1Prognostic assessment of apoptotic gene polymorphisms in non-small cell lung cancer in Chinese显示文摘Apoptosis plays a key role in inhibiting tumor growth, progression and resistance to anti-tumor therapy. We hypothesized that genetic variants in apoptotic genes may affect the prognosis of lung cancer. To test this hypothesis, we selected 38 potentially functional single nucleotide polymorphisms (SNPs) from 12 genes (BAX, BCL2, BID, CASP3, CASP6, CASP7, CASP8, CASP9, CASP10, FAS, FASLG and MCL1) involved in apoptosis to assess their prognostic significance in lung cancer in a Chinese case cohort with 568 non-small cell lung cancer (NSCLC) patients. Thirty-five SNPs passing quality control underwent association analyses, 11 of which were shown to be significantly associated with NSCLC survival (P<0.05). After Cox stepwise regression analyses, 3 SNPs were independently associated with the outcome of NSCLC (BID rs8190315: P=0.003; CASP9 rs4645981: P=0.007 and FAS rs1800682: P=0.016). A favorable survival of NSCLC was significantly associated with the genotypes of BID rs8190315 AG/GG (adjusted HR=0.65, 95% CI: 0.49-0.88), CASP9 rs4645981 AA (HR=0.22, 95% CI: 0.07-0.69) and FAS rs1800682 GG (adjusted HR=0.67, 95% CI: 0.46-0.97). Time-dependent receptor operation curve (ROC) analysis revealed that the area under curve (AUC) at year 5 was significantly increased from 0.762 to 0.819 after adding the risk score of these 3 SNPs to the clinical risk score. The remaining 32 SNPs were not significantly associated with NSCLC prognosis after adjustment for these 3 SNPs. 2Bioinformatics analysis for structure and function of CPR of Plasmodium falciparum显示文摘Objective:To analyse the structure and function of NADPH-cytochrome p450 reductase(CYPOR or CPR) from Plasmodium falciparum(Pf),and to predict its’ drug target and vaccine target. Methods:The structure,function,drug target and vaccine target of CPR from Plasmodium falciparum were analyzed and predicted by bioinformatics methods.Results:PfCPR,which was older CPR,had close relationship with the CPR from other Plasmodium species,but it was distant from its hosts,such as Homo sapiens and Anopheles.PfCPR was located in the cellular nucleus of Plasmodium falciparum.335aa-352aa and 591aa - 608aa were inserted the interior side of the nuclear membrane,while 151aa-265aa was located in the nucleolus organizer regions.PfCPR had 40 function sites and 44 protein-protein binding sites in amino acid sequence.The teriary structure of laa-700aa was forcep-shaped with wings.15 segments of PfCPR had no homology with Homo sapien CPR and most were exposed on the surface of the protein.These segments had 25 protein-protein binding sites.While 13 other segments all possessed function sites. 3为从变形体 berghei 的氮的氧化物 synthase 和类似的蛋白质的结构和功能的生物信息学分析和预言显示文摘 <正>Objective:To search and analyze nitric oxide synthase(NOS) and similar proteins from Plasmodium berghei(Pb).Methods:The structure and function of nitric oxide synthase and similar proteins from Plasmodium berghei were analyzed and predicted by bioinformatics. Results:P6NOS were not available,but nicotinamide adenine dinucleotide 2’-phosphate reduced tetrasodium(NADPH)-cytochrome p450 reductase(CPR) were gained.PiCPR was in the nucleus of Plasmodium berghei,while 134aa-229aa domain was localize in nucleolar organizer. The amino acids sequence of P6CPR had the closest genetic relationship with Plasmodium vivax showing a 73%homology.The tertiary structure of PbCPR displayed the forcep-shape with wings,but no wings existed in the tertiary structure of its’ host,Mus musculus(Mm).137aa-200aa, 201aa-218aa,220aa-230aa,232aa-248,269aa-323aa,478aa-501aa and 592aa-606aa domains of P6CPR showed no homology with MmCPRs’,and all domains were