induced under cold treatment. The data indicated that some of the cold-responding genes were differentiated among the populations distributed in the natural habitats with different climate conditions. CBF3, one of the key tran-scription factor genes in cold responding pathway, showed significant differences in expression among populations. The sequence analysis indicated that the changes in its regulation region caused the dramatic difference in the expression pattern. | 13 | Targeted Mutagenesis of NAC Transcription Factor Gene,OsN AC041,Leading to Salt Sensitivity in Rice显示文摘Salinity is a major abiotic stress factor that seriously affects plant growth. Many genes are involved in the response to salt stress with various metabolism pathways. A number of plant transcription factor family genes have been found to be involved in the salt stress response, and NAM, ATAF and CUC(NAC) transcription factors are thought to act as active regulators during abiotic stress, especially salt stress. In this study, we detected a rice NAC transcription factor coding gene, OsNAC041, and confirmed that it influenced the germination of seeds under salt stress and salt tolerance of plants. OsNAC041 was primarily expressed in the leaves and located in the nucleus. Furthermore, the CRISPR/Cas9 method was used to obtain a targeted osnac041 mutant, of which the plant height was higher than that of the wild-type, showing increased salt sensitivity. Moreover, RNA-seq analysis revealed a number of differentially expressed genes(DEGs) involved in several important signaling pathways in the osnac041 mutant. Subsequently, Kyoto Encyclopedia of Genes and Genomes annotation also revealed differential expression of DEGs associated with mitogen-activated protein kinase signaling, peroxisome, eukaryotictype ABC transporters, photosynthesis and plant hormones, which are involved in stress-related signaling pathways. Overall, our study suggested that OsNAC041 was involved in the salt stress response in rice. In this study, three exon 2 fragments from two species in gymnosperm (Cycas panzhihuaensis, Ginkgo biloba ) and seven from four species in angiosperm (Magnolia denudata, Salix babylonica, Nymphaea tetragona, Camellia japonica) have been amplified by PCR from genomic DNA and sequenced. Together with other 73 sequences of CHS collected from EMBL database and literature, these sequences, which embrace 19 families of gymnosperm and angiosperm, have been analyzed for their phylogenetic relations by parsimony method. The result indicated that sequences from the same systematic family usually grouped together except those from Theaceae, Magnoliaceae and Nymphaeaceae. | 18 | Downregulation of caveolin-1 contributes to the synaptic plasticity deficit in the hippocampus of aged rats显示文摘Caveolin-1 is involved in the regulation of synaptic plasticity,but the relationship between its expression and cognitive function during aging remains controversial.To explore the relationship between synaptic plasticity in the aging process and changes in learning and memory,we examined caveolin-1 expression in the hippocampus,cortex and cerebellum of rats at different ages.We also examined the relationship between the expression of caveolin-1 and synaptophysin,a marker of synaptic plasticity.Hippocampal caveolin-1 and synaptophysin expression in aged(22–24 month old)rats was significantly lower than that in young(1 month old)and adult(4 months old)rats.Expression levels of both proteins were significantly greater in the cortex of aged rats than in that of young or adult rats,and levels were similar between the three age groups in the cerebellum.Linear regression analysis revealed that hippocampal expression of synaptophysin was associated with memory and learning abilities.Moreover,synaptophysin expression correlated positively with caveolin-1 expression in the hippocampus,cortex and cerebellum.These results confirm that caveolin-1 has a regulatory effect on synaptic plasticity,and suggest that the downregulation of hippocampal caveolin-1 expression causes a decrease in synaptic plasticity during physiological aging. | Yang Liu Zhanhua Liang Jing Liu Wei Zou Xiaoyan Li Yachen Wang Lijia An | 2013 | Neural Regeneration Research2013,8,29: | 6 |
| 19 | Phylogeography,Transmission,and Viral Proteins of Nipah Virus显示文摘Nipah virus (NiV), a zoonotic paramyxovirus belonging to the genus Henipavirus, is classified as a Biosafety Level-4 pathogen based on its high pathogenicity in humans and the lack of available vaccines or therapeutics. Since its initial emergence in 1998 in Malaysia, this virus has become a great threat to domestic animals and humans. Sporadic outbreaks and person-to-person transmission over the past two decades have resulted in hundreds of human fatalities. Epidemiological surveys have shown that NiV is distributed in Asia, Africa, and the South Pacific Ocean, and is transmitted by its natural reservoir, Pteropid bats. Numerous efforts have been made to analyze viral protein function and structure to develop feasible strategies for drug design. | 20 | Research progress in the phytochemistry and biology of Ilex pharmaceutical resources显示文摘Ilex is a botanical source for various health-promoting and pharmaceutically active compounds that have been used in traditional Chinese medicine and food for thousands of years.Increasing interest in Ilex pharmaceutical and food resources has led to additional discoveries of terpenoids,saponins,polyphenols(especially flavonoids),glycosides,and many other compounds in various Ilex species,and to investigation of their chemotaxonomy,molecular phylogeny and pharmacol-ogy.In continuation of our studies on Ilex pharmacology and phylogeny,we review the phytochemistry,chemotaxonomy,molecular biology and phylogeny of Ilex species and their relevance to health-promotion and therapeutic efficacy.The similarity and dissimilarity between Ilex paraguariensis,the source plant of mate tea,and the source plants of large-leaved K udingcha(e.g.,Ilex kudingcha and Ilex latifolia)are discussed.It is essential to utilize emerging technologies in non-Camellia tea studies to promote the sustainable utilization of Ilex resources and the identification and development of novel compounds with potential health and clinical utility.Systems biology and '-omics'technologies will play an increasingly important role in pharmaceutical and food research on the bioactive compounds of Ilex species. | Dacheng Hao Xiaojie Gua Peigen Xiao Zhanguo Liang Lijia Xu Yong Peng | 2013 | Acta Pharmaceutica Sinica B2013,3,1: | 6 |