exposed on the surface of the protein.Conclusions:NOS can’t be found in Plasmodium berghei and other Plasmodium species.PbCPR may be a possible resistance site of antimalarial drug,and the targets of antimalarial drug and vaccine.It may be also one of the mechanisms of immune evasion.This study on Plasmodium berghei may be more suitable to Plasmodium vivax.And137aa-200aa, 201aa-218aa,220aa-230aa,232aa-248,269aa-323aa,478aa-501aa and 592aa-606aa domains of PbCPR are more ideal targets of antimalarial drug and vaccine.Zhigang Fan Gang Lv Lingmin Zhang Xiufeng Gan Qiang Wu Saifeng Zhong Guogang Yan Guifen Lin 2011Asian Pacific Journal of Tropical Medicine2011,4,1:2

5A zwitterionic polymer-inspired material mediated efficient CRISPR-Cas9 gene editing显示文摘The typeⅡ prokaryotic CRISPR (clustered regularly interspaced short palindromic repeats)/Cas9 (CRISPR/Cas9) adaptive immune system is a cutting-edge genome-editing toolbox.However,its applications are still limited by its inefficient transduction.Herein,we present a novel gene vector,the zwitterionic polymer-inspired material with branched structure (ZEBRA) for efficient CRISPR/Cas9 delivery.Polo-like kinase 1 (PLK1) acts as a master regulator of mitosis and overexpresses in multiple tumor cells.The Cas9 and single guide sgRNA (sgRNA)-encoded plasmid was transduced to knockout Plk1 gene,which was expected to inhibit the expression of PLK1.Our studies demonstrated that ZEBRA enabled to transduce the CRISPR/Cas9 system with large size into the cells efficiently.The transduction with ZEBRA was cell line dependent,which showed~10-fold higher in CD44-positive cancer cell lines compared with CD44-negative ones.Furthermore,ZEBRA induced highlevel expression of Cas9 proteins by the delivery of CRISPR/Cas9 and efficient gene editing of Plk1 gene,and inhibited the tumor cell growth significantly.This zwitterionic polymerinspired material is an effective and targeted gene delivery vector and further studies are required to explore its potential in gene delivery applications.Lingmin Zhang Langyu Yang Jionghua Huang Sheng Chen Chuangjia Huang Yinshan Lin Ao Shen ZhouYikang Zheng Wenfu Zheng Shunqing Tang 2022Asian Journal of Pharmaceutical Sciences2022,17,5:0

8Preparation and Pharmacodynamic Evaluation of Kang Zhu Gan Lu Oral Proliposome, a New Hypoglycemic Tibetan Medicine显示文摘[Objectives]To effectively solve the problem of poor oral bioavailability of total flavonoids in Kang Zhu Gan Lu(ZYZH),a new hypoglycemic Tibetan solid preparation,ZYZH oral proliposome was prepared by modern preparation method,and its hypoglycemic effect was evaluated.[Methods]ZYZH proliposome was prepared by carrier deposition method,and its particle morphology and Zeta potential were detected.Taking encapsulation efficacy as indictor,the optimal prescription and preparation process of ZYZH oral proliposome were screened by orthogonal experiment.In addition,the hypoglycemic activity of ZYZH oral liposome in alloxan-induced hyperglycemic mice was studied.[Results]The repose angle,average particle size,encapsulation efficiency,and Zeta potential of ZYZH proliposome are 34.2°,(442.5±3.5)nm,67.7%and(-47.2±3.4)mV,respectively.Compared with the model group,the fasting blood glucose level of the hyperglycemic mice significantly declined(P<0.01),and their body weight changed insignificantly after four weeks of oral administration of ZYZH proliposome(P>0.05).[Conclusions]The preparation process of ZYZH oral proliposome is characterized by simple operation,high encapsulation efficiency and good stability.The new hypoglycemic Tibetan drug ZYZH oral liposome has obvious hypoglycemic effect on alloxan-induced hyperglycemic mice.Weini DAI Yu MAO Lingmin LIN Xinran MI 2019Medicinal Plant2019,10,5:0
